  • 소비자와 가공업체가 원하는 다양한 색깔의 양갱과 앙금 제품 개발에 붉은색 팥은 제품 다양화에 한계가 있었다. 이에 흰앙금 제조가 가능한 황백색 종피의 품질이 우수한 팥 품종을 개발하기 위하여 노력한 결과, 국립식량과학원에서 흰앙금제조가 가능한 신품종 '흰나래' 팥을 개발하였다.

Differences in Yields, Antioxidant Compounds, and Antioxidant Activity of Ethanolic Extracts among 11 Adzuki Bean Cultivars (Vigna angularis L.) Cultivated on a Somewhat Poorly Drained Paddy Field (논 재배 팥 품종별 수량구성요소 및 에탄올 추출물의 항산화 성분 비교)

  • This study investigated the changes in yields, antioxidant compounds, and antioxidant activities among 11 adzuki bean cultivars cultivated in a paddy field with somewhat poorly drained soil. The adzuki bean cultivars were cultivated in the paddy field from 2015 to 2016 in Milyang, Gyeongsangnam, Korea. Average soil moisture content was 16.5% in 2015 and 31.0% in 2016 at the experimental site during adzuki bean cultivation. As the soil moisture content increased, most of the adzuki bean cultivars showed deceases in stem height, first setting pod node, number of pods, 100 seed weight, and yield. Chungju-pat produced the greatest yields among the 11 cultivars in 2015 and 2016, whereas Hongeon had the smallest yields. Adzuki bean yields after paddy field cultivation was lower for all cultivars than for adzuki bean yields from the same cultivars after upland field cultivation. Chungju-pat and Chilbo-pat showed the smallest decreases in yields after paddy field cultivation, whereas Hongeon and Kumsil showed the greatest yield reductions. There were no significant differences in proximate composition. Some mineral components ($P_2O_5$, Ca, and Mg) were statistically different across cultivars. Chungju-pat had the highest Ca and Mg contents, but antioxidant components (polyphenol and flavonoids) and antioxidant activities (ABTS and DPPH) were highest in Saegil and lowest in Jungbu-pat. PCA and clustering analyses, based on the growth, yield, and antioxidant component measurements, performed to identify which variables contributed the most to separating adzuki bean cultivars or to grouping cultivars with similar characteristics. These analyses showed that the antioxidant components and antioxidant activities had the most influence on grouping cultivars together. Among the 11 cultivars, Saegil was statistically different from the other cultivars, but the other 10 cultivars were not significantly different under paddy field cultivation. Soil moisture content affected adzuki bean yield and antioxidant component contents. An increase in soil moisture led to a decrease in yield, but an increase in antioxidant components. These results provide information that will improve the selection of an appropriate adzuki bean cultivar for use in paddy fields.

Identification and Characterization of Protease-Resistant Proteins from Adzuki Beans (소화 효소 저항성을 지니는 팥 단백질의 성질 규명)

  • It is already known that adzuki beans (Vigna angularis) are able to control appetite. Therefore, this study tested the proteins isolated from adzuki beans for their protease resistance and interaction with the intestinal mucosa. The major proteins from adzuki beans were found to be resistant to the digestive enzymes pepsin and pancreatin, and were identified using 2D-SDS-polyacrylamide gel electrophoresis and mass spectrometry. The major adzuki proteins were easily fractionated by treating the soluble protein extract with 10mM $CaCl_2$, and were found to contain lactotransferrin, a homologous protein to the dynein light chain domain, proteinase inhibitor, and proteins with unknown functions. From a tissue binding assay using mouse intestinal tissue sections, the major protein fraction showed weak, yet significant and specific binding to the mucosa layer of the small intestine. Thus, the current results suggest that adzuki proteins are resistant to digestive enzymes, which enables them to survive protease digestion in the intestinal tract, plus they may interact with the intestinal mucosa layer. Therefore, the molecules responsible for controlling appetite in adzuki beans are presumably protease-resistant proteins that interact with the intestinal mucosa or delay digestion in the digestive tract.

A Comparison of the Components and Biological Activities in Raw and Boiled Red Beans (Phaseolus radiatus L.) (생팥과 증자팥의 성분 및 생리활성 비교)

  • In the course of study for the development of functional food using red beans (azuki beans, Phaseolus radiatus L.), the ethanol extracts from raw-red bean (RRB) and boiled-red bean (BRB) were prepared, and the components and various biological activities of both were compared. It was observed that the extraction yield, and the total polyphenol content, of the BRB were 1.2 times higher than that of the RRB. However, the contents of total flavonoid, total sugar and reducing sugar in the BRB were 30, 27.9 and 30.8% respectively when compared with those of RRB. In relation to antioxidative activity, both RRB and BRB exhibited moderate DPPH anion, ABTS cation, and nitrite scavenging activities and reducing power, though in all cases RRB demonstrated stronger activities than BRB. The extracts of RRB and BRB did not reveal any antimicrobial activities. In a ${\alpha}$-amylase inhibitory activity assay, RRB was higher than BRB, while BRB showed higher ${\alpha}$-glucosidase inhibitory activity than RRB. A strong and particular activity was observed in an anti-thrombosis activity assay of RRB. The extract of RRB demonstrated strong inhibitions against prothrombin and blood coagulation factors, with moderate thrombin inhibition. However, the extract of BRB did not exhibit any significant anti-thrombosis activity. Our results indicate that RRB has different, but useful biological activities, and loss or elimination of the biologically active substances in RRB occurs during the production of BRB. Therefore, to develop more functional foods from red beans, a study of suitable boiling, heating and drying processes is essential, and the efficient re-use of boiled waste water from the boiling process is necessary. These results could be applied to the further development of functional red bean beverages and sweat red bean pastes.

