A Study on the Manufacture of Healing Textile Material with Characteristics of Dermatitis Symptom Relief (피부염 증상완화특성을 갖는 힐링섬유소재 제조에 관한 연구)

  • 피부염 증상 완화특성을 갖는 힐링섬유소재 제조를 연구하였다. 면소재, 폴리에스테스/나일론 소재에 피부염 증상완화특성이 있다고 알려진 기능성물질을 바인더, 수지 등의 혼합물 페이스트로 만들어 스크린프린팅법을 사용하여 실험을 시도하였다. 제조한 힐링섬유시편소재의 항균특성은 면소재의 경우 99.9% 의 우수한 항균특성을 나타냈다. 암모니아가스의 소취성능은 시간경과에 따라 최대 68%의 소취율을 보였다. 타소재와의 오염견뢰도는 평균 4.5 ~ 5등급, 변퇴세탁견뢰도는 5등급, 마찰견뢰도는 3 ~ 4.5 등급, 일광견뢰도는 5등급을 나타냈다. 의약품 등의 독성시험기준에 따른 힐링소재의 피부자극성을 평가하기 위해 NZW토끼의 피부에 24시간 동안 부착한 후 72시간 동안 사망률, 일반증상, 체중변화 및 피부자극성을 시험한 결과 사망토끼는 관찰되지 않았고, 모든 시험동물에서 특이할 만한 이상증상은 관찰되지 않았다. 체중측정결과 모든 동물에서 정상적인 체중증가를 보였다. 시험소재의 피부자극성을 관찰한 결과 피부자극성이 관찰되지 않아 비자극성(None Irritant)섬유소재로 판단되어 피부염증상완화 특성을 갖는 힐링섬유소재 제조의 실용화 가능성을 연구하였다.

Protein Characteristics of Ovotransferrin Under the pH and Temperature and Its Anti-microbial Activity (Ovotransferrin의 pH 및 온도에 따른 단백질 및 항균 특성)

  • Protein function of ovotransferrin with various pH and temperature, and its antimicrobial characteristics were determined. Foaming ability of ovotransferrin was high in alkali condition (pH 11), then diminished as time follows. In acidic condition (pH 3.0), very little amount of foam was produced and disappeared promptly in 30min. However, neutral condition (pH 7.0) was revealed as the best area for foam production and foam stability of ovotransferrin. Temperature effect on foam stability of ovotransferrin showed that the highest foam was produced at 60℃. Ovotransferrin was shown weak antimicrobial activity against E. Coli, S .typhi, P. aerug and Candida albicans at dose of 12.5mg/ml and 25mg/ml. Anti-microbial effect of ovotransferrin with either lysozyme or albumine on pathogenic bacteria and fungi shows that the most effective dose was 25mg/ml, especially on S. typhi and C. albicans.

Characterization of Antimicrobial Substance Produced by Lactobacillus sp. HN 235 Isolated from Pig Intestine (돼지 장관으로부터 분리한 Lactobacillus sp. HN 235 균주가 생산하는 항균물질의 특성)

  • In order to develop probiotics which may be a viable alternative of antibiotic use in pig industry, five bacterial strains (Lactobacillus sp. HN 52, 92, 98, 235 and AP 116) possessing antimicrobial properties were selected from 500 strains isolates of pig intestines. The bacteriocin produced by Lactobacillus sp. HN 235 displayed a relative broad spectrum of inhibitory activity against all Enterococcus strains, Pseudomonas aeruginosa, Listeria monocytogenes and Clostridium perfringens using the spot-on-lawn method. The production of antimicrobial substance started in the middle of the exponential growth phase, reached maximum levels (6,400 AU/mL) in the stationary phase, and then declined. Bacteriocin activity remained unchanged after 30 min of heat treatment at $95^{\circ}C$ and stable from pH 2.0 to 10 for 1 h, or exposure to organic solvents; however, it diminished after treatment with proteolytic enzymes. The molecular weight of the bacteriocin was about 5 kDa according to a tricine SDS-PAGE analysis.

