  • Histological study of eel was carried out to determine the effect of drying on muscular tissue and fat movement. The results are as follows: (1) Changes of muscular tissue during drying may he divided into two stages early and late. In the early stage there was apparent dehydration of connective tissue while in the late stage muscle fibre dehydration appears to occur. (2) Movement of fat during drying depends upon division and contraction of the myocommata caused by inner structural changes. This phenomenon appeared to be more apparent in the late rather than the early drying stage.(3) After freeze-drying traces of ice crystals were detected around the muscle cells. These ice crystals remained even after rehydration. (4) When muscular tissue was heated the connective tissue melted while muscle fiber dehydrated and solidified.

Investigation of Factors Contributing to Childhood Soft-Tissue Injuries in the Face (소아의 안면 연부조직 손상에 관한 연구)

  • The purpose of this study was to retrospectively investigate the contributing factors in facial soft tissue injuries among children. A record-based analysis was carried out on 126 patients with facial lacerations who visited I hospital in the dept of plastic surgery from April 2010 to May 2011. Through all age group showed a significant difference in incidence rate for male and female.(p<.05). Overall, 49% of injuries occurred outdoors and the most frequent cause of injury was slip-down (42%), followed by being struck (40%). In general, the frequency of injuries peaked summer(30.2%) and increased between 12pm and 4pm. 105 patients (83.3%) arrived at the emergency room within an hour. The forehead(26%) was the most commonly injured area. The depth of soft tissue injuries was mostly subcutaneous(54%), and 1~2cm was the length of injury with the highest incidence(46%). Most children (40%) needed local anesthesia and only 4 children (1.53%) suffered associated fractures. Facial injuries require special attention because of their potential threat in inducing permanent scar or disfigurement. For the safety of children, the preventive measures to reduce facial soft tissue injuries must be improved.

Relative Biocompatibility Evaluation of Anodized Titanium Specimens in vivo and in vitro (In vivo와 in vitro 에서 양극산화 처리한 타이타늄 시편의 상대적인 생체적합성 평가)

  • To evaluate the biocompatibility of untreated and anodized titanium specimens, the specimens were implanted in the subcutaneous tissue of the abdominal region of female mice for two weeks. The reaction of connective tissue to specimens was histologically studied. The implants were encapsulated by fibrous connective. tissue consisting of fibroblast, fibrocyte and other cellss including neutroophil, macrophage and giant multinucleated cell. some newly formed blood vessels were located in the fibrous capsule surrounding the implant. Giant multinucleated cells were observed at the fibrous capsule adjacent to the implant. Kind of cell types and the thickness of fibrous capsules were examined quantitatively. Most of cell types located in the fibrous capsule were fibroblast and fibrocyte. The average thickness of fibrous capsules for the anodized specimens was much thinner than that of the untreated titanium specimen. Biocompatibility of titanium specimens were also studied by using cell culture method. The number of MG-63 cells was significantly increased on the anodized titanium specimens in vitro experiment. Our observations suggest that anodized titanium specimens are more effective for the improvement of biocompatibility in vivo and in vitro.

Preparation and Characterization of Ipriflavone-Loaded Poly(L-lactide-co-glycolide) Scaffold for Tissue Engineered Bone (조직공학적 골을 위한 애프리플라본을 함유한 다공성 지지체의 제조 및 그 특성)

  • Ipriflavone (IP), a non-hormonal isoflavone derivative, has been shown to interfere with bone remodeling by inhibiting bone resorption and stimulating bone formation. IP consistently increased the amount of Ca incorporated into the cell layer by mesenchymal stem cells (MSCs). In this study, we developed the novel IP loaded poly(L-lactide-co-glycolide) (PLGA) scaffolds for the possibility of the application of the tissue engineered bone. IP/PLGA scaffo1ds were prepared by solvent casting/salt leaching method and were characterized by porosimeter, scanning electron microscopy, determination of residual salt amount, differential scanning calorimetry, and X-ray diffractometer, respectively. IP/PLGA scaffolds were implanted into the back of athymic nude mouse to observe the effect of IP on the osteoinduction compared with control PLGA scaffo1ds. Thin sections were cut from paraffin embedded tissues and histological sections were stained H&E, von Kossa, and immunohistochemical staining for Type I collagen and osteocalcin. It can be observed that the porosity was above 91.7% and the pore size was above 101 $\mu\textrm{m}$. Control scaffo1d and IP/PLGA scaffo1ds of 50% IP were implanted on the back of athymic nude mouse to observe the effect of IP on the induction of cells proliferation for 9 weeks. The evidence of calcification, osteoblast, and osteoid from the undifferentiated stem cells in the subcutaneous sites and other soft connective tissue sites having a preponderance of stem cells has been observed. From these results, it seems that IP plays an important role for bone induction in IP/PLCA scaffolds.

