Antibiotic Properties of an Entomopathogenic Fungus, Beauveria bassiana, on Fusarium oxysporum and Botrytis cinerea (살충성 진균 Beauveria bassiana의 Fusarium oxysporum과 Botrytis cinerea에 대한 항균활성)

  • 식물병원균 Botrytis cinerea와 Fusarium oxysporum에 대한 생물학적 방제의 기초자료를 얻기 위하여 살충성 진균 Beauveria bassiana의 식물병원균에 대한 균사성장저해, 포자발아 억제, 균사와 포자의 형태변화 등의 효과를 살펴보았다. 평판배지 상에서 두 식물병원균의 균사생장이 저해되었으며, 저해효과는 배지종류에 따라 달랐는데 B. cinerea의 경우 PDA배지에서 가장 크게 저해되었으며 F. oxysporum의 경우에는 TSA배지에서 가장 크게 저해되었다. B. bassiana는 $25^{\circ}C$에서 6일간 배양했을 때 F. oxysporum에 대한 항균력이 가장 높았으며 동시에 최대의 균체량을 생산하였다. 또한 B. bassiana의 배양여액을 식물병원균에 30% 농도로 첨가하여 배양했을 때 식물병원균의 포자 발아율은 B. cinerea, F. oxysporum에서 각각 30%(control: 88.2%), 10.0%(control: 78.6%)로 낮아졌으며 발아 개시 시간도 4~8시간 지연되었다. 현미경을 통한 미세구조관찰에서는 10%의 B. bassiana배양여액을 첨가했을 때 F. oxysporum의 포자 크기가 1/2~1/3으로 줄어들었으며 균사와 격막의 형태도 비정상적으로 변하여 균사외막과 격막-격막 사이의 구분이 불명확해졌다.

Leaf Blight of Perilla Caused by Alternaria alternata (Alternaria alternata에 의한 들깨 잎마름병)

  • A new leaf blight was found on the perilla leaves at the major perilla-cultivating areas such as Kangdong in Busan and Miryang in Kyungnam province. Symptoms of the disease initially appeared on the edge of perilla leaves showing black necrosis and drying, and the infected leaves were finally fell down. The SD1 isolate showing strong pathogenicity and forming abundant conidial spores on the diseased lesions was isolated. Among the tested media, mycelial growth was abundant on PDA (Potato Dextrose Agar) medium at $25^{\circ}C$ under dark condition, but conidial formation was greater on V8A (V-8 juice A8ar) medium than that on PDA medium. Optimal temperatures for mycelial growth and conidial formation on PDA medium were respectively $25^{\circ}C$ and 3$0^{\circ}C$. The rate of conidial germination and the elongation of germ tube were more effective in 10% tomato juice than those in PDB (Potato Dextrose Broth) and sterile water. In 10% tomato juice, the rate of conidial germination and the length of germ tube were 100% after incubation for 24k and 535.2${\mu}{\textrm}{m}$ after incubation for 36hr, respectively. According to the result of pathogenicity, it revealed that conidial suspention with 10% tomato juice was the most effective for pathogenicity test showing as 100% of disease incidence, and the symptoms caused by artificial inoculum were same as those of naturally infected perilla. In this study, the SD1 isolate according to the results of morphological characteristics, the incubation characteristics and pathogenicity was firstly identified A. alternata, and named as leaf blight of perilla.

  • Coprinus cinereus resistant strain against validamycin was successfully isolated. Germination rate of oidiospores, branching pattern, and localization of actin protein of the resistant strain were compared with normal strain. The resistant strain showed better germiantion rate of oidia (about 20 fold), more frequent branching, and even actin localization on validamycin plate where actin content was severely reduced in case of normal strain.

  • The commercial product "pyto-patch" prepared as nano sized silver particle less than 5 nm, has effective antifungal activity against Collectotrichum gloeosporioides, Botrytis cinerea, Sclerotinia sclerotiorum in vitro. As a fungal growth inhibiton mechanism, it can reduce spore germination rate and mycelial growth. As s promising fungicide, Phyto-patch can control anthracnose effectively. The spore of C. gloeosporioides dipped in 5 ppm phyto-patch dilute suppressed germination rate to 13.2%, and mycelial growth stopped for 15 days at 10 ppm. The spore postinoculated on 10 ppm phyto-path smeared PDA surface could not germinate for 3 days and prohibit pathogen infection effectively. In field test, the anthracnose development of 4 ppm phyto-patch treated plot was less than 7% after 21 days compaired to 40% of it in untreated plot. In heavy rainfall season, pepper anthracnose effectivly contrrolled by regular 10 ppm phytopatch spraying every 7 days. The diseased pepper fruit decreased to 5.8% compaired to 94.6% in untreated plot. During drying period, the diseased pepper fruit havested in phyto-patch treated plot was 24.2%, but pepper fruit havested in untreated plot destroyed to 100% within 3 days. The nano silver particle coated on multching textile prevented late blight of pepper effectively and disease occurance delayed about month.