  • Some physicochemical properties and molecular structure of cow pea, mung bean and acorn starches(mook-forming starches) and red bean, wheat and sweat potato starches(mook-unforming starches) were investigated. Amylose contents of cow pea, mung bean and acorn starch were higher than the others. Cow pea starch was similiar to mung bean starch in gelatinization characteristics by Brabender amylogram but cold viscosity of red bean starch and peak viscosity of sweet potato starch were especially high. Whereas viscosity of wheat starch was low in whole temperature range. Amylose molecules of larger molecular size$(above\;5{\times}10^5\;molecular\;weight)$ of three mook-forming starches were more than shoes of red bean and wheat starch. Chain distribution ratios$(DP\;35{\sim}55\;to\;DP\;10{\sim}20)$ of cow pea, mung bean and acorn amylopectin were higher than thoes of red bean, sweet potato and wheat amylopectin.

A Study of Whitening Functional Activity Verification from Red Bean (Phaseolus angularis) Shell Extract (팥 껍질 추출물의 미백 기능성 활성 검증)

  • Whitening effect of the extract of Phaseolus angularis (PA) shell was investigated for its potential application as a functional ingredient for cosmetic products. The tyrosinase inhibitory effect, which is related to whitening, was 76.4% at a concentration of 1,000 ㎍/ml. Cell viability of melanoma cells was measured to test toxicity of the PA shell extract at concentrations showing at least 90% viability. In addition, western blot and RT-PCR assays of the PA shell extract showed concentration-dependent decreases of MITF, TRP-1, TRP-2 and tyrosinase protein and inhibitory effect of mRNA expression. These findings suggested that extract from PA shell has great potential as a cosmeceutical ingredient with whitening effect.

Variation in Physicochemical Characteristics and Antioxidant Activities of Small Redbean Cultivars (팥 품종의 이화학적 특성 및 항산화 활성 변이)

  • This research was conducted to evaluate the physicochemical properties, antioxidant components, and their activities for more taking advantage of small redbean cultivars. Seed size, 100 seeds weight, and hardness on the 8 cultivars were measured. The free sugar and crude protein contents were evaluated using HPLC and protein analyzer, respectively. Amylose content, antioxidant components, and activities were analyzed by spectrophotometer. The range of 100 seeds weight and hardness were 12.55-18.81 g and 9,527.38-14,341.25 gf, respectively. Total free sugar, amylose, and crude protein were showed 22.49-31.07 mg/g, 13.53-15.67%, and 21.27-23.30%, respectively. The cultivar 'Hongeon' was higher antioxidant component and activity more than others. In clustering the cultivars based on the results, the tree showed four major clades. The 'Huinnarae' group was high in total free sugar and amylose content. The 'Hongeon' group were high in 100 seeds weight, antioxidant component. and activity, while amylose content was lower than that in the other groups. The results of the cultivars can be utilized for research of functional materials. The findings of this study will provide valuable information for expansion of functional food industry related on small redbean.

Water-Soluble Components of Small Red Bean and Mung Bean Exposed to Gamma Irradiation and Methyl Bromide Fumigation (감마선과 Methyl Bromide 처리된 팥과 녹두의 수침에 따른 용출 특성)

  • Comparative effects of gamma irradiation and methyl bromide (MeBr) fumigation on water-soluble components of small red bean and mung bean were investigated. The levels of soluble solid and reducing sugar of soaked water at $20^{\circ}C$ for 16 h definitely increased as irradiation dose increased, which was more apparent in small red bean than in mung bean. But, their levels of MeBr-fumigated sample showed a similar result to those of the control, except for soluble solid of small red bean. The pH of soaked water was insignificantly different among treatments. The decrease in lightness (L value) and the increases of redness (a value) and yellowness (b value) were remarkable as irradiation dose increased, which was more significant in fumigated samples. The ${\Delta}E$ values of fumigated sample showed almost equaled to those of 10 kGy in red bean and 30 kGy in mung bean, respectively. Gamma irradiation caused the increase in free amino acids of soaked water, that mainly consisted of asparagine, glutamic acid, and valine, while fumigation reduced their contents. The extractables in soaked water, such as soluble solid and reducing sugar, showed a highly positive correlation with irradiation dose applied.

A New Small Redbean Cultivar 'Geomguseul' with Lodging Resistance and High-Antioxidant Activity (쓰러짐에 강하고 항산화 활성이 높은 팥 '검구슬')

  • A new small red bean cultivar, 'Geomguseul', was artificially crossed between 'Chilbo' and 'SA8412-3-1-4-3-3-2-3' in 1998, fixed excellent agronomic characters by pedigree breeding method, and selected for the further trials with the name of 'Miryang 10'. It was prominent and showed good result, such as high grain quality, lodging resistance, and high-antioxidant activity in the regional adaptation yield trials (RYT) for three years from 2009 to 2011 and released for the public consumption as the name of 'SA9801-24-4-3-1' in 2011. 'Geomguseul' has a semi-determinate growth habit, yellow flower, green embryonic axis, black seed coat, white hilum, and small spherical seed (12.2 grams per 100 seeds). The average yield of 'Geomguseul' was 2.01 MT/ha in the regional yield trials (RYT) carried out for three years from 2009 to 2011 which was a little higher than that (1.98 MT/ha) of the check variety, 'Chungju'.
