양식장 배양수를 통한 Prymnesium sp.(Haptophyta)의 혼입과 그에 의한 Isochrysis gabana의 증식 양상

  • Haptophyta(이하 하프토조로 표기)는 그 일부종이 toxin을 가지며, 더욱이 이들 종들이 원인 생물로 발생하는 bloom으로 인한 경제적 피해 발생 등으로 인해 최근 여러 나라에서 활발한 연구가 이루어지고 있다. 남해안 일대의 몇몇 어ㆍ패류 양식장에서 이용하고 있는 식물먹이생물 및 동물먹이생물의 배양수조 내에서 배양을 목적으로 하는 먹이생물 이외의 혼재 생물의 종류를 조사하던 중 Haptophyta의 Prymnesium속 생물이 혼재하고 있는 것을 발견하였으며, 혼재된 Prymnesium은 양식 패류의 인공 종묘 생산 과정에서 사육 치패의 먹이로 이용되는 식물먹이생물 Isochrysis galbana의 증식을 크게 억제하였고, 더욱이 Prymnesium이 혼재된 Isochrysis galbana를 인공 종묘생산 중이던 치패에 투이한 결과 치패 모두가 폐사에 이르는 것을 관찰하였다. 이에 Isochrysis galbana의 배양 수조내에 Prymnesium이 혼입 하였을 경우 Isochrysis galbana의 증식양상을 알아보기 위해, Isochrysis galbana의 세포밀도를 5,440,000cells/ml로 하고 Prymnesium의 세포밀도를 10,000cells/ml로 하여 혼합 배양한 결과 배양개시 1일 후의 각 세포밀도는 Isochrysis galbana가 1,040,000cells/ml로 급격히 저하하였으며, 이와는 달리 Prymnesium은 50,000cells/ml로 증가하였다. 이들 종 각각의 증가 및 감소추세는 계속되어 배양개시 5일 후에는 Isochrysis galbana가 530,000cells/ml로 처음 배양당시 세포수의 90% 이상 감소하였고, Prymnesium은 43,333cells/ml로 4배 이상 증가하였다. 이 실험에서 Isochrysis galana의 세포수가 감소하는 이유는 정확히 알 수는 없으나, 하프토조의 일부 종에서는 광합성을 통한 유기물합성 이외에도 외부로부터의 DOC(dissolved organic carbon)를 직접 배양수로부터 취하거나 또는 영양염 제한 조건에서는 food particle을 섭취하는 경우가 있는 것으로 보고되고 있다. 한편, 이들 Haptopyta가 양식장에서 이용되어지는 해수를 통해 유입되는지를 알아보기 위해 모래여과조를 통과한 해수를 플랑크톤 배양용 배지의 첨가 없이 배양한 결과, Haptophyta의 Prymnesium, Chrysochromulina 및 Phaeocystis 둥의 수 종이 출현하였으며, 일부 종의 경우는 일정기간 지속적으로 배양되어짐을 알 수 있었다.

Heavy Metals in Shellfishes Around the South Coast of Korea (우리나라 남해안산 패류의 중금속 함량)

  • The levels of heavy metals (total-mercury, cadmium, lead and chromium) in shellfishes, such as oyster (Carassostrea gigas), blue mussel (Mytilus edulis), arkshll (Scapharca broughtonii) and little neck clam (Ruditapes philippinarum), were determined. The samples were collected from 11 stations for oyster and 12 stations for blue mussel in the South Coast of Korea, and little neck clam and arkshell were collected from Koheung and the Kangjin Bay, respectively, during 1999 to 2000. The average concentrations of heavy metal in 4 different species of shellfishes analyzed were ranged from 0.007 to 0.009 ${\mu}g/g$ for total-mercury; 0.232 to 0.559 ${\mu}g/g$ for cadmium; 0.107 to 0.220 ${\mu}g/g$ for lead; and 0.116 to 0.364 ${\mu}g/g$ for chromium on wet basis. The highest level of total-mercury, lead, cadmium, and chromium in the samples analyzed were all below the quarantine limit of the Korean regulation and guideline established by the U.S.Food and Drug Administration for human consumption. The levels of heavy metals in the samples were negligible, which could be endogenous. It was elucidated that oyster was able to accumulate 2-times more cadmium than blu mussel in this study.

