Hemagglutinative Activity of Lectin Isolated from Shiitake, Lentinula edodes (표고버섯에서 분리한 렉틴의 적혈구 응집활성)

  • Since lectin condense more easily cancer cell than normal cell, it has been investigated very actively. Recently, a lot of researchers gave attention to lectin in natural products especially because lectin has effects on T-cell activation and anticancer activity, and specificity on polysaccharide. The specificity is useful to confirm kind of polysaccharide of the cell surface and to study the polysaccharide. In this research, we purified lectin from shiitake, Lentinula edodes, and then characterized it. The molecular weight of the lectin was 23 kDa, and it was stable only under the $40^{\circ}C$ and in a alkaline solution. As for the specificity of polysaccharide, the lectin had specificity on galactose, fucose, glucose, lactose and N-acetyl-D-galactosamine. In addition, it was confirmed to be a glycoprotein.

Effect of Mushrooms on the Growth of Intestinal Lactic Acid Bacteria (버섯의 장내 유산균 증식 효과)

  • The objective of this study was to evaluate the effect of mushrooms on the growth of intestinal lactic acid bacteria. Bifidobacterium breve and the total intestinal flora of human and rats were inoculated in the general anaerobic medium which contained each mushroom water extract. Except Pleurotus ostreatus and Flammulina velutipes, the mushroom extracts induced the growth of lactic acid bacteria by decreasing pH of the broth. The pH decreasing effect was excellent especially with Lentinus edodes, Agarocus bisporus and Coriorus versicolor. This effect was due to the increase in the number of Bifidobacterium in the intestinal bacterid. This growth of lactic acid bacteria effectively inhibited the bacterial enzymes, ${\beta}-glucosidase,\;{\beta}-glucuronidase$ and tryptophanase, of intestinal bactetria.

An investigation of insect pest on the Mushroom (Lentinus edode, Pleurotus ostreatus) in south region of Korea.) (한국남부 표고버섯 및 느타리버섯 재배지에 분포된 해충상에 관한 연구)

  • As the pest of the Letinus edoeds, five insect species and six animal species are investigated injuring the fresh mushroom. Of these, insect and animals, Achorutes armatus, Scaphidium amurense and Philomycus confusa caused severe damage to mushroom fruitbody, and a greate deal of damage was done to mushroom lamella by mites (Rhizoglyphis sp., and Histiostoma sp.). As the pest of Lentinus edodes bed logs, two insect species from immature bed logs, eight insects and one isopod species from matured bed logs were investigated. Of those, Moechotypa diphysis was found to be the most injurious species from both immature and matured bed logs. As the pest of Pleurotus ostreatus, six insects and the two mite species were investigated from sporophore. Of those, Lycoriella sp. and Mycophilla sp. are considered to be very serious. while two insects and five mite species were investigated from the mushroom bed. Of those, one insect (Lycoriella sp.) and two mites (Histiostoma feroniarum and Throglyphus bankslogior) found to be abundant and caused serious damage.

The Study of Anti-mutagenic Activity of Various Additive Materials on Pot-stew (부재료 첨가에 따른 찌개의 항변이원성 검색)

  • This study was performed to determine the mutagenicity of pork and ham pot-stew and the anti-mutagenicity of various additive materials on pot-stew by the Ames test using Salmonella typhimurium TA100. Boiled kimchi didn't show mutagenicity and effectively inhibited the mutagenicity induced by 4-NQO and Trp-p-1. But boiled pork and ham showed mutagenicity dose-responsively and pork's mutagenicity was higher than that of ham. On the mutagenicity of boiled pork and ham, the inhibition of kimchi was most effective and when scallion and galic was added with mushroom showed synergic effect. Boiled ham made in USA did not show mutagenicity different from ham made in Korea because of the addtion of ascorbic acid and when mutagen was added it's mutagenicity was lower than that of ham made in Korea.

