Studies on the Preparation of Food Proteins from Castor Bean Protein (피마자 단백질의 식품화를 위한 연구)

  • Detoxified and deallergenized castor bean protein isolate was prepared from defatted castor bean pomace for use in animal feedstuffs and human foods. Succinylation and acetylation of the ${\varepsilon}-amino$ groups of the protein improved markedly the water solubility of the protein at $pH\;7{\sim}8$. The results of the amino acid analysis of the protein isolate revealed that the sulfur-containing amino acids and L-lysine were limiting amino acids and that succinylation and acetylation caused some little loss of the amino acid content. The L-methionine enriched plastein was synthesized from the protein isolate or the acylated protein isolates and DL-methionine ethyl ester by one step process with papain. By this method the extent of incorporation of L-methionine was about 50%. Pepsin hydrolyzed both unmodified and modified protein isolates at the same rate (about 92%). Tryptic hydrolysis, however, was less for the succinylated protein isolates (about 42%) and less for the acetylated protein isolates (about 26%). The protein efficiency ratio of L-methionine enriched protein isolate (about 2.5 weight %) was 90% that of reference casein. The protein efficiency ratio values of succinylated (88%) and acetylated (84%) protein isolate were 55 and 69% of reference casein, respectively.

Antimicrobial and Antioxidant Activity of Hot Water and Ethanol Extracts of Ricinus communis L. Leaves and Fruits (피마자(Ricinus communis L.) 잎과 열매의 열수(熱水) 및 에탄올 추출물의 항균 활성과 항산화 효능)

  • RFW and RFE of castor fruit selected as part of the development of natural antimicrobials and antioxidants yielded 15.8% and 18.4% respectively. In the results of measuring the antimicrobial activity through paper disc method, the antimicrobial activity of castor fruits in ethanol extracts appeared. Especially, the activity was excellent in P. aeruginosa and S. aureus, and antimicrobial activity of C. was 1.5mm up to 16 hours. However, the proliferation of C. was observed again after 24 hours. In the MIC experiment results of RFE, S. aureus and P. aeruginosa showed 96% and 93% of antimicrobial activity, respectively. The DPPH radical scavenging activity of RLW and castor leaf ethanol extract showed $1.8{\pm}0.6%$ and $2.1{\pm}0.7%$ free radical scavenging activity at $1000({\mu}g/m{\ell})$. This study is expected to provide basic data for the development of antimicrobial agents and antioxidants using natural products.

Effect of Soil Salinity and Flooding on Plant Growth and Yield of Rape-Castor Bean Cropping System in the Newly Reclaimed Tidal Land of Western Seaside of Korea (서해안 신간척지에서 유채-피마자 작부체계시 토양염농도 및 침수가 생육 및 수량에 미치는 영향)

  • Double cropping system of oilseed crops introduced rape and castor bean was studied in the newly reclaimed tidal land of Korea. Surface soil EC to reach at 50% of growth reduction to the tallest height of crops was estimated $4dS\;m^{-1}$ for castor bean and $6dS\;m^{-1}$ for rape by logarithmic function. The castor bean cultivated during rainy summer much more suffered serious growth and yield reduction by flooding damage in the Hwaong and Yeonsangang reclaimed lands having low soil conductivity and finer textured soil than in the Iweon reclaimed land having higher soil conductivity and coarse sandy textured soil. Rape cultivated during dry winter-spring much more suffered serious growth and yield reduction by high soil salinity come from re-salting process. 50% yield reduction was estimated at $2.0\;2.5dS\;m^{-1}$ of surface soil EC by logarithmic function. Consequentially, it was concluded that oilseed production for energy by double cropping system of rape and castor bean might be possible under good controling soil salinity below $4dS\;m^{-1}$ for castor and $3dS\;m^{-1}$ for rape in the newly reclaimed land of Korea.

  • To treatment of Ricinus cotyledons with ABA, induced several proteins with molecular weights of 53, 54, 56, 58 and 73.5kD. 54 and 56kD among those proteins resulted in increased more when ABA concentrations in external media are increased. The molecular weight of 35, 49, 53, 54, 62, 65 and 79kD of proteins are induced by ABA treatment of roots. The induced proteins are not the same a those by cold treatment exception of 73.5kD of cotyledons and 62kD of roots. 49, 58 and 79kD of proteins are important to research in future because of the induction of proteins in the presence of cydoheximide(CH) which is blocked the synthesis of proteins.

