In conventional activated sludge process, COD fractions in wastewater are important parameters, significantly. Depending on characteristics of influent COD fractionation, activated sludge process requires a major change of a process operation to ensure meeting a stricter standards. In order to validate and evaluate the accuracy of the traditional COD fractionation methodologies, readily and slowly biodegradable COD was mixed using glucose and peptone as a sole carbon source in a synthetic wastewater, respectively. In this research, prediction of the COD fraction was made using the OUR(Oxygen Utilization Rate) and the NUR(Nitrate Utilization Rate) experiments. The result showed that COD fractions calculated by OUR experiment were similar to the composition of synthetic wastewater. On the other hand, it was found that an error was generated during the NUR experiment. This error was due to the intracellular storage period for storage microorganisms such as PAOs, and the error in COD fraction was observed about 8-14 % in terms of Total COD.
A bacterial strain KH-95 producing a high concentration of red pigment was isolated from the soil. The strain KH-95 was identified as a strain of Serratia sp. based on morphological and physiological characteristics. The optimal temperature and initial pH range for the production of pigment were 28$^{\circ}C$ and 7.0-8.0, respectively. The red pigment was purified through solvent extraction and silica gel column chromatography. Analyzing the structure of this pigment by instrumental analysis, it was identified as prodigiosin-like compound. In optimization of carbon and nitrogen sources, all carbon sources tested in this work inhibited the production of pigment except oils. Casein fumed out to be the most suitable nitrogen source for pigment production. Other nitrogen sources such as yeast extract, beef extract and peptone showed good cell growth but potently inhibited the production of pigment.
Two bacterial strains isolated from soil (Bacillus subtilis strains: PS2 and RFO41) were evaluated to determine their promoting effect on the growth of tomato seedling under axonic and pot conditions. The production of phytohormone, such as indole-3-acetic acid, indole-3-butyric acid, gibberellin and zeatin by these two strains was investigated as possible mechanisms for plant growth stimulation. Both PS2 and RFO41 were shown to produce various phytohormones, and. the production of phytohormones was stimulated by the addition of peptone-rich brain heart broth medium. In addition, these bacteria exhibited high levels of phosphatase activity, which ranged from 2.18 to $2.7\;{\mu}\;{\rho}-nitrophenol/ml/hr$. PS2 and RFO41 were applied to the pot test for growth of tomato seed with phosphate. Root and shoot lengths of germinated tomato after 15 days were 45.5% and 36.5% longer than that of control in RFO41 treated samples, respectively. Baciller sp. PS2 and RFO41 may have a potential for biofertilizer in the agriculture.
Pleurotus ostreatus No.42, known as the ligninolytic basidiomycetes, showed production of MnP and Lac, but did not show any LiP acitivity in static culture, grown in GPYW liquid medium. Maximum production of MnP (80U/flask) was observed on day 11 of culturing in this medium. Chromatographic purification of MnP included the use of Sepharose CL-6B and Mono-Q. The major MnP isozyme purified by column chromatography was observed to be a 36.4 KDa (single band on SDS PAGE). The 19-amino acid sequence from the N-terminal was determined by protein sequencing to be ATCADGRTTANAACCVLFP. The N-terminal sequence of the major MnP isozyme of P. ostreatus No.42 was found to be the same as a previously reported sequence of an MnP3 isozyme from this fungus.
The results of field experiments with mesocosms on Lake Baikal, containing natural plankton assemblage, and laboratory experiments with microcosms containing Daphnia magna and Chlorella vulgaris demonstrated decrease of the structural exergy of the communities after the addition of allochtonous compounds peptone, diesel oil, o-diphenol, $CdCl_2$ to mesocosmsassemblage, phenol, $CoCl_2$ and $CuSO_4$ to micro-cosms. Structural exergy changes were more expressed than changes of components biomasses and total biomass of the community. Comparison of exergy content for benthos in cleanand affected by the discharges of Baikalsk Pulp and Paper Combine also showed sufficient docrease of structural exergy in polluted area. It points to the possibility of the use of structural exergy as ecosystem health reflecting parameter.
In this study, Pleurotus ostreatus No.42 was cultured in glucose-peptone-yeast-wheat bran medium using a previously reported novel rotary draft tube bioreactor. Versatile peroxidase (VP), a lignin-degrading enzyme, was isolated from a pellet-type mycelium culture grown in the medium for seven days. The VP was purified by sequentially applying ultra-filtration, DEAE-Sepharose CL-6B column, and Mono Q column. SDS-PAGE analysis revealed the molecular weight of VP to be 36.4 KDa with an isoelectric point of 3.65. The amino acid sequence was confirmed as VTCATGQTT. The purified VP was observed to possess the property of not only oxidizing Mn ions but also decomposing veratryl alcohol, a non-phenolic compound. The catalytic ability of VP is a subject for future research.
