Antibacterial Effect and Cytotoxicity of Desensitizer Containing Antimicrobial (항균물질을 함유한 지각과민처치제에 대한 항균효과 및 세포독성)

  • Dentinal hypersensitivity is a type of dental pain that occurs when various stimuli are applied to exposed dentin lesions. If the symptoms of dentinal hypersensitivity continue, the dentin is exposed and the hypersensitivity may become a periodontal disease or root caries due to bacterial infection. Therefore, the clinical goal of the present study is to reduce the pain of the patient suffering from dentinal hypersensitivity by developing antimicrobial hypersensitivity treatments and to improve oral hygiene. We prepared chlorhexidine, tetracycline, cetylpyridinium chloride, gallic acid loaded desensitizer by adding 0.1%, 0.5%, 1.0%, and 2.0% chlorhexidine, tetracycline, cetylpyridinium chloride, gallic acid to desensitizer (Micro Prime; Denville and Hurri Seal; Beutlich), and antibacterial effect, and cytotoxicity. The antibacterial test by using Staphylococcus aureus (SA. ATCC 6538, FDA 209) showed that the antibacterial effect of all experimental groups was significantly higher than that of control group (p<0.05). Cytotoxicity test by using agar diffusion assay indicated that Micro Prime showed mild toxicity, Hurri Seal showed severe toxicity and this cytotoxicity is supposed to be caused by one of desensitizer components showing weak antibacterial effect.

Characterization of Bacteriocin from Bacillus subtilis cx 1 (Bacillus subtilis cx1이 생산하는 박테리오신의 특성)

  • A new bacteriocin produced by Bacillus subtilis cx1, was partially purified and characterized. The bactericoin from B. subtilis cx1 was stable in the range of pH 2.5-9.5. B. subtilis csx1 retained its antimicrobial activity to long-term exposure at $-20^{\circ}C$ and $-70^{\circ}C$. However, B. subtilis cx1 was inactivated completely within 15 min over $60^{\circ}C$ and lost 50% of its antimicrobial activity within 15 min at $50^{\circ}C$, B. subtilis cx1 was inactivated by protease, trypsin, proteinase K and carboxypeptidase, which indi-cates its protein nature. Direct detection of the antimicrobial activity on Tricine -SDS-PAGE suggested an apparent molecular mass of about 9,500 dalton.

Antimicrobial activity by Paenibacillus elgii DS381 and its antimicrobial substances against microbial residents on human skin and pathogenic bacteria (인간 피부 상재균과 병원성 세균에 대한 Paenibacillus elgii DS381과 그 항균물질의 항균활성)

  • This study was carried out to evaluate effects of antimicrobial substances produced by isolated soil bacteria. Among two thousands of bacterial isolates Paenibacillus elgii DS381 exhibited high antimicrobial activities against several microbial residents on human skin and pathogenic bacteria. DS381 showed 15.3~26.0 mm inhibition zone diameter against all target bacteria and yeast in agar well diffusion test. Antimicrobial peptide produced by DS381 indicated low minimum inhibitory concentration (0.039-5.000 mg/ml). DS381 produced biosurfactant such as lipopeptide, and surface tension of culture supernatant of DS381 reduced from 60.0 to 40.3 mN/m. DS381 also showed $1.56{\pm}0.13U/ml$ of chitinase activity. These results suggest that Paenibacillus elgii DS381 may be utilized as an efficient biocontrol agent against some important human skin microbes and pathogenic bacteria.

Study on the Screening and Development of Antibiotics in the Mushrooms -The Screening of Fungal Antibiotics in Basidomycetes (I)- (버섯 중 항균물질의 검색 및 개발에 관한 연구 -곰팡이에 대한 항균물질의 검색 (1보)-)

  • In order to study antibiotic activities of basidomycetes (mushroom), 68 species of mushroom were extracted with petroleum ether, 80% ethanol, and distilled water in that order. A total of 204 extracts were obtained. Antibiotic activities against Microsporum gypseum were observed from the petroleum ether extracts of Abortiporus DGU-L6 mushroom, and the water extracts of Clitogbe DGU-7 mushroom. Antibiotic activity against Aspergillus niger were observed from the 80% ethanol extracts of Cortinarius DGU-51 and Marasmminus DGU-L67 mushroom. The petroleum ether extracts of Hetero DGU-L25 mushroom showed various antibiotic activities, particularly strong activities against M. canis. and the minimum inhibitory concentration (MIC) was $40\;{\mu}g/ml$. The extracts also showed antibiotic activities against A. niger (KCTC 2025), A. niger (KCTC 2118), A. versicolor (KCTC 2120), A. flavus (KCTC 2117), M. gypsem, Pyricularia oryzae, and Trichopyto mentagrophytes, and MIC for each fungus was $600\;{\mu}g/ml,\;500\;{\mu}g/ml,\;800\;{\mu}g/ml,\;100\;{\mu}g/ml,\;600\;{\mu}g/ml,\;200\;{\mu}g/ml,\;and\;600\;{\mu}g/ml$, respectively.

