Relationship between Vaginal Cytology and Reproductive Hormone during the Estrous Cycle and Optimal Mating Time in Beagle Dogs (비글개에서 발정 주기 및 교배 적기 동안 질세포상과 번식 호르몬의 관계)

  • This study was conducted to examine relationship between vaginal cytology and reproductive hormones during the estrous cycle and to provide basic data to estimate for ovulation time and optimal mating time in 6 beagle dogs The duration of proestrus, estrus and diestrus were $8.5{\pm}1.4,\;10.0{\pm}1.4\;and\;54.0{\pm}2.8$ days at pregnant respectively, and $7.9{\pm}2.1,\;9.5{\pm}0.7\;and\;62.0{\pm}11.3$ days at non-pregnant respectively. The duration of interestrous intervals were $246.2{\pm}24.5$ days at pregnancy, and $175.3{\pm}34.5$ days at non-pregnancy. The duration of interestrous intervals at pregnancy was longer than that of non-pregnancy. A characteristic features of vaginal cytology during the estrous cycle were the high proportion of superficial cell, anuclear cell and erythrocyte in proestrus and estrus, parabasal cell, small intermediate cell and leukocyte in diestrus, and parabasal cell and small intermediate cell in anestrus, respectively. Cornification index (CI) in proestrus and estrus were significantly higher than that of CI in diestrus and anestrus. Plasma progesterone concentration was below 1.0 ng/ml at the first day of vulval bleeding at pregnancy and non-pregnancy, and then it was above 2.0 ng/ml at Day -2 in all bitches. When plasma progesterone concentration was first increased above 4.0 ng/ml, it was the second day after the first day of male acceptance. Plasma progesterone concentration showed above 40 ng/ml on Day $20{\sim}22$ in all bitches, and then it was gradually decreased until Day 35. Plasma progesterone concentration at pregnancy was higher than that of non-pregnancy from Day 35 to Day 63. Plasma estradiol-$17\;{\beta}$ concentration was above 9.0 pg/ml at the first day of vulval bleeding, and it showed 26.4 pg/ml on Day -2. When it was timed from the first day of male acceptance (Day 0), plasma estradiol-$17{\beta}$ concentration showed a peak on Day 0 and plasma progesterone concentration was first increased above 4.0 ng/ml on Day 2 which was the third day after plasma estradiol-$17{\beta}$ peak. CI was first increased above 80 and 90% on Day -1 and Day 1, respectively. CI was maintained above 80% from Day -1 to Day 8 (10 days) and above 90% from Day 1 to Day 6 (6 days), respectively. CI was maintained above 80% from Day 0 to Day 8 (9 days) and above 90% from Day 1 to Day 6 (6 days), respectively. Plasma progesterone concentration was first increased above 4.0 ng/ml on the second day after the day which CI was first increased above 90%. In conclusion, beagle bitches ovulated on the second day after the day which CI was first increased above 90% and on the day which plasma progesterone concentration was first increased 4.0 ng/ml, and it was estimated that the optimal mating time was the day which the second day after CI was first increased above 90% and plasma concentration was between $2{\sim}25ng/ml$. The measurement of plasma progesterone was used to determine of and accurate ovulation time and the optimal mating time, but vaginal cytology is low-priced and simple method to estimate estrous cycle, optimal mating time and ovulation time.

A Study on the Mineral Content of Calcium-fortified Foods in Korea (우리나라의 칼슘강화식품의 무기질 함량에 관한 연구)

  • This study was done to analyze the contents of minerals, to compare the measured values of calcium and the labeled values in food labeling and to analyze the ratio of calcium to other minerals in 43 calcium-fortified Food products sold in markets in Seoul, Korea. Content of minerals such as Ca, P, Mg, Na, K, Fe, Cu, Zn was measured by atomic absorption or colorimetric method after dry-ashing or wet-ashing. The measured values of calcium were ranged 65.5~343.9% of the labeled values in 43 calcium-fortified products. In 21 calcium-fortified food products, the measured calcium values were ranged 120~160% of the labeled values, and in three drinks those were less than 80% of the labeled, which is not acceptable to the food regulation. The ratios of Ca:P were 2.63$\pm$1.99 (mean$\pm$SD) in grain Products, 1.79$\pm$0.39 in Ramyuns, 2.80$\pm$0.53 in retort pouch food products and 8.35$\pm$12.87 in drinks. The Ca:Fe ratios were 126.33$\pm$44.36 in grain products, 130.65$\pm$34.67 in Ramyuns, 120.31$\pm$71.15 in retort pouch food products and 700.25$\pm$553.70 in drinks. The ratios of Ca:Mg were 11.86$\pm$5.40 in grain products, 9.29$\pm$1.34 in Ramyuns, 9.09$\pm$2.09 in retort pouch food products and 32.50$\pm$41.35 in drinks. The P:Mg ratios were 4.11$\pm$1.54 in grain products, 4.17$\pm$0.67 in Ramyuns, 2.58$\pm$0.45 in retort pouch food Products and 2.59$\pm$2.50 in drinks. These results suggest calcium contents and the ratio of calcium contents to other minerals in calcium-fortified food products should be strictly controlled.

