작업환경을 위한 TLV의 근거 - PERFLUOROBUTYL ETHYLENE(1)

  • Perfluorobutyl ethylene(PFBE)은 유기용제 혼합물에 주로 사용되는 산업화학물질로 동물에게 경구, 피부, 눈 그리고 흡입 노출되는 경우 급성 독성이 매우 낮다. Perfluorobutyl ethylene을 흰쥐에게 하루 6시간, 일주일에 5일씩 2주간을 0, 500 ppm, 5,000 ppm, 50,000 ppm을 흡입 노출시킨 연구에서 최대무작용량(no-observed-adverse effect level, NOAEL)으로 500 ppm이 결정되었으며 5,000 pm에서는 단핵 백혈구가 약간 증가하였다. 50,000 ppm에서도 병리학적 변화는 없었다. 그 후의 흡입연구는 하루 6시간, 28일간 연속적으로 흰쥐 암수에게 400 ppm, 2,000 pm, 10,000 ppm의 농도로 perfluorobutyl ethylene 증기를 노출시켰다. 최종 노출 후 흰쥐를 희생시켜 관찰한 결과, 체내 기관 무게변화를 포함한 어떠한 변화도 관찰되지 않았다. 그러나 10,000 ppm에 노출된 수컷 흰쥐 5마리 중 4마리는 작은 다중간세포질 액포가 발견되었다. Perfluorobutyl ethylene 증기 10,000 ppm에 노출된 수컷 흰쥐는 부분적으로 평균 혈액응고 촉진 시간이 감소하였다. 이 연구에서는 최대무작용량을 2,000 ppm으로 결정하였다. Perfluorobutyl ethylene의 TLV-TWA 100 ppm은 노출 후 악영향을 최소화하는데 충분한 농도이며 "피부흡수(Skin)", "감작제(SEN)" 그리고 발암성에 대한 경고주석을 설정하기에는 유용한 연구가 충분하지 않다.

  • Toxic epidermal necrolysis (TEN) is a rare, acute and life-threatening cutaneous drug reaction. TEN is characterized by the sudden onset of extensive necrosis in the epidermis and frequent mucous membrane involvement. The pathogenesis has not yet been elucidated. In addition, no particular treatment for TEN has been established. We report a case of TEN in a 14-year-old-boy, which might have been caused by steroids with enalapril treatment for membranous nephropathy. He recovered after intravenous immunoglobulin therapy.

  • Lymph node tissues of BALB/C mouse treated with DNCB were immunohistochemically observed to investigate the activation of cell mediated immunity in lymph node of murine with allergic contact dermatitis. The inguinal region of BALB/C mice were sensitized by one application of $25{\mu}l$ of 5% 2,4-dinitrochlorobenzene(DNCB) onto an abdominal skin and 2 weeks later, the mice were challenged with $4{\mu}l$ of 2.5% DNCB. The inguinal lymph node were obtained at hour 24, 48, and 72 after 2nd DNCB treatment and embedded with paraffin, and then stained by following ABC method that used monoclonal antibody including L3T4(CD4), Ly2(CD8), IL-2R(CD25). The distribution of helper T lymphocytes, cytotoxic T lymphocytes and IL-2 receptors began to increase at hour 24 after after 2nd DNCB treatment and these increase appeared in paracortical area and medullary sinius. These increase were greatest at hour 48. These results indicated that the IL-2 secretion began to increase by activation of helper T lymphocytes in lymph node of DNCB re-exposure area and subsequently to activate suppress T lymphocytes.

Anti-Wrinkle Effect of Schizandra chinensis Baillon Fermented with Lactobacillus plantarum (오미자 유산균 발효물의 주름개선 효과)

  • To identify new active anti-wrinkle ingredients, this study investigated the anti-wrinkle effects of Schizandra chinensis Baillon fermented with Lactobacillus plantarum (SCF) by assessment of cytotoxicity of human dermal fibroblast, collagen biosynthesis, matrix metalloproteinase-I (MMP-1) inhibition and elastase inhibition. S. chinensis was fermented with L. rhamnosus for 1 day at $37^{\circ}C$. The cytotoxicity of SCF was evaluated by a cytopathic effect reduction method. Effects on collagen biosynthesis and matrix metalloproteinase-I (MMP-1) of SCF were evaluated by previous reported method using procollagen type-IC peptide EIA kit and Matrix Metalloproteinase-1 Biotrack activity Assay Kit, respectively. Elastase inhibition assay was conducted by reaction of enzyme and substrate using N-Suc-$(Ala)_3$-nitroanilide as the substrate. As the results, SCF didn't show cytotoxicity against human dermal fibroblast at concentration of $100{\mu}g/mL$. Also, SCF was increased collagen synthesis and showed inhibitory effect of MMP-1 (p < 0.05). In the elastase inhibition assay, the $IC_{50}$ of SCF was $36.4{\mu}g/mL$. Therefore, our results indicated that SCF possesses anti-wrinkle effects and can be used practically for anti-wrinkle care of skin.

