Effect of Ascorbic Acid Supplementation on Hepatic Microsomal and Mitochondrial Cytochrome P450 System in Diabetic Rats (비타민 C의 보강이 당뇨쥐의 간 소포체와 미토콘드리아의 Cytochrome P450계에 미치는 영향)

  • This study was performed to investigate whether ascorbic acid can modulate the induction of CYP2E1 and prevent the lipid peroxidation which may cause diabetic chronic complication. Diabetes was induced by intraperitoneal injection of streptozotocin to 5-week-old male Sprague-Dawley rats(150~170g). Normal and diabetic group was randomly divided into three groups each; Control(CON, no supplementation), SUP1 (50mg/d ascorbate supplementation) and SUP2(250mg/d ascorbate supplementation). Ascobic acid was prepared daily by dissolving in drinking water and supplied for 4 weeks. There was no difference in hepatic microsomal and mitochondrial P450 contents between normal and diabetes. Hepatic microsomal N-nitrosodimethylamine(NDMA) demethylase activity, which repre-sents contents of CYP2E1, was elevated in diabetes, but not significantly. The NDMA demethylase activity of diabetic SUP2 group was significantly lower activity than that of the diabetic CON group. However, no difference in hepatic mitochondrial NDMA demethylase activity was observed between the diabetes and the normal group. The result suggests that the induction of CYP2E1 in diabetes can be alleviated by ascorbic acid supplementation at the dose of 50mg1d. In addition, ascorbic acid supplementation showed dose-dependent reduction of hepatic microsomal TBARS contents in diabetic rats.

Effects of Ethanol Extract of Prunus mume on the Antioxidative System and Lipid Peroxidation on Ethanol-Induced Hepatotoxicity in Rat Liver (매실 추출물이 알코올 투여 흰쥐의 항산화계 및 지질과산화에 미치는 영향)

  • To investigate the antioxidative effects of Prunus mume ethanol extract on the ethanol-induced hepatotoxicity in rat liver, Sprague-Dawley rats weighing 120∼160 g were divided into 5 groups; normal group(NOR), Prunus mume ethanol extract 200mg/kg treated group(PME), ethanol(10 mL/kg, 35%) treated group(ETH), Prunus mume ethanol extract 200 mg/kg and ethanol treated group (PML) and Prunus mume ethanol extract 400 mg/kg, and ethanol treated group(PMF), respectively. The antioxidative activity in vitro was reduced in order of EtOAC>n-hexane>water> chloroform fraction. The growth rate and feed efficiency ratio decreased by ethanol administration were gradually increased to the adjacent level of NOR by administering Prunus mume ethanol extract. It was observed that activities of catalase, superoxide dismutase(SOD), xanthine oxidase and glutathione peroxidase(GSH-Px) of liver and alanine aminotransferase(ALT) and asparate aminotransferase(AST) of serum were elevated by ethanol administration. Besides, Prunus mume ethanol extract markedly decreased elevated activites of catalase, GSH-Px, ALT and AST, except in activites of SOD and xanthine oxidase compared to ETH. Also, the depleted content of GSH by ethanol was increased similar to NOR level by administering Prunus mume ethanol extract. These results suggested that Prunus mume ethanol extract has a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.

Effect of Dietary Anti-oxidant Supplementation on Telomere Length and Egg Quality in Laying Hens (산란계에 항산화물질 급여가 텔로미어 함량 및 난질에 미치는 영향)

  • There are many kinds of anti-oxidant materials in natural plant resources. The Siberian ginseng and Eucommia are well known as anti-oxidant and medicinal plants. To investigate the effect of their anti-oxidant-like activity on telomere quantity and egg quality, diets containing Siberian ginseng leaf and Eucommia leaf at 0.5% and 1% were given Hyline Brown commercial laying hens during two periods of age: 20 to 30 wks and 60 to 70 wks. The amount of telomere in lymphocyte, liver, ovary, heart and lung was analyzed by quantitative fluorescence in situ hybridization using telomeric DNA probe. Egg weight, albumin height, Haugh unit, egg yolk color, egg shell color, egg shell thickness, egg shell weight and egg shell density were measured to analyze egg quality. The chickens consuming diets Siberian ginseng and Eucommia had higher telomeric DNA in lymphocytes than control chickens in younger layers whereas no significant differences were detected in all target cells analyzed from older layers. Egg quality was increased in younger hens with dietary supplementation as determined by egg weight, albumin height and Haugh unit but there were no effects in older hens. These results imply that dietary supplementation of Siberian ginseng and Eucommia in layers improves bio-activity and egg quality at early laying stage.