Biodisc Tissue-Engineered Using PLGA/DBP Hybrid Scaffold (DBP/PLGA 하이브리드 담체를 이용한 조직공학적 바이오 디스크 개발)

  • Demineralized bone particle (DBP) has been used as one of the powerful inducers of bone and cartilage tissue specialization. In this study, we fabricated DBP/PLGA scaffold for tissue engineered disc regeneration. We manufactured dual-structured scaffold to compose inner cylinder and outer doughnut similar to nature disc tissue. The DBP/PLGA scaffold was characterized by porosity, wettability, and water uptake ability. We isolated and cultured nucleus pulposus (NP) and annulus fibrosus (AF) cells from rabbit intervertebral disc. We seeded NP cells into the inner core of the hybrid scaffold and AF cells into the outer portion of it. Cellular viability and proliferation were assayed by 3-(4,5-dimethylthiazole-2-yl) -2,5- diphenyltetrazolium -bromide (MTT) test. PLGA and PLGA/DBP scaffolds were implanted in subcutaneous of athymic nude mouse to observe the formation of disc-like tissue in vivo. And then we observed change of morphology and hematoxylin and eosin (H&E). Formation of disc-like tissue was better DBP/PLGA hybrid scaffold than control. Specially, we confirmed that scaffold impregnated 20 and 40% DBP affected to proliferation of disc cell and formation of disc-like tissue.

Congenital Dropsy in Korean-native Calf (한우 송아지의 선천성 전신수종)

  • A 4-year old Korean-native cow of third parturition delivered an deformed calf with congenital anasarca and hypoplasia of the extremity at Muan County in Chonnam Province. At parturition, the dam showed amniotic fluid approximately two times more than that at the normal and also occurred severe dystocia. The anomaly died before parturition was 82 cm long and weighed 25 kg. It was difficult to recognize the neck and face of the anomaly due to severe edema and the stretched tongue through mouth was found. The shape of the extremity was recognizable and the systemic hypoplasia of the body with marked edema was found. At necropsy, there was a large amount of serous fluid both in pleural and peritoneal cavity. Also the fluid was contained some of yellowish granular materials and diffusely was fulfilled in all subcutaneous tissue. There was a partial opening in diaphragm which was resulted in the protrusion of approximately half of liver toward peritoneal cavity. There were two kidneys (15${\times}$21 and 13${\times}$18 cm) on the left which were enlarged and relatively larger than the right one (13${\times}$9 cm). In addition, there were found slight to moderate hepatomegaly and splenomegaly with slight congestion, and diffuse edema of intestinal serosa. Lung (32${\times}$49 mm) with marked hypoplasia was smaller than the size of heart (56${\times}$45mm). Serological test to the dam showed relatively high antibody titer (>64) to Akabane virus which might be involved in the outbreak of the deformed Korean-native calf as one of many causative agents.

Detection of Radial Pulse Wave by Photoplethysmogram (광전용적맥파를 이용한 요골동맥 맥파 검출)

  • Photoplethysmogram is widely used to measure heart rate and arterial blood oxygen saturation in human. This paper describes radial pulse waves recorded by photoplethysmogram with 940 nm wavelength infra red light. Radial pulse waves were varied according to the recording site. When recorded on the skin over radial artery, the radial pulse wave was inverted, comparing to the photoplethysmogram at fingertip. The mechanism of inverted pulse wave seemed to be caused by the change of the blood volume in the subcutaneous tissue between radial artery and the skin, which was reduced during systolic period and increased during diastolic period of the cardiac cycle. These results suggest that radial arterial wall may reflect infra red ray.