Monitoring and Risk Assessment of Pesticide Residues for Circulated Agricultural Commodities in Korea-2013 (국내 유통 농산물의 잔류농약 모니터링 및 위해평가-2013년)

  • The purpose of this study is the establishment of scientific processes for making food safety policies. Thus, we investigated pesticide residue level of the agricultural commodities from market, and performed risk assessment. Fifteen agricultural items are chosen based on the frequency of Korean consumption. The samples were collected from 9 cities where populations are more than one million. Total 283 active ingredients were monitoring ( total sample number =232). Single-analysis of target pesticides was for three kinds of possible growth regulators and the multicomponent analysis was for 280 kinds of pesticides, a total of 283 species were selected to perform the pesticide residues. Before monitoring the analytes, the improvements of the analytical methods were done by method validations under the CODEX analytical method development guidelines and can produce metrics that represent the international standards applied in accordance with the guidelines. In addition to residual pesticides detected during monitoring we compare the ADI to EDI values using detected result and dietary consumption data which is extracted from annual market basket survey. The 163 samples were non-detected in the total 232 samples so it means that every agricultural commodity will residual pesticides-free in 70.3%. The detected residual pesticides showed for a total of 69 cases (29.7%). Two of samples violate Korean MRL (0.9%). The ratio of EDI compared to ADI resulted in only from 0.00087 to 0.902%. In result, we can assume that all detected residual pesticides are very safe level and current policies of Korean pesticides control may be working.

Mapping of the Righteous Tree Selection for a Given Site Using Digital Terrain Analysis on a Central Temperate Forest (수치지형해석(數値地形解析)에 의한 온대중부림(溫帶中部林)의 적지적수도(適地適樹圖) 작성(作成))

  • The study was conducted to make a map for selecting righteous tree species for each site by digital terrain analysis. We set an algorithmic value for each tree species' characteristics with distribution pattern analysis, and the soil types were digitized from data indicated on soil map. Mean altitude, slope, aspect and micro-topography were estimated from the digital map for each block which had been calculated by regression equations with altitude. The results obtained from the study could be summarized as follows 1. We could develope a method to select righteous tree species for a given site with concern of soil, forest condition and topographic factors on Muju-Gun in Chonbuk province(2,500ha) by the terrain analysis and multi-variate digital map with a personal computer. 2. The brown forest soils were major soil types for the study area, and 29 tree species were occurred with Pinus densiflora as a dominant species. The differences in site condition and soil properties resulted in site quality differences for each tree species. 3. We tried to figure out the accuracy of a basic program(DTM.BAS) enterprised for this study with comparing the mean altitude and aspect calculated from the topographic terrain analysis map and those from surveyed data. The differences between the values were less than 5% which could be accepted as a statistically allowable value for altitude, as well as the values for aspect showed no differences between both the mean altitude and aspect. The result may indicate that the program can be used further in efficiency. 4. From the righteous-site selection map, the 2nd group(R, $B_1$) took the largest area with 46% followed by non-forest area (L) with 23%, the 5th group with 7% and the 4th group with 5%, respectively. The other groups occupied less than 6%. 5. We suggested four types of management tools by silvicultural tree species with considering soil type and topographic conditions.

