Electrochemical Properties of Molydenum (V) Complexes with Multidentate Ligands Containing Nitrogen or / and Oxygen Donor Atom (질소, 산소 주게원자 리간드를 가진 몰리브덴 (V) 착물의 전기화학적인 성질)

  • Reactions of $(Et_4N)_2[MoOCl_5]$ with multidentate ligands containing nitrogen or/and oxygen donor atom (EDTA, DTPA, IDA, CyDTA, OX) produce a series of binuclear molybdate (V) complexes. The prepared Mo (V) complexes has been identified by Elemental Analysis, Infrared Spectra, Proton Magnetic Resonance Spectra, and Electronic Spectra. The electrochemical reduction mechanism has been studied by Cyclic voltammetry, Controlled Potential Coulometry, and Spectrophotometry in pH 3.571-10.375 acetate, borate, phosphate/sodium hydroxide, phosphate, ammonium/ammonia buffers. The cyclic voltammogram of the Mo-EDTA, DTPA, IDA, CyDTA complexes at pH < ca. 6.00 have shown two reduction waves. The first reduction wave shows two electron process and the second reduction wave shows two electron process. The cyclic voltammogram of the Mo-EDTA, DTPA, IDA, CyDTA complexes at pH < ca. 8.00 has shown one reduction wave. This reduction wave show four electron process. The cyclic voltammogram of the Mo-OX complex at pH < ca. 7.2 has shown one reduction wave. This reduction wave show four electron process.

Growth of Candida albicans Biofilm is Inhibited by Salvia miltiorrhiza (단삼에 의한 Candida albicans 바이오필름 발달의 억제)

  • Candida albicans is an opportunistic human pathogen that causes infections. Candidiasis is often related to antifungal resistance because the pathogen has the ability to form biofilms. In a previous study, we found that the Salvia miltiorriza ethanol extract demonstrated anticandidal activity by altering membrane permeability and inhibiting the cell wall synthesis in C. albicans. Our results here demonstrate that $78{\mu}g/ml$ of the S. miltiorriza extract significantly diminished the early stage biofilms formed by 10 clinical C. albicans isolates by 51.3%; this was analyzed by 2,3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide salt (XTT) reduction assay. The effect of the S. miltiorrhiza extract on the adhesion of C. albicans cells to polystyrene plates and germ tube formation was examined via microscopic investigation. Although the density of the adhered cells was remarkably reduced up on incubation with $39{\mu}g/ml$ S. miltiorrhiza extract, germ tube formation by C. albicans was rarely affected. Quantitative real-time PCR analysis showed that the S. miltiorrhiza extract downregulated the expression of C. albicans hypha-specific genes, EAP1 by 34.7% (p < 0.001), ALS1 by 45.0% (p < 0.001), ALS3 by 48.1% (p < 0.001), and ECE1 by 21.3% (p = 0.006), respectively. Our data suggest that the S. miltiorrhiza ethanol extract significantly inhibited the early stage of biofilm formation by C. albicans by interfering with cell adhesion, by downregulating EAP1, ALS1 and ALS3, and presumably by modifying the cell wall and membrane structure.

  • With the recent advent of IoT technology, automatic pet feeders are being distributed so that owners can feed their companion animals while they are out. However, due to behaviors of pets, the method of measuring weight, which is important in automatic feeding, can be easily damaged and broken when using the scale. The 3D camera method has disadvantages due to its cost, and the 2D camera method has relatively poor accuracy when compared to 3D camera method. Hence, the purpose of this study is to propose a deep learning approach that can accurately estimate weight while simply using a 2D camera. For this, various convolutional neural networks were used, and among them, the ResNet101-based model showed the best performance: an average absolute error of 3.06 grams and an average absolute ratio error of 3.40%, which could be used commercially in terms of technical and financial viability. The result of this study can be useful for the practitioners to predict the weight of a standardized object such as feed only through an easy 2D image.

Cancer Chemopreventive Compounds of Red Ginseng Produced from panax ginseng C.A. Meyer (고려인삼으로 제조된 홍삼중의 화학적 암 예방성분)

  • Fresh Panax gineng C.A. cultivated in Korea(Korean red ginseng) was found to be ineffective as anticarcinogenic or cancer preventive in experimental animal model or in human case-control and cohort study. However, when treated with heat, the fresh ginseng, white ginseng were highly effective cancer preventives. Four compounds including 20(S)-ginsenoside Rh$_1$(Rh$_1$), 20(S)-ginsenoside Rh$_2$(Rh$_2$), 20(S)0-siwenoside Rg$_3$(Rg$_3$) and sinsenoside Rg$\sub$5/ were consequently purified from Korean red ginseng, and they were tested by Yun\`s 9 week medium-term anticarcinogenicity test model. Rg$_3$ and Rg$\sub$5/ statistically significantlydecreased the incidence of benzo(a)pyrene-induced mouse lung tumor, Rh$_2$showed tendency of decrease, and Rh1 showed no effect. It is, therefore, concluded that Rg$_3$ and Rg$\sub$5/ are active anticarcinogenic components in res ginseng and they either singularly or synergistically act in the prevention of cancer.

