Ex vivo Expansion and Clonal Maintenance of CD34+ Selected Cells from Cord Blood and Peripheral Blood (제대혈 및 말포혈로부터 분리한 CD34 양성 세포의 체외 증폭 및 클론 유지)

  • Purpose : Because of the unavailability of marrow transplantation, umbilical cord blood (CB) is increasingly being used. We evaluated the potential of ex vivo expansion and clonality in CD34+ cells separated from cord blood source and mobilized peripheral blood (PB) in a serum-free media. Methods : The CD34+ cells, selected from CB and mobilized PB, were expanded with hematopoietic growth factors. They were then cultured for burst-forming units of erythrocytes (BFU-E), colony-forming units of granulocytes and monocytes (CFU-GM) and colony-forming units of megakaryocytes (CFU-Mk) at culture days 0, day 4, day 7, and day 14 with various growth factors. Results : The CB-selected CD34+ cells showed significantly higher total cell expansion than those from the PB at day 7 (2 fold increase than PB). The CB-selected CD34+ cells produced more BFU-E colonies than did the PB on culture at days 7 and at day 14. Also, the CB-selected CD34+ cells produced more CFU-Mk colonies than did the PB on culture at day 4 and at day 7. Conclusion : The ex vivo expansion of the CB cells may be promising in producing total cellular expansion, CFU-Mk and BFU-E compared with PB for 7 to 14 days. The growth factors combination including megakaryocyte growth and development, flt3-ligand and interleukin-3 showed more expansion in the view of total cells and clonal maintenance compared with less combination.

Effects of matured silkworm hemolymph on suppressing melanin synthesis (누에 숙잠 혈림프의 멜라닌 생성 억제 효과)

  • In this study, matured silkworm hemolymph was studied for inhibitory effect on melanin formation, tyrosinase activities. A matured silkworm hemolymph mass purification method was developed and some of suppressing melanin formation effects were investigated, too. As a result, the advanced purification method was higher in yield rate than existing one by 53%. A free radical scavenging activity of hemolymph was higher than sericin on the market($IC_{50}$, $202.9{\pm}33.1$) and lower than vitamin C($0.57{\pm}0.02$) by $6.4{\pm}0.3{\mu}g/m{\ell}$. An inhibitory effect on melanin synthesis was higher in hemolymph compared to Morus bark and arbutin by 9.15% and 11.56%, respectively. An inhibitory effect on tyrosinase activity was higher in hemolymph than Morus bark and lower than arbutin. Effect of hemolymph $45{\mu}g/m{\ell}$ on suppressing of tyrosinase was almost 90%. Thus, these results suggest that hemolymph of matured silkworm may have beneficial properties as a material for cosmetics.

Correlation of Cord Serum Adiponectin and IGF-I with Fetal Growth in Healthy Term Neonates (제대혈 Adiponectin과 IGF-I이 태아성장에 미치는 영향)

  • Purpose : The purpose of this study was to evaluate the relationship between cord serum adiponectin (APN) and IGF-I concentrations and fetal growth. Methods : Umbilical cord serum APN and IGF-I concentrations were measured in healthy term singleton deliveries (n=72). The association of cord serum APN and IGF-I concentrations was evaluated in relation to birth weight, height, head circumference, gender, ponderal index, placental weight, feto-placental (F/P) weight ratio, maternal weight gain, and maternal body mass index (BMI). Results : The mean cord serum APN was 29.2${\pm}$10.46 $\mu$g/mL. The cord serum APN and birth weight demonstrated a bell-shape relationship. The cord serum APN concentration was higher in females than males (P=0.001). The cord serum APN was negatively correlated with maternal BMI (r=-0.301, P=0.027), but the mean cord serum APN concentration was not correlated with birth height, birth head circumference, ponderal index, placental weight, F/P ratio, or maternal weight gain. The mean cord serum concentrations of IGF-I was 51.26${\pm}$21.54 ng/mL. The cord serum IGF-I concentration was positively correlated with birth weight (r=0.312, P=0.009), but not birth height, ponderal index, placental weight, F/P weight ratio, or maternal BMI. Conclusion : APN demonstrated a bell-shaped relationship with birth weight in healthy term infants. IGF-I was highly correlated with fetal growth, especially birth weight.

  • Objectives : Yogaktong is the genernal term of lowback and sciatic pain. Simpley this is not for the syndrom or illiness but for the symptoms. In this study, the effect of Acupuncture and Moxibustion Treatment by Analysing Su-point(背兪穴) compared with X-ray measuring for three patients with Yogaktong(腰脚痛) were evaluated. Methods : After treatment with the Acupuncture and Moxibustion Treatment by Analysing Su-point(背兪穴) compared with X-ray measuring for one time, we evaluated the effect by the figure of x-ray measuring and complaints of patients. Results : In three cases, the change in the figure declined dramatically and symptoms of patients got better after one treatment. Conclusions : Acupuncture and Moxibustion Treatment by Analysing Su-point(背兪穴) compared with X-ray measuring was effective. It will be attempted to the patients with it.

