PHARMACEUTICAL COMPOSITION OF VELVET ANTLER (녹용의 약리학적 성분)

  • 알려진 녹용의 효능을 과학적으로 증명하고 활성성분을 이용하여 신의약품 개발에 기여하고자 한다. 본 연구실에서는 녹용의 가장 대표적인 효능인 조혈작용을 하는 성분을 구조 분석하여 MADG임을 밝히었다. MADG는 조혈촉진기능뿐만 아니라 대식세포 증식기능 그리고 비장세포성장을 촉진하는 등 면역활성 조절제로써의 기능도 나타내었다. 그리고 녹용에 존재하는 인지질 중에서 포화지방산을 가지고 있는 phophatidylcholine도 면역활성을 촉진시키었다. 항진균작용을 하는 성분을 구조 분석한 결과 lysophophatidylcholine임을 밝히었으며 그 유도체인 CHJ-ester가 숙주세포에는 해를 주지 않으면서 항진균작용을 하는 것을 관찰하였다. 그리고 골다공증치료효과를 검색하기 위해 녹용 정제성분으로 연구한 결과 파골세포로의 분화를 100$\%$ 저해함을 밝히므로 녹용이 골다공증 치료효과가 있음을 증면하였다. 녹용성분의 유도체인 SCOH가 파골세포분화를 억제함을 밝혀 현재 골다공증치료제로써의 가능성을 조사하고 있다. 본 연구실에서는 1999년부터 녹용연구를 시작하여 현재까지 10여 개의 활성성분을 분석한 바 있다. 현재 조혈세포촉진작용, 면역증진작용, 골다공증치료작용, 대식세포 증식작용, 항진균작용, 항간질작용에 대한 성분 분석 및 그 약리 기전을 연구하고 있다.

Isolation of Bacillus sp. SW29-2 and Its Antifungal Activity against Colletotrichum coccodes (Bacillus sp. SW29-2의 분리 및 Colletotrichum coccodes에 대한 항진균 활성)

  • Antifungal bacterium against Colletotrichum coccodes causing black dot disease of potatoes and anthracnose of tomatoes was isolated from sewage sludge. The isolate showed a 99% sequence homology of partial 16S rRNA of Bacillus methylotrophicus CBMB205 and Bacillus amyloliquefaciens subsp. plantarum FZB42. The isolate was identified as Bacillus sp. SW29-2, using the neighbor-joining phylogenetic tree, BlastN sequence analysis, and morphological and cultural characteristics. Bacillus sp. SW29-2 is an aerobic, Gram-positive, endospore-forming bacterium, of which the morphological and physiological characteristics were the same as those of type strain B. lichniformis CBMB205, except for the cell growth of over 4% NaCl. The cell growth of the temperature and the initial pH of the medium was shown at $18-47^{\circ}C$ (opt. ca. $38^{\circ}C$) and 3-9 (opt. ca. 6.0), respectively. The inhibition size (diameter) of Bacillus sp. SW29-2 against four strains of C. coccodes ranged from 23 to 29 mm. Also, the isolate showed antifungal activity against penicillium rot-causing Penicillium expansum in apples. Thus far, any report on the antifungal activity of Baciilus spp. against C. coccodes has not been found. These results suggest that the Bacillus sp. SW29-2 isolate could be used as a possible biocontrol agent against C. coccodes, and further applied to other plant pathogenic fungi.

Antimicrobial Effects of Chamaecyparis obtusa Essential Oil (편백정유의 항균효과)

  • Essential oil of Chamaecyparis obtusa showed antimicrobial on relatively broad spectrum of bacterial and fungal species. Staphylococcus epidermidis was highly sensitive to the essential oil but Streptococcus aureus and Streptococcus mutans were not. Vibro parahemolyticus, Pseudomoas aeruginosa and Pseudomoas putida showed sensitivity at the concentration higher that 400 ppm, Thegrowth of a pathogenic yeast Candida albicans was inhibited by the essential oil above 200ppm. The radialgrowth of several filamentous fungi was also inhibited The antifungal activity of the essential oil was effective on two plant pathogens Fusarium oxysporum and Altenaria mali. These results sug-gest that essential oil of Chamaecyparis obtusa has an antimicrobial activity by inhibiting bacterial and fungal species.