Four-Week Topical Toxicity Studies of SDK in Beagle Dogs and Sprague-Dawley Rats (비글개와 랫드에서 SDK시제품(가칭)의 4주간 피부도포 반복투여 독성시험에 관한 연구)

  • SDK (skin decontamination kit) is new skin decontaminant which is developed by ADD (Agency for defence development). In this study, four-week toxicity of SDK was investigated using beagle dogs and Sprague-Dawley rats. The beagle dogs and Sprague-Dawley rats were dressed topically seven days per week for 28 days, with dosage of 0, 0.25, 0.8 and 1 g/kg/day. respectively. Animals treated with SDK did not cause any death and show any clinical signs. They did not show any significant changes of body weight, feed uptake and water consumption. They were not significantly different from the control group in urinalysis, ocular examination and histopathological examination. In hematological and serum biochemical assay, there were no-dose-defendent changes. Therefore, SDK was not indicated to have any toxic effect in the beagle dogs and Sprague-Dawley rats when it was dressed topically below the dosage 1 g/kg/day for four weeks.

Comparison of Eye Irritation Potency with Skin Irritation and Cytotoxicity Potency of Anti-wrinkle Agents (주름방지용 화장품원료의 안점막 자극성과 피부자극성 및 세포독성과의 비교)

  • In the present study, we examined eye irritation oj six anti-wrinkle agents (ascorbic acid, glycolic acid, all trans-retinoic acid, ginseng extract, retinol, EB). We also compared eye irritation with skin irritation and cytotoxicity in HaCaT cells by these agents. The highest eye irritation was found in glycolic acid, but all trans-retinoic acid showed the highest skin irritation. The rank of eye irritation was not correlated with the cytotoxicity of agents. This result shows that eye irritation potency by these agents were not correlated with skin irritation potency, and cytotoxicity in HaCaT cells.

  • 본 논문은 피부미백에 관한 실험적 연구로서, 봉선화(Impatiens balsamina Linne)의 성분 중에서 하이드록시-나프타-퀴논(hydroxynaphthoquinone)이 멜라닌 생성억제 효능, 색소침착 저해 효과 등의 미백 활성이 있는 것으로 밝혀졌다. 피부흑화(melanogenesis)는 멜라닌 색소생성세포(melanocyte)에서 자외선 등의 자극에 대한 방어기작으로 멜라닌 생성 활동이 증가되고 이로 말미암아 만들어진 다량의 멜라닌이 각질형성세포(keratinocyte)로 전이되어 피부 표피층에 축척된 결과이다. 비록 멜라닌이 피부에 보호작용을 하나 피부의 과색소 침착은 기미, 주근깨, 피부염증 후의 피부흑화, 노인성 색소반점 등을 일으키며 이로 인해 피부미용 상의 불편뿐만 아니라 정신적으로 부정적인 영향을 미쳐 사회활동에 불편을 초래하기도 한다. 멜라닌 생성 과정은 아미노산의 일종인 티로신(tyrosine)에 티로시나제(tyrosinase)라는 효소가 작용하여 도파(DOPA), 도파퀴논(dopaquinone)으로 바뀐 후 비효소적인 산화 반응을 거쳐 만들어 지며, 이것이 피부 내에 이상 침착하여 기미, 검버섯 등이 생기는 것이라고 알려져 있다. 이와 같은 색소침착, 기미, 반점 등의 완화, 예방 및 치료에는 멜라닌 생성을 억제하는 물질, 예를 들면 하이드로퀴논(hydroquinone), 알부틴, 비타민 C 및 그 유도체 등이 개발되어 사용되고 있으나 이 중, 하이드로퀴논은 일단 효과가 인정되고 있지만 감작성이 있기 때문에 일반적으로 사용이 제한되고 있다. 아스콜빈산은 쉽게 산화되어, 이를 배합한 제품에는 변색, 변취가 되는 문제를 야기하고, 식물추출물 유래의 물질들은 식물의 산지에 따라 효능의 차이가 심하여 제품의 균질성이 유지되기 어렵다. 이러한 이유로 많은 멜라닌 생성을 억제하는 물질을 개발하기 위한 연구들이 계속 진행하고 있다. 2-히드록시-[1,4]나프토퀴논 유도체는 다음과 같은 구조를 나타낸다. 상기 구조에서 R은 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬로서, 바람직하게는 C1 내지 C10의 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬이고, 보다 바람직하게는 C1 내지 C5의 포화 혹은 불포화된 직쇄 또는 분지쇄의 알킬이다. 실험에 활용된 화합물은 다음과 같은 반응식으로 얻어졌다. 2-히드록시-[1,4]나프토퀴논과 포타슘카보네이트(K2CO3)를 아세토니트릴에 용해한 후, 저온수조로 냉각하고 알킬할라이드를 적당한 당량비로 천천히 적하하여 히드록시기와 에테르(ether) 반응을 통하여 얻어질 수 있다. 상기 반응식에서 R은 C1~10의 포화 또는 불포화된 알킬기로서 직쇄형 또는 분지쇄형 모두 가능하다. 상기 화합물인 2-히드록시-[1,4]나프토퀴논(2-hydroxynaphthoquinone)은 배양된 쥐의 멜라노마 세포에 대하여 하이드로퀴논과 대등한 멜라닌 생성 억제효과를 보였다. 하이드로퀴논은 저농도에서 강력한 멜라닌 생성 억제효과를 보이지만 상기 화합물들은 보다 낮은 농도에서도 세포독성을 나타내지 않으며 하이드로퀴논보다 높은 멜라닌 생성 억제 효과를 보였다.