Multi-station joint inversion of receiver function and surface-wave phase velocity data for exploration of deep sedimentary layers (심부 퇴적층 탐사를 위한 수신함수와 표면파 위상속도를 이용한 다측점 자료의 복합 역산)

  • In this study, we propose a joint inversion method, using genetic algorithms, to estimate an S-wave velocity structure for deep sedimentary layers from receiver functions and surface-wave phase velocity observed at several sites. The method takes layer continuity over a target area into consideration by assuming that each layer has uniform physical properties, especially an S-wave velocity, at all the sites in a target area in order to invert datasets acquired at different sites simultaneously. Numerical experiments with synthetic data indicate that the proposed method is effective in reducing uncertainty in deep structure parameters when modelling only surface-wave dispersion data over a limited period range. We then apply the method to receiver functions derived from earthquake records at one site and two datasets of Rayleigh-wave phase velocity obtained from microtremor array surveys performed in central Tokyo, Japan. The estimated subsurface structure is in good agreement with the results of previous seismic refraction surveys and deep borehole data. We also conclude that the proposed method can provide a more accurate and reliable model than individual inversions of either receiver function data only or surface-wave dispersion data only.

Purification and Properties of Sunflower Seed $\alpha$-Galactosidase by Affinity Chromatography. (Galactomannan 이용에 관한 연구;Affinity Chromatography법에 의한 해바라기씨 유래 $\alpha$-Galactosidase의 정제 및 성질)

  • An ${\alpha}$-D-galactosidase (${\alpha}$-D-galactoside galactohydrolase, EC 3. 2. 1. 22) from sunflower seed was purified by affinity chromatography using N-$\varepsilon$-aminocaproyl-${\alpha}$-D-galactopyranosylamine coupled to sepharose and its properties were examined. The specific activity of the purified enzyme, tested with p-nitrophenyl-${\alpha}$-D-galactopyranoside as substrate, was 291.66 units/mg protein, representing an 115-folds purification of the original crude extract. The final preparation obtained from by Sephadex G-25 chromatography showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight was determined to be 42,000 by SDS-polyacrylamide gel electrophoresis. The purified galactosidase showed maximum activity at pH 4.5 and 55$^{\circ}C$, and was stable in the pH and temperature ranges of 4.0 to 5.0 and 30 to 55$^{\circ}C$, respectively. The enzyme activity was inhibited by Ag$\^$2+/, Hg$\^$2+/ and Co$\^$2+/. The enzyme activity was not affected considerably by treatment with other metal compounds. The enzyme liberated galactose from melibiose, raffinose, copra galactomannan, guar gum and locust bean gum by TLC, and also the hydrolysis rate of substrate was compared by HPLC.

Suitability evaluation of magnesium substituted biphasic calcium phosphates prepared by coprecipitation method (공침법을 이용한 마그네슘이 첨가된 biphasic calcium phosphate의 적합성 평가)

  • Magnesium-substituted BCP (biphasic calcium phosphate) powders were prepared by incorporating small amounts of magnesium into the structure of different hydroxyapatite (HAp)/${\beta}$-tricalcium phosphate (${\beta}$-TCP) ratios through coprecipitation method. A series of magnesium substitutions ranging from 0, 0.5, and 1.0 wt%, which are comparable to the measured magnesium contents, were performed. The obtained powders were characterized by the following analytical techniques: X-ray diffraction analysis (XRD), Thermo Gravimetric Analyzer (TGA) and Fourier transform infrared spectroscopy (FT-IR). The results have shown that substitution of magnesium in the calcium-deficient apatites resulted in the formation of biphasic mixtures of different HAP/${\beta}$-TCP ratios after heating above $1000^{\circ}C$. The 1.0 wt% magnesiumsubstituted-BCP were soaked in Hank's solutions after 2 weeks to observe the morphology of the biocement, especially needle-like hydroxyapatite crystals and to estimate the length and diameter of nanoneedle crystals.

Monitoring of nervous necrosis virus in fertilized eggs of walleye pollock (Gadus chalcogrammus) (명태(Gadus chalcogrammus) 수정란에서 신경괴사증바이러스(nervous necrosis virus) 모니터링)

  • We previously monitored nervous necrosis virus (NNV) in brain samples of artificially produced walleye Pollock (Gadus chalcogrammus) seedlings, with a low prevalence (1.8%, 1/55) but no clinical symptoms. Given that this virus is considered one of the most serious viral threats for almost all marine aquaculture fish species and characterized by both vertical and horizontal transmission, it would be interesting to monitor NNV in the fertilized eggs as well. We collected fertilized walleye pollock eggs from the farms located in Goseong during January to March, 2017. Approximately 50 mg of eggs were periodically taken from 4 each different batches, and 37 different pooled sample sets in total were made during sampling period. RNA was extracted from the eggs by using Trizol and cDNA was synthesized for RT-PCR for detecting NNV. Primers and PCR conditions are the same as previously described. As a result, NNV was not detected from any of the sample sets by one step PCR (0%, 0/37), suggesting NNV may not be a threat in walleye pollock aquaculture in Korea at present time. However, continuous monitoring for NNV should be conducted because introducing a new species into aquaculture industry involves potentials of disease outbreak and NNV is already known to cause outbreaks in gadoid fishes.

