Effects of Albizziae Cortex Pharmacopuncture Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity (합환피(合歡皮) 약침액(藥鍼液)의 사람 피부아세포의 콜라게나제 활성 및 프로콜라겐 합성과 티로시나제 활성에 미치는 영향)

  • 목적 : 본 연구는 합환피약침액(合歡皮藥鍼液)이 사람 피부 섬유아세포의 콜라게나제 활성 및 프로콜라겐 합성에 미치는 영항과 티로시나제 활성에 미치는 효과를 측정하고자 실시하였다. 방법 : HS68 사람 정상 섬유아세포에 UVB 조사 후 합환피(合歡皮) 약침액(藥鍼液)가 type I procollagen 생성과 콜라게나제 효소활성에 미치는 효능과 티로시나제 효소활성에 미치는 효능을 평가하였다. 결과 : 합환피약침액(合歡皮藥鍼液)은 UVB 조사된 세포의 콜라게나제 효소활성을 통계적으로 유의하게 억제하였고, 티로시나제 활성을 통계적으로 유의하게 억제하였다. 그러나 티로시나제 억제활성의 정도는 미백효능으로 활용하기에 약간 약한 경향이 있었다. 결론 : 합환피약침액(合歡皮藥鍼液)의 콜라게나제 억제효능은 주름개선 약침치료에 활용이 가능할 것으로 생각된다.

  • 본 연구는 육미지황환(六味地黃丸) 약침액(藥鍼液)가 사람 피부 섬유아세포의 콜라게나제 활성 및 프로 콜라겐 합성에 미치는 영항과 티로시나제 활성에 미치는 효과를 측정하고자 실시하였다. 방법 : HS68 사람 정상 섬유아세포에 UVB 조사 후 육미지황환(六味地黃丸) 약침액(藥鍼液)가 type I procollagen 생성과 콜라게나제 효소활성에 미치는 효능과 티로시나제 효소활성에 미치는 효능을 평가하였다. 결과 : 육미지황환(六味地黃丸) 약침액(藥鍼液)은 UVB 조사된 세포의 콜라게나제 효소활성을 통계적으로 유의하게 억제하였고, 티로시나제 활성과 L-DOPA oxidation 활성 또한 통계적으로 유의하게 억제하였다. 그러나 티로시나제와 L-DOPA oxidation 억제활성의 정도는 미약하였다. 결론 : 육미지황환(六味地黃丸) 약침액(藥鍼液)의 콜라게나제 억제효능은 주름개선 약침 치료에 활용이 가능할 것으로 생각된다.

Whitening Effect of the Extracts from Juglans mandshurica (가래나무 추출물의 미백효과)

  • Juglans mandshurica belongs to the family Juglandaceae is known to contain a wide range of pharmacological activities including anti-cancer, anti-inflammation, astringent, and anti-human immunodeficiency virus-type 1 (HIV-1). Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. In this study, to investigate the whitening activity of the extracts from Juglans mandshurica, we measured effects on a tyrosinase activity, a melanogenesis, and a tyrosinase synthesis in the B16/BL6 melanoma cells and an antioxidant activity. The extracts significantly scavenged a 1,1-diphenyl-2-picrylhydrazyl (DPPH) and a superoxide anion radicals in a dose-dependent manner with a $SC_{50}$ value of $20\;{\mu}g/mL$ and $25\;{\mu}g/mL$, respectively. Also, the tyrosinase activity and melanogenesis were significantly inhibited by the extracts. Furthermore, the synthesis of tyrosinase protein was significantly decreased by the extracts in enzyme-linked immunosorbent assay. Double blind study on the clinical efficacy of a cream containing 2% of the extracts showed that the extracts have a significant skin whitening effect. Therefore, this study demonstrates that the extracts from Juglans mandshurica may be useful as a potential agent for skin whitening.

