Mast cells play an important role in early and late phase allergic reactions through allergen and IgE-dependent release of histamine, proteases, prostaglandins, and several multifunctional cytokines. In this study, we investigated whether Cudrania tricuspidata extract (CTE) suppresses IgE-mediated allergic responses in mast cells, an allergic animal model, and its mechanism of action in mast cells. We found that CTE inhibited IgE-mediated degranulation and cytokine production in rat basophilic leukemia (RBL)-2H3 mast cells and bone marrow-derived mast cells (BMMC), as well as passive cutaneous anaphylaxis (PCA) in mice. With regard to its mechanism of action, CTE suppressed the activating phosphorylation of spleen tyrosine kinase (Syk), a key enzyme in mast cell signaling processes and that of LAT, a downstream adaptor molecule of Syk in $Fc{\varepsilon}RI$-mediated signal pathways. CTE also suppressed the activating phosphorylation of mitogen-activated protein (MAP) kinases and Akt. The present results strongly suggest that the anti-allergic activity of CTE is mediated through inhibiting degranulation and allergic cytokine secretion by inhibition of Syk kinase in mast cells. Therefore, CTE may be useful for the treatment of allergic diseases.
This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.
Atypical antipsychotics are more effective than typical antipsychotics and have fewer side effects such as tardive dyskinesia and extrapyramidal symptoms; therefore, prescriptions of atypical antipsychotics are increasing. However, recently, it has been reported that atypical antipsychotics have a higher incidence of diabetes, hyperglycemia, and obesity than typical antipsychotics. Atypical antipsychotics induce obesity-inhibiting appetite-related receptors such as serotonin and dopamine. Decreased exercise due to improving psychotic symptoms, and genetic characterictics can also cause weight gain. Hyperglycemia and hypoglycemia were another metabolic problem related to treatment with atypical antipsychotics. The mechanisms of hyperglycemia were mainly related obesity, decreased anorexigenic hormones, and increased insulin resistance in multiple organs. There are also reports that genes related to diabetes have an effect on the incidence of diabetes mellitus treated with atypical antipsychotics. On the other hand, although it is not clear why hypoglycemia occurs, it documented in case reports all over the world. There are more reports of atypical antipsychotics than typical antipsychotics and these are frequently reported in Asians. Further research on the mechanism of hypoglycemia related to atypical antipsychotics is strongly recommended.
This study aimed to determine the anti-obesity effect of adding Wolfiporia extensa Ginns (W) to fermented pollack skin products in an obesity-induced animal model. The experimental groups were the normal diet group (C), high-fat diet group (HF), dried pollack skin (H1), fermented pollack skin (H2), and W of 0.1 (F2-WL), 0.3 (F2-WM), and 0.5 (F2), respectively. It was confirmed that adding W to fermented pollack skin reduced blood triglycerides, total cholesterol, and LDL levels, while increasing HDL levels. Wolfiporia extensa Ginns was effective in controlling weight and improving blood lipids in a dose-dependent manner. In histological analysis, findings of fatty liver induced by a high-fat diet were improved by the addition of H2 and W. Size and density of fat globules in the epididymis were decreased. In addition, the concentration of TNF-α was increased in the high-fat diet group, but decreased by the addition of fermented pollack skin and W. In conclusion, adding fermented dried pollack skin and Wolfiporia extensa Ginns was effective for weight control and blood lipid improvement. Thus, the use of by-products in functional foods is expected to have a high value in the future.
