The Chemical Composition of Taraxacum officinale Consumed in Korea (한국산 서양민들레(Taraxacum officinale)의 화학적 조성)

  • For centuries, dandelion(Taraxacum officinale) is widely used as a food and a medicinal herb. However, there is inadequate documented information on the chemical composition of T. officinale consumed in korea. This study was attempted to analyze proximate component, amino acid, free sugar, mineral and fatty acid composition in T. officinale to establish the value as an useful food material. Moisture, ash, crude protein and fat contents in leaf are 7.85, 11.35 21.6 and 5.12%, respectively. Moisture and ash contents of root were 7.73 and 4.82%. Crude protein and fat contents of root were 11.8 and 1.73%, respectively. The major free amino acid was asparagine, arginine, serine and proline etc. The total contents of free amino acid in leaf and root were 1476.98 and 2710.06 mg%, respectively. The major free sugar in T. officinale was glucose, fructose, sucrose and maltose. Sucrose was also detected in a large amount. The major fatty acid of T. officinale are palmitic acid, stearic acid, oleic acid, linoeic acid and linolenic acid. The fatty acid compositions of leaf and root were different each other. Linolenic acid(63.6%) was the major fatty acid of the leaf and linoleic acid(56.4%) was the major fatty acid of the root. Most of mineral contents were high in leaf and root. Especially K content was higher in leaf than root.

Changes in Physicochemical Properties of Actinidia arguta (Siebold & Zucc.) Planch. ex Miq. by Blanching, Drying, and Fermentation (다래순의 데침, 건조 및 발효 조건에 따른 이화학적 특성 변화)

  • This study investigated changes in physicochemical characteristics by drying and fermentation in order to utilize Actinidia arguta. Moisture content of A. arguta was 85.81%. Major sugar and organic acids were sucrose, succinic acid, and citric acid. A. arguta contained 19 kinds of amino acids, including 8 kinds of essential amino acids such as valine, methionine, isoleucine, leucine, threonine, phenylalanine, tryptophan, and lysine. For total organic acids, sample fermented for 17 h at $50^{\circ}C$ under a relative humidity of 80% showed 29,026.53 mg/100 g total organic acids. For total free sugars, cold-dried sample showed the highest level at 6,560.86 mg/100 g, which decreased to 2,386.73 mg/100 g after blanching. For the ratio of essential amino acids, freeze-dried sample showed a content of 11.66%, which increased 4-fold up to 40.71~55.50% with fermentation. Both GABA and vitamin U were highest after 17 h of fermentation (110.29 mg and 6.78 mg/100 g fresh weight, respectively). A. arguta contains a variety of free amino acids that increase in amount after fermentation and thus is expected to be developed as a functional food and substitute tea.

Content Analyses of Fiber, Protein and Amino Acids of Fully Ripe Fruits of Korea Native Squash, Cucurbita moschata Poir (한국재래종 호박 완숙과의 섬유질, 단백질 및 아미노산 함량 비교분석)

  • We studied active substances like crude cell wall components, crude protein, composing amino acids and free amino acids including orinithine cycle-related amino acids such as asparagine, ornithine and citrullin in fully ripe fruits of Korean native squash, Cucurbita moschata Poir. Crude protein content of 'Jeju 2' was the highest with $2,830\;{\mu}g/g$, while 'Sangju' was the lowest with $1,319\;{\mu}g/g$. Regarding the contents of crude cell wall components, fruit 'Kanghaw' was the highest with 2,961 mg% while 'Namhea' was the lowest with 1,582 mg%. Pectin contents of crude cell wall components were the highest in 'Kanghaw' (2,198 mg%) followed by 'Jeju 2' (2,178 mg%) and 'Jeju l' (1,461 mg%). The main contents of amino acids in squash were glutamic acid, aspartic acid, lysine, leucine and valine, which comprised to be more than 50% of total amino acid contents. Especially, in 'Jeju 2' aspartic acid and threonine were not detected. In fully ripe fruits, a total of 34 kinds of free amino acids were detected including 8 kinds of essential amino acids (histidine, isoleucine, leucine, lysine, phenylalanine, methionine, threonine and valine). More than 50% of the total free amino acids were aspartic acid and asparagine, and also all varieties were detected in ornithine, citrullin, and arginine, which are related to Ornithine cycle. There was a big difference in the contents of arginine in all varieties whereas the contents of ornithine and citrullin were very similar. 'Teaan' 29.34% was 7 times higher than 'Namhea' 4.30% in regards to arginine contents.