Study on the Effects of the Extracts from Sophora Japonica L. Flowers, Fruits and Branches on Improvements in Skin Condition (회화나무 꽃, 열매, 가지 추출물의 피부개선 효과에 관한 연구)

  • The purpose of this research was to examine the effect of the ehanol extracts of Sophora japonica L. (S. japonica). flowers, fruits and branches on skin enhancement with assessing anti-oxidative, whitening, and wrinkle enhancement effects. Results showed that l,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activities were $17.68{\pm}1.59{\sim}51.40{\pm}1.04$, $27.48{\pm}0.22{\sim}50.89{\pm}0.13$ and $30.79{\pm}0.55{\sim}45.17{\pm}0.83%$, respectively, in 50~1,000 mg/L of concentrations. The capacities of inhibiting tyrosinase of ethanol extracts from S. japonica. flowers, fruits and branches were $0.27{\pm}0.12{\sim}11.38{\pm}0.57$, $0.27{\pm}0.02{\sim}0.82{\pm}0.27$ and $0.09{\pm}0.16{\sim}0.55{\pm}0.27%$, respectively. The capacities of preventing porcine pancreatic elastase (PPE) were $3.70{\pm}1.23{\sim}7.28{\pm}1.01$, $3.06{\pm}2.13{\sim}13.03{\pm}2.99$ and $6.00{\pm}0.96{\sim}9.71{\pm}0.44%$, respectively, in the case of 50~1,000 mg/L of concentrations. It is concluded that the effects of S. japonica. flowers, fruits and branches on skin improvement are varied significantly.

Whitening Effect of Angelica keiskei Koidzumi Extract by Inhibition of Endothelin-1 Production and Melanogenesis (Endothelin-1 생성 저해와 멜라닌생성 저해에 의한 신선초 추출물의 미백 효과)

  • In order to investigate the potential of a Angelica keiskei Koidzumi extract and its fractions as an active ingredient for whitening cosmetics, we prepared Angelica keiskei Koidzumi extract(70% ethanol), and the aqueous suspension was successively extracted with hexane, ethyl acetate(EA), and n-butanol fraction. We measured their inhibitory effects on mushroom tyrosinase and melanin synthesis in B16 melanoma cells and normal human kerationocytes in vitro. They did not show inhibitory activity against mushroom tyrosinase and the melanin synthesis except hexane and EA fractions. Hexane and EA fractions markedly inhibited cellular tyrosinase activity at a lower concentration(25 and 5 ${\mu}g/mL$, respectively) than arbutin(250 ${\mu}g/mL$). We also quantified the released amount of endothelin-1(ET-1), a mitogen of melanocyte, and interleukin-$1{\alpha}$(IL-$1{\alpha})$, a mediator of UVB-induced inflammation. Hexane and EA fractions did not affect IL-$1{\alpha}$ production, but they decreased UVB-induced ET-1 production in normal human keratinocytes in a dose dependant manner. As a result, hexane and EA fractions could inhibit the melanogenesis through the inhibition of UVB-induced ET-1 production in normal human keratinocytes. This result suggests that hexane and EA fractions of Angelica keiskei Koidzumi extract could be used as an active ingredient for cosmetics.

Synthesis of Novel Kojic Acid Derivatives and Their Tyrosinase Inhibitory Activities (새로운 코직산 유도체의 합성과 티로시나제 저해활성)

  • Four derivatives of kojic acid were synthesized and their inhibitory activities against tyrosinase were evaluated. The C-2 hydroxymethyl and C-7 hydroxyl of kojic acid were replaced by carboxylate and amine, respectively. These derivatives were coupled to L-phenylalanine, producing two amide compounds. The carboxylate derivative (3), its amide compound (5), and the amine derivative (7) were weak inhibitors. The amide compound (9) where amine derivative (7) coupled to L-phenylalanine showed strong inhibitory activity ($IC_{50}=24.6{\;}{\mu}M$) comparable to kojic acid.

Cosmetic Effect of Mixed Plant Extracts Including Saururus Chinensis, Morus Bombycis Stem and Morus Papyrifera Stem (삼백초, 뽕나무 줄기, 닥나무 줄기 등을 함유한 복합수목추출물의 화장품소재 특성)

  • In this study, plant extracts including 25% Saururus chinensis, 20% Morus bombycis stem and 20% Morus papyrifera stem were tested to use as possible cosmetic agent. As cosmetic effect test, cell toxicity test, elastase inhibition assay, tyrosinase inhibition assay, anti-oxidation assay and temperature stability test were done as cosmetic assays. Plant extract showed low cell toxicity, excellent elastase inhibition and anti-oxidation effect, however, tyrosinase inhibition effect was lower. Skin, lotion and essence formulation was made using plant extracts and temperature stability test was done. Skin and essence indicated good stability, however, lotion showed phase separation and severe viscosity variation, which means lotion formulation change. From the research, plant extracts including Saururus chinensis, Morus bombycis stem and Morus papyrifera stem were suitable for possible cosmetic agent.

