A study on the antioxidant activity of products of caramel-type-browning reaction (Caramel 갈색화 반응 생성물의 항산화성에 관한 연구)

  • The study was carried out to compare the antioxidant activities of products from caramel-type-browning reaction of xylose(XY), glucose(GL), sucrose(SU), glucose + citric acid (GLCA), glucose + sodium citrate(GLSC), glucose + glycine(GLGC) heated at 80, 120 or 140$^{\circ}C$ for 24 hr. 1. The hydrogen donating ability (HDA) of browning reaction products was generally enhanced as the browning temperature and time increased. The HDAs of the browning reaction products heated at 80$^{\circ}C$ for 24 hr were in the order of GLSC (0.387) > GLSC (0.362) > GLCA (0.301) > GL (0.299) > XY (0.290) > SU (0.281). But they were in the order of GLSC (0.543) > SU (0.328) > GL (0.309) > GLGC (0.325) > XY (0.298) > GLCA (0.275) under the condition of heating at 140$^{\circ}C$ for 24 hr. 2. The antioxidant activities of the anhydrous ethanol extracts of the browning mixtures were inferior to that of TBHQ as measured in com oil, but SU was superior to tocopherol in its antioxidant activity. All the browning mixtures showed antioxidant activities when heated at 80$^{\circ}C$; however, only SU and GLCA showed the activites at 120 or 140$^{\circ}C$. And the antioxidant activity of the SU extract was higher than that of TOCO. The antioxidant activities of the ethanol extracts were in the order of TBHQ > GLCA > GLGC > TOCO > SU > XY > GL > GLSC > control at 80$^{\circ}C$, TBHQ > SU > TOCO > GLCA > control > GLSC> XY > GL > GLGC at 120$^{\circ}C$, and TBHQ > SU > TOCO > GLCA > control > GLSC > GLGC > XY > GL at 140$^{\circ}C$.

Antioxidant Activity and Inhibitory Effect of Taraxacum officinale Extracts on Nitric Oxide Production (서양민들레 부위별 추출물의 항산화활성 및 nitric oxide 생성저해효과)

  • In the current study, antioxidant properties of extracts from different parts of Taraxacum officinale were determined by measuring the radical scavenging activity of 1,1-diphenyl-2-picryl hydrazyl(DPPH) and 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). The antioxidant activity of aerial and root parts of this plant was measured in extracts obtained from various solvents, e.g., water, 50, 70, 100% ethanol, and hot-water. The hot-water extracts had the highest radical scavenging activities. A further study was conducted to determine the total antioxidant activity of hot-water extract fractions obtained from different solvents, e.g., chloroform, ethyl acetate, n-butanol, and water. The ethyl acetate fraction of this extract displayed the strongest DPPH and ABTS radical scavenging activity and the highest total phenolic contents. The inhibitory effect of individual solvent fractions on the production of nitric oxide in LPS-stimulated RAW 264.7 cells was measured. The results indicated that the ethyl acetate fraction of aerial and roots part extracts significantly reduced nitric oxide productions.

Protective Role of Curcuma longa L. Extracts on Hydrogen Peroxide-Induced DNA Damage in Human Leukocytes (산화적 스트레스로 유도된 인체 백혈구 DNA 손상에 대한 울금 추출물의 보호효과)

  • Curcuma longa L. (CL) is widely used as a spice and coloring agent in several foods, such as curry and mustard, as well as cosmetics and drugs. In this study, we investigated the protective effects of CL extracted with various solvents [methanol (MC), ethanol (EC), acetone (AC)] on $H_2O_2-induced$ DNA damage in human leukocytes along with total polyphenol contents (TPC) and antioxidant properties. The antioxidant effects of CL were determined by measuring 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (RSA) and superoxide dismutase (SOD)-like activity. The preventive effect of CL on oxidative stress-induced DNA damage and DNA repair capacities were assessed using comet assay. MC showed the highest TPC (11.17 g gallic acid equivalents/100 g) and antioxidant properties among the solvent extracts. The $SC_{50}$ for DPPH RSA was MC: 35.0 > AC: 45.8 > EC: $57.8{\mu}g/mL$ and SOD-like activity was MC: 46.6 > EC: 141.5 > AC: $296.4{\mu}g/mL$. In the comet assay, the $ED_{50}$ value of MC showed the highest inhibition ($86.7{\mu}g/mL$) of $H_2O_2-induced$ DNA damage, followed by AC ($110.0{\mu}g/mL$) > EC ($115.8{\mu}g/mL$). Analysis of the percentage of damaged cells showed that repair capacity significantly decreased at 4, 8, and 12 h from $H_2O_2-induced$ oxidative stress in each extract. After 12 h, level of DNA damage recovery was similar to the negative control level. These results suggest that CL has potential antioxidant activity and a protective effect against oxidation-induced DNA damage, and the methanol extract of CL was the most effective.

