The development of antibody titers and crossreaction between Sarcocystis and Toxoplasma were investigated by means of IF A test and ELISA in pigs experimentally infected with $1.5{\times}10^6$ S. suicanis sporocysts and 10,000 T. gondii oocysts, respectively. The intact and soluble Sarcocystis antigens were prepared from the bradyzoites harvested by peptic digestion of infected pork. The intact and soluble Toxoplasma antigens were prepared from the tachyzoites in mouse peritoneal cavity. IgG antibodies in pigs infected with Sarcocystis and Toxoplasma, respectively were detected first at 2 weeks post infection on both IF A test and ELISA. The antibody titer to Toxoplasma reached its maximum at 6 weeks post infection and decreased thereafter. The antibody titer to Sarcocystis reached its maximum terminally. The cross-reaction titer in pigs infected with Toxoplasma against Sarcocystis antigen was up to 1 : 16 in IFA test and up to 1 : 32 in ELISA. The titer in control group was below 1 : 4 in both reactions.
Antigenic components reacting with IgE and IgG antibodies were localized in muscular layer of adult and of larva, sparganum. But the antigenic components inducing IgG were localized at tegument and parenchyma in addition to muscular layer in adult and sparganum. Also in sparganum, the surface of calcareous corpuscles of parenchyma showed immunoreactivity to IgG antibody. However antigenic components inducing IgE antibody were not localized in tegument and parenchyma, but in adult worm, we observed the immunopositive reaction at the lining of vitelline follicles in mature proglottis and on surface of egg shell within uterus of graved proglottis. By the method of immunogold-labelling, we observed the location of antigenic particles in tegument of sparganum. The density of antigenic particles inducing IgG was higher than that of antigen particles inducing IgE in syncytial tegument, tegument cells. A total of 43 and 36 protein bands were resolved from crude extracts of adult and sparganum, respectively, by SDS-PAGE. 34 bands from crude extracts of adult and larva were migrated to same positions. By EITB, 21 bands of 44 bands in adult were recognized with IgG antibody, and also 21 bands of 36 bands in sparganum. 13 bands of them were common antigenic components both in the adult worm and sparganum. Because 19 bands of 44 bands in adult worm were reacted with IgE antibody, they were IgE antigenic component. In sparganum, 13 bands were IgE antigenic components. 9 bands of them were common antigenic component inducing IgE antibody in both a-dult and sparganum. 3 bands of antigenic component recognized by IgE and IgG antibody were nonspecific antigen in both adult and sparganum of Spirometra erinacei.

Streptococcus mutans is known to be a major causative organism of human dental caries. The development of a vaccine against dental caries involves identification of appropriate antigens of mutans streptococci against which protective immune responses can be mounted, and the selection of a method of immunization that will generate sustained levels of protective antibodies. Antigens receiving most attention include streptococcal surface proteins that are involved in attachment to tooth surfaces and glucosyltransferases (GTF) that synthesize adhesive glucans from sucrose. The induction of antibody responses to orally administered antigens is often difficult due to digestive destruction of antigens and immune tolerance. Here we report the induction of antibody responses to an anti-caries vaccine containing retinoic acid (RA). Subcutaneous immunization with formalin-fixed bacteria or GTF supplemented with RA induced higher serum IgM and IgA responses to GTF compaired to oral adminstration. Antisera induced by Ingbritt strain showed partial cross-reaction with LM-7 strain, but not with OMZ175. These results suggest that subcutaneous immunization with GTF combined with an immunomodulator, RA, may be applied to anti-caries vaccine.
Monoclonal antibodies (Mabs) were produced against crude scolex extract of T solium metacestodes, and applied to ELISA-inhibition test for improving the specificity of serodiagnosis of human cysticercosis. Four hybridomas secreting species-specific anti- cysticercal Mabs (Cya-1, Cya-7, Cya-28 and Cya-31) were selected. Each Mab reacted on antigenic components of 25.5 kDa (Cya-1), 28 kDa (rcya-7), 87.5 kDa (Cya-281), and 12.5 kDa (Cya-31). IFA showed that Cya-1 was located at the calcium corpuscles, and Cya-7 at the loose connective tissue of T soLium metacestode scolex. Cya-28 and Cya-31 reacted on the tegument of the scolex. By conventional ELISA, 23 out of 28 (82.1%) cysticercosis patients were found serologically positive, but 1 out of 9 (11.1%) sparganosls cases and 6 out of 31 (19.4%) paragonlmiasis cases showed false positives. By ELISA-Inhibition test using species-specific anti-cysticercal Mab Cya-7, 19 out of 28 (67.9%) cystlcercosis cases were found serologically positive, but there were no false positives In other parasitic infections.
인체감염이 다발하는 폐흡충증의 혈청학적 진단에서 항원으로 사용하는 성충추출액은 여러단계의 질환 이행과정을 진단하는 항원으로서 문제가 있다. 이를 해결하려면 먼저 추출액내의 성분단백질의 성상을 파악할 필요가 있다. 이 연구에서는 폐흡충 성충 추출액으로 면역시킨 BALB/c mice의 비장세포와 SP2/0 형질세포종 세포를 세포융합하여 제작한 단세포군항체를 이용하여 친화성크로마토그래피로 폐흡충 성충의 구성단백질의 일부를 순수분리하고 성상을 관찰하였다. 그 결과는 다음과 같다. 1. 세포융합으로 PFCK-21, PFCK-44, PFCK-136, PFCK-189 등 4종류의 단세포군 항체를 얻었다. 그중 PFCK-21과 PFCK44는 17k Da, PFCK-136은 23, 46, 92 kDa 단백질에 반응하였고 PFCK-189는 여러종류의 단백질에 반응하였다. 2. PFCK-44 단세포군항체를 고리로 친화성 크로마토그래피를 실시하여 분리한 성분 단백질은 disc-PAGE상 4번째에 위치하고 분자량이 17 kDa로 알려진 단밸질이었다. 이 단밸질은 17 kDa의 monomer로 판단하였다. 면역조직화학염색을 실시한 결과 이 단밸질은 장관 상피세포에 반응하고 있었다. 3. PFCK-136 단세포군항체로 순수분리한 단밸질은 disc-PAGE상 1번 단밸질(440 kDa)이었으며 환원성 SDS-PAGE에서는 23 kDa 단밸질이었다. 면역조직화학염색으로 이 단세포군항체는 충란내 세포에 강하게 반응하고 있었다.