Pesticide Residues Survey and Safety Evaluation for Perilla Leaf & Lettuce on the Garak-dong Agricultural & Marine Products Market (가락동 농수산물도매시장 반입 들깻잎과 상추의 잔류농약 실태 및 안전성 평가)

  • This study was conducted to monitor residual pesticides in perilla leaf & lettuce and to assess their risk to human health. The total number of perilla leaf & lettuce were 4,063 and 2,248 respectively and these products were collected at the Garak-dong Agricultural & Marine Products Market sold by auction from 2010 to 2012. Multi-residual analysis of 285 pesticides were performed by GC-ECD, GC-NPD, HPLC-DAD and HPLC-FLD. In perilla leaf, 61 pesticides were detected and detection rate was 20.0%. 28 pesticides were detected in case of lettuce and detection rate was 4.8%. In order to do risk assessment by perilla leaf & lettuce consumption, estimated daily intake of residual pesticides were determined and compared to acceptable daily intake, referring to hazard index (HI values). The range of % HI values of perilla leaf was from 0.000% to 0.049%. The range of % HI values of lettuce was from 0.000% to 0.095%. These results show that the risk caused by pesticide residues in perilla leaf & lettuce intake were very low and these vegetable intake was safe.

Development of a Window Program for Searching CpG Island (CpG Island 검색용 윈도우 프로그램 개발)

  • A CpG island is a short stretch of DNA in which the frequency of the CG dinucleotide is higher than other regions. CpG islands are present in the promoters and exonic regions of approximately $30{\sim}60$% of mammalian genes so they are useful markers for genes in organisms containing 5-methylcytosine in their genomes. Recent evidence supports the notion that the hypermethylation of CpG island, by silencing tumor suppressor genes, plays a major causal role in cancer, which has been described in almost every tumor types. In this respect, CpG island search by computational methods is very helpful for cancer research and computational promoter and gene predictions. I therefore developed a window program (called CpGi) on the basis of CpG island criteria defined by D. Takai and P. A. Jones. The program 'CpGi' was implemented in Visual C++ 6.0 and can determine the locations of CpG islands using diverse parameters (%GC, Obs (CpG)/Exp (CpG), window size, step size, gap value, # of CpG, length) specified by user. The analysis result of CpGi provides a graphical map of CpG islands and G+C% plot, where more detailed information on CpG island can be obtained through pop-up window. Two human contigs, i.e. AP00524 (from chromosome 22) and NT_029490.3 (from chromosome 21), were used to compare the performance of CpGi and two other public programs for the accuracy of search results. The two other programs used in the performance comparison are Emboss-CpGPlot and CpG Island Searcher that are web-based public CpG island search programs. The comparison result showed that CpGi is on a level with or outperforms Emboss-CpGPlot and CpG Island Searcher. Having a simple and easy-to-use user interface, CpGi would be a very useful tool for genome analysis and CpG island research. Development of Error Analysis Program for Phase-based Respiratory Gating Radiation Therapy (위상기반 호흡연동 방사선치료 시 오차 분석 프로그램 개발)

  • The respiratory gating radiation therapy which Irradiates only in the stable respiratory period with analyzing the periodic motion of a reflective marker on the patient's abdomen has been applied to the precise radiation treatment in order to minimize the effect of organ motion induced by the respiration. This respiratory gating system establishes irradiation region using the amplitude-based or phase-based method. Although phase-based method Is preferred because of the stability in the real treatment conditions, it has some limits to explain the exact correlation between the marker motion and organ motion. Even when the variation of amplitude which can introduce target motion considered as an error is produced, the phase-based method has the possibility to irradiate including the error positions. In this study, the error analysis program was developed for the verification of the tumor position's variation correlated with the variation of marker's amplitude which can be occurred during a phase-based respiratory sating treatment. The analysis program was tested with a virtual treatment record file and with a record file using moving phantom which were modified considering the irregular amplitude's variation simulating the real clinical situations. In both cases, accurate discrimination of error points and error calculation were produced. When the treatment record files of a real patient were analyzed with the program, the accurate recognition and calculation of the error points were also verified. The analysis program developed in this study will be applied as a useful tool for the analysis of errors due to the irregular variation of patients' respiration during the phase-base respiratory gating radiation treatment.