Detection and Analysis of the Potential Risk of VHSV in Bivalves in Korea (한국 이매패류 내 VHSV 검출 및 잠재적 위험성 분석)

  • VHSV is a major viral agent that affects freshwater and marine fish, causing serious economic losses in aquaculture in the world. Due to their filter-feeding activity, bivalve mollusks may act as viral transmitters after accumulation of the fish viruses released into seawater from infected fish. Amplification by RT-PCR was carried out to investigate the presence of VHSV in pacific oysters (Crassotrea gigas) and blue mussels (Mytilus edulis), inhabiting regions around aquatic farms in Korea. Primers designed from conserved regions of VHSVs allowed us to detect four different types of VHSV in a single PCR. Twenty two of the eighty four samples showed positive results of VHSV in a 2-step RT-PCR. Using six positive samples from three different regions in Korea, we cloned and sequenced the glycoprotein (G) gene (467-bp long) of VHSVs. Genetic analysis of the VHSVs detected in shellfish in various geographical areas of Korea showed highly restricted results to VHSV type Iva. This was in agreement with the reports showing only a single genotype of VHSV (Iva genotype) in outbreaks in cultured or wild fish in Korea. Consequently, we investigated VHSVs carried by bivalve mollusks inhabiting the vicinity of aquatic farms, and revealed correlationship between the type of viral accumulated in shellfish by filter-feeding, and those detected in disease outbreaks in fish.

Patterns in Benthic Polychaete Community and Benthic Health Assessment at Longline and Bottom Culture Shellfish Farms in Gangjin Bay, Namhae, Korea (남해 강진만 수하식 및 살포식 패류양식장의 다모류군집구조 양상과 저서생태계 건강도 평가)

  • This study was conducted to investigate the changes in the structure of benthic communities resulting from aquaculture activities and to assess the benthic health status of surface sediment in Gangjin Bay, a region known for concentrated shellfish aquaculture on the southern coast of Korea. Survey stations were divided into longline culture, bottom culture, and non-cultivation areas. The spatiotemporal distribution of physiochemical factors such as the grain size, water temperature, salinity, and total organic carbon in Gangjin Bay showed no significant differences between sampling stations. However, the species number, density, and diversity were relatively lower at the sampling stations in the bottom culture areas than at the other stations throughout the entire survey period. Cluster analysis and principal coordinates analysis also clearly distinguished the benthic communities in the bottom culture areas from those in the other sampling areas. At the sampling stations in the longline culture and non-cultivation areas, Scolectoma longifolia and Sigambra tentaculata, which are indicator species of organically enriched areas, appeared as dominant species. However, excluding some stations influenced by physical factors such as the water depth and current speed, the occupancy rate was not high. The health assessment results, conducted using the fisheries environment assessment method, revealed good conditions with Grades 1 and 2 across the entire area. However, an examination of the spatiotemporal changes in benthic communities and the benthic health index indicated that the benthic environment in the bottom culture areas was affected by physical disturbances.

Pollution Status of Surface Sediment in Jinju Bay, a Spraying Shellfish Farming Area, Korea (살포식 패류양식해역인 진주만 표층 퇴적물의 오염도)

  • We investigated the concentrations of acid volatile sulfide (AVS), ignition loss (IL), total organic carbon (TOC), total nitrogen (TN), and metallic elements (As, Cd, Cr, Cu, Fe, Hg, Mn, Pb, and Zn), in August 2015, to determine the spatial distribution and pollution status of organic matter and metals in the surface sediment of Jinju Bay, a spraying shellfish farming area, Korea. The concentrations of organic matter and metallic elements were significantly higher in the southern part of the bay than in the mouth and center of the bay. The C/N ratio (5.7~8.0) in the sediment represents the dominance of organic matter of oceanic origin in the surface sediment of the study area. The concentrations of AVS, TOC, and metals (As, Cd, Cr, Cu, Hg, Pb, and Zn) were much lower than the values of the sediment quality guidelines applied in Korea. Based on the results of the pollution load index (PLI) and ecological risk index (ERI), the metal concentrations in the surface sediment of Jinju Bay have a weakly negative ecological effect on benthic organisms although the sediments with high metal pollution status are distributed in the southern parts of the bay, with high dense shellfish farming areas. Thus, the surface sediments in Jinju Bay are not polluted with organic matter and are slightly polluted with metallic elements.