  • These studies were carried out to investigate the effects of the addition of amino acids and FBS as source of exogenous nitrogen fixation added to medium on in vitro production of blastocyst derived from bovine follicular oocytes. The base medium was TCM-199 solution for in vitro maturation(IVM) of bovine follicular oocytes and Fer-TALP solution for in vitro fertilization(IVF) and YS solution for in vitro culture(IVC). IVC used the fertilized oocytes of 24-hr culture (day 1) after IVF. Rmbryos were cultured in drop-culture that contained 25 embryos per 10 ${mu}ell$. The results obtained are as follows: 1 The developmental rates of fertilized oocytes to blastocyst that developed from YS solution with NEAA derived from MEM alone were higher than those of YS solution without NEAA. 2. The developmental rates of fertilized oocytes to blastocyst that developed from YS solution with EAA derived RPMI 1640 alone were significantly higher than those of YS solution without EAA (p<0.05). 3. When added to EAA on day 5 after NEAA supplementation on day 1, the developmental rates of hatched blastocyst and blastocyst to hatched blastocyst were improved. 4. When removed to EAA on day 3, day 4 and day 5 from medium after NEAA and EAA supplementation on day 1. the developmental rates of blastocyst to hatched blastocyst were reduced. 5. When added to FBS as source of exogenous nitrogen fixation, the developmental rates of blastocyst and hatched blastocyst that developed from the later culture higher(day 5) than those of the early culture.

Varietal Difference of Protein Content and Amino Acid Composition in Sesame (참깨품종의 단백질함량과 아미노산 조성)

  • To obtain the basic information an seed quality improvement in sesame, protein content of 114 varieties and amino acid composition of 12 varieties was analyzed. Protein content showed the vaietal difference ranged 20.6-30.2% and the mean was 24.72%. The highest variety in protein content was PI158066 (30.2%) originated from U.S.A. Protein content of Korean local varieties were highest among original group analyzed. Seed coat texture and seed coat color affected to protein content so, smooth type was higher than rough type in protein content, and black seeded varieties showed the hight protein content. Amino-acid composition of sesame was uneque in balance and higher than FAO reference. Total amino-acid of variety PI258372 was highest as 25.03%. Essential amino-acid (EAA) /total amino-acid(TAA) ratio of sesame was higher as 42-58.2% than soybean, corn, rice, peanut. Korean local variet 'Samcheck' showed best quality in amino-acid composition as 58.2% in EAA/TAA ratio with high tyrocin and lysine. Total amino acid content was high in order of Korean local '||'&'||'gt; introduced '||'&'||'gt; Korean bred varieties.

  • Potato is the 4th important crop along with rice, wheat and maize. It contains high quality of starch with relatively high content of vitamin C and protein. However, there is a nutritionally limiting factor due to a low level of sulphur-containing essential amino acid including methionine and cysteine. Recently, recombinant DNA technology and metabolic engineering with genes involved in the bio-synthetic pathway have been applied to enhance the level of these essential amino acids. In this report, it has been discussed about the current status and bottleneck on the development of transgenic potato containing high level of sulphur-containing essential amino acids.

Change of amino acids contents of Gastrodia elata Blume with harvest times and seed tuber (수확시기와 자마의 특성별 천마의 아미노산 함량 변화)

  • Gastrodia elata Blume, an achlorophyllous orchid plant, has been used in traditional medicine and harvests in spring and fall. Here we investigated the quantitative changes of amino acids in G. elata by harvest times and seed tubers. In the results, we found that there was not big difference in contents of total amino acids but the distribution of amino acids differed depending on harvest times. 19% of total amino acids were asparagin and valine, an essential amino acid, occupied 10% within total amino acids in the tubers harvested in October. Among amino acids in the tubers harvested in April, serine occupied 23% and arginine, an essential amino acid, occupied 10% within total amino acids in tuber. Interestingly, the use of sexually propagated seed tubers produced high concentration of total amino acids compared to vegetatively propagated seed tubers. As for sexually propagated seed tubers, essential amino acids contents similar to the tubes harvested in October and nonessential amino acids contents similar to the tubers harvested in April. In this study, we found that amino acids contents in G. elata tubers altered depending on various cultivation practices. Therefore if these results can be applied to food industry, the value of G. elata as a natural food resource will be enhanced to a great extent.