Effect of the Ethanol Extract of Cassia tora L. on Antioxidative Compounds and Lipid metabolism in Hepatotoxicity of Rats-induced by Ethanol (결명자 에탄올추출물이 알코올 투여 흰쥐의 항산화물질 및 지질대사에 미치는 영향)

  • This study was done to investigate the effects of ethanol extract of Cassia semen (Cassia tora L.) on the activities of hepatic oxygen free radicals metabolizing enzymes and blood lipid profile in rats of hepatotoxicity induced by ethanol. Sprague-Dawley male rats weighing 100~160 g were divides into 5 groups; control grouts (CON), Cassia semen ethanol extracts (200 mg/kg) treated group (CEL), ethanol (10 mL/kg, 35%) treated group (ETH), Cassia semen ethanol extracts (200 mg/kg) and ethanol treated group (CE1 ) and Cassia semen ethanol extracts (400 mg/kg ) and ethanol treated group (CE2), respectively. Compared with ETH, the growth rate of CE1 and CE2 were to be increased tendency, and in blood levels of total cholesterol, LDL-cholesterol and the activities of alanine aminotranferase and asparate aminotranferase elevated by ethanol were significantly decreased (p<0.05). It was observed that the activities of superoxide dismutase, catalase, xanthine oxidase and glutathione peroxidase of rat liver increased by ethanol, were more decreased by the treatment of Cassia semen ethanol extract than the only ethanol-treated group. The content of glutathione depleted by ethanol treatment was increased in CE1 and CE2. TBA-reactants of liver increased by ethanol were decreased in CE1 and CE2, compared with ethanol-treated group. These results suggested that ethanol extract of Cassia semen may influence upon the ability of oxygen free radical detoxication and lowering of blood lipid level on ethanol-induced hepatotoxicity in rat.

Antioxidative Activities of Aronia melanocarpa Fruit and Leaf Extracts (아로니아 베리 열매 및 잎 추출물의 항산화 활성)

  • In this study, the antioxidative effects of Aronia melanocarpa fruit and leaf extracts were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of the ethylacetate and aglycone fractions of fruit extracts were 16.29 ${\mu}g/mL$, and 12.29 ${\mu}g/mL$, respectively. The free radical scavenging activity of fruit extract was higher than that of leaf extracts. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethylacetate and aglycone fractions of fruit extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay showed 2.86 ${\mu}g/mL$, and 1.80 ${\mu}g/mL$, respectively. ROS scavenging activity of the aglycone fraction of fruit extracts was similar to that of L-ascorbic acid (1.50 ${\mu}g/mL$). The ROS scavenging activity of fruit extracts was higher than that of leaf extracts. The cellular protective effects of aglycone fraction of fruit extracts (${\tau}_{50}$ = 72.3 min) on the $^1O_2$-induced cellular damage of human erythrocytes especially were increased in a concentration dependent manner (5 ~ 50 ${\mu}g/mL$). ${\tau}_{50}$ (72.3 min) of the aglycone fraction showed 1.9 times higher than (+)-${\alpha}$-tocopherol (38 min), known as lipophilic antioxidant at 10 ${\mu}g/mL$. These results incidicate that A. melanocarpa fruit extracts have higher antioxidant effects than leaf extracts and could be applicable to functional cosmetics materials for antioxidants by protecting skin exposed to solar UV radiation against ROS including $^1O_2$.