Establishment of Environmental Assessment using Sediment Total Organic Carbon and Macrobenthic Polychaete Community in Shellfish Farms (퇴적물 총유기탄소와 저서 다모류 군집을 이용한 패류양식장의 환경평가 기준설정)

  • In this study, the seasonal field survey was conducted in Goseong Jaran Bay(22 stations), Geoje Hansan Bay(15 stations) and Jinhae Bay(18 stations). We analyzed the sediment environmental parameters(Chemical Oxygen Demand, Ignition Loss, Acid Volatile Sulfides, Total Organic Carbon) and biotic parameters of macrobenthic polychaetes(number of species, density, diversity, evenness). It had a good correlation between total organic carbon and polychaete diversity(R=0.61, P<0.01), and we made a decision them as representative environmental indices. As a result of that, regarding the criteria in the assessment of farm environment, we suggest concentrations of total organic carbon : Peak Point = 15 mg/g dry, Warning Point = 26 mg/g dry, Contaminated Point = 31 mg/g dry and polychaete diversity : ~2.6(Good), 2.6~2.1(Moderate), 2.1~1.2(Poor) and 1.2~(Bad). This could be a scientific basis to establish the environmental standards for fishery management.

  • 1. The basic investigations on soil textures, water holding capacity, pH, exchangeable calcium, and organic matter contents in four species bed were conducted in order to improve the natural flat for the bivalve beds. 2. It was statistically significant that there were differences in the soil textures among all beds of four species, D. japonica, C. sinensis, M. veneriformis, and T. philippin-arum. 3. There was no significant difference in water holding capacity, pH, and exchange-able calcium contents among different bivalve beds statistically, 4. It was highly significant that there were diffences in soil organic matter content among all groups of beds except that of D. japonica - C. sinensis. 5. It is considered that that soil textures and soil organic matter contents are most important factors to restrict the distribution of each species bivalve within a certain limited area of tidelands.

큰구슬우렁이, Glossaulax didyma의 생식주기

  • 큰구슬우렁이 (Glossaulax didyma)는 복족강 (Gastropoda), 중복족목(Gastropoda), 구슬우렁이과 (Naicidae)에 속하고 내만의 조간대에서 수심 10m 가까이의 진흙과 모래가 섞인 조하대애 묻혀 살며 일본과 우리나라 전역에 분포한다 (유, 1976). 큰구슬우렁이는 이매패나 복족류의 껍질에 구멍을 뚫어 섭식하는 육식성패류로 패류양식장 등의 해적생물로 알려져 있다 (유, 1976). 그러나 이들은 식용으로 이용되고 있어 지방에 따라 배꼽고둥, 배꼽, 우렁이, 잠빼기, 말띠, 미소라, 개소랑, 명지고둥, 가내기, 해구 등으로 폭넓게 불려진다. 이와 같이 식용으로 널리 이용되고 있는 큰구슬우렁이는 산업종으로 중요한 위치에 있어 자원관리적 측면에서 이들의 생식생물학적 조사가 조속히 이뤄져야 할 종이다. The characteristic of photosynthetic pigments distribution of the sediment in the shellfish farm (패류 양식장 퇴적물의 광합성색소 분포 특성)

  • This study researched and compared the sedimentation environment and photosynthetic pigments at Gomso (GS) -bay, Yoja (YJ) -bay and KangJin (KJ) -bay in May, 2012. It was shown that KJ-bay consist of C (clay) and M (mud), and GS-bay consists of Zs (silty sand) and Sz (sandy silt). Averagely, IL of YJ-bay was 4.98%, KJ-bay 6.10%, and GS-bay 1.45%. As for COD concentration, there were no places that exceeded Japanese sediment contamination standard 20 mg/g-dry. As for AVS concentration, in case of KJ-bay, two places exceeded Japanese sediment contamination standard 0.2 mg/g-dry. If we look into the average C/N ratio of bays, YJ-bay showed 8.50, KJ-bay 6.60, and GS-bay 5.52, thus all of them showed the characteristic of oceanic origin. As for the plankton make-up classified by photosynthetic pigments, diatom was dominant, and both ratios of pigment and C/chlorophyll. a showed the relatively lower distribution at GS-bay than at KJ-bay and YJ-bay. It is judged that this is related to the characteristic of flow-in, and it implies that predation process and decomposition by organic matters actively occurs at YJ-bay and KJ-bay.

진해만내 진주담치 양식장에서의 생산력 평가

  • 진주담치는 전세계 온대 수역에 분포하는 법세계종(Cosmopolitan species)으로 연안을 따라 l0m 이내 수심에 대량 번성함으로서 이젠 오손생물(Fouling organism)로 알려진 종이다. 하지만 양식도 활발히 진행되고 있어 1990년대에는 35,000 여톤이 생산되어 패류 양식생산량의 10% 내외를 차지하기도 한다. 진해만에서의 진주담치는 오랜 기간 양식이 진행되면서 시기에 따라 다량의 유생이 분포하는 지역으로 특별한 종묘 생산 없이도 양식이 원활한 지역이다. (중략)
