Histochemical Studies of the Goblet Cell of the Oviduct in Korean Ring-necked Pheasants(Phasianus colchicus karpowi) (한국산 꿩 난관의 술잔세포에 관한 조직화학적 연구)

  • The study was designed to investigate the aspects of the goblet cell of the oviduct in laying Korean ring-necked pheasants by histochemical methods (PAS, alcian blue-PAS, alcian blue pH 2.5, alcian blue pH 1.0, and aldehyde fuchsin-alcian blue stain). Twelve laying Korean ring-necked pheasants were used. The results were summarized as follows. The goblet cell of the oviduct of the Korean ring-necked pheasant contained sulfated acid mucosubstances and neutral mucosubstances in the infundibulum. The compounds of sulfated acid mucosubstances, nonsulfated acid mucosubstances, and neutral mucosubstances are present in goblet cells of the magnum, vagina and openings of the tubular gland of the uterus of the oviduct in the Korean ring-necked pheasant. The goblet cell of the isthmus and uterus of the oviduct of the Korean ring-necked pheasant contained neutral mucosubstances except openings of the tubular gland of the uterus. The histochemical characteristics of secretory granules of goblet cells in oviduct of the Korean ring-necked pheasant were not related to the position of eggs in oviduct.

  • The morphological study was carried out to classify the spermatogenic germ cells of the seminiferous tubule in Korean Native Pheasant during the breeding season. The results were as follows : 1. The spermatogonia can be classified into the three types of A, In(intermediate) and B. 2. The primary spermatocyte can be classified into the five types as preleptotene, leptotene, zygotene, pachytene and diakinesis. 3. The maturing processes of nucleus of spermatid can be divided into seven steps. The round shape of the spermatid was changed to the elongated form during the spermiogenesis. This observation may be useful to the study of the breeding cycles in the Korean Native Pheasant.

Morphological Study on the Post-hatching Development of the Pineal Gland in Korean Pheasant (Phasianus colchicus Karpowi) (한국산 꿩 송과샘의 부화후 발달에 관한 형태학적 연구)

  • The pineal gland of the bird occupies a key position in the phylogenetic evolution of this organ. Therefore, the purpose of this study was to investigate the developmental changes of the pineal gland during post-hatching period in Korean pheasant. The pheasants were sacrificed at 1-day-, 1-month-, 2-month-, and 6-month-old after hatching. The morphological characteristics of a pineal glands were determined in all pheasants using light microscope, and transmission electron microscope. Connective tissue originated from the capsule divided the pineal parenchyma into incomplete lobules. The parenchyma was consisted of pinealocytes and supportive cells. These parenchymal cells were arranged in the forms of solid lobules as well as incomplete follicles. At the follicular lumen, membraneous lamellar complexes and blob -like structures were present. Pinealocyte, a predominent cell type, had euchromatic nucleus, and showed the segmental organization. The bulbous apical portion had scanty free ribosomes and occasional cilia associated with basal bodies. The constricted neck, transitional portion from apical to pericarya had junctional complexes with adjacent supportive cells, and had microtubules. Cell body contained abundant mitochondria, well-developed Golgi complex, rough endoplasmic reticulum (RER) and free ribosomes. Basal processes extended from the base of the cell soma toward the basal lamina and contained 60∼90 nm dense cored vesicles. Supportive cells, another major type of the parenchyma, were characterized by the dense and elongated nucleus, and contained moderate number of mitochondria, RER, developed Golgi complex, free ribosomes and a few dense bodies in the perinuclear cytoplasm. Slender processes of supportive cells interposed between the pinealocytes and often bordered the basal region of the parenchyma. These results indicate that the pinealocytes of the pheasant are not rudimentary photoreceptor cells, and appear to have secretory function. Further studies will be required to confirm the morphological characteristics of pineal gland in adult pheasant during breeding and nonbreeding season.

Accumulation of Heavy Metals in Forest Birds from Korea - Especially in Mt. Chiri - (한국산(韓國產) 산림조류(山林鳥類)의 중금속(重金屬) 축적(蓄積) - 지리산지역(智異山地域)을 중심(中心)으로 -)

  • The heavy metal contents accumulated in the birds organ were analyzed by use of 10 species of birds to investigate the environmental pollution of the birds inhabited in Mt. Chiri. Heavy metal concentrations in bird organs, e.g. muscle, liver, kidney, lung and heart were analyzed for 10 species of the birds. 1. Contents of some heavy metal elements such as Fe, Zn, Cu and Mn were similar level compared with any other district. But contents of some heavy metal elements shch as Pb, Cd and Hg were comparatively lower level than those in any other district. 2. The contents of heavy metal accumulation in the muscle of the bird organs were detected much more from the migrants than the resident birds ; the resident bird, Phasianus colchicus were detected to contain $0.01{\mu}g/g$ og Hg and $0.05{\mu}g/g$ of Cd, but in the migrant bird, Turdus dauma they were detected to contain $0.17{\mu}g/g$ of Hg and $0.08{\mu}g/g$ of Cd. 3. The contents of lead were scarcely detected in the muscle of 10 species of birds, but it was highly detected from the hunting (meat-eating) birds such as Phasianus colchicus ($0.17{\mu}g/g$) and Streptopelia orientalis ($0.11{\mu}g/g$). Generally, the concentration of heavy metals of the analyzed birds was more mercury than Cadmium.