  • This study was conducted to investigate the effects of methionine(Met) and selenium(Se) levels on alcohol metabolic enzyme system in rats. Sprague-Dawley male rats were fed on diets containing one of the three levels of Met(0, 3, 9g/kg diet) with or without Se(0.45mg/kg diet). Alcohol was administrated with 25%(v/v) ethanol orally at the same time once a day in alcohol group and isocaloric sucrose was administrated to the control group. The rats were sacrificed after 5 and 10 week of feeding periods. Alcohol dehydrogenase(ADH) and microsomal ethanol oxidizing system(MEOS) activities of hepatic tissuedom were increased more in alcohol treated groups than control group. Increment of activities preinated in simultaneous deficiency of dietary Met and Se(LMet-Se+EtOH) group. Aldehyde dehydrogenase (AIDH) activity was decreased more in alcohol treated groups than control group and significantly decreased in Met and Se supplemented(NMet+Se+EtOH) group. Hepatic cytochrome P-450 content and xanthine oxidase(XO) activity were significantly increased in alcohol treated groups Compared to control group and predominated in Met deficiency(LMet) group and excessive Met administration (HMet) group. Superoxide dismutase(SOD), catalase, glutathione S-transferase(GST) activities tended to increase by alcohol administration, the degree of increase predominated in 10 week. The activity of glutathione peroxidase(GSH-Px) was decreased in alcohol groups and tended to increase in proportion to the level of dietary Met.

Radioprotective Effect of Post-treatment with Rutin on γ-Irradiation-induced Cellular Damage in Mice (감마선 조사로 유도된 세포 손상 마우스에서 루틴 투여 후의 방사선 방호 효과)

  • This study aimed to investigate the therapeutic effect of rutin against whole-body ${\gamma}$-irradiation in BALB/c mice. BALB/c mice were randomly divided into four groups and exposed to 6 Gy ${\gamma}$-irradiation. One hour later, mice were orally administered rutin (50 and 100 mg/kg) for seven consecutive days. ${\gamma}$-Irradiation (6 Gy) resulted in cellular damage as manifested by elevated levels of plasma hepatic marker enzymes and lipid peroxidation in liver tissue, accompanied with decreased spleen and thymus indices, and white blood cell count. In addition, ${\gamma}$-irradiation significantly decreased the levels of antioxidant enzymes such as superoxide dismutase, glutathione peroxidase and catalase. Rutin treatment significantly protected against ${\gamma}$-irradiation-induced cellular damage, which was evident by the improvement in the status of most of the investigated parameters. Therefore, rutin has beneficial effects against radiation-induced damage.

Antibacterial and Antioxidative Activity of Lespedeza cuneata G. Don Extracts (비수리 추출물의 항균 및 항산화 활성)

  • In this study, the antibacterial activity and the antioxidative effects, inhibitory effects on tyrosinase of Lespedeza cuneata G. Don extracts were investigated. MIC value of ethyl acetate fraction from L. cuneata G. Don on P. ovale (0.125%) showed that the antibacterial activity of the ethyl acetate fraction was higher than methyl paraben. The aglycone fraction of L. cuneata G. Don (14.63 ${\mu}g$/mL) showed the most prominent the free radical (1,1-diphenyl-2-picryl-hydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of L. cuneata G. Don fraction on $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The aglycone fraction of L. cuneata G. Don (0.07 ${\mu}g$/mL) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of L. cuneata G. Don on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The L. cuneata G. Don extracts suppressed photohemolysis in a concentration dependent manner (1 ~ 50 ${\mu}g$/mL). The inhibitory effects ($IC_{50}$) of L. cuneata G. Don extracts on tyrosinase were determined with ethyl acetate fraction (104.83 ${\mu}g$/mL) and aglycone fraction (27.55 ${\mu}g$/mL) of L. cuneata G. Don extract. These results indicate that L. cuneata G. Don extract/fractions can function as high potential as bactericide against the pathogenic bacteria and antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Extract/fractions of L. cuneata G. Don could be applicable to new functional cosmetics for antiaging, antioxidant, and antibacterial activity.

