과학원로에게 듣는다 - 우리 재래 닭 복원해 낸 가금학의 대부 '오봉국'

  • 닭과 함께 한 80년 세월, 지난 1957년에 미국 미네소타대학에서 석사학위를 한 후, 귀국 길에 모교에서 개량 보존하고 있던 미국산 종계와 종란을 국내에 도입, 6.25 전쟁으로 황폐화한 우리나라 닭의 육종을 개량, 증식함으로써 국민 먹거리와 농가 소득 증대에 기여하였고, 1997년 멸종 위기에 놓여있던 '우리 재래닭' 4종류를 탄생시켜낸 오당(梧堂) 오봉국(嗚鳳國) 박사(86)를 서울 서초구에 있는 대한민국학술원에서 만났다. 오봉국 박사는 서울대학교 농대 교수로 40년간 재직하면서 150여 편의 연구논문을 발표하고 후학들을 이끌어 왔으며, '한국가금발달사'를 저술하여 우리나라의 가금학을 집대성했고, 서울대 농대 학장, 한국가금협회장 등을 역임했다. 평안남도 진남포 출생이며, 국민훈장 모란장을 수훈했다.

Genetic Polymorphism of Avian Leukosis Virus Host Receptors in Korean Native Chickens and Establishment of Resistant Line

  • Avian leukosis virus (ALV) is a highly contagious retrovirus that causes tumors and has resulted in great economic loss worldwide owing to its high transmission rate. Various ALV viral subgroups exist, with infections occurring via specific host receptors. The susceptibility or resistance of avian species to the ALV-A and K subgroups is determined by the host receptor, the tumor virus locus A (tva) gene, while that to ALV-B depends on another host receptor, the tumor virus locus B (tvb) gene. The resistance alleles of tva and tvb have primarily been identified in China, but none have beendetected in Korea. We analyzed the frequencies of tva and tvb genotypes in White Leghorn (WL), Korean Ogye (KO), and Korean native chicken (KNC) breeds, and assessed the resistance to ALV subgroups. In WL, both tva and tvb had various genotypes, including susceptibility and resistance alleles, whereas in KO, tva and tvb resistance alleles were dominant. In KNC, tva susceptibility and resistance alleles were mixed, whereas tvb resistance alleles were dominant. In addition, we showed that there were differences in the splicing pattern of tva transcripts and the expression level of tvb transcripts within breeds. Finally, we confirmed that ALV resistance depended on KO and KNC genotypes by in vitro infection of chicken embryonic fibroblasts with ALV. These results highlight that some KO and KNC individuals are naturally resistant to ALV subgroups A, B, and K, and will facilitate the preservation of economically superior traits through selective breeding.

Distribution of Telomeric DNA in Korean Native Chicken Chromosomes (한국 재래닭 염색체의 텔로미어 분포 양상)

  • Telomeres are nucleoprotein structures at the ends of chromosomes consisting of DNA sequences arranged in tandemly repeated units $(TTAGGG)_n$. However, this hexamers can also occur at non-telomeric sites in some avians and vertbrate. This study was carried out to present the distribution of telomeric DNA sequences in Korean Native Chicken chromosomes. The fluorescence in situ hybridization technique using a telomeric DNA probe was performed to the metaphase spreads of chicken early embryonic cells. Telomeric DNA signals were detected at the ends of chromosomes including macrochromosomes and microchromosomes. In chicken, surprisingly, chromosome 1 showed very distinct interstitial telomeric DNA hybridization patterns which located two interstitial sites in the p-arm at 1p11 and 1p23, and one in the q-arm at 1q32. In chromosome number 2 and 3 also displayed interstitial telomeric signals (ITS) in the long arms at 2q24 and 3q32, respectively. The pattern of telomeric DNA distribution in Korean Native Chicken chromosomes was in agreement with a previously reported in Gallus domesticus. The relative amount of telomeric DNA sequences in each macrochromosomes ranged from 4.6% to 16.3%. Distribution of telomeric DNAs at the end of p-arm was much more than that of q-arm in almost chicken chromosomes. The distribution of ITS in chicken chromsomes implicate to tandem chromosome fusions that might have occurred during the process of karyotype evolution.

