Antibacterial Effect and Cytotoxicity of Desensitizer Containing Antimicrobial (항균물질을 함유한 지각과민처치제에 대한 항균효과 및 세포독성)

  • Dentinal hypersensitivity is a type of dental pain that occurs when various stimuli are applied to exposed dentin lesions. If the symptoms of dentinal hypersensitivity continue, the dentin is exposed and the hypersensitivity may become a periodontal disease or root caries due to bacterial infection. Therefore, the clinical goal of the present study is to reduce the pain of the patient suffering from dentinal hypersensitivity by developing antimicrobial hypersensitivity treatments and to improve oral hygiene. We prepared chlorhexidine, tetracycline, cetylpyridinium chloride, gallic acid loaded desensitizer by adding 0.1%, 0.5%, 1.0%, and 2.0% chlorhexidine, tetracycline, cetylpyridinium chloride, gallic acid to desensitizer (Micro Prime; Denville and Hurri Seal; Beutlich), and antibacterial effect, and cytotoxicity. The antibacterial test by using Staphylococcus aureus (SA. ATCC 6538, FDA 209) showed that the antibacterial effect of all experimental groups was significantly higher than that of control group (p<0.05). Cytotoxicity test by using agar diffusion assay indicated that Micro Prime showed mild toxicity, Hurri Seal showed severe toxicity and this cytotoxicity is supposed to be caused by one of desensitizer components showing weak antibacterial effect.

Antifungal and Insecticidal Activity of Ohyang (Five Medicinal Plants) (오향(五香) 성분의 살균 및 살충효과)

  • The antibacterial and insecticidal properties of ethanol extracts and volatile components extracted from Eugenia caryophyllata, Boswellia carterii, Agastache rugosa, Aristolochia contorta, and Aquilaria agallocha were evaluated. The ethanol extract and volatile component of E. caryophyllata showed strong antimicrobial effect against all strains (Mucor hiemalis, Aspergillus niger, Penicillium funiculosum, Trichoderma viride) and the volatile component of B. carterii showed antimicrobial effect against all strains except T. viride. The ethanol extract of E. caryophyllata and A. contorta showed $100\%\;and\;32\%$ mortality against Reticulitemes spertus kyushuensis Morimoto for 48 hours and 72 hours, respectively. In the case of volatile component, E. aryophyllata showed $100\%\;and\;20\%$ mortality against R. spertus and Lyctus linearis GOZE, respectively. The main constitute, eugenol $(92\%)$ among nine components from volatile component of E. aryophyllata were identified as antibacterial active substance.

Antimicrobial Activities of 'Formosa' Plum at Different Growth Stages against Pathogenic Bacteria (생육 시기에 따른 자두류 중 후무사의 식중독균에 대한 저해효과)

  • To determine the antimicrobial activity of methanol extracts form 'Formosa' plum against 4 kinds of pathogenic bacteria, the fermosa were got at different growth stages and were extracted using methanol, respectively. The Formosa methanol extracts treated with 5.0 mg/disc showed the highest antimicrobial activity against 4 kinds of pathogenic bacteria and those of Formosa 1-4(immature fruit), which thin out 10∼25 days before foal harvest, showed higher antimicrobial activity against gram positive and gram negative microorganisms than Formosa 5-6(mature fruit). Especially, the methanol extracts of Formosa 1 and 2 were exhibited the strongest growth inhibiting activities to these bacteria. The minimal inhibitory concentrations(MIC) of immature Fomosa methanol extracts was 320 $\mu\textrm{g}$/mL against Escherichia coli 0157:H7 and 160 $\mu\textrm{g}$/mL against Staphylococcus aureus respectively. The MIC of immature Formosa methanol extracts to Salmonella typhimurium and Listeria monocytogenes were 640 $\mu\textrm{g}$/mL. These results suggest that methanol extracs of immature formosa can be used as an effective natural antimicrobial agent in food.

  • 배지의 영향에 대하여 검토하기 위하여 SDA, MHA, NA 및 BHIA를 사용하였으며, 시험균주 중 Pseudomonas aeruginosa에 대하여만 NA에서의 MIC 값이 낮게 나타났을 뿐 나머지 균주에 대하여는 배지에 따른 항균력 변화가 관찰되지 않았으며, 배지의 pH를 5.4-8.4까지 변화시켰을 때 pH 변화에 따른 DA-1131의 유의적인 항균력 변화와 접종균량에 따른 항균력 변화가 관찰되지 않았다. Methicillin-resistant Staphylococcus aureus(MRSA)에 대하여는 DA-1131과 vancomycin 간의 상승 및 상가작용이 관찰되었고, P. aeruginosa, Klebsiella pneumoniae, Enterobacter cloacae 및 Serratia marcescens 등의 균종에 대하여는 ciprofloxacin의 상가작용이 확인되었다. Sisomicin은 P. aeruginosa에 대한 상승 및 상가작용이 인정되었으나 ceftazidime은 DA-1131에 의한 길항작용이 관찰되었다.