This study was carried out to investigate dietary effects of illite on humoral immune response against Salmonella typhimurium flagella in Hyline Brown laying hens. Total of twenty eight 36-week-old hens were divided into two groups; The first was fed commercial diet as control and the other was fed diet containing 2 % illite. Each group was divided into 2 sub-groups and then performed immunization of Salmonella flagella with different adjuvants which were Freund's adjuvant and croton oil. The rates of egg-production in all groups were normal range but no difference was found between illite-treated and untreated groups. The serum antibody titers of Freund's adjuvant-used subgroup in illite fed groups were significantly increased, especially from 6-to 9-week after $1^{st}$ immunization compared to those of control groups (p<0.05). And the antibody titers of croton oil-used subgroup in illite-fed groups were also significantly increased, especially at 4-, 6- and 7-week (p<0.05). The results demonstrated that the feeding illite stimulated the immune response against S. typhimurium flagella antigen in laying hens and suggest that the supplementation of illite to the poultry diets may support protective effects against bacterial infections such as Salmonellosis.
Antigenic analysis of Herpes simplex type 2 virus was performed and its major antigen was localized using an immunoelectron microscopy. Antigens of 32, 43, 59 and 69 kDa were constantly expressed during the course of infection for 48 hr in the infected Vero cell. An antigen of 51 kDa was turned out to be the major one in inducing a immune response in Western-blot analysis. The 51 kDa antigen was localized on the surface of HSV-2 by immunoelectron microscopy using colloidal golds and anti-HSV 2 polyc1onal antibody. Immunofluorescence assay indicated that viral antigens were found throughout the infected cell and, especially, on the surface of the cell.
The applicability of indirect immunoftuorescent antibody test (IFAT) was compared with enzyme-linked immunosorbent assay (ELISA) in sera from 163 cases of confirmed neurocysticercosis, 101 other neurologic and parasitic diseases and 100 normal controls. As antigen, frozen sections of a Taenia solium metacestode from a human brain was used in IFAT and cystic fluid was used in ELISA. For the detection of specific IgG antibody, IFAT was equally sensitive (89.6%) and specific (85.1%) as ELISA. The antibody titers by IFAT were correspondingly increased with mean absorbance of ELISA. The corresponding rate of positivity in the two techniques was 90.8%. Except for the difficulty in detecting antibodies in cerebrospinal fluid (CSF), IFAT was concluded to be very useful for the serodiagnosis of human neurocysticercosis.

In this study, PAT protein of genetically modified maize was prepared from the recombinant E. coli strain BL21 (DE3), and mice were immunized with the recombinant PAT protein. After cell fusion and cloning, two hybridoma cells (PATmAb-7 and PATmAb-12) were chosen since the monoclonal antibodies (Mabs) produced by them were confirmed to be specific to PAT protein in the indirect enzyme-linked immunsorbent assay (ELISA) and western blot tests. There were no cross-reactions of either Mabs to other GM proteins or to the extracts of non-GM maize. The ELISA based on the PATmAb-7 can sensitively detect 0.3 ng/g PAT protein in corn. These results indicate that the developed Mabs can be used as bio-receptors in the development of immunosensors and biosensors for the rapid and simple detection of GM corn adulterated in foods.
돼지 생식기 호흡기 증후군 바이러스의 nucleocapsid와 반응을 하는 SDOW17 단크론항체를 이용하여 중성 포르말린에 고정시킨 자연감염된 포유자돈의 폐장에서 면역조직화학법을 이용하여 돼지 생식기 호흡기 증후군 바이러스 항원을 확인하였다. 서울대학교 수의과대학 병리학교실에 의뢰된 포유자돈들 중에서 병리조직학적으로 폐장에서 간질성 폐렴이 관찰된 포유자돈 7두를 임의로 선택하여 본 실험을 실시하였다. 간질성 폐렴의 병변으로 많은 수의 대식세포 침윤을 동반한 폐포벽 두께의 증가와 제II형 폐포세포의 비후가 관찰되었다. 검사한 7두 포유자돈중에서 6두에서 돼지 생식기 호흡기 증후군 바이러스에 대한 항체를 enzyme-linked immunosorbent assay에 의해 확인하였다. SDOW17 단크론항체를 이용한 면역조직화학염색과 간질성 폐렴의 대식세포에서 돼지 생식기 호흡기 증후군 바이러스의 항원을 검출하였고, 항원은 (주로)대식세포의 세포질에서만 진한 갈색의 양성반응이 관찰되었다. 이상 검사결과 돼지 생식기 호흡기 증후군 바이러스는 폐장의 간질과 폐포강에 분포되어 있는 대식세포에서 주로 증식하는 것으로 판명되었다. 본 실험에서 사용한 면역조직화학법은 돼지 생식기 호흡기 증후군 바이러스 감염여부를 바이러스 분리 또는 혈청검사 없이 진단하는데 사용할 수 있는 유용한 진단방법으로 판명되었다.