  • Kimura's disease is a mass producing uncommon chronic inflammatory process of unknown cause. It is more common among Orientals and affects particularly the young male. Sites of predilection include the head and neck regions, primarily the subcutaneous tissue and dermis. It's clinical course is benign in nature. The treatment modalities for this disease are surgical excision, steroid therapy and radiation therapy. We experienced a case of soft tissue mass in the right upper arm. It was painless and relatively movable. We performed marginal excision of the mass, which was turned out to be Kimura's disease on microscopic examination. In this case, disease recurrence was not found fifteen months after the operation.

Accidental Extrusion of Sodium Hypochlorite during Endodontic Treatment in a Primary Tooth (유치의 근관 치료 중 차아염소산나트륨의 치근단 유입으로 인한 합병증)

  • Although sodium hypochlorite is the most frequently used canal irrigant during endodontic treatment, its complications are not as well recognized as its effectiveness. This report demonstrates that sodium hypochlorite extrusion during endodontic treatment can cause severe complications. A 5-year-old boy experienced immediate pain and swelling, ecchymosis in surrounding tissues, and profuse bleeding from the root canal during endodontic treatment, because of accidental extrusion of sodium hypochlorite. The patient was hospitalized, and analgesics and antibiotics were prescribed. Accidental extrusion of the irrigating solution occurs more frequently in teeth with immature apices, root resorption, and apical perforations; therefore, caution is needed. When such complications occur, proper management and medications are needed.

취장 외분비기능 부전에서 합성 단백 분해효소 억제 물질에 대한 취조직 재생 및 기능 변동 연구

  • 취장 외분비 기능 부전은 임상적으로 영양결핍, 발육부전, 지방변동을 유발하나 이의 치료로는 결려된 소화효소 보충 등 보존적 요법만 시행될 뿐 근본적인 치료법은 없는 실정이다. 지난해 과제는 합성 단백분해 억제물질인 Camostat이 취장의 비대와 중식을 일으키며 또한 단백분해 효소분비를 증가시킴을 보고한 바 있다. 이를 토대로 최근 보고된 실험적 취장 외분비기능 부전 모델을 이용하여 camostat의 효과를 검색하고자 하였으며 아울러 이를 CCK효과와 비교하였다. 실험동물로는 몸무게 200 g 안팎의 수컷 Sprague-Dawley계 흰쥐를 사용하였으며 취장기능 부전은 oleic acid (25 $\mu$/100 g bw)를 취관내 주입하여 유발하였다. Camostat은 200 mg/kg씩 위내 투여(i.g.)하였으며 CCK(CCK-8)는 5 $\mu\textrm{g}$/kg씩 하루 2회 피하주사하였고 투여기간은 각각 3, 7, 14 일간으로 하였다. 각 약물 투여 후 취장 외분비 기능과 조직학적 검색을 실시하여 다음과 같온 결과를 얻었다. 1. Oleic acid의 취관내 주입으로 흰쥐의 취장 무게, 조직내 효소단백 함량 및 효소분비량이 현저히 감소되었고 조직학적으로 심한 위축과 섬유화를 관찰할 수 있었으며 이는 주입 후 기간이 지남에 따라 계속 진행하였다. 2. 취장기능 부전 유발 흰쥐에서 camostat 처치로 조직내 효소단백 함량 및 효소분비가 증가 되었으며 이는 14일간 처치군에서 뚜렷하였다. 3. 취장 기능부전 유발 횐쥐에서 camostat 처치는 조직학적으로 기능적인 외분비 조직이 유지되었으며 이는 3일군에서 특히 뚜fut하였다. 4. 취장기능부전 유발 횐쥐에서 CCK 처치효과는 camostat 처치효과와 비슷하였다. 이상의 결과로 보아 oleic acid 주입은 취장기능부전 연구에 유용한 실험모델로 생각되며 합성 단백 분해 효소 억제제인 Camostat은 취장외분비 기능 부전의 진행을 억제하고 어느정도 그 기능을 호전시킬 수 있으며 이는 CCK유리에 기인한다고 생각한다.
