저질내의 와편모조류 휴면포자 계수방법 비교 연구

  • 해양저질에서 휴면포자의 분포와 분포량에 대한 조사는 미래의 와편모조류 적조발생 시기나 장소를 예측할 수 있게 한다. 그러므로 저질내의 휴면포자를 찾고 분리하는 기술이 기본적으로 수행되어야 하며 지금까지 여러 방법이 개발되어 왔다. 이 연구의 목적은 기존의 휴면포자 계수방법들을 비교함으로써 연구의 효율을 제고 하고자 하는데 있다. 이 실험에서는 새만금 해역의 동일정점 저질내에 존재하는 휴면포자를 형광 염색하여 계수하는 Primuline 염색법, 해수, 휴면포자, sodium polytungstate의 비중차이를 이용하여 밀도구배원심법으로 분리하는 방법, 그리고 10% HCl로 탄산질을 제거하고 47% HF로 규산질을 제거한 뒤 휴면포자를 계수하는 고전적 계수방법 (palynological technique)을 사용하여 저질 내의 휴면포자를 분리 비교하였다. Primuline 염색법을 이용하였을 경우 Alexandrium famarense/catenella complex, Alexandrium sp. (spherical), Gonyaulax verior, Gonyaulax scrippsea, Gonyaulax spinifera complex등의 일부 종만 염색되어 관찰되었다. sodium polytungstate를 비중 1.3 g$cm^{-3}$으로 조정한 후 휴면포자를 분리한 밀도구배 원심법은 생물에 거의 무해한 것으로 알려져 있어서 발아가 가능해 휴면포자와 영양세포간의 관계를 규명할 수 있다는 장점이 있으나 계수에 있어서는 재현성이 떨어졌다. 반면 Palynological technique은 발아는 되지 않지만 석회질 벽을 가지는 휴면포자를 제외한 대부분의 종들이 관찰되었다. 이 결과는 실험 목적별로 휴면포자 계수방법을 달리 할 수 있음을 보여준다. 즉 발아실험을 할 경우에는 밀도구배원심법을, Alexandrium과 같은 특정종의 계수목적시에는 Primuline 염색법을, 정점 내 정확한 종 분포 파악을 위해서는 palynological technique을 사용하는 것이 효과적일 수 있다. 하지만 종분포 파악시에는 시간이 많이 소요되므로 좀더 신속하고 정확한 실험을 위해 분자생물학적인 기법들이 요구된다.

  • Spore production mechanism of selected lignicolous basidiomycetes was inhibited by soaking their sporocarps in water for varying periods. Depending on the duration of immersion, some species resumed spore discharge, while in others it was completely ruined. This was related to the nature and consistency of these sporocarps. All the species studied absorbed water quite readily into their sporocarps. The loss of this water through dehydration was more rapid in smaller and delicate species than the bulky and robust ones.

The Destruction of Bacterial Spores Upon Compressional Pressure (타정 압력에 의한 세균포자의 파괴현상)

  • The tolerance of useful bacterial spores to the conditions of tablet making, specifically, the destruction of bacterial spores upon compressional pressure was investigated. The damage of bacterial spores occurred mainly during the tabletting. The bacterial spores obeyed a logarithmic destruction rate upon compressional pressure. The spore destruction rate was dependent upon the strains of microorganism. The Decimal Reduction Pressure, designated as P-value, were $2.9\;ton/cm^2$, $2.6\;ton/cm^2$ and $2.1\;ton/cm^2$ for the spores of Bacillus subtilis, Bacilus coagulans and Clostridium butyricum, respectively, and $1.7\;ton/cm^2$ for the vegetative cell of Streptococcus faecalis. The spore destruction upon compressional pressure was influenced by the type of filler. The P-value of the spore of B. coagulans was $2.8\;ton/cm^2$ in the lactose filler, but $2.0\;ton/cm^2$ in the starch filler. The number of viable spores was inversely proportional to the hardness and density of tablet, in case that the same type of filler was used. The starch filler, which resulted in the lower hardness and lower density of tablet, caused higher spore destruction rate compared with the lactose filler.

Pathological Studies on the New Microsporidia K79 Isolated from the Silkworm, Bombyx mori L. I. Purification and Serological Discrimination of Microsporidian Spores. (가잠에서 분리된 새로운 미포자충 K79의 병리학적 연구 I. 미포자충 포자의 정제 및 식별)

  • The present study was made to establish the purification method of mature microsporidian spores by iso-density equilibrium technique using percoll and the serological discrimination by an indirect fluorescent antibody technique. The purification of new microsporidian spores took effect in the three steps purification method(precentrifugation-percoll iso-density equilibrium centrifugation-rising). There were clear differences in the size of spores between the new microsporidia and N. bombycis. The spores of N. bombycis is short elliptical of 2.07 in a ratio of length to width in diameter while that of new microsporidia is characterized with long elliptical shape which shows a ratio of 2.76 in length to width in diameter. The specific antigens of new microsporidia K79 spores was showed in the spores wall by the indirect fluorescent antibody reaction, and it was affected by the antibody against N. bombycis which antiserum was diluted in 1:20. It means that the new microsporidia K79 is serologically not identical to N. bombycis.