Optimizing Conditions for Streptomyces chibaensis J-59 Glucose Isomerase Production Using Response Surface Methodology (반응표면분석에 의한 방선균 Streptomyces chibaensis J-59 포도당 이성화효소의 생산 최적화)

  • Using response surface methodology(RSM), the various conditions(agitation speed, air flow, glucose concentration) in jar fermentor culture were investigated to find the optimum conditions for maximum enzyme production. Central-composite-design was used to control the variable constant in the experiment. The glucose isomerase production of Steptomyces chibaensis J-59 was mostly affected by the air flow rate and glucose concentration. The estimated optimum conditions were as follows: 1% birchwood xylan, 1.5% CSL, 0.1% $MgSO_4{\cdot}7H_2O$, 0.012% $CoCl_2{\cdot}6H_2O$, pH 7.0; air flow, 2.2vvm; agitation speed, 587rpm; glucose concentration, 0.586%. Experimental values(7.43GIU/ml) for the enzyme production obtained from the given optimum conditions had a almost resemblane to response values(7.67GIU/ml) predicted by the RSM. The jar fermentor culture by the RSM produced xylose isomerase about 2.7 times as much as the baffled flask culture.

Studies on the analysis of phytin by the Chelatometric method (Chelate 법(法)에 의(依)한 Phytin 분석(分析)에 관(關)한 연구(硏究))

  • Phytin is a salt(mainly calcium and magnesium) of phytic acid and its purity and molecular formula can be determined by assaying the contents of phosporus, calcium and magnesium in phytin. In order to devise a new method for the quantitative analysis of the three elements in phytin, the chelatometric method was developed as follows: 1) As the pretreatment for phytin analysis, it was ashfied st $550{\sim}600^{\circ}C$ in the presence of concentrated nitric acid. This dry process is more accurate than the wet process. 2) Phosphorus, calcium and megnesium were analyzed by the conventional and the new method described here, for the phytin sample decomposed by the dry process. The ashfied phytin solution in hydrochloric acid was partitioned into cation and anion fractions by means of a ration exchange resin. A portion of the ration fraction was adjusted to pH 7.0, followed by readjustment to pH 10 and titrated with standard EDTA solution using the BT [Eriochrome black T] indicator to obtain the combined value of calcium and magnesium. Another portion of the ration fraction was made to pH 7.0, and a small volume of standard EDTA solution was added to it. pH was adjusted to $12{\sim}13$ with 8 N KOH and it was titrate by a standard EDTA solution in the presence of N-N[2-Hydroxy-1-(2-hydroxy-4-sulfo-1-naphytate)-3-naphthoic acid] diluted powder indicator in order to obtain the calcium content. Magnesium content was calculated from the difference between the two values. From the anion fraction the magnesium ammonium phosphate precipitate was obtained. The precipitate was dissolved in hydrochloric acid, and a standard EDTA solution was added to it. The solution was adjusted to pH 7.0 and then readjusted to pH 10.0 by a buffer solution and titrated with a standard magnesium sulfate solution in the presence of BT indicator to obtain the phosphorus content. The analytical data for phosphorus, calcium and magnesium were 98.9%, 97.1% and 99.1% respectively, in reference to the theoretical values for the formula $C_6H_6O_{24}P_6Mg_4CaNa_2{\cdot}5H_2O$. Statical analysis indicated a good coincidence of the theoretical and experimental values. On the other hand, the observed values for the three elements by the conventional method were 92.4%, 86.8% and 93.8%, respectively, revealing a remarkable difference from the theoretical. 3) When sodium phytate was admixed with starch and subjected to the analysis of phosphorus, calcium and magnesium by the chelatometric method, their recovery was almost 100% 4) In order to confirm the accuracy of this method, phytic acid was reacted with calcium chloride and magnesium chloride in the molar ratio of phytic: calcium chloride: magnesium chloride=1 : 5 : 20 to obtain sodium phytate containing one calcium atom and four magnesium atoms per molecule of sodium phytate. The analytical data for phosporus, calcium and magnesium were coincident with those as determine d by the aforementioned method. The new method employing the dry process, ion exchange resin and chelatometric assay of phosphorus, calcium and magnesium is considered accurate and rapid for the determination of phytin.