Aroma Compounds and Antimicrobial Effect of Garlic from Different Areas in Korea (국내 산지별 마늘의 향기성분 및 항균활성)

  • Physico-chemical characteristics such as width, weight, color and aroma compounds of garlic from 9 different areas were analyzed. Also, antimicrobial activity was tested for their juices. Width of garlic bulb was larger in growing worm-season garlics (Jeju, Namhae, Hapcheon, Hampyeong and Muan) than those of cool-season garlics (Taean, Seosan, Uiseoung) and imported from China. But numbers of nuts were smaller in growing cool-season garlics than others. L color levels of garlics were 80.73~87.40 and a color level have not significantly difference in all samples, b color level was lower in growing Namhae ($20.79{\pm}1.20$) and Muan ($20.91{\pm}1.62$). 25~30 peaks obtained from aroma compounds analysis of various garlics. Among these compounds, 17 kinds of aroma compounds were identified by GC-MS. And, we showed that 15 kinds of compounds were sulfur containing aroma compounds. Diallyl disulfide was contained 34.90~60.54% in various garlic and the highest contents than other compounds in total compounds. It was detected the highest in China garlic but the lowest in growing Muan garlic. All of garlic samples showed antimicrobial activity against Strep. mutans, B. subtilis, E. coli. V. parahaemolyticus, Asp. flavus, and C. albicans. Especially antimicrobial activity was stronger against E. coli, and C. albicans. Antimicrobial activity was enhanced by sample addition volume and the highest in Namhae garlic sample.

Antifungal and Proteolytic Activity and Auxin Formation of Bacterial Strains Isolated from Highland Forest Soils of Halla Mountain (한라산 고지대 토양에서 분리한 미생물의 항균 및 단백질분해 활성, 오옥신 생산 특성)

  • Bacterial strains were isolated from forest soils of Halla mountain, Jeju island in Korea. The soil samples were collected at each altitude of 100m from 1,000 m above sea level. Total 398 strains were isolated and tested for their physiological characteristics of antagonistic and proteolytic activities, and auxin production. Among the isolates, 172 strains were selected as antifungal strains showing antagonistic activity against at least one of 8 plant fungal pathogens (Alternaria alternata, Botrytis cinerea, Collectotrichum acutatum, Fusarium oxysporum, Phytophthora capsici, Pythium ultimum and Sclerotinia sclerotiorum). In addition 203 strains for proteolytic activity and 26 strains for auxin production were characterized for further study. Je28-4 (Rhodococcus sp.) were showed 80% of control value against tomato gray mold in vivo. Thus, it is suggested that soil bacteria isolated from forest soils of Halla mountain can be important sources of bioactive compounds for improving plant growth or promising biocontrol agents.

Physicochemical properties and biological activity of three-year-old and seven-year-old Platycodon grandiflorum extracts (3년근 도라지와 7년근 도라지의 이화학 특성 및 생리활성)

  • This study was designed to evaluate the physicochemical properties and biological activities (antioxidant activity and antimicrobial activities) of three and seven-year-old Platycodon grandiflorum extracts. Three and seven-year-old Platycodon grandiflorum contained crude saponins, free amino acids and minerals. Water extracts of the three and seven-year-old plants were prepared using reflux extraction methods. The total polyphenol contents (TPC), total flavonoid contents (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and antimicrobial effects of the extracts were determined. The seven-year-old Platycodon grandiflorum extract had higher TPC ($5.08{\pm}0.07TAEmg/g$) and TFC ($3.80{\pm}0.07QUEmg/g$). DPPH radical scavenging activity ($IC_{50}$ value: $288{\pm}3.88{\mu}g/mL$) and ABTS radical scavenging activity ($IC_{50}$ value: $568{\pm}2.09{\mu}g/mL$). The three and seven-year-old Platycodon grandiflorum extracts exhibited a strong antimicrobial effect against three kinds of bronchus disease-inducing bacteria; the seven-year-old Platycodon grandiflorum extracts showed a stronger antimicrobial effect than the three-year-old Platycodon grandiflorum extracts.

Studies on Physiological and Functional Properties of Methanol Extract from Chicken Bile (닭 쓸개 methanol 추출물의 생리기능적 특성에 관한 연구)

  • Methanol extracts and powder of chicken bile were evaluated to determine antimicrobial, electron-donating, nitrite-scavenging, and inhibitory abilities against angiotensin I-converting enzyme (ACE). HPLC revealed taurochonodeoxycholic acid (TCDOA) and taurocholic acid (TCA) were major bile salts, at 5,893 and 395 mg/100g, respectively. Methanol extracts showed inhibitory effect on growth of Bacillus cereus, Salmonella typhimurium, Staphylococcus aureus, and Vibrio parahaemolyticus, whereas no effect on Escherichia coli. Electrondonating and nitrite-scavenging abilities increased significantly with increasing amount of bile samples. Electrondonating activity of dried powder was higher than that of methanol extracts, whereas nitrite-scavenging activity showed opposite trend. Both samples showed positive inhibitory activity of ACE. Methanol extracts showed higher activity than that of freeze-dried powder at high level of bile sample (5 and 10%).