We studied the antioxidant, the anti-inflammatory, antithrombotic, and antiobesity activities of seven different kinds of edible plants. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ABTS radical scavenging activities were determined as a measurement of antioxidant activity. NO production inhibition by the macrophage cell line (Raw 264.7) treated with lipopolysaccharide (LPS) was carried out to assess anti-inflammatory activity. Thrombin inhibitory activity was measured for its antithrombotic function and inhibition of 3T3-L1 cell differentiation was evaluated as a measurement of antiobesity activity. Total phenolic components and total flavonoid contents were measured to determine functional materials in medicinal plants. Common sage, Japanese lady bell, and hairy agrimony showed high antioxidant activity ($IC_{50}$) of less than $100{\mu}g/mL$. All samples used in this study showed anti-inflammatory activity. Common sage, hairy agrimony, and hooker chives showed antithrombotic effects. Hairy agrimony showed the highest antithrombotic effect (98.1%). Common sage, Japanese lady belly, hairy agrimony, and hooker chives showed reduced 3T3-L1 cell differentiation and hooker chives strongly inhibited lipid accumulation in the cells compared to other medicinal plants. Common sage and hairy agrimony contained more than 1 mg GAE/g of phenolic compounds and more than 1 mg CE/g of flavonoids. Functional activities were different by plant part and extraction method from each sample. These results suggest that common sage, Japanese lady belly, hairy agrimony, and hooker chives may be used as healthy food sources with antioxidant, anti-inflammatory, antithrombotic, and antiobesity activities, and appropriate extracting methods from each plant need to be developed.
알코올에 의한 지방간(지방변성증)은 에탄올 대사에 의해 감소되는 당량의 과도한 발생에 의해 유발된다. 일반적으로 만성적인 에탄올 투여는 간 지질의 합성을 증가시키는 sterol regulatory element-binding protein 1c (SREBP-1c)를 조절함으로써 지방변성증을 유발시킨다. SPEBP-1c에서 에탄올의 영향은 간에서의 지방대사를 조절하는 NAD+ 의존적 단백질 탈아세틸화효소인 mammalian sirtuin-1 (SIRT-1)에 의해 조정된다. 홍삼은 항당뇨와 항비만 효과를 위해 아시아에서 광범위하게 사용되는 한약재이다. 홍삼의 약리학적 치료학적인 효과는 진세노사이드와 같은 생물활성 성분에 의해 주로 일어난다. 따라서 우리는 마우스 간세포주인 AML-12 세포에서 SREBP-1과 SIRT-1에대한 한국홍삼 추출물의 조절효과를 평가하였다. 알코올과 홍삼추출물(0-1,000 μg/ml)을 AML-12 세포주에 처리하고, 지방소립을 Oil red O 염색법으로 확인하고, western blots을 사용해 SIRT-1과 SREBP-1의 발현을 확인하였다. 에탄올을 처리한 세포에서 홍삼추출물은 SIRT-1과 SREBP-1c의 발현을 회복시켰다. 또한 에탄올이 처리된 세포에서 홍삼추출물과 진세노사이드 Rb2와 Rd가 SREBP-1을 유의적으로 감소시키는 것으로 확인 되었다. 결과적으로 홍삼과 활성 진세노사이드 성분인 Rb2와 Rd가 SIRT-1과 간 지질대사를 변화시키는 SREBP-1c의 아세틸화의 조절을 통해 알코올에 의한 간지방변성을 억제하는 것을 확인하였다.
최근 식생활의 변화에 따라 당뇨병, 비만 등의 만성대사질환의 발병률이 증가함에 따라 예방과 치료를 위한 물질연구가 많이 진행되고 있다. 본 연구에서는 선행 연구에서 항당뇨 효능이 확인된 Vigna nakashimae (VN) 메탄올 추출물로부터 VN 클로로포름층 분획물과 물층 분획물을 얻어 각 분획물의 항당뇨 효능과 분자적 기전을 살펴보았다. 각각의 VN 분획물을 제 2형 당뇨병질환모델인 db/db 마우스에 경구투여하여 VN 분획물의 혈당 및 지질 대사 변화 등을 측정하였다. VN 클로로포름층 분획물은 공복 혈당과 당화혈색소를 물층 분획물보다 더 효과적으로 감소시켰다. 또한 내당능도 개선하였으며, 혈중 지방산과 중성지방도 감소시켰다. VN 클로로포름층 분획물은 HepG2와 C2C12세포에서 인산화된 AMPK와 AMPK 하위 유전자인 CPT-1의 유전자 발현을 증가시켰다. VN 클로로포름층 분획물은 AMPK의 활성화와 일치하게 HepG2에서 당 생성 효소인 PEPCK와 G-6Pase의 발현을 감소시켰으며, C2C12에서는 당 유입을 증가시켰다. 이상의 결과는 VN 클로로포름층 분획물은 AMPK의 활성화를 통해 공복혈당을 감소시켰으며, 이러한 효과는 물층 분획물보다 더 효과적이었다.