Comparison of Effect of Water and Ethanolic Extract from Roots and Leaves of Allium hookeri (물과 주정을 이용한 삼채 뿌리와 잎 추출물의 항산화 활성 및 항염증 효과 비교)

  • The aim of this study was to evaluate the antioxidant activities and anti-inflammatory effects of water and ethanolic extracts from Allium hookeri roots and leaves. Antioxidant activities of Allium hookeri extracts were determined based on various radical scavenging activities using an ESR spectrophotometer, ferric reducing antioxidant power, 2,2'-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) radical scavenging activity, and oxygen radical absorbance capacity assays. In addition, we investigated anti-inflammatory effects of Allium hookeri extract on lipopolysaccharide (LPS)-induced inflammation in RAW264.7 cells. We also explored the effects of extract from Allium hookeri root on suppression of pro-inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines such as IL-6 and TNF-${\alpha}$ against LPS-induced activation of RAW264.7 cells. Our results demonstrated superior antioxidant activity for leaf extract of Allium hookeri compared to extract from root of Allium hookeri. On the other hand, root extract of Allium hookeri showed better anti-inflammatory activity compared to leaf extract. Our study suggests that Allium hookeri extract exhibits strong antioxidant activity and anti-inflammatory effects and can be developed as a potential therapeutic candidate for diseases involving oxidative stress and inflammation.

Composition of Phenolic Acids and Flavonoids and Skin Care Cosmetic Antioxidant Activity of Akebia quinata Fruit Extracts (연복자 추출물의 Phenolic acids와 Flavonoids의 조성 및 화장품 항산화 활성)

  • In order to verify the effect of using functional cosmetic ingredients for skin beauty, the composition and content of some phenolic acid and flavonoids in the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds were analyzed, and the skin care antioxidant activity was investigated. The results are as follows. Total polyphenol and flavonoid contents were found to be higher in pericarp extract than seed extract in both hot water and 80% methanol extract. In the analysis of the composition and content of phenolic acid and flavonoids, two kinds of phenolic acids were identified in the hot water extract of pericarp, 6 kinds of phenolic acids were identified in the 80% methanol extract, and one kind of flavonoid. Two types of phenolic acids were identified in the hot water and 80% methanol extract of seeds, respectively. DPPH and ABTS radical scavenging activities tended to increase in proportion to the treatment concentration in both hot water and 80% methanol extracts, and antioxidant activity was found to be high. Therefore, from the above results, it was found that the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds contained various kinds of phenolic acid and flavonoids, and the antioxidant activity was also high. It is believed to be of value as a natural antioxidant in skin care cosmetic ingredients.

Antioxidant Properties in Water and 70% Ethanol Extracts of Houttuynia Cordata Thunb (어성초의 추출물에 따른 항산화력 연구)

  • Houttuynia cordata Thunb is a famous traditional Chinese medicine. Also it has rich in functional and bioactive substances with outstanding immunopotentiation, anti-cancer, antimicrobial, and anti-aging effects. This study was evaluated water and 70% ethanol extracts of Houttuynia cordata Thunb using in vitro system antioxidant properties methods. The scavenging DPPH and ABTS radicals of 2 mg/mL 70% ethanol extract were 91.8 and 54.8%, respectively. Also, The FRAP assay of this showed increase in activity. In results, the antioxidant properties of 70% ethanol extract increased significantly compared to water extract(p < 0.05). Therefore, 70% ethanol extract of Houttuynia cordata Thunb may contribute to antioxidant property.

