Comparison of Changes in Cell Wall Characteristics during Storage in 'Niitaka' and 'Pingguoli' Pear Fruits (동양배 '신고'와 '사과배' 과실의 저장 중 세포벽 특성의 변화 비교)

  • The characteristics of fruit ripening and cell wall modifications of two oriental pear fruits were compared during storage. The loss of fresh weight was lower in 'Pingguoli' than 'Niitaka', probably due to the difference in wax accumulation and skin structure. 'Pingguoli' produced much higher amount of ethylene and showed climacteric ripening pattern, but this response was not found in 'Niitaka'. A significant difference in soluble pectin contents was found, thus, the amount of water soluble pectins were much higher in 'Niitaka' at harvest but this difference was not significant at later stage of storage. A severe loss of water soluble pectins in 'Niitaka' was confirmed whereas a similar tendency was not found in CDTA soluble ones. Even the amount of $Na_2CO_3$ soluble pectins was relatively low, 'Pingguoli' contained approximately 2 times higher than 'Niitaka' and both cultivars showed an increasing tendency in $Na_2CO_3$ soluble polymers at later stage of storage. No significant difference in alkali soluble polysaccharides (hemicelluloses fraction) was found between both cultivars. There was a significant change in gel filtration profiles regardless of cultivars, especially in water soluble pectins resulting from severe depolymerization probably due to degradation of higher molecular weight pectins and loss of their side chains. In gel filtration profiles of $Na_2CO_3$ soluble pectins, neutral sugars were evenly distributed regardless of molecular size of pectic polymers but the decrease of high molecular weight fraction was found. In comparison of alkali soluble polymers, a similar trend was found, that is, there was no difference in gel filtration profiles but 'Niitaka' seemed to have longer side chains in alkali soluble polysaccharides in both 4% and 24% of KOH soluble fractions.

Preparation of γ-oryzanol-loaded pectin micro and nanocapsules and their characteristics according to particle size (감마오리자놀 함유 칼슘-펙틴 미세 및 나노캡슐의 제조와 입자 크기에 따른 캡슐특성)

  • ${\gamma}-Oryzanol-loaded$ calcium-pectin micro- and nanocapsules were prepared by ionic gelation to improve oxidation stability and the effect of particle size on capsule properties was investigated. The physical properties were influenced by preparation conditions such as concentrations of pectin, $CaCl_2$, ${\gamma}-oryzanol$, and hardening time. Particle sizes of micro- and nanocapsules that showed the maximum encapsulation efficiency and sustained release were $2.27{\pm}0.02mm$ and $347.7{\pm}58.1nm$, respectively. Microcapsules showed higher encapsulation efficiency ($50.73{\pm}1.98%$) than nanocapsules ($17.70{\pm}2.04%$), while nanocapsules showed more sustained release and higher stability than microcapsules. Release of ${\gamma}-oryzanol$ from both microand nanocapsules, which was low in gastric environments and promoted in intestinal environments, showed suitable characteristics for oral administration. Furthermore, antioxidant activity of ${\gamma}-oryzanol$ against autoxidation of linoleic acid was prolonged by both micro- and nanoencapsulation in a ferric thiocyanate test. Therefore, micro- and nanoencapsulation using pectin can be effective for improving biodelivery, stability, and antioxidant activity of ${\gamma}-oryzanol$.

Comparison of Pectin Hydrogel Collection Methods in Microfluidic Device (미세유체 장치에서 수거 방법에 따른 펙틴 하이드로겔 입자의 특성 비교)

  • This study investigated the effect of different collection methods on physical properties of pectin hydrogels in microfluidic synthetic approach. The pectin hydrogels were simply produced by the incorporation of calcium ions dissolved in continuous mineral oil. Then, different collection methods, pipetting, tubing, and settling, for harvesting pectin hydrogels were applied. The settling method showed most uniform and monodispersed hydrogels. In the case of settling, a coefficient of variation was 3.46 which was lower than pipetting method (18.60) and tubing method (14.76). Under the settling method, we could control the size of hydrogels, ranging from $30{\mu}m$ to $180{\mu}m$, by simple manipulation of the viscosity of pectin and volumetric flow rate of dispersed and continuous phase. Finally, according to the characteristics of simple encapsulation of biological materials, we envision that the pectin hydrogels can be applied to drug delivery, food, and biocompatible materials.