Cord Blood Adiponectin and Insulin-like Growth Factor-I in Term Neonates of Gestational Diabetes Mellitus Mothers: Relationship to Fetal Growth

  • Purpose: The purpose of this study was to evaluate the relationship between cord blood adiponectin and insulin-like growth factor (IGF)-I and their effect on fetal growth and insulin resistance in mothers with gestational diabetes mellitus (GDM). Methods: Cord blood adiponectin and IGF-I were compared between mothers with GDM (GDM group, N=53) and controls (non-GDM group, N=101). Neonates were classified into three groups of small for gestational age (SGA, N=26), appropriate for gestational age (AGA, N=97), and large for gestational age (LGA, N=31) by birth weight. The association between cord adiponectin and IGF-I levels was evaluated in relation to maternal and neonatal clinical data. Results: Cord adiponectin was lower in the GDM group than in the non-GDM group (P<0.001). There was no significant difference in cord adiponectin among the SGA, AGA, and LGA groups in the GDM group (P=0.228). The cord adiponectin of AGA in the GDM group was significantly lower than that in the non-GDM group (P<0.001). The most powerful predictor affecting cord adiponectin was the result of maternal 75 g oral glucose tolerance test. The cord IGF-I values between the GDM group and the non-GDM group were not different (P=0.834). Neonates with the heavier birth weight had the higher cord IGF-I levels. The most powerful predictor affecting cord IGF-I was birth weight and the next was maternal parity. Conclusion: Both cord blood adiponectin and IGF-I were associated with fetal growth, but IGF-I was a more general and direct factor affecting fetal body size, and adiponectin seemed to have more association with insulin sensitivity than growth.

제대혈을 활용한 유전자 캐릭터 제작 및 DNA 정보 데이터베이스 구축

  • 유전자 감식법은 1980년대 후반에 미국에서 처음으로 법의학적으로 적용된 이래, 강력범죄, 친자확인 및 항공기 추락사고, 화재 등의 대형 참사의 신원 확인에 큰 공헌을 하였다. 유전자감식을 위해서는 높은 다형성을 보이는 STR (short tandem repeats) 좌위의 타이핑을 주로 이용한다. 한편, 최근 줄기세포 (stem cell)의 중요성이 인식되면서 폐기되던 제대혈의 보관 사업이 활성화되고 있다. 본 연구에서는 제대혈의 보관시 STR 타이핑의 유전 정보를 표기한 유전자 캐릭터를 동시에 제공하여 불의의 사고시 신원확인이 가능하게 하고, 아울러 이산가족에 대한 DNA정보 데이터베이스 구축 및 검색 프로그램을 개발하였다.

  • 산조 가야금의 경우 몸체의 뒷판에 세 개의 구멍(공명혈)이 있다. 이 구멍은, 몸체를 일정한 부피를 가진 용기로 보았을 때, 그 용기의 입구의 단면적, 다시 말해, 헬름홀츠 공진기 (Helmholtz Resonator) 입구의 단면적과 같다. 본 논문에서는 이 공명혈의 역할을 알아보기 위해, 1) 우선 기성의 산조 가야금 몸체의 세 공명혈을 모두 막은 상태, 일부만 막은 상태, 열려있는 상태에서의 몸체의 충격응답 전달함수를 측정, 분석하여 개략적으로 알아보았고, 이어서 2) 뒷판에 구멍이 전혀 없는 실험용의 산조 가야금 몸체의 제작을 의뢰하여, 뒷판 중앙에 구멍을 점차적으로 넓게 뚫어가면서, 각 단계별로 몸체의 충격응답을 측정하여, 구멍의 크기와 몸체의 충격응답 전달특성 변화 간의 상관관계를 분석 해보았다.

Non-invasive cardiac output estimation based upon a mathematical model of the aorta; comparsion with thermo-dilution method in 13 patients (대동맥의 수리 모델을 사용한 새로운 비관혈적 심박출량 계산방법)

  • 13명의 환자에 있어서 새로운 비관혈적 방법에 의한 심박출량을 계산하고, 열희석에 의한 방법으로 측정한 심박출량과 비교하였다. 본 논문에서 사용한 비관혈적 펄스 전달 방법은 대동맥 모델에서의 최적 매개변수 방법을 사용하였다. 모델의 최적 매개변수는 맥압의 첫번째 고주파 전달함수에서 측정치와 계산치의 차이를 최소화하는 조건에 의하여 계산하였다. 전달함수의 2개의 펄스는 각각 경동맥동과 대퇴동맥에서 Piezo-resistive Pulse Microphone을 사용하여 측정하였고, 대동맥의 직경은 초음파 방법을 사용하여 측정하였다. 13명의 환자에게서 16가지 측정을 해서 열희석과 비관혈적 방법은 0.89의 상관계수를 보여주었다.