Isolation and Characterization of Oligotrophic Strains with High Enzyme Activity from Buckwheat Sokseongjang (메밀 속성장 유래 효소활성 우수 저영양성 균주 분리 및 특성)

  • Bealmijang is a short-term fermented regional product that is prepared with soybean and extra ingredients. In this study, starter strain candidates were screened from Bealmijang for fermented soybean paste products. Twenty one bacterial strains producing extracellular enzymes (amylase, cellulase, protease, xylanase and lipase) were isolated from Bealmijang, buckwheat sokseongjang. The isolates were assessed for fibrinolytic and antibacterial activities, and salt tolerance. Strain HJ18-4, identified as Bacillus subtilis (AB601598) by biochemical properties (89.6%) and 16S rDNA sequencing (100%), showed the highest enzymatic, fibrinolytic, and antibacterial activities among the isolates. Although the growth of HJ18-4 was inhibited by the increase of NaCl concentration, the growth still exceeded that of B. subtilis KACC 10114 at 5% and 10% NaCl. These results suggest that B. subtilis HJ18-4 is suitable as a starter for soybean paste manufacture.

Characterization and Antimicrobial Activity of Lactic Acid Bacteria Isolated from Vaginas of Women of Childbearing Age (가임기 여성의 질에서 분리한 젖산 세균인 Lactobacillus plantarum UK-3의 특성 및 항균활성)

  • The purpose of this work was to examine the antimicrobial activity derived from the lactic acid bacterium, UK-3 isolated from the vaginas of women of childbearing age. Various physiological and biochemical properties of this strain were characterized. Both the BIOLOG system and phylogenetic analysis using 16S rRNA sequencing were utilized for identification, and the strain was designated as Lactobacillus plantarum UK-3, and registered in GenBank as [JK266589]. Growth rate, production of organic acids (e.g., lactic acid and acetic acid), and pH during growth were monitored. The maximum concentrations of lactic acid and acetic acid were approximately 684.11 mM and 174.26 mM, respectively, and pH changed from 7.0 to 3.7 after 72 h of incubation. High performance liquid chromatography was used to confirm lactic acid and acetic acid production. Significant antimicrobial activity of the concentrated supernatant was demonstrated against various Gram-positive (e.g., Staphylococcus aureus, Staphylococcus epidermidis, Methicillin-resistant Staphylococcus aureus, Enterococcus faecalis, Neisseria species., Listeria monocytogenes), Gram-negative bacteria (e.g., Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis), and yeast (e.g., Candida albicans) by the plate diffusion method. As a result, the concentrated L. plantarum UK-3 cultures had lower acidity and inhibited the growth of all microorganisms tested, whereas the growth of L. acidophilus was not affected.

Physiochemical and Antimicrobial Activity of Garlic Cultivar (품종별 마늘의 이화학적 특성 및 항균활성)

  • Physicochemical and antimicrobial activities of 12 different garlic cultivars were investigated. Width and weight of California late cultivar (60.44mm, 53.73g) was the biggest and heaviest but Changyoung cultivar (44.04mm, 25.15g) was the smallest and lightest among the variety of garlic. The range of L, a and b color characteristics of garlic surface from different variety were 84.13~90.56, -1.10~0.77 and 18.24~26.61, respectively. Shear force was the lowest in California early, but 94-12-2 cultivar ($4211.35cm/kg^2$) was higher than another cultivars. Soluble solid range was 6.40~11.33 %brix, and Changyoung cultivar was the highest than the others, significantly. pH of garlics from different cultivar were 5.57~6.53. Total thiosulfinate content of California late cultivar (146.05mM/g) was higher, but Italy cultivar (93.23mM/g) was lower than the others. Total pyruvate content was the highest in Yugo cultivar ($162.50{\mu}M/g$) and the lowest in California early cultivar ($147.41{\mu}M/g$).