  • Antifungal activities of cinnamaldehyde derivatives against various fungi were investigated using paper disc diffusion method. Among the derivatives tested, ${\alpha}-chlorocinnamaldehyde$ was stronger than cinnamaldehyde in antifungal activity and was effective in inhibiting the growth of the representative fungi of dermatomycosis with minimum inhibitory concentration of $9.76{\sim}19.5\;{\mu}g/ml$. A comparison of antifungal activity of cinnamaldehyde derivatives revealed that antifungal action of cinnamaldehyde might be related to a basic structure of acrolein.

  • Bacillus lichemiformis Kll, a plant growth promoting rhizobacterium was reported as a producer of auxin, siderophore, as well as antifungal cellulase under some culture conditions. In vitro test, B. licheniformis Kll represented excellent antagonistic ability against Fusarium oxyspoum (KACC 40037), and showed broad spectrum against other phytopathogenic fungi. B. licheniformis Kll had cellulolytic activity toward not only carboxymethyl-cellulose (CMC) but also insoluble cellulose, such as fungal cell wall cellulose, filter paper (Whatman No. 1), and Avicel. In addition, we confirmed antifungal substance production by butanol-extract methods. The strain produced optimally the antifungal substance when it was cultivated at pH 9.0, 30${\circ}$C for 4 days on nutrient medium. The biological control mechanisms of B. lichemiformis Kll were caused by antifungal substance, cellulase and siderophore against phytopathogenic fungi.

Biological Control of Pythium Root Rot by Radiation Induced Mutant, Bacillus lentimorbus WJ5a17 (방사선유도 돌연변이체 Bacillus lentimorbus WJ5a17에 의한 Pythium Root rot의 방제)

  • To control pythium root rot, Bacillus lentimorbus WJ5a17 with high anti-fungal activity against Pythium ultimum was induced from B. lentimorbus WJ5 by gamma radiation ($^{60}Co$). The biocontrollers of FWJ5 and FWJ5a17 were formulated ($1.0\times 10^{11}$) with B. lentimorbus WJ5 and WJ5a17, respectively, The population density of FWJ5 and FWJ5a17 maintained highly up to $1.0\times 10^{9}$ CFU $g^{-1}$ in nursery and field soils until 30 days after treatment. P. ultimum spores germination were inhibited 71.0% and 81.4% by FWJ5 and FWJ5a17, respectively. Pythium root rot of yea pepper, Chinese cabbage and radish were significantly (p < 0.05) controled by one time treatment of FWJ5 and FWJ5a17.

  • This study was performed to examine the possibility of Fig extracts for the natural product research. The Fig extracts method were wash, dry and extracted and prepared into two parts; fruit, leaf. The extracts were prepared using alcohol and their chemical structures were estimated by column chromatography, GC-mass spectroscopy, IR, $^1H$-nmr data-base. In order to apply the extracts to hair-care cosmetics, antimicrobial and antifungal ability of the extracts were accomplished using broth serial dilution method. and In order to customer's interest satisfy and test to Fig's industrial propriety of accompany with table luxuries change, The author suggested that to test Fig's industrial property for hand made of transparent and solid soap contained of Fig extracted materials

Antimicrobial Activity of Extracts from Different Parts and Essential Oil from Pinus densiflora on Skin Pathogens (소나무 부위별 추출물 및 essential oil의 피부상재균에 대한 항균 활성)

  • This study was carried out to examine the antimicrobial activity of 70% ethanol and hot water extracts from different parts and essential oil in Pinus densiflora on skin pathogens such as Staphylococcus epiermidis, S. aureus, Propionibacterium acnes, Candida albicans, C. tropicalis, and Trichophyton rubrum. The antimicrobial activities of extracts and essential oil were tested by paper disc assay and minimum inhibitory concentration test. The ethanol extract of pine pollen showed antimicrobial activity against C. tropicalis and T. rubrum. Ethanol extract of pine needle and pine gnarl showed antimicrobial activity against S. aureus, C. tropicalis, and P. acnes. Essential oil from pine needle exhibited antimicrobial activity against C. albicans, C. tropicalis, and P. acnes. The minimum inhibitory concentration of ethanol extracts of pine needle and pine gnarl against P. acnes and C. tropicalis ranged from 0.002% to 0.0063%. The lowest minimum inhibitory concentration (0.025%) against C. albicans and C. tropicalis was obtained from essential oil. These results indicate that ethanol extracts of pine pollen, pine needle, pine gnarl, and essential oil of pine needle could be applicable to control the skin infection pathogens. Especially, ethanol extract of pine gnarl had a broad spectrum of antimicrobial activity and pine extracts and essential oil exhibited higher antimicrobial activity with Candida sp. and P acnes.