Comparison of antioxidant activities of sea buckthorn (Hippophae rhamnoides) leaf extracts at different ethanol ratios (비타민나무(Sea Buckthorn, Hippophae rhamnoides) 잎 에탄올 농도별 추출물의 항산화활성 비교)

  • This study was conducted to investigate the potential of sea buckthorn leaf (SBL) as a natural antioxidant. SBL was extracted using different ethanol ratios (0, 40, 80, and 100%), and the resulting extracts were examined for their antioxidant contents and antioxidant activities using various methods. The 40% and 80% ethanol extracts (EEs) showed higher total polyphenol contents of 285.06 and 285.64 mg GAE/g respectively, than those in other solvents, and as the ethanol concentration increased, other antioxidant contents of the extracts increased. The 40% and 80% EEs recorded the highest radical scavenging activities against DPPH, superoxide-, ABTS+, nitrite scavenging activity (NO-), and reducing power (RP). The NO- and RP of the SBL EEs were higher than those of ascorbic acid or tannin, which are known as representative antioxidants. The results of this study, suggest that SBL is a potential source of natural antioxidants.

구기자(Lycium chinence Miller) 첨가 요구르트의 항산화 활성과 ACE 저해 및 ${\alpha}-glucosidase$ 저해 효과

  • 구기자, 구기엽, 지골피 추출물을 이용한 요구르트를 제조하여 항산화효과, ACE (angiotensin converting enzyme) 저해 효과 및 ${\alpha}-glucosidase$ 저해 활성을 시험하였다. 구기자 첨가 발효유의 Antioxidants activity는 물 추출물보다 methanol 추출물에서 더 많은 항산화 활성능을 보였으며 특히 구기엽 요구르트에서 83.85%의 높은 활성능을 보였다. 구기자와 지골피 요구르트에서는 각각 47%와 54%내외의 활성능을 나타냈다. 구기자 첨가 발효유의 ACE(angiotensin converting enzyme) 저해 효과는 원재료에서는 물 추출물이 angiotensin converting enzyme 저해능이 높았고, 구기자 요구르트의 경우 물 추출물과 methanol 추출물 모두가 angiotensin converting enzyme 저해능이 높은 것으로 나타났다. 구기자 첨가 발효유의 ${\alpha}-glucosidase$ 저해 활성은 구기자, 구기엽 및 지골피 4% 첨가 요구르트에서 methanol 추출물의 경우 각각 8.2%, 5.6%, 7.3%의 ${\alpha}-glucosidase$ 저해능을 나타냈는데 이들 모두 원재료 보다는 활성 저해능이 큰 것으로 나타났으나 물 추출물의 경우에는 활성 저해능이 거의 나타나지 않았다.

정향추출물의 항산화 효과

  • 정향을 물, 메탄올, 에테르로 각각 추출하여 각 추출물별로 총페놀함량, 전자공여능(electron donating ability, EDA), 과산화물가와 에멀젼 시스템에서의 항산화효과를 비교, 검토하였다. Follin-Dennis법으로 구한 물, 메탄올 및 에테르 추출물의 총페놀 함량은 각각 2.34, 6.98, 5.83 mg/100g으로 메탄올 추출물에서 가장 큰 함량을 보였다. 정향의 물, 메탄올 및 에테르 추출물의 전자공여능은 각각 86%, 78%, 68%이었다. 리놀레인산 기질에서의 항산화효과는 메탄올 및 에테르 추출물에서 높게 나타난 반면, 에멀젼 시스템에서는 물 추출물이 가장 우수한 것으로 나타났다.   • This study was designed to investigate the antioxidant effects of 10 kinds of medicinal plants and vegetable extracts and total extracts. The cytotoxicity was measured by MTS assay, and the antioxidant activity was measured by DPPH free radical scavenging activity and Riboflavin-derived superoxide inhibitory activity (SQA). As a result, cytotoxicity was safe for all 10 medicinal plants, vegetable extracts and total extracts. DPPH free radical scavenging ability was observed in Cinnamomum cassia Blume, Eugenia caryophyllata Thunb. Arctium lappa, Total extract was excellent, and Riboflavin-derived superoxide inhibitory activity (SQA) was found in Cinnamomum cassia Blume, Arctium lappa, Prunus mume Sieb. et Zucc., Excellent, but total extract showed the best antioxidant effect. As a result of comparing the antioxidant effects of medicinal plants and vegetables using traditional ingredients, the antioxidant activity was increased when used as a mixture than when used alone. It is considered that it can be used as an antioxidant functional material, and it is expected to be of value when developing antioxidant material in the future.