Potent whitening effects of rutin metabolites (루틴 대사체의 미백 효능)

  • The aim of this research was to investigate the whitening effects of rutin and rutin metabolites including 3,4-dihydroxyphenyl acetic acid (DHPAA), 3-hydroxyphenyl acetic acid (HPAA), 3,4-dihydroxytolene (DHT) and homovanillic acid (HVA). The potent whitening effect of rutin and rutin metabolites were determined by mushroom tyrosinase inhibition assay and expressed as the half maximal inhibitory concentration ($IC_{50}$) against tyrosinase activity in vitro. The HVA showed the highest inhibitory effect ($IC_{50}=37.10{\mu}M$) of tyrosinase activity, followed by DHPAA ($IC_{50}=45.87{\mu}M$), HPAA ($IC_{50}=50.96{\mu}M$), rutin ($IC_{50}=57.98{\mu}M$), and DHT ($IC_{50}=66.09{\mu}M$), respectively. To evaluate cell cytotoxicity, MTT assay was performed with JB6 P+ mouse epidermal cells and expressed as a relative percentage of untreated control. The results showed that rutin and rutin metabolites had no cytotoxic effects on JB6 P+ cells up to $100{\mu}M$ except for DHT (up to $50{\mu}M$). These results suggests that rutin metabolites may be utilized as a potential tyrosinase inhibitors and the whitening agents for the future.

Inhibitory Effects of Fractions from Glycine soja Siebold et Zucc. on Melanogenesis in B16F10 Melanoma Cells (B16F10 멜라닌 세포에서 약콩(Glycine soja Siebold et Zucc.) 분획 추출물의 멜라닌 생성 저해 효과)

  • This study was performed to cytotoxicity, tyrosinase inhibition activity, intracellular melanin contents to verify the whitening effect of fraction from Glycine soja Siebold et Zucc. (G. soja). Using western blotting, tyrosinase expression in B16F10 melanoma cells and expression levels of tyrosinase related protein-1 (TRP-1) and protein-2 (TRP-2) were examined. As a result, all of the fractions showed a high cell viability over 82% at the concentrations of 0.125, 0.25, 0.5, 2.0 mg/mL. When the whitening effects of fractions from G. soja were tested using B16F10 melanoma cells treated with the ${\alpha}$-melanocyte stimulating hormone (${\alpha}-MSH$), the EtOAc fractions inhibited tyrosinase and melanogenesis effectively. The result of protein expression measurement using western blot showed that TRP-1, TRP-2 and tyrosinase protein expression in B16F10 melanoma cells treated with extracts decreased. Therefore, it is concluded that the fractions from G. soja have whitening effect by inhibiting protein related melanogenesis.

Screening of Inhibitors against Tyrosinase Activity from Natural Products (천연물로부터 티로시나제 활성 저해제의 검색)

  • Tyrosinase plays an important role in the process of melanin biosynthesis, and it is a biochemical target enzyme for skin-whitening agents and the remedy for disturbances in pigmentation. We have screened 25 natural plant extracts for inhibitory effects against DOPA oxidase activity of tyrosinase. For the inhibitory effect against DOPA oxidase activity of tyrosinase, the recombinant human tyrosinase and the purified mushroom tyrosinase were used. Each of the dried plants extracted with methanol, and then the extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol and water, respective The methylene chloride fractions of Angelica tenuissima, Nardostachys chinensis, Bombyx mop and Saposhnikovia divaricata, and the n-butanol fraction of Bombyx mori notably inhibited the human tyrosinase activity as well as mushroom tyrosinase activity (more than 90% inhibition). This study suggests that the above extracts have a potential as whitening agents as single ingredients or in combination with other of the extracts.