Chemical Compositions of the Four Lines of Korean Native Chickens (4계통 재래종 닭고기의 화학적 특성)

  • This study was conducted to compare nutritional composition and taste-related compounds of breast and thigh meats from four lines of Korean native chickens (KNC) Yeonsan Ogye, Hyunin Black, Hwangbong, and Hoengseong Yakdak. White Leghorn (WL) was used as a control. Fifteen male chickens (three chickens in each line) were grown under same condition and slaughtered at 53 weeks old. The contents of Fe and K in KNC, especially Hwangbong breast meat and thigh meat of Hyunin Black and Hoengseong Yakdak, were higher than WL. The contents of Na were lower in KNC compared with WL regardless of parts (p<0.05). Vitamin A contents were higher in thigh meat of WL and Hyunin Black, and vitamin $B_1$ contents were lower in Hoengseong Yakdak than other lines. Vitamin $B_3$ were higher in breast meat of four lines of KNC and thigh meat of Hoengseong Yakdak than WL. Total amino acid contents were higher in breast meat of KNC than WL. The level of good-tasting amino acids were significantly higher in breast meat of Hoengseong Yakdak and WL, and thigh meat of Hyunin Black and WL than other lines of chickens (p<0.05). The ratio of good-tasting amino acids to bitter tasting amino acids was higher in breast meat of Hoengseong Yakdak and thigh meat of Hyunin Black than WL. IMP contents were higher in Hyunin Black and WL than other lines of chickens. Based on these results, it can be concluded that four lines of KNC may have superior nutritional quality and taste when compared with WL.

Estimation of Genetic Characteristic and Cumulative Power of Discrimination using the Microsatellite Markers in Korean Native Chicken (Microsatellite Marker를 사용한 재래 닭 품종 유전적 특성 및 개체 식별력 분석)

  • To estimate the genetic characteristics and cumulative power of discrimination (CPD) Korean Native Chicken. We used a total of 195 genomic DNAs from four breeds population (Korean Native Red chicken: R, Korean Native Yellow chicken: Y, Korean Native Black chicken: L, Ogal chicken: S). Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The lowest distance (0.05) was observed between the R and L strains and the highest distance (0.158) between the L and S strains. Korean native chicken strains (R, Y, K) have each other comparatively near genetic distance. Cumulative power of discriminate (CPD) was 99.999% by including the 10 microsatellites loci individual identification system. And then matching probability in that two different individuals incidentally have same genotype was estimated to $0.36{\times}10^{-7}$. The system employing the 10 markers therefore provided to be applicable to individual identification in Korea native chicken.

Epistatic Effects of Six Candidate Genes on Fatty Acid Composition in Korean Native Chicken

  • Fatty acid composition is an important economic trait that affects meat flavor. Several genes that influence fatty acid composition in meat have been investigated. In a previous study, we identified 51 significant SNP × SNP interactions (P≤0.05) between nine SNPs of six candidate genes (DEGS1, ELOVL6, FABP3, FABP4, FASN, and SCD) on meat fatty acid composition in Korean native chicken. This study further investigated the patterns of the SNP × SNP interactions to understand how they affect the fatty acid content in thigh and breast meat of Korean native chicken. The significant epistatic effects of SNP combinations showed various patterns for each fatty acid trait. The results of this study suggest that the respective additive effects of each SNP on polygenic traits, such as fatty acid composition, should be considered in combination with the epistatic effect of SNP combinations in animal breeding programs. The findings of this study have provided new genetic information for improving meat quality, especially the fatty acid composition, of Korean native chicken.

  • 1960년 4월 부모님이 농사를 천직으로 알고 농촌이 살아야 나라가 산다고 본인에게 역설하셨다. 닭의 울음은 한 국가지도자의 탄생을 알리는 고고한 외침소리다. 마찬가지로 각자 자신의 목표달성도 닭울음 소리의 영감에서 출발한다. 어언 50년 동안 전국농업기술자협회에서 정신혁명, 기술혁명, 생활혁명으로 영농기술의 선도주자로 활동해 왔다. 고 상허 유석창 박사, 고 성천 류달영 박사, 백포 정장섭 회장, 강춘성 회장, 현재 윤천영 회장과 본인과 같이 37년 이란 세월이 흐르고 줄기차게 열어온 협회대회, 농민대학 영농교육, 도시민 귀촌 농업교육, 권익신장을 위한 농정활동, 도농녹색교류, 농업의 지속발전 도시민과 상생하는 국민농업 운동 확산을 계속하고 있다.