This study was designed to investigate the optimum manufacturing condition of germanium-fortified yeast, and the binding properties of germanium (Ge) in germanium-fortified yeast. The nutritional optimum conditions were glucose 3.0 (w/v) %, yeast extracts 0.3 (w/v) % and peptone 0.5 (w/v) %, and the amounts of yeast cells were 67.4 mg/ml. And, the standard germanium-fortified yeast was produced under the condition at the ratio of yeast cell and germanium solution was 1 : 0.5 (50%), pH 6.5 and $35-40^{\circ}C$ during fermentation. In results of the identification, binding of germanium-protein showed structural difference between the inorganic Ge $(GeO_2)$ added during fermentation process and germanium-fortified yeast. Therefore, germanium-fortified yeast made by biosynthetic technology formed structurally safe organic germanium during fermentation process. Germanium-fortified yeast can be applied as a new functional material far the improvement of health, the prevention and treatment of chronic degenerative disease like cancer, and the enforcement of immune system.
As a way to determine the optimal culture conditions for the production of ${\gamma}$-linolenic acid by Mucor sp. KCTC 8405P, the influence of different carbon and nitrogen sources, initial pH, and C/N ratio of medium was investigated. Glucose was found to be the best carbon source in terms of lipid content and ${\gamma}$-linolenic acid yield. Ammonium sulfate and organic nitrogen sources such as urea and peptone resulted in relatively increased lipid and ${\gamma}$-linolenic acid production. The highest accumulation of lipid was obtained at a C/N ratio of 56.6 using glucose and (NH$_4$)$_2$SO$_4$ as carbon and nitrogen source, respectively. It was found that the lipid content increased significantly with increasing initial pH of medium up to pH 9.0. The influence of mixed carbon source on the ${\gamma}$-linolenic acid yield was also investigated. High accumulation of lipids, 315 mg/100 ml medium, and 13-14% of ${\gamma}$-linolenic acid content in the cellular lipid were obtained in a shaking culture containing 3% of glucose and 2% sodium acetate as carbon source and 0.1% of (NH$_4$)$_2$SO$_4$ as nitrogen source at pH 8.0.
Fruit bodies of Ganoderma lucidum have been used for most pharmacological studies, but pharmacological effects are likely variable because the habitats and strains of Ganoderma lucidum are different. Therefore, their fermentation is required to produce constant and reliable pharmacological constituents from Ganoderma lucidum. During the studies of medium for industrial application. it was found that ginseng root residues, remaining after being extracted with ethanol, were a good carbon source for a fermentation of Genoderma lucidum and a corn steep liquor was also economical for the nitrogen source. Yield of the mycelial cultured in ginseng root residues and corn steep liquor was 2.5 times higher than that in glucose and peptone, known as a conventional medium of Ganoderma lucidum. The polysaccharide content of the extracts from the cultured mycelia was higher than that from fruit bodies, but protein content was vice versa. Extracts of the cultured mycelia were more effective and lasting than extracts of the fruit bodies in decreased hypertention of spontaneously hypertensive rats (SHR).
A soil enrichment LYF-1 culture from a contaminated site, which could reductively dechlorinate 900 $\mu$M (ca. 150 mg/L) of tetrachloroethylene (PCE) stoichimetrically into cis-1,2-dichloroethylene (cis-DCE), was established and characterized. The enrichment culture can use yeast extract, peptone, formate, acetate, lactate, pyruvate, citrate, succinate, glucose, sucrose, and ethanol as electron donors for dechlorination of PCE. Addition of NO$_2$$^{[-10]}$ and NO$_3$$^{[-10]}$ as alternative electron acceptors showed complete inhibition of PCE dechlorination, but S$_2$O$_3$$^{-2}$ , SO$_3$$^{-2}$ and SO$_4$$^{-2}$ had no significant effect on PCE dechlorination. The enrichment culture was attached to ceramic media in an anaerobic fixed-bed reactor. The fixed-bed reactor showed more than 99% of PCE degradation in the range of PCE loading rate of 0.13-0.78 $\mu$moles/L/hr. The major end product of PCE dechlorination was cis-DCE.