Antioxidative Activity and Component Analysis of Phellinus linteus Extracts (상황버섯 추출물의 항산화 및 성분분석)

  • In this study, the antioxidative effect, antibacterial, inhibitory effects on tyrosinase, inhibitory effects on elastase and component analysis of Phellinus linteus (P. linteus) extracts were investigated. The ethyl acetate fraction of P. linteus extracts ($2.94\;{\mu}g/mL$) showed the highest free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of P. linteus extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.0072\;{\mu}g/mL$) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of P. linteus extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. linteus extracts showed cellular membrane protective effects in a concentration dependent manner ($5{\sim}50\;{\mu}g/mL$). The inhibitory effect ($IC_{50}$) on tyrosinase of P. linteus extract was the highest at 50 % ethanol extract ($6.34\;{\mu}g/mL$), and the inhibitory effect ($IC_{50}$) on elastase of P. linteus was the highest at ethyl acetate fraction ($14.08\;{\mu}g/mL$). TLC, HPLC chromatogram and LC/ESI-MS of the ethyl acetate fraction obtained from P. linteus extracts were identified interfungin A (PL RPT-1a). These results indicate that extract/fractions of P. linteus can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging ROS, and protect cellular membranes against ROS. Extract/fractions of P. linteus can be applicable to new cosmeceuticals for antioxidant, antiaging, antiwrinkle and whitening.

Antioxidative and Antiaging Effects of Platycarya strobilacea Extract and Clinical Trial (굴피나무 추출물의 항산화, 항노화 효과 및 인체 시험)

  • In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of Platycarya strobilacea bark extracts. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract / fractions of Platycarya strobilacea was in the order: 50% ethanol extract ($6.75{\mu}g/mL$) < deglycosylated aglycone fraction ($6.62{\mu}g/mL$) < ethyl acetate fraction ($4.15{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Platycarya strobilacea extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity was ethyl acetate fraction (OSC50, $0.56{\mu}g/mL$) < 50% ethanol extract ($0.02{\mu}g/mL$) < deglycosylated aglycone fraction ($0.01{\mu}g/mL$). The deglycosylated aglycone fraction showed the most prominent scavenging activity. The protective effects of extract / fractions of Platycarya strobilacea on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract (50%) suppressed photohemolysis in a concentration dependent manner, particularly ethyl acetate fraction exhibited the most prominent cellular protective effect (${\tau}_{50}$, 717.27 min at $10{\mu}g/mL$). The inhibitory effect of Platycarya strobilacea extracts on tyrosinase were investigated to assess their whitening efficacy. Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. The inhibitory effect ($IC_{50}$) on tyrosinase of some Platycarya strobilacea extracts was 50% ethanol extract ($243.98{\mu}g/mL$) < ethyl acetate fraction ($153.87{\mu}g/mL$) < deglycosylated aglycone fraction ($137.53{\mu}g/mL$). Also, The inhibitory effect of elastase ($IC_{50}$) of some Platycarya strobilacea extracts was 50% ethanol extract ($31.01{\mu}g/mL$) < ethyl acetate fraction ($14.42{\mu}g/mL$) < deglycosylated aglycone fraction ($1.48{\mu}g/mL$). The cream containing the ethyl acetate fraction of Platycarya strobilacea extracts was formulated. The skin hydration, transepidermal water loss, and the whitening effects were investigated after topical application of the cream. The skin hydration of cream containing extract was increased by $2{\sim}8%$ than the placebo cream, transepidermal water loss was decreased. The cream containing extract suppressed the melanogenesis of skin by 9.55% than the placebo cream. These results indicate that extract / fractions of Platycarya strobilacea can function as antioxidants in biological systems, particularly skin exposed to UV radiation by anti-oxidative activity and protect cellular membranes against ROS. The inhibitory effect on elastase and tyrosinase, and the increase of skin hydration and the whitening effect of the cream containing extract could be applicable to new functional cosmetics for antiaging.
