Gene Expression of Detoxification Enzymes in Tenebrio molitor after Fungicide Captan Exposure (살진균제인 캡탄 처리 후 갈색거저리의 해독효소 유전자 발현)

  • The application of fungicides is indispensable to global food security, and their use has increased in recent times. Fungicides, directly or indirectly, have impacted insects, leading to genetic and molecular-level changes. Various detoxification mechanisms allow insects to eliminate reactive oxygen species (ROS) toxicity induced by agrochemicals including fungicides. In the present study, we analyzed the mRNA expression levels of detoxifying enzymes in Tenebrio molitor larvae following exposure to non-lethal doses (0.2, 2, and 20 ㎍/µL) of a fungicide captan. Transcripts of peroxidases (POXs), catalases (CATs), superoxide dismutases (SODs), and glutathione-s-transferases (GSTs) were screened from the T. molitor transcriptome database. RT-qPCR analysis showed that TmPOX5 mRNA increased significantly 24 h post-captan exposure. A similar increase was noticed for TmSOD4 mRNA 3 h post-captan exposure. Moreover, the expression of TmCAT2 mRNA increased significantly 24 h post-treatment with 2 ㎍/µL captan. TmGST1 and TmGST3 mRNA expression also increased noticeably after captan exposure. Taken together, these results suggest that TmPOX5 and TmSOD4 mRNA can be used as biomarkers or xenobiotics sensors for captan exposure in T. molitor, while other detoxifying enzymes showed differential expression.

간의 지방섭취세포의 구조와 기능

  • FSC는 상시지방적을 보유하는 특성을 가지고 있으므로 지방섭취세포(fat-storing cell, FSC)라고 명명되어으며 유동에 노출하는 성세포, 내피세포와 달리 이 세포의 유동면은 교원직유와 내피세포에서 유래하는 윤랑선에 의하여 피복된다. 더욱이 이 세포는 각종 척추동물에 존재한다는 사실이 확인 되었으며 그 지방적은 동물의 종류에 따라 특유한 형을 구비하는 동시에 개체차가 없다. FSC는 이물탐식성이 없으며 지방적을 함유한 성세포와는 위치적, 기능적으로 다르다는 것이 증명되었다. 발생학적으로는 간엽세포에 속하며 유사분제에 의하여 증식능력을 가지고 있다. 이 세포에는 glycogen이 증명되며 모든 지방세포의 실험이 나타나는 성적에서와 유사하게 항시 지방을 합성하여 종의 특유한 형을 가진 지방적으로서 저장됨을 시사하였다. 전현적관륜에 의하여 FSC가 유동주위강(disse腔) 속에 있어 성세포, 내피세포와는 달리 유동에 노출하지 않고 항상 내피세포층과 교원직유에 의하여 유동에서 격리됨을 추정하였다. 이로 인하여 많은 학자들에 의하여 FSC가 승인을 받았다. 오늘날에는 유동을 둘러싸고 있는 상재세포가 내피세포, 성세포 및 FSC의 3종임이 인증이 되었다. 이 세포의 미세구조상의 특징은 기저막이 없는 공허한 세포로 보이나 항상지방적(空胞)을 보유하고 잘 발달한 조면소포체를 가지며 유동내피면에 따라 분기확산하는 돌기를 가지고 있다. 그리고 세포체는 유동주위강을 달리는 무수신경직유의 varicosity (사립체 및 신경결합소포를 함유한 신경종말)와의 사이에 adrenalin 작동성 synaps를 형성한다. FSC의 기능적 의의는 ㄱ, 간소엽내 교원직유형성에 참여 ㄴ. 유동내피하 돌기는 내피를 외측에서 지지하고 보강하며 또한 유동을 둘러싸고 연장하는 돌기는 수축하여 유동강을 축소한다. ㄷ. 지방을 저장하여 간세포의 energe원을 공급하며 VitaminA를 그 속에 저장한다. ㄹ. 간의 해독작용에 관여한다.