본 연구는 고지방식이에 동결 건조한 된장, 청국장, 고추장 및 쌈장 등의 전통 장류를 첨가하여 4주 동안 사육한 후 체중, 피하지방함량, 장기별 무게, 장기별 지질 및 콜레스테롤 함량을 측정하여 항비만 효과를 살펴보았다. 고지방식이군에 비해 모든 장류 첨가군에서 체중과 식이효율이 감소되었고, 특히 된장을 첨가한 식이군에서 체중감소 효과가 가장 높았다. 지방조직 중량은 된장과 쌈장을 첨가한 식이군에서 가장 낮았으며 간, 신장주위 지방조직의 중성지질 및 콜레스테롤 농도는 장류 첨가식이군에서 낮아졌다. 특히 된장과 쌈장 첨가식이군의 간 무게는 고지방 식이를 동시에 섭취시켰지만 정상군의 수준으로 감소되었다. 간의 콜레스테롤 농도는 청국장을 제외한 다른 장류군에서 감소하였고, 신장주위 지방조직의 콜레스테롤 농도도 고지방식이군에 비해 감소하였다. 혈액에서의 중성지방과 콜레스테롤 모두 고지방식이군에 비하여 장류를 첨가한 식이군에서 감소하는 경향을 보였는데 된장과 쌈장 첨가식이군에서 가장 큰 효과를 나타내었다. 결국 이 연구에서 고지방식이에 의한 항비만효과는 된장, 쌈장, 고추장, 청국장 순이었고 된장이 가장 효과가 컸었다.
간의 지질 축적과 인슐린 저항성은 비알콜성 지방간 환자에게서 증가한다. Piperine은 후추(Piper nigrum)와 필발(인도산 후추, P. longum)의 주요 성분으로 항암, 항비만, 항 당뇨병, 항염증 및 항산화 등의 생리활성이 보고되었다. 그러나 piperine의 인간 간세포 HepG2 세포에서 지질 축적과 인슐린 저항성의 억제제로서의 연구는 보고된 바가 없다. 본 연구의 목적은 지질 축적 및 인슐린 저항성에 대한 piperine의 효과를 palmitate처리된 HepG2 세포에서 잠재적인 분자 기전을 밝히는 것이다. 그 결과 piperine처리군은 지질 함량을 감소시켰고, 지방 형성 표적 유전자인 SREBP-1c와 FAS의 발현을 억제함으로써 palmitate처리된 세포내 지질 축적을 감소시켰다. 게다가 piperine처리군은 지방산 산화에 관련된 CPT-1과 인산화된 ACC 및 인산화된 IRS-1 (Tyr632)와 Akt의 레벨을 증가시켰다. 또한, piperine처리군은 인산화된 IRS-1 (Ser307)의 레벨을 감소시켰다. 결론적으로 palmitate처리된 HepG2 세포에서 piperine은 SREBP-1와 FAS발현의 감소 및 CPT-1과 ACC 인산화의 증가 및 인산화된 IRS-1(Try632)와 Akt 신호전달 경로를 조절함으로써 지질 축적 및 인슐린 저항성을 개선함을 확인하였다. 따라서 piperine의 지질 축적 및 인슐린 저항성을 예방하는 약물로써 가능성이 제시되었다.