  • Antimicrobial activity of dandelion(Taraxacum platycarpum D.) was investigated. Methanol extract of dried dandelion was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, L. monocytogenes, S. aureus, E. coli and V. parahaemolyticus at $500\mu\textrm{g}/disc$. Ethylacetate fraction was further fractionated into 13 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 4, 5 and 6 had high antimicrobial activity. These were mixed again, re separated and five fractions were obtained. Among them, No. 2 fraction had the highest inhibitory effect on the microorganisms, which was then separated into six fractions. In the 3rd fractionation, No. 3 fraction was identified as benzoic acid by HPLC, $^{1}H-NMR$ and GC MS.

Sensitivity Test on the Food Poisoning Bacteria of the Garlic Extract (마늘추출물의 식중독균에 대한 항균검사)

  • Garlic (Allium sativum, L.) has a long history of reputed value and actual use for its medicinal, antimicrobial, and pesticidal properties. This study was conducted to find possible developments to natural food preservatives from garlic extracts. Garlic extracts were prepared from fresh crushed garlic and diluted with sterilize distilled water. The sensitivity test was conducted to investigate the antimicrobial effect of garlic extract against six strains of food poisoning bacteria such as Staphylococcus aureus, Escherichea coli, Salmonella typhimurium, Vibrio parahemolyticus, Bacillus subtilis, and Listeria monocytogenes. The results of sensitivity test were then compared with those of Vancomycin and Imipenem which were used as treatment for G. (+) bacteria and G. (-) bacteria, respectively. All of the 6 tested strains exhibited strong or moderate activity. V. parahemolyticus exhibited susceptible only in undiluted solution, but not in diluted garlic extracts.

  • Methylelaiophylin generated superoxide radicals in Bacillus subtilis and showed antibacterial activity against a broad range of gram positive bacteria. The inhibition of DNA synthesis was more sensitive than one of RNA synthesis. A recombination-deficient mutant strain of B. subtilis was 2-fold more sensitive than a wild strain, and this sensitivity was reduced in the presence of an antioxidant, dithiothreitol. Methylelaiophylin generated superoxide radicals in B. subtilis lysates, and this suggests that the antibacterial activity of methylelaiophylin is related to the generation of active oxygen species in the cells.

Antibacterial Effect of Tea Tree Ingredient for Denture Cleaners (티트리 성분 함유 의치세정제의 항균 효과)

  • The purpose of this study was to evaluate the antibacterial effect of tea tree oil in denture cleaners. A self-curing denture resin was used to make the experimental specimen ($12mm{\times}2mm$). A saline solution was used as the control. To observe surface changes after cleaning, the microhardness and color of the experimental specimen's surface were analyzed. For the antibacterial activity test, Candida albicans was used. The microhardness and color of the surface remained unchanged after cleaning. The result of the antibacterial activity test revealed that the tea tree oil-containing solutions had a more enhanced antibacterial effect than did the saline solution. Therefore, these results suggest that the tea tree oil-containing solution is a promising denture cleaners.

  • This study was conducted to select the promising antifungal microoganisms for biological control of wet bubble, Mycogone perniciosa. Ninty one isolates of fungi, 342 isolates of bacteria, and 556 actinomycetes were isolated from mushroom composts and soils, were subjected to primary screening test on agar medium base for their antimicrobial spectra. Among them, 12 bacteria and 71 actinomycetes were selected. Among the antibiotic producing microoganisms, 5 cultures were selected on the basis their antibiotic activities on casing soil with Benlate nontolerence M. perniciosa. Finally, AJ-117, AJ-136 and AK-139 were selected as microoganism with antifungal activity against two strains of M. perniciosa.

Characterization of a heat-resistant antimicrobial peptide secreted by Bacillus subtilis A405 (Bacillus subtilis A405 균주가 생성하는 내열성 항균 peptide의 특성 검정)

  • An antimicrobial peptide producing bacterium, Bacillus subtilis A405, was screened and identified among 700 of antagonistic bacteria. The heat-resistant antimicrobial peptide, AMP-405, was purified from the broth culture of B. subtilis A405 through $20{\sim}40%$ ammonium sulfate precipitation and ultrafiltration. The AMP-405 exhibited strong antimicrobial activities against Botrytis cinerea, Cercospora sp., Fusarium oxysporum, Penicillium digitatum, Celletotrichum gloeosporioides, Rhizoctonia solani, Pythium ultimum, Pyricularia oryzae, Escherichia coli, Pseudomonas spp. and Candida albicans. The molecular weight of the peptide was about 3.0 kDa determined by SDS-PAGE, Native-PAGE and Tris-Tricine gradient electrophoresis, and composed of 9 kinds of amino acid such as aspartic acid, glycine, serine, glutamine, valine, leucine, isoleucine, proline, tyrocine. To determine the efficiency of AMP-405 as a potential maintenance of fruits freshness, cherry tomato was srored at $25^{\circ}C$ for 2 weeks after treatment with $50{\mu}g/ml$ of AMP-405 and $10^{5}$ spores/ml of Botrytis cinerea simultaneously. Treatment with AMP-405 resulted in significantly less infection by Botrytis cinerea, than the treatment with tap water as a control.