  • This study was carried out to investigate the distribution and morphological properties of mast cells in the ileum of Korean pheasant. In the ileum mast cells were located in the lamina propria of mucoase. The cells were mainly oval with irregular cell surface occasionally spindle-like or triangular in shape. Some mast cells had one or more tail-like long cytoplasmic processes. All mast cells had many blue granules stained by toluidine blue in the cytoplasm.

  • Protothalipine colorless needle crystal, mp $195-6^{\circ}$, $C_{21}H_{25}NO_{5}$) and jatrorrhizine chloride (Base A, orange needle crystal, mp $198^{\circ}$) are identified by chemical, spectral analysis, and comparison with authentic sample. Base B (reddish crystal, mp $192-4^{\circ}$) might be a artifact substance, which was changed from the jatrorrhizine chloride. Base C (yellowish needle crystal, mp $228~230^{\circ}$) and Base D (reddish needle crystal, mp $211~213^{\circ}$) were assumed to be thalifendine chloride, desoxythalidastine chloride by phytochemical and spectral analysis respectively.

  • 1997년 4월부터 1998년 4월까지 경상북도 내연산 일대의 관속식물상 조사를 실시한 결과, 관속식물은 100과, 350속, 538속, 1아종, 92변종, 10품종, 1교잡종 총 642종류(texa)를 확인하였으며, 이 중 211종류는 금번조사에서 새롭게 추가하였다. 특히 고란초, 부들, 창포, 꽃창포, 노랑무늬붓꽃, 산작약, 둥근잎꿩의 비름, 꿩의 바람꽃, 연잎꿩의다리, 참좁쌀풀 등의 희귀 및 멸종위기 식물을 확증표본과 함께 보고함으로써 본 지역 식물상 조사에 큰 의의가 있다고 판단된다. 본 조사에서 확인한 희귀 및 멸종위기 식물은 총 29종류로서, 환경부 법정보호식물이 3종류, 환경부에서 실시하고 있는 전국자연환경조사에서 제시한 특정식물종 중 정밀생태종은 27종류, 그리고 산림청 희귀 및 멸종위기 식물은 15종류로 나타났으며, 둥근잎꿔의 비름, 망개나무, 노랑무늬붓꽃 등의 분포 등에 대해 논하였다.

  • Frozen korean traditional poultries(Manyker, Korean native fowl, Ogol fowl and Pheasant) were thawed at $5^{\circ}C$, and $20^{\circ}C$ in the air, and using a microwave oven. Effect of thawing methods on the quality of Korean poultry was investigated. The pH of the poultry muscle was lowered after thawing in all treatments. The pH of the muscle thawed by microwave oven was the lowest. The quantity of drip released after thawing was the least in a microwave thawing. The quantity of drip in Maniker and Pheasant was loss than Ogol fowl. The relationship between pH and water binding capacity showed highly significant, and the drip quantity after thawing increased as the pH decreased. Leg muscle has higher strength than breast muscle in all the Korean poultry. Shear strength of all the poultry muscle showed closely relative related to the pH. As the pH lowered, the shear strength of the muscle was also decreased.

  • The image analyser was used for the measurement of the morphological changes of the spermatogenic cells and seminiferous tubules in the pheasant during the breeding and non breeding season. 1. The seminiferous tubules were enlarged 2.57 times during the breeding season than during the non breeding season. Only Sertoli cells and spermatogonia in the seminiferous tubules can be found during the non breeding season. 2. There is no significantly difference in the ratio of nucleus area against that of cell size in the spermatogonium between breeding and non breeding season. The ratio of area was 28.71% and 29.11%, respectively. However, the enlargement of spermatogonium was noticed during the non breeding season. 3. The highest value of the ratio of the nucleus area against that of cell size among the germ cells was measured 37.40% in the pachytene phase of the spermatocyte during the breeding season. 4. The ratio of nucleus area against that of cell size in the spermatid was 22.53%.

The Anatomical Study on the Distribution of Oviductal Vessels in Korean Native Pheasants (한국산 꿩 난관의 혈관분포에 관한 해부학적 연구)

  • The distribution of oviductal vessels of eight Korean native pheasants was observed after Latex casts of vessels. Latex was injected into thoracic aorta and posterior vena cava of these birds for cast preparation. The results were as follows: 1. The arteries suppling oviduct of the Korean native pheasants contained A. oviductalis cranialis, media, caudalis and vaginalis. The veins drainaging oviduct were stretched along the following ateries and all of the oviductal vessels were situated on the left side of the body. 2. The anterior oviductal artery arose from the pubic artery of the left external iliac artery and distributed to the infundibulum and the magnum of the oviduct. 3. The middle oviductal artery arose from the left sciatic artery and distributed to the isthmus and shell gland of the oviduct. 4. The caudal artery arose from the left internal pudendal artery of the median sacral artery and distributed to the caudal part of uterus and the cranial part of vagina. 5. The vaginal artery arose from the left internal pudendal artery of median sacral artery and distributed to the caudal part of vagina.