  • This study was conducted to investigate agents inducing conidial germination of an entomopathogenic fungus, Beauveria bassiana KK5. Different chemicals including carbohydrates were mixed with conidia of B. bassiana and incubated on water agar for 12 hours. Fructose, mannose and skim milk were useful for spore germination compared to other chemicals. Bioassays against green peach aphids were conducted with the fungal conidia suspended in 1% fructose, mannose and skim milk. Of them, a mixture of skim milk plus conidia of B. bassiana KK5 showed the highest mortalities against $3^{rd}$ instar of green peach aphid.

  • In order to establish a dose limits for subsequent induced mutation research itl Pyricularia oryzae, X-ray sensitivity of the conidia and the vegetative hypae of the fungus, race N-1, was investigated. Conidia of the fungus irradiated with X-rays reduced significantly in spore germination inversely with radiation doses. A severe suppression of conidia germination in about $80\%$ was found at the dose of 120kR, and the rests of the conidia produce very short and lysed germ tubes. A stimulated effect was observed in the elongation of hyphae from the conidia of 10 kR irradiation at initial stage of the growth. The radiosensitivity of hyphae was exremely higher than that of conidia with the increase of radiation doses. It was also recognized that the frequency of X-ray induced mutation in pathogenicity was directly proportional to radiation doses.

Several Factors Affecting Mass Production of Microlepia strigosa (Thunb.) C. Presl Sporophytes (돌토끼고사리 포자체의 대량생산을 위한 몇 가지 요인)

  • This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation and seedling growth in Microlepia strigosa (Thunb.) C. Presl. Spore germination and prothallus development were promoted by low concentrations of Knop medium nutrient solution. The optimal medium for prothallus propagation and antheridium formation was 2X MS medium with 3% sucrose. The activated charcoal content of the medium did not affect prothallus proliferation. Among the various combinations of culture soil (bedding soil, peat moss, perlite and decomposed granite), a mixture of bedding soil, peat moss and decomposed granite at a ratio of 1 : 1 : 1 (v : v : v) had a positive effect on sporophyte formation. The most efficient conditions for promoting the growth of whole plants (sporophyte seedlings) were 50 - cell plug trays filled with a mixture of bedding soil and decomposed granite at a 2 : 1 (v : v) ratio.

Spore Germination and Prothallium Development Conditions of Lygodium japonicum (Thunb.) Sw. (실고사리(Lygodium japonicum (Thunb.) Sw.) 포자발아와 전엽체 발달조건)

  • This study was conducted to determine the optimal conditions of growth medium, temperature, and light quality for efficient propagation of Lygodium japonicum spores. The rate of spore germination and prothalium development was high in Knop and 1/8MS and 1/4MS media, which had low mineral content; in particular, the germination rate exceeded 74%, and the germinated spores developed into heart-shaped prothallia. However, in Knop‘s medium with the lowest mineral content, a rapid prothallium senescence was observed; in 1/4MS medium, prothallium development was delayed. Germination rate increased with the increase in temperature and reached its maximum, 86.7%, at 30℃; however, at this temperature, the prothallia were thinner and abnormal development of rhizoids was observed compared to normally developed prothallia and rhizoids at 25℃. Therefore, the results suggested that the optimal temperature for L. japonicum spore germination was 25℃. The rate of germination was also measured under different light conditions, and the highest rate of 90.6% was observed under LED red light compared to fluorescent (77.2%) or LED blue (5.4%) lights. The germinated spores developed into heart-shaped prothallia under LED red light; however, 15 days after seeding, prothallium development decreased and the became elongated. In contrast, a normal and continuous development of heart-shaped prothallia was observed under fluorescent light.

  • This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation, and seedling growth in Polystichum braunii (Spenn.) $F{\acute{e}}e$. The rate of spore germination and early prothalium development was high in Knop (41.2%), which had low mineral content. The optimal medium for prothallus propagation and sexual organ formation was 2MS medium (2% sucrose). Among the various mixtures of cultivation soil (bedding soil, peat moss, perlite, and decomposed granite), a mixture of bedding soil and decomposed granite at a ratio of 2:1 (v:v) had a positive effect on sporophyte formation (276.3 ea/$7.5m^2$). The most efficient conditions for promoting the growth of sporophytes were pots filled with only bedding soil.

Improvement of Cultivated Mushroom Agaricus bisporus by Means of Single Basidiospore Selections (양송이(洋松茸)의 단일담포자(單一擔胞子) 분리(分離)에 의(依)한 신계통(新系統) 선발(選拔)에 관한 연구(硏究))

  • Germination of the single-basidiospore of Agaricus bisporus, mycelial characteristics of the single spore culture and its fruit body formation were studied. Germination of the single basidiospore was the best on water agar. Of 6372 spores isolated, 1622 spore were germinated and the germination rate was 26 percent. Cultures from single basidiospores showed two distinct mycelial types on the compost extract sucrose agar: one was the strandy type and the other was the fluffy. The strandy type culture produced more sporophores than the fluffy. Of 778 strandy isolates screened, nine cultures yielded 5 percent higher than the origine. No. 1567 and 1708 were selected for the commercial cultivation.