Effect of Glue Plant(Codium Fragile) on Physicochemical Characteristics of Dongchimi During Fermentation (청각채가 동치미의 발효 중 이화학적 특성에 미치는 영향)

  • The effect of glue plant(Codium fragile) on the Dongchimi fermentation was investigated by measuring physicochemical properties during fermentation at $10^{\circ}C$. Dongchimi was prepared with various levels(0, 0.5, 1.0 and 1.5%) of glue plant(Codium fragile). The pH of Dongchimi decreased slowly in all samples during fermentation. Total acidity of Dongchimi increased gradually during fermentation and total acidity of Dongchimi with glue plant(Codium fragile) was higher than that of control. Redox potentials decreased until 30 days of fermentation but increased thereafter. Reducing sugar content increased in the initial stage of the fermentation periods, and then decreased gradually. The reducing sugar content of Dongchimi with glue plant(Codium fragile) was higher than that of control. The content of total vitamin C was much higher in dongchimi with glue plant(Codium fragile). In color measurement, lightness value decreased gradually. However, redness and yellowness values increased gradually during fermentation but decreased thereafter. The content of hot water soluble pectin(HWSP) decreased as the fermentation proceeded, but the content of hydrochloric acid soluble pectin(HClSP) and sodium hexametaphosphate soluble pectin(NaSP) increased.

Chemical Components in Different Parts of Pumpkin (늙은 호박의 부위별 화학성분)

  • The contents of free sugars, organic acids, amino acids, total carotenoids and pectin of pumpkin were analysed. Weight percentage of flesh was 84% of total weight. Rind and funicular attachment of seed were 10% and 3.5%, respectively. Major free sugars in pumpkin were fructose, glucose, sucrose. In flesh, fructose and glucose were the major free sugars, corresponding to 87% of total free sugars . Total sugar content in the flesh was three times higher than that in the funicular attachment of seed. Contents and composition of free amino acids were quite different according to the different parts of pumpkin. Non-volatile organic acids in pumpkin was malic, succinic and fumaric acid. The major acid in the flesh and funicular attachment of seed was malic acid and the contents in these parts were 85.2mg% and 226.5mg%, corresponding to 63% and 70% of the total organic acids, respectively. Eigh-tyseven percent of total carotenoids in the pumpkin was in the funicular attachment of seed, amounting 65.3mg%. However flesh and rind contained 6.6mg% and 3.3mg%, respectively. Water soluble pectin consisted 58% of the total soluble pectin in different parts of pumpkin, and soluble Pectin content was higher in funicular attachment of seed than in flesh. The major neutral sugars of the pectin in flesh and funicular attachment of seed were galactose and glucose consisting 45% and 36.5% of total neutral sugars.

Changes of Texture in terms of the Contents of Cellulose, Hemicellulose and Pectic Substances during Fermentation of Radish Kimchi (숙성 기간에 따른 무우 김치의 텍스쳐와 섬유소, 헤미셀루로오스, 펙틴질의 함량 변화)

  • The changes of dietary fiber and texture of radish Kimchi fermented at $17{\sim}20^{\circ}C$ were investigated. During the fermentation period, hardness and brittleness of radish Kimchi were decreased. The changes were more marked in the brittleness. But the contents of cellulose and hemicellulose were not changed considerably. The contents of AIS was decreased with fermentation. During the fermentation, hot soluble pectin was increased and protopection was decreased but the amount of changes was small. The texture of radish Kimchi was affected by contents of pectic subsrances, especially protopectin. The coefficient of correlation between hardness and protopectin contents was 0.85.(=r)

  • The structure of fresh, salted and heated petiole tissue from Chinese cabbage was examined with optical microscope, and the pectin of the tissues was fractionated by successive extraction. The pectin in Chinese cabbage consisted mainly of high methoxyl pectin(pA) and low methoxyl pectin(pB). The pA content was converted to pB markedly by salting and heating at $60{\sim}70^{\circ}C$ for 30 min while heating the cabbage above 80 decreased pB content. The firmness of heated tissue was highly correlated with pB content(r=0.996). Cytorrhysis test showed that pore size of the tissue became smaller by salting and heating at temperature below $80^{\circ}C$.