Effects of Korean Green Tea, Oolong Tea and Black Tea Beverage on the Antioxidative Detoxification in Rat Poisoned with Cadmium (한국산 녹차, 우롱차 및 홍차가 카드뮴에 중독된 흰쥐 간조직의 항산화적 해독작용에 미치는 영향)

  • In order to investigate the effect of Korean green tea, oolong tea and black tea beverage on the antioxidative detoxification in cadmium(Cd) poisoned rat liver, male Sprague-Dawley rat weighing 143$\pm$3.2g were divided into control and experimental groups. The experimental groups were fed standard diet containing 40ppm Cd and were given distilled water(CD), 5% black tea(BT), oolong tea(OT) and green tea(GT), respectively. Tea beverages were extracted from 5G dry leaves of teas in 100ml hot distilled water by the treatment at 85$^{\circ}C$ for 3 min. Liver xanthine oxidase(XOD) activity was increased by the administration of Cd except GT group. Liver superoxide dismutase(SOD), glutathione peroxidase(GSH-px), glutathione S-transferase(GST) activities were decreased by te administration of Cd but did not decreased by the administration of green tea(in GT group). Vitamin E and reduced glutathione contents were significantly decreased in Cd administered groups. Liver lipid peroxide value in Cd administered groups were increased compared to control group, but was not increased in GT group. Serum glutamic oxaloacetic transaminase(GOT) activities in CD, OT, BT groups were higher than control, but that in GT group was similar to control group. Serum glutamic pyruvic transaminase(GPT) activity was not significantly different among various groups. It was concluded that green tea might alleviate peroxidative damage in Cd-administered rat liver by reinforcing antioxidative detoxification system.

Effect of Korea Ginseng Root on Detoxification of Heavy Metal, Mercury by Fusarium oxysporum (고려인삼이 Fusarium oxysporum에 의한 중금속 수은의 해독작용에 미치는 영향)

  • Extracts of Panax ginseng root significantly induced tolerance of Fusarium oxysporum to heavy metal, mecury, as the fungal mycelial growth was less inhibited by mercury chloride on potato dextrose medium(PDA) amended with ginseng root than on the PDA with no ginseng amendment. The most favorable concentration of ginseng root powder in detoxification of mercury chloride was 1%. The induced tolerance of F. oxysporum to mercury chloride appeared to be rather due to absorption of ginseng components, and was not related to stimulation of mycelial growth of the fungus per so by ginseng treatment. Ginseng components responsible for inducing tolerance of the fungus to mercury were involved in the water fraction of the ginseng root extract, although the water fraction had no effect on enhancement of the mycelial growth on the medium without mercury chloride. The hexane fraction of ginseng root extract, by which the mycelial growth was stimulated, was not related to the inducement of the tolerance to mercury chloride. However, more tolerance to mercury chloride was noted in PDA with both the water and hexane fractions combined than with either of the two fractions. Six-year-old ginseng roots were more effective in detoxification of mercury chloride than 4-year-old ginsng roots, and American ginseng (P quinquifolium) had no or little effect on inducing tolerance of the fungus to mercury chloride. This method may be used to screen other natural materials for test in the detoxification of mercury chloride.

  • The enzymatic studies on the methylglyoxal metabolism in yeast and bacterial cells indicated that organisms are equipped with the common and manifold systems for the detoxification of methylglyoxal. Among these systems, the glyoxalase I is the most important route for methylglyoxal detoxification. The molecular structure of glyoxalase I is apparently distinct from the enzyme sources, and zinc ion is an essential cofactor in enzyme activity. The gene for Pseudomonas putida glyoxalase I functioned as a scavenger of methylglyoxal and regulated the cell size of the bacterium. Comparison of the nucleotide sequence of the P. putida glyoxalase I gene with the N-terminal amino acid sequence of the purified enzyme revealed that the N-terminal methionine residue was removed after translation. Possible physiological role of glyoxalase I was also discussed.

  • Isoenzymatic analysis related with cadmium adaptation and detoxifying mechanism were carried out upon Rhizopus oryzae. When cadmium was added into R. oryzae culture, activities of malate dehydrogenase (MDH) and glucose phosphate isomerase (GPI) related with carbohydrate metabolizing pathways were stimulated. Novel isoenzyme CAT-2 related with removing intracellular toxic peroxides, was induced lately and derepressed very highly. On the other hand, lactate-catabolizing enzymes such as lactate dehydrogenase (LDH) and alcohol dehydrogenase (ADH) were repressed. These results strongly suggest that, under cadmium stress, much of derepression of enzymes relating with central metabolism such as TCA cycle that produces high yield of energy and relating with removal of toxic peroxides should be necessary.