Effects of Ehwa Makgeolli Containing Oriental Herbs on Skin Whitening and Wrinkles (한방 이화주의 미백 및 피부 주름 개선 효과)

  • Extrinsic skin aging is characterized by the loss of skin tone and resilience, irregular pigmentation, and deep wrinkles. The aim of this study was to investigate the effects of Ehwa Makgeolli containing oriental herbs (Glycyrrhiza uralensis Fisch., Lycium chinense MILL., Morus alba L., and Saururus chinensis Baill) on skin whitening and wrinkling in human skin cells. We prepared Makgeolli extracts (HEE) with 70% ethanol. HEE significantly inhibited in vitro mushroom tyrosinase activity and reduced the cellular and secreted melanin content of mouse melanoma melanocytes (B16F1 cells). HEE down-regulated the protein expression of tyrosinase related protein (TRP)-1/-2, a key player in melanogenesis. Treatment with HEE in human keratinoctyes (HaCaT cells) inhibited the proteolytic activities of matrix metalloproteinase (MMP)-2/-9 in a dose-dependent manner and dramatically reduced the expression of MMP-2/-9. In addition, HEE attenuated lipopolysaccharide (LPS)-induced nitric oxide production in murine macrophages (RAW264.7 cells). These results indicate that HEE may be a great cosmeceutical ingredient for its whitening, anti-wrinkle, and anti-inflammatory effects.

Effects of Molecular Weights of Sodium Hyaluronate on the Collagen Synthesis, Anti-inflammation and Transdermal Absorption (히알루론산나트륨의 분자량 크기에 따른 Collagen 합성, 항염증 및 피부 흡수에 미치는 영향)

  • In this study, we examined the effects of various molecular weights (1, 10, 50, 100, 660, and 1500 kDa) of sodium hyaluronate (HA), which were prepared by enzyme hydrolysis, on the collagen synthesis, anti-inflammation and skin absorption. These HA did not significantly affect the viability of human dermal fibroblast Hs68 cells. Among them, 1500 kDa, 50 kDa HA most significantly increased collagen production by 59%, and 50% in the Hs 68 cells, respectively. Whereas 1500 and 660 kDa HA hardly pass through mouse transdermis membrane, lower molecular weights (1, 10, or 50 kDa) of HA showed time-dependent increase in skin permeation. HA of 50 kDa showed highest anti-inflammatory effects by reducing nitric oxide and tumor necrosis factor-${alpha}$ production in the RAW 264.7 cells, comparing to other HA (1, 10, and 100 kDa HA). Recently, there is no report about anti-wrinkle and anti-inflammatory effects and skin permeation of different molecular weights HA (1, 10, 50, 100, 660 and 1500 kDa), which were produced by enzyme hydrolysis. These results suggested that 50 kDa HA can be potent candidates for the development of effective anti-aging and anti-wrinkle cosmetic agents. The results of this study demonstrated that among those HA with different molecular weights, 50 kDa HA showed highest anti-inflammatory activity, significant capability to induce collagen synthesis and high level of skin permeation.

A Study on Animal Skin Irritation Measurement of Ozoneized Olive Oil for Cosmetic Ingredients (화장품원료를 위한 오존화 올리브오일의 동물 피부자극 측정에 관한 연구)

  • This study has attempted to use ozone for the treatment of skin diseases as research results that ozonated olive oil has an excellent therapeutic effect on skin diseases are known. However, there is hardly any accurate data in Korea. Usually, animal tests related to cosmetics are not performed, but toxicity tests were conducted because they were absolutely necessary. In general, there are not many cases of measuring actual data through animal tests for the purpose of confirming the performance of cosmetics, but in the case of toxicity tests, it is recommended to accurately measure skin reactions, so this experiment was conducted. In this experiment, in order to evaluate the skin irritation of ozonated oil (high concentration) on the rabbit, the test substance was applied to the back of the rabbit for 24 hours, and then mortality, general symptoms and skin irritation were evaluated. Experimental Results As a result of evaluating the treatment site of the test substance after a certain period of time, no skin irritation was observed in all animals.