Growth Characteristics of Castor Bean (Ricinus communis L.) Collections (피마자 수집종의 생육특성)

  • This study was carried out to evaluate ecological growth characteristics of forty castor bean collections for biodiesel utilization. The emergence date and it's rate of collections was 15-16 days and 82.5%, respectively. It took 63-68 days from emergence to flowering. Stem length was ranged from 173 to 251 cm with an average 211.8 cm. The average stem diameter was 24.2 cm, and showed the high occurrence as 52.5% at the 20.4-24.9 cm. The node number was widely distributed as 13-22 and the average node number was 18.9. The seed numbers were widely distributed as 7-33ea, and the average liter weight of seed was 502.9 g of which minimum weight was 432.3 g and maximum weight was 572.5 g, and the gap between maximum and minimum was 140.2 g. It showed high difference. The 100 grain weight was distributed as 20.3 g-38.3 g which showed high differences between small grain and large grain collections. The seed numbers per plant were varied as 72 at the minimum and 1,011 at the maximum number, and average seed number was 465.2. The total yields of seed per plant were varied as 20 g-266.2 g, and the average yields were 128.1 g.

  • This paper is the tenth contribution towards taxonomic studies on Cercospora and allied genera, and contains ten species of Korean cercosporoid fungi; viz., Cercospora coreopsidis, C. hydrangeae, C. physalidis, C. ricinella, C. volkameriae, Pseudocercospora abelmoschi, P. cruenta, P. varia, Pseudocercosporella paridicola, and Ramularia grevilleana var. grevilleana. Morphological characteristics of taxonomic value are described and illustrated for these species to contribute towards a mycological monograph of Korean cercosporoid fungi.

Studies of Eri-Silk Cultring in Korea (한국피마잠사개발에 대한 연구)

  • Eri-silkworm is known as a tropical insect where as poly-voltine type in that area. It eats caster oil plant leaves which are cultivated as an every year cultivatable seed oil use in this country, even though it grows for many years in tropical countries. That is why, farmers have freedom for its cultivation in any year if they want. Therefore, eri-silkworm rearing service is flexible for its diet procurment as wish of farmer. The eri-cocoon price or economical fluctuation may be reactable for the rearing work not like as mulberry cocoon. Fortunately, it also eats cynthia tree leaves. Standing from such a easy condition, the authors have studied about this problem since 1963 to develope a culturing method of eri-silkworm rearing in this country and the authors brought out the matters to be produced as an industry scale. Here, the authors summarized their works of the results covering with thirty three work tables. The obtained results are as follows.

  • Fatty acids in seed oil from plants are essential for human nutrients and have been used for industrial purpose. The growing demands of seed oil as food resources and feedstocks for industrial uses have attempted to modify fatty acid composition and to increase oil content in transgenic plants. However, production of unusual fatty acids in transgenic plants are limited, which is not synthesized the level same as original plants. This bottleneck was common for production of several unusual fatty acids in transgenic plants and suggests that there is different for substrate preference in oil metabolic pathway enzymes between host oil plants and original wild plants. Review of acyltransferases involved in acyl-editing and seed oil accumulation of oil plant and wild-plant producing unusual fatty acids will design strategies to maximize the production of unusual fatty acids in transgenic plants. In here, we identified eleven acyltransferase genes in castor based on sequence homology, which will be useful to increase hydroxy unusual fatty acids in transgenic plants.

Antioxidaitve and Antibacterial Activities of Endemic Plants Extracts in Korea (국내 자생 식물 추출물의 항산화 활성 및 항균효과)

  • Antioxidative and antibacterial activities of endemic plants extracts in Korea were investigated. Hydrogen radical scavenging activity was 99.72% in an elm tree, 99.725 in a Job's tears, 99.575 in an eggplant, and 94.025 in a barrenwort. Antibacterial activity of ethanol extract from 23 different species of wild plants were determined. The gull nut was showed the strongest antibacterial activities (16.0-19.0 mm) and also showed high antibacterial activities in a pine neddles, a gulmyungja, a wild ginger plants, a ginko (9.5-11.5 mm). Only extracts of Saurrranceae extract showed the antibacterial activity in Bacillus subtilus, Stapylococcus aureus, Escherichia coli, Samonella entetotidis. An antibacterial activities were showed weakly in an esungcho and an eggplant. A ground ivy showed Antibacterial activity on Stapylococcus aureus. A barren wort showed in antibacterial activity on Stapylococcus aureus, and E. coli. This results were suggested that many edemic plants resources contains antioxidative and antibacterial substances.

  • The characteristics of enzymatic methanolysis of castor oil were investigated as a clean technology. Among 16 lipases tested in this study, Novozym 435 showed the highest activity in methanolysis. Solvents had different effects on the methanolysis of castor oil according to weight percent (wt%) of Novozym 435. Heptane showed best activity with 1 wt% of Novozym 435, while isopropyl ether gave the best yield of ricinoleic acid methyl ester with 0.5 wt% of that. Ricinoleic acid methyl ester was obtained in 86% of yield through the methanolysis of castor oil catalyzed by Novozym 435 (1.0 wt%) during 24hr.