The Effect of old Antler on the Galactosamine-induced Hepatotoxicity in Rate (Galactosamine에 의해 유도된 녹각추출물이 간장해에 미치는 영향)

  • This study was conducted to investigate the effect of old antler extracts on galactosamine-induced liver injuries in rats. Male rats of Sprague-Dawley strain with average weight of 110$\pm$10g were fed on diets containing three kinds of old antler extracts(water extract, neutral extract and ether extract) for four weeks. Galactosamine(400mg/kg body weight) was injected intraperitoneally at the same time every week in galactosamine treatment groups. Cytochrome P-450 content was decreased in galactosamine treatment groups and increased by old antler extracts administration. Glutathione-peroxidase activity was increased in water extract group. Hepatic glutathione content was not observed significant differences by the old antler extracts administration. Lipid peroxide content was higher in the galactosamine treatment groups than that of the control group and decreased in galactosamine administerd groups after pretreatment with water extract. Total lipid, triglyceride and total cholesterol contents of liver were decreased in old antler extracts administerd groups and decreased in water extract group.

  • The purpose of this study is to suggest guidelines for drawing and keeping customers through knowing function of herbal and food. Customers have known herbal foods to high nutritional foods, prevention of high blood pressure, heart disease, and obesity. Also, people recognized highly garlic which has function prevention cancer, brown seaweed which is controlled blood pressure, and ginkgo nut which is helpful for asthma, detoxication. They know specifically function of herbal materials such as mugwort, ginseng, and royal golly. The guidelines are as followed. The first, seasonal herbal tea provide in stead of water like medicinal foods. The second, various cereals, Chinese herbal materials, and vegetables are added to the main rice. So it can make to eat everyday like nutrition rice which is mixed with garlic and ginkgo. The third, specific function food' and herbal's should be explained on the menu. Therefore, it make attract customer's curious and interest.

Protective Effect of DWP-04 Against Hepatotoxicity Induced by D-galactosamine (흰쥐에서 DWP-04가 D-galactosamine에 의해 유도된 간독성의 보호효과)

  • This study was conducted to investigate the biological activity and hepatoprotective effect of DWP-04 [DDB : selenium yeast: glutathione (31.1 : 6.8 : 62.1(w/w/w)] in D-galactosamine (GaIN) intoxicated rats. The DWP-04 (50, 100 or 200 mg/kg) or its vehicle was orally administered everyday before the start of GaIN injection (400 mg/kg, ip) for two weeks and animal decapitated for 24 hrs after GaIN­injected. The activities of serum enzymes, markers of liver function, were increased in the GaIN group compared to normal group and significantly lowered in the DWP-04 pretreated group than in the GaIN group. Hepatic lipid peroxide level and activities of phase 1 enzymes were significantly higher than those of GaIN group compared to normal group and lower in the DWP-04 pretreated group than in the GaIN group, and phase II enzyme activities in liver were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Total hepatic glutathione content and glutathione biosynthesis enzymes were lower in the GaIN group than in the normal group and were increased in the DWP-04 pretreated group than in the GaIN group. Therefore, the current results indicated that DWP-04 administration alleviated the GaIN-induced adverse effect through enhancing the antioxidant enzyme activities.

Protective Effect of Oenanthe javanica Extract on the Carbon Tetrachloride-Induced Hepatotoxicity in Mice (미나리추출물이 사염화탄소에 의한 마우스 간손상에 미치는 영향)

  • The present work was undertaken to investigate the protective mechanism of Oenanthe iavanicu n-butanol extract on the carbon tetrachloride-induced hepatotoxicity in mice. It was observed that a striking enhancement of serum alanine aminotransferase and hepatic lipid peroxide content after carbon tetrachloride administration were markedly decreased by the presentment of Oenanthe javanica extract for 5 days. It was also observed that the hepatic aniline hydroxylase, catalase, glutathione S-transferase activity and glutathione content were not changed by the injection of Oenanthe javanica extract for 5 days. Whereas, hepatic xanthine oxidase activity was inhibited by the treatment of Oenanthe javanica extract for 5 days. After treatment with Oenanthe javanica extract, xanthine oxidase activity was decreased with dose and time-dependent manner as compared to control group. However, hepatic xanthine oxidase activity was not affected by the addition of Oenanthe javanica extract in vitro. These results suggest that the inhibition of hepatic xanthine oxidase activity by the injection of Oenanthe javanica extract is believed to be a possible protective mechanism for the carbon tetrachloride-indured hepatotoxicity in mice.