  • To investigate the antibacterial activities of 53 species of mushrooms for Gram-positive bacteria, bioactive substances from mushrooms were extracted with petroleum ether, 80% ethanol, and distilled water. The 14 extracts of 12 species of mushrooms, largely with 80% ethanol, showed antibacterial activities against 6 species of Gram-positive bacteria selectively. Especially, the extracts of Thylopilus PS-5, Pulveroboletus PS-7, and Russula PS-9 showed various antibacterial activities against several bacteria. Minimal inhibitory concentration (MIC) of the 80% ethanol extracts from Pulveroboletus PS-7 and Boletus PS-8 was $300\;{\mu}g/ml$ against Bacillus subtilis, and that of the 80% ethanol extracts from Trichaptium PS-3 was $500\;{\mu}g/ml$ against Micrococcus luteus. MIC of other extracts with antibacterial activities was above $1000\;{\mu}g/ml$.

Isolation and Identification of the Antagonistic Microorganisms Against Streptococous spp. Causing Dental Caries in Korean Soy Sauce (한국재래간장으로부터 구강질환 방제균의 선발 및 동정)

  • The antagonistic microorganisms against Streptococcus sanguis, S. salivarius and S. mutans causing the dental caries of oral diseases were isolated from Korean traditional soy sauce. Twenty five strains were isolated by pairing culture, paper disc culture and dual culture methods. The isolate NG 06 strain was observed with various cultural and physiological test, and $Biolog^{(R)}$ Bacterial Identification System. The strain was identified as Bacillus racemilacticus. The isolate NG 16 strain was confirmed to Gram-positive, rods, endospore production, utilization of melibiose, casein hydrolysis and starch hydrolysis. Also the second strain NG 16 was identified as $\beta$. amyloliquefaciens.

  • The antimicrobial properties of methanolic extract (PML) from Prunus nune byproducts after liquor manufacturing weremeasured using various putrefactive and food spoilage microorganisms. PML showed remarkable antimicrobial effects against various putrefactive and food spoliage microorganisms when used at 500g/mL. The antimicrobial properties were stable for 30 min at 100C and at pH 3 11. PML seems to be a natural antimicrobial agent with high effectiveness, and shows both thernal and pH stability. In addition, the mode of antimicrobial action suggests that the active components may synergistically perturb microbial membrane functions.

Characterization of the Bacteriocin from Enterococcus faecium CJNU 2008 (Enterococcus faecium CJNU 2008 균주 생산 박테리오신의 특성 규명)

  • Bacteriocin is a proteinaceous compound produced by microorganisms showing antimicrobial activities. In this study, the physicochemical properties of the bacteriocin produced by Enterococcus faecium CJNU 2008 strain were characterized. Partially purified bacteriocin showed stabilities against heat treatments at $100^{\circ}C$ for 30 min and $121^{\circ}C$ for 15 min and against solvents treatments such as methanol, ethanol, acetone, acetonitrile and chloroform. The bacteriocin also exhibited stabilities against lipase and ${\alpha}-amylase$ treatments but the stability was abolished at protease treatment, indicating that the antimicrobial agent from E. faecium CJNU 2008 was a proteinaceous bacteriocin. The bacteriocin also showed bactericidal mode of action against Listeria monocytogenes. The molecular mass of the bacteriocin was estimated to be under 6.5 kDa by a tricine-SDS-PAGE analysis. The bacteriocin was purified by HPLC. Further studies toward biochemical analysis of the bacteriocin are needed in near future.

Interaction of Enrofioxacin-Colistin Combination and LPS-Neutralization of the Different Antibiotic Classes In Vitro (시험관내에서 Enrofloxacin과 Colistin의 병용투여시 상호작용과 항균물질들의 독소중화능)

  • In the present study, we investigated the neutralization activity of various antimicrobials against lipopolysaccharide (LPS) as well as interaction between two antimicrobials (enrofioxacin and coilstin) using checkerboard method. The neutralization activity of antimicrobials used in the test was assayed by means of LAL chromogenic test after reaction of LPS with colistin, enorfioxacin, ampicillin, polymyxin B, oxytetracycline, streptomycin, and erythromycin. As the results, the neutralization activity of coltstin and polymixin B had a more stronger than that of tested other antimicrobials. In bacterial culture broth, the best neutralization activity of the antibiotics was also shown to coltstin and polymixin B. Meanwhile, It was shown to have synergism between enorfloxacin and coltstin on the basts of FIC (fractional inhibition concentration). The FIC of enorfioxacin-colistin combinations was 0.50-1.03 to Staphylococcus aureus R-209, 1.03-1.06 to Salmonella typhimurium, 0.75-1.25 to Bordetella Bronchtseptica and 1.02-1.25 to E. coli K88ab. On the basts of the above results, the present study may be of clinical usefulness in the choice of an antibiotic therapy for severe sepsts in animals.

  • Quaternary ammonium compounds(QACs) are some of the most commonly used antimicrobials. Antimicrobial polymers have been used as biomedical devices. In this Study, We synthesized new methacrylate monomers including quaternary ammonium moieties for preparation of contact lens, And also, we tested for antimicrobial activities against S. aureus, E. coli and P. aeruginosa. They showed much higher antimicrobial activities against all than the polymers of no including quaternary ammonium.