  • 사과 겹무늬썩음병의 주 전염원은 가지 병반에서 형성되는 자낭포자 및 병포자이므로, 가지에서의 포자형성 또는 포자의 분산을 화학적 또는 물리적 방법으로 저해할 수만 있다면 효과적으로 병을 방제할 수 있을 것으로 생각했다. 전염원을 줄이기 위한 첫 번째 시도로 1992년 이른봄 가지에 형성된 사마귀를 호미로 제거한 결과, 병포자의 분산과 과실발병 억제효과에 있어서 무처리와 유의적인 차이가 인정되지 않았다. 두 번째 시도로 포자의 분산을 막기 위해 1993년 이른봄에 겹무늬썩음병에 걸린 가지를 비닐필름으로 감고 5월부터 9월까지 20일 간격으로 살균제를 살포한 결과, 통계적으로 유의성이 인정되는 병 방제효과가 있었고, 나무의 생육에 있어서도 콜크층이 이상 비대하는 것 이외에 별다른 이상이 없었다. 이 개념을 더욱 발전시켜 1994년 이른봄에는 병든 가지를 수용성 고분자 물질인 polyvinyl alcohol로 도포하고 수배 분산 포자의 수를 조사한 결과, 포자의 분산량이 크게 줄어져 실용화의 가능성이 시사되었다.

The Effects of Cyclic AMP, Theophylline and Caffeine on Sporulation of Aspergillus niger in the Defined Media (검정곰팡이(Aspergillus niger)의 포자형성(胞子形成)에 미치는 cAMP, 테오필린 및 카페인의 영향(影響)에 관한 연구(硏究))

  • Aspergillus niger van Tieghem was cultured by the method of synchronous and submerged culture. The sporulation occurred through the culture and its life cycle and differentiation were completed in the experiments. The effects of cAMP, theophylline and caffeine on the sporulation of A. niger were investigated. In the sporulation medium, the sporulation was stimulated by addition of cAMP and its optimum concentration was $10^{-4}M$. In the sporulation medium, the sporulation was stimulated by addition of theophylline and its optimum concentration was 10 mg/ml. In the sporulation medium, the sporulation was stimulated by addition of caffeine and its optimum concentration was 300 mg/ml. Theophylline added to the sporulation medium together with cAMP enhanced the promotion effect of cAMP on sporulation. Caffeine added to the sporulation medium together with cAMP enhanced the promotion effect of cAMP on sporulation. In the sporulation medium, the sporulation was stimulated by addition of neither AMP nor ATP. In the potassium acetate medium, cAMP, theophylline and caffeine stimulated the sporulation, respectively.

Five Sporocarpic Species of Glomus Found in Korea (한국에서 발견된 Glomus의 포자과를 형성하는 종)

  • Five sporocarpic species of Glomus were found at the four sites of the different plant's cultivations in Korea ; G. clavisporum, G. liquidambaris, G. rubiforme, G. sinuosum, and G. taiwanense. The morphological characteristics of these species collected were observed and compared under light microscopes. Three species G. clavisporum, G. liquidambaris and G. taiwanense were similar with each other in the aspects of having no peridium and the clavate spores orderly arranged within sporocarps. G. sinuosum was different from the other in the aspects of having a thick and sinuous peridium and the orderly arranged spores within sporocarps. However G. rubtforme was different from the other four species in aspects of sporocarps haying ovoid spores arranged irregulary.

  • A new microsporidia (S85) was isolated from the silkworm, Bombyx mori L. in Suwon, Korea in 1985. The electron microscopic observation of the fine structure of the spore was studied for the characterization of the new microsporidia and its taxonomic position. The sporewall was composed of three different electron dense layers and the spore surface was rouph and wrinkled. The polaroplast has two parts which differed in the arrangement of the lamellae. The polarfilament was isofilar with thirteen coils and it was consisted of eight concentric layers of different electron density. The spore contained a single nucleus and the pansporoblast appeared as a thin ballon-like structure surrounding the spore. The membrane was generally smooth and shown occationally wrinkled.

  • High ionic strength is expected to enhance dissociation of Ca-DPA from spores and to contribute to a detrimental effect on spore stability or on spore heat resistance with a combined treatment of gamma-radiation. From this study, this hypothesis has become apparent as as follows; 1) Ca-DPA dissociation contributes to loss of stability of bacterial spores with respect to heat resistance, survival during storage, and 2) the cytoplasmic membrane plays a role in maintaining the stability of DPA-Ca-spore complex, apparently by serving as a permeability barrier.

Agar media containing various amount of tomato juice were tested to determine the degree of conidial production of Cercospora beticola. Non-sporulating culture on potato dextrose agar readily sporulated on agar media containing various amout of tomato juice 48 hours after transfer. Considerably small amount of sporulation occurred on agar media containing 10% of tomato juice. Sporulation increased considerably on media containing more than 20% of tomato juice; the higher the proportion of tomato juice in a medium, the greater the number of spores produced.