Oak Mushroom Cultivation on Larch(Larix leptolepis Gorden) sawdust and its economic efficiency (낙엽송 톱밥을 이용(利用)한 표고버섯 재배(栽培)와 경제성)

  • To enhance the utilization of Larch(Larix leptolepis Gorden) wood, the sawdusts were tested as cultivating media for Lentinus edodes(Mori 290) by varying the composition of the media. The economic efficiency for producing mushrooms from larch sawdusts was evaluated. The results are as follows. 1. Larch sawdust was as much efficient as oak sawdust in the growth of the mycelia and production of good quality of oak mushrooms. 2. The mushrooms were able to be harvested starting six months after beginning the cultivation. No differences were found among the qualities of the mushrooms produced by various treatments. 3. The contents of lignin(28.6-28.7%) in the fresh sawdust of larchwood are higher than those in the residues of the used media(18.2%). But lignin content of 1% NaOH extracts(30.4-34.4%) in fresh sawdust of larch was lower than those in the residues(68.0%). 4. The production of mushroom was relatively high to be 29.7-31.5%(760-780g.)/2.5kg pack) of mushroom weight. The net profit estimated was 1,475Won per pack.

Thiamin Requirements for Vegetative Growth and Fruit Body Formation of Lentinula edodes

  • The effects of thiamin on vegetative mycelial growth and fruit body formation of Lentinuia edodes were investigated in basal peptone-glucose liquid medium in relation to the uptake of thiamin. Thiamin was essential for fruit body formation, and the minimum requirements for thiamin were estimated to be approximately 10 ${\mu}g$/L. The vegetative mycelial growth was little influenced by the addition of thiamin in the range of 1.5 ${\mu}g$~1.5 mg/L. While the mycelium was successively transferred to fresh peptone-glucose-agar medium three times, the repression of mycelial growth was not significant. Even in cases using vitamin-free casamino acid or glutamic acid as a nitrogen source instead of peptone, a thiamin deficiency for mycelial growth did not occur as a result of transferring the mycelia to fresh media. Almost all of the thiamin contained in the media accumulated in the mycelia during the first 3 weeks of a 9-week incubation. These results suggest that only trace amounts of thiamin are required for vegetative mycelial growth in Lentinula edodes and that almost all thiamin added to a basal medium will be used for fruit body formation.

Cultural characteristics and formation of fruiting body in Lentinula edodes (표고버섯의 배양적 특성 및 자실체 형성에 관한 연구)

  • Lentinula edodes is known by oak mushroom. It has been favored as delicious and nutritious food and the low-calorie food with a high nutritional value. It is also functional food since it contains a material well-known for its medicinal benefits. Since the growth and quality of oak mushrooms are sensitively affected by environmental conditions, an adequate environmental control is very essential to improve the yield and quality under protected cultivation. The main objectives of the study were to investigate cultural characteristics of mycelial growth and in vitro fruiting of Lentinula edodes. The optimum culture media for mycelial growth of L. edodes were PDA and MYA. Similarly, optimum temperature was $25^{\circ}C$. Malt extract(2%) and yeast extract(0.2%) were optimum carbon and nitrogen sources. Optimal culture period was 110~120 days in sawdust medium. Mycelial growth in medium(61 mm/7 days) Quercus mongolica extract the most good. Among different five log types, highest mycelial growth and fruiting productivity were observed in Quercus variabilis sawdust(20.9%).

Profit Analysis in Using Small Diameter Log of Quercus species (참나무류 소경재 활용용도에 따른 수익성 분석)

  • This study aims to compare the profitability of using small diameter logs of Quercus species in terms of net profit (ratio), net income (ratio). The small logs of Quercus species could be used for the bed logs for shiitake mushroom and Phellinus linteus, sawdust, charcoal, and wood chip. The data of cost and revenue are collected by a face-to-face survey method from the associated factories. Our results show that the profitability for Phellinus linteus production is highest because its price has been formed high due to low market competition mainly resulted from capital service cost and production technique requirement. The next is followed by Shiitake mushroom, sawdust, wood chip, and charcoal. The study also reveals that profitability for long returning periods of investment is higher than that of short returning periods.