Comparison and Analysis of Photon Beam Data for Hospitals in Korea and Data for Quality Assurance of Treatment Planning System (국내 의료기관들의 광자 빔 데이터의 비교 분석 및 치료계획 시스템 정도관리자료)

  • Purpose: Photon beam data of linear accelerators in Korea are collected, analyzed, and a simple method for checking and verifying the dose calculations in a TPS are suggested. Materials and Methods: Photon beam data such as output calibration condition, output factor, wedge factor, percent depth dose, beam profile, and beam quality were collected from 26 institutions in Korea. In order to verify the accuracy of dose calculation, ten sample planning tests were peformed. These Include square, elongated, and blocked fields, wedge fields, off-axis dose calculation, SSD variation. The planned data were compared to that of manual calculations. Results: The average and standard deviation of photon beam quality for 6, 10, and 15 MV were $0.576{\pm}0.005,\;0.632{\pm}0.004,\;and\;0.647{\pm}0.006$, respectively. The output factors of 6 MV photon beam measured at depth of dose maximum for $5{\times}5cm,\;15{\times}15cm,\;20{\times}20cm\;were\;0.944{\pm}0.006,\;1.031{\pm}0.006,\;and\;1.055{\pm}0.007$. For 10 MV photon beam, the values were $0.935{\pm}0.006,\;1.031{\pm}0.007,\;1.054{\pm}0.0005$. The collected data were not enough to calculate average, the output factors for 15MV photon beam with field size of $5{\times}5cm,\;15{\times}15cm,\;20{\times}20cm\;were\;0.941{\pm}0.008,\;1.032{\pm}0.004,\;1.049{\pm}0.014$. There was seven institutions $e{\times}ceeding$ tolerance when monitor unit values calculated from treatment planning system and manually were compared. The measured average MU values for the machines calibrated at SAD setup were 3 MU and 5 MU higher than the machines calibrated at SSD for 6 MV and 10 MV, respectively except the wedge case. When the wedges were inserted, the MU values to deliver 100 cGy to 5 cm depends on manufactures. When the same wedge angle was used, Siemens machine requires more MUs then Varian machine. Conclusion: In this study, photon beam data are collected and analyzed to provide a baseline value for chocking beam data and the accuracy of dose calculation for a treatment planning system.

Development and validation of an analytical method for fungicide fenpyrazamine determination in agricultural products by HPLC-UVD (HPLC-UVD를 이용한 살균제 fenpyrazamine의 시험법 개발 및 검증)

  • Fenpyrazamine which is a pyrazole fungicide class for controlling gray mold, sclerotinia rot, and Monilinia in grapevines, stone fruit trees, and vegetables has been registered in republic of Korea in 2013 and the maximum residue limits of fenpyrazamine is set to grape, peach, and mandarin as 5.0, 2.0, and 2.0 mg/kg, respectively. Very reliable and sensitive analytical method for determination of fenpyrazamine residues is required for ensuring the food safety in agricultural products. Fenpyrazamine residues in samples were extracted with acetonitrile, partitioned with dichloromethane, and then purified with silica-SPE cartridge and eluted with hexane and acetone mixture. The purified samples were determined by HPLC-UVD and confirmed with LC-MS and quantified using external standard method. Linear range of fenpyrazamine was between $0.1{\sim}5.0{\mu}g/mL$ with the correlation coefficient (r) 0.999. The average recovery ranged from 71.8 to 102.7% at the spiked level of 0.05, 0.5, and 5.0 mg/kg, while the relative standard deviation was between 0.1 and 7.3%. In addition, limit of detection and limit of quantitation were 0.01 and 0.05 mg/L, respectively. The results revealed that the developed and validated analytical method is possible for fenpyrazamine determination in agricultural product samples and will be used as an official analytical method.

Monitoring of Pesticide Residues and Risk Assessment on Agricultural Products Marketed in the Northern Area of Seoul in 2013 (2013년 서울북부지역 유통농산물의 잔류농약 모니터링 및 위해성 평가)

  • The aim of this study was to investigate pesticide residues in 2,877 market vegetables in the northern area of Seoul in 2013. Pesticide residues in the samples were analysed by multiresidue method for 285 pesticides using GC-ECD/NPD and HPLC-DAD/FLD. 385 samples(13.4%) were detected with pesticide residues at or below MRL, and 15 samples(0.5%) were found to detect pesticide residues exceeding MRL. The most frequently detected samples were sedum(63.6%), chamnamul(45.8%), leek(44.5%) and green&red pepper(30.8%). Among the 15 violated samples, leek(5 cases) and welsh onion(4 cases) showed the highest violation rate. A total of 74 samples(18.5%) contained multiple pesticide residues in one vegetable. Procymidone, chlorofenapyr and cypermethrin were the pesticide most frequently found. As a tool of risk assessment through the consumption of pesticide detectable agricultural products, the ratio of estimated daily intake (EDI) to acceptable daily intake (ADI) was calculated into the range of 1.05~28.61%. The results have meant that there was no health risk through dieting commercial agricultural products detected with pesticide residues.