  • The manufacturing characteristics and physicochemical components analysis of Bokbunja jelly made with the addition of different amounts of materials (Bokbunja extract, sugar, gelatin, and pectin) were investigated. There were no significant changes in the pH, total acidity and sugar content of all Bokbunja jelly prepared with different amount of materials. L-value of color in the Bokbunja jelly increased in proportion to additive amount of gelatin. a-value and b-value of color in the Bokbunja jelly increased in proportion to additive amount of pectin. The value of hardness, adhesiveness, and chewiness in the Bokbunja jelly increased in proportion to the additive amount of gelatin. The value of springiness, cohesiveness, and gumminess were increased in proportion to additive amount of petin. The highest of overall acceptability values in the sensory test for color, sweetness, springiness, chewiness, and overall favorite were 5.80, 5.32, 5.27, 5.83, and 6.02, respectively, with the addition of 10% gelatin and 10% pectin.

Effects of Dietary Fiber and Fat Sources on Lipid Contents of Serum and Tissues in Rats Fed Cholesterol Diet (식이섬유와 지방의 종류가 고콜레스테롤식을 섭취한 흰쥐의 혈청과 조직의 지질성분에 미치는 영향)

  • The purpose of this research was to investigate the effects of 10%(w/w) of various dietary fiber, pectin(P), lignin(L), cellulose(C) and fiber-free (F) on lipid content of serum, liver and intestinal tissues in rats fed 10% coix oil (C)-or lard(L)-1% cholesterol diets for four weeks. The lard pectin(LP) group was the lowest In body weight gain and feed intake. Liver weight was not affected by dietary fiber and fat sources, and intestinal weight was higher in pectin(CP, LP) groups than in the other groups. Serum cholesterol was significantly lower in pectin(CP, LP) groups than in fiber-free(CF, LF) groups, and was higher in lard fiber-free(LF) than In the other groups(p<0.05). Serum triglycerides was significantly lower in coix oil pectin(CP) and lignin(CL, LL) than in fiber-free(CF, LF) groups. Liver total lipid and cholesterol content was significantly lower In pectin than In lard fiber-free group, and liver triglycerides was lower in coix oil pectin(CP) than in fiber-free groups (p<0.05). Lipid content of intestinal tissue was not affected by dietary fiber source. But total lipid and triglyceride content of intestinal tissues was significantly lower, and cholesterol content was higher in coix oil than in lard group(p<0.05). These results indicate that pectin and lignin reduced serum cholesterol. Pectin also decreased total lipid, cholesterol and triglyceride contents of liver in rats fed either coix oil-or lard-cholesterol diets whereas lignin did not affect liver lipid content.

Optimization of Ingredients for the Preparation of Chinese Quince (Chaenomelis sinensis) Jam by Mixture Design (모과잼 제조시 혼합물 실험계획법에 의한 재료 혼합비율의 최적화)

  • This study was performed to find the optimum ratio of ingredients in the Chinese quince jam. The experiment was designed according to the D-optimal design of mixture design, which included 14 experimental points with 4 replicates for three independent variables (Chinese quince paste $45{\sim}60%$, pectin $1.5{\sim}4.5%$, sugar $45.5{\sim}63.5%$). A mathematical analytical tool was employed for the optimization of typical ingredients. The canonical form and trace plot showed the influence of each ingredient in the mixture against final product. By use of F-test, sweetness, pH, L, b, ${\Delta}E$, and firmness were expressed by a linear model, while the spreadmeter value, a, and sensory characteristics (appearance, color, smell, taste, and overall acceptability) were by a quadratic model. The optimum formulations by numerical and graphical method were similar: Chinese quince paste 54.48%, pectin 2.45%, and sugar 53.07%. Optimum ingredient formulation is expected to improve use of Chinese quince and contribute to commercialization of high quality Chinese quince jam.
