시클로덱스트린과 아스피린의 포접화합물의 구조와 가수분해에 관한 연구 (Structure and Hydrolysis Study of Inclusion Complex of Cyclodextrin and Aspirin)

  • Specific molecular recognition of cyclodextrin and aspirin was determined. A stable 1:1 inclusion complex was established in solution. The distinct structure of inclusion complex was elucidated by FT-IR, FAB-MS, UV, 1H NMR, and 13C NMR spectroscopy. Based on the 1H NMR data, a time-averaged conformation of $\alpha$-cyclodextrin exhibited significant catalytic activity toward the hydrolysis of aspirin in alkaline solution.

  • The structural specificity and the chemical dynamics between ${\beta}-cyclodextrin$ and aspirin were studied by FT-IR UN, $^1H$ NMR. $^{13}C NMR$. and FAB-MS spectroscopy in solution and solid state, A stable solid inclusion complex was prepared by the recrystallization method, From the spectral changes of the host and guest molecules, orientational preference for binding in the cyclodextrin cavity was determined.

  • 본 연구에서는 이중결합을 가진 불포화화합물로 제조나 저장 시 열과 산화 (빛)에 의해 쉽게 파괴되어 활성이 감소하여 응용범위에 한계가 있는 아스타잔틴의 안정성 향상을 위하여 나노리포좀 제형기술을 이용하였다. 제형안정성 평가를 통하여 리포좀 제조조건 및 조성비를 확립하였고, 포접나노리포좀을 제조하여 포접전의 아스타잔틴과 안정성을 비교 검토하였다. 아스타잔틴을 포접하여 포접나노리포좀 제조 시포접전의 아스타잔틴에 비해 열에 대한 안정성이 2배 정도 향상되고, UV 안정성 또한 3배 향상된 것을 확인하였다. 이상의 결과로부터 아스타잔틴을 포접한 포접나노리포좀은 안정성이 우수한 기능성 원료로서 산업적으로 용도를 넓히고 부가가치를 더욱 향상시킬 수 있을 것으로 기대된다.

  • 각종 CD와 여러 종류의 화합물, pH indicator, biostain, 아미노산, 천연배당체, 그리고 fatty acid 간의 inclusion complex 형성능을 비교하였다. Fatty acid가 각종 CD에 대한 포접형성능과 선택성 이 높아 CD 분리용 ligand로 적합함을 알았다. Fatty acid의 포접능과 선택성은 탄소쇄의 길이와 밀접한 상관관계를 나타내어 capric acid는 ${\alpha}$- CD와 palmitic acid는 ${\beta}$-CD와 높은 선택성을 보였다. Complex형성에 미치는 반융조건인 흔합 농도비, pH, ionic strength, 그리고 온도의 영향을 검토하였다. 적정 혼합 농도비는 CD가 50mM 그리고 lig and 인 fatty acid가 50mM 일 때 였으며 , 온도가 중 요한 형성조건으로 작용하였다. 포접되지 않은 CD 와 fatty acid가 포접 된 CD의 분자구조를 X-ray diffraction과 IR spectrum을 사용하여 비교 검토하였으며, DSC로 inclusion complex 형성반응의 en­t thalpy의 변화값(${\gamma}H$)을 측정하여 발열반응임을 확인하였다.

TMA-물계 포접화합물의 과냉각 억제에 대한 연구 II (A Study on Supercooling Repression of TMA-Water Clathrate Compound II)

  • This study aims to find out cooling characteristics of TMA(Tri-Methyl-Amine, ($CH_{3})_{3}N$) 25wt%-clathrate compound with ethanol($CH_{3}CH_{2}OH$) such as supercooling, phase change temperature and specific heat. For this purpose, ethanol is added as per weight concentration and cooling experiment is performed at $-6{\sim}-8^{\circ}C$, cooling heat source temperature, and it leads the following result. (1) Phase change temperature is decreased due to freezing point depression phenomenon. Especially, it is minimized as $3.8^{\circ}C$ according to cooling source temperature in case that 0.5w% of ethanol is added. (2) If 0.5wt% of ethanol is added, average supercooling degree is $0.9^{\circ}C $ and minimum supercooling is $0.8,\;0.7^{\circ}C$ according to cooling heat source temperature. The restraint effect of supercooling is shown. (3) Specific heat shows tendency to decrease if ethanol is added. It is $3.013{\sim}3.048\;kJ/kgK$ according to cooling heat source temperature if 0.5wt% of ethanol is added. Phase change temperature higher than that of water and inhibitory effect against supercooling can be confirmed through experimental study on cooling characteristics of TMA 25wt%-water clathrate compound by adding additive, ethanol. This can lead to shorten refrigerator operation time of low temperature thermal storage system and improve COP of refrigerator and efficiency of overall system. Therefore energy can be saved and efficiency can be improved much more.

  • The purpose of this study is to investigate the propriety of TMA clathrate as a cold storage medium. Particularly, this is to examine the extent of subcooling improvement when the additives are added to the TMA clathrate, because water used cold storage material has low phase change temperature and subcooling. This study has been analysed and compared with TMA 30 wt% clathrate how phase change temperature, subcooling and specific heat in the various concentrations are changed. This results view to be improved phase change temperature and subcooling of TMA 30 wt% as a cold storage medium, when it had some additive. Additional1y, it is found that the additive must be controlled under available solution limit and study for new additive must be lasted to know its effect.

Octyldiinethyl p-aminobenzoate와 \brta -Cyclodextrin$의 포접화합물에 관한 연구 (A Study on the Inclusion Complexation of Octyldimethyl p-aminobengoate with \brta -Cyclodextrin$)

  • Inclusion complex formation of octyldimethl p-aminobenzoate with $\beta$-cyclodextrin in aqueous solution and in the solid state was studied by the solubility method, spectroscopic(UV, FT-lR) and X -ray diffractometry. The solid complex of octyldimethy p-aminobenzoate with $\beta$-cyclodextrin was obtained in molar ratio of 1 : 2(guest/host). A spatial relationship between host and guest molecule was clearly reflected in the magnitude of the apparent stability constant (K') and in the stoichiometry of the inclusion complex. Furthermore, a typical type Bs phase-solubility diagram was obtained for octyldimethyl p-aminobenzoate and p -cyclodextrin in water at $25^{\circ}C$. The results indicated that the solubility of the guest molecule was higher by the formation of $\beta$-cyclodextrin inclusion complex.