염절임 가지(시키부, Shikibu)의 저장방법에 따른 성분 변화

  • 본 연구는 수출방법의 개선과 절임 가공식품의 개발하고자 일본 수출용 시키부(Shikibu) 염절임 가지의 저장방법과 저장중에 당류, 유기산 및 아미노산의 변화를 분석하였다. 총당 및 환원당 함량은 저장방법에 관계없이 저장기간이 길어질수록 조금씩 감소하였고, 침지 저장한 것보다는 진공포장하여 저장한 것이 변화가 적었다. 가지에서 분리 정량된 유기산은 acetic acid, citric acid, lactic acid, malic acid 그리고 succinic acid 총 5종이었으며 acetic acid와 malic acid의 함량이 가장 높았으며, 저장기간이 길어질수록 저장 초기에 비해 acetic acid의 함량은 증가하였는데 반해 malic acid의 함량은 감소하였다. 가지의 주요한 구성아미노산은 valine, leucine, aspartic acid, glutamic acid, proline, alanine 등이었으며, 필수아미노산 함량은 저장기간이 길어질수록 저장 초기에 비해 진공포장하여 저장한 방법에서는 감소하였다가 증가하였는데 반해 침지하여 저장한 방법에서는 계속 증가하였고 저장 20일째에는 진공포장한 방법보다 l00mg/100g 정도의 차이를 나타내었다. 유리아미노산은 aspartic acid, alanine, cystine과 proline등의 함량이 높았고, 저장 기간이 길어질수록 저장초기에 비해 유리아미노산의 함량이 증가하였는데, 특히 진공 포장하여 저장한 가지에 비해 침지 저장 한 가지에서 그 함량이 높았다. 또한 아미노산 유도체의 함량은 phosphoserine, taurine, ${\gamma}$-aminoisobutyric acid 및 hydroxyproline의 함량이 높았고, 저장중에 phosphoserine의 함량은 감소하였는데 반해 ${\gamma}$-aminoisobutyric acid는 진공포장하여 20일 동안 저장하였더니 생가지 보다 5배 이상의 증가를 보였다. 스키부품종의 절임가지의 저장방법과 저장기간에 따른 성분의 변화를 고려할 때 축양품종과 동일하게 진공포장하는 것이 절임가지의 품질유지할 수 있는 것으로 생각된다.

Quality Improvement in Perilla III. Varietal Differences of Protein Content and Amino Acid Composition in Perilla (들깨의 성분개량 연구 III. 들깨 품종의 단백질 함량과 아미노산 조성)

  • Korean local collected strains of perilla were analyzed on their protein and amino-acid contents in the seed. The strains analyzed were 80 of which grown in the experimental field of Crop Experiment Station at Suwon in 1991. This experiment was performed to obtain the basic informations on the varietal differences of protein and amino-acid contents and further applied in the breeding project for quality improvement of perilla. The seed protein contents of 80 perilla varieties ranged from 17.9% to 24.6% and the average content was 24.6%. There were no differences in protein contents according to the maturity and 1,000 seed weight, but differed by the seed coat color showing 1.4% higher content in light gray than that of drak brown, which may suggest that the seed coat color could be used as a marker gene for breeding high protein varieties. While the total essential amino-acid contents of collected perilla ranged from 36.41% to 44.85%, the mean of 40.55%, the non-essential amino-acid contents was 47.23%~60.01% with the mean of 55.29%. Moreover, there was high positive correlation between the essential amino-acid contents and seed coat color. The essential amino-acids of perilla has an exceptionally high content of arginine, leucine, but methionine indeed being almost marginal. Of the essential amino-acids, lysine, methionine and valine were contained higher compared with the FAO recommendation level.

Effects of Amno Acids Supplemented to a Low- protein Broiler Diet (저단백질 브로일러 사료에 아미노산 첨가효과)

  • Three experiments were conducted to show effects of adding amino acids to a broiler starter diet (0-3 weeks of age) low in protein. A diet of 22% protein with adequate levels of all essential amino acids was used as a reference diet. Supplementing an 18% protein diet with adequate amounts or methionine and lysine significantly improved performance of chicks. However, adding excessive amounts of these amino acids was detrimental, indicating that the excess aggravated deficiency or the third amino acid. Supplemental arginine tended to alleviate this aggravating effect.
