A Study on the R&D Trend and Patent Analysis of Treatments for Degenerative Brain Diseases (퇴행성 뇌질환 치료제의 연구개발 및 특허동향 분석)

  • Degenerative brain diseases including Alzheimer's diseases, Parkinson's diseases increase in frequency with age. They are amongst the most costly and devastating diseases to patients and their families. Therefore developing therapies for degenerative brain diseases is of the highest priority. Recently therapeutics for these diseases have undergone scrutiny by many clinical trials according to the advances of cellular and molecular neurobiology. This review is focused on studies investigating the current therapeutic strategies already undergone different stage of clinical trials and recent R&D trend by nations through patent analysis on treatments for degegerative brain diseases.

  • The incidence of neurodegenerative diseases in the older population has increased in recent years. A considerable number of studies have been performed to characterize these diseases. Imaging analysis is an important biomarker for the diagnosis of neurodegenerative disease. Objective and reliable assessment and precise detection are important for the early diagnosis of neurodegenerative diseases. Artificial intelligence (AI) using brain MRI applied to the study of neurodegenerative diseases could promote early diagnosis and optimal decisions for treatment plans. MRI-based AI software have been developed and studied worldwide. Representatively, there are MRI-based volumetry and segmentation software. In this review, we present the development process of brain volumetry analysis software in neurodegenerative diseases, currently used and developed AI software for neurodegenerative disease in the Republic of Korea, probable uses of AI in the future, and AI software limitations.

The role of p62 in ceramide induced neuronal cell death (Ceramide에 의한 신경세포 사멸과정에서 p62의 역할)

  • p62 is a key component of protein aggregates found in brains of neurodegenerative diseases in which oxidative stress is involved in the pathogenesis. p62 was induced in SH-SY5Y, a neuroblastoma cell line, by hydroxydoparnine or $C_2-ceramide$ known to be related to neurodegenerative diseases. The over-expression of p62 showed the neuroprotective effect against the ceramide induced cell death. In addition, p62 became insoluble and cleaved forms as time proceeded after the ceramide treatment, suggesting the mechanism by which p62 is associated with aggregates in neurodegenerative diseases.

Recent Updates on PET Imaging in Neurodegenerative Diseases (퇴행성 뇌질환에서 PET의 발전과 임상적 적용 및 최신 동향)

  • Over the past decades, the immense clinical need for early detection methods and treatments for dementia has become a priority worldwide. The advances in PET biomarkers play increasingly important roles in understanding disease mechanisms by demonstrating the protein pathology underlying dementia in the brain. Amyloid-β and tau deposition in PET images are now key diagnostic biomarkers for the Alzheimer's disease continuum. The inclusion of biomarkers in the diagnostic criteria has achieved a paradigm shift in facilitating early differential diagnosis, predicting disease prognosis, and influencing clinical management. Furthermore, in vivo images showing pathology could become prognostic as well as surrogate biomarkers in therapeutic trials. In this review, we focus on recent developments in radiotracers for amyloid-β and tau PET imaging in Alzheimer's disease and other neurodegenerative diseases. Further, we introduce their potential application as future perspectives.

  • 최근 교통사고의 증가와 평균수명의 연장으로 인한 노인 인구의 급증으로 중추신경계의 기능에 손상을 초래하는 각종 질환들 즉 뇌졸중, 외상성 뇌손상, 뇌성마비 및 퇴행성 뇌질환 등이 더욱 많아지고 있다. 이러한 질환들은 뇌기능 장애로 인한 운동, 감각 및 인지능력 저하를 일으켜 보행과 일상생활 동작수행에 큰 장해를 초래하게 된다. 특히 보행능력의 장해는 일상생활 및 사회생활에 필요한 기동력을 제한하므로 결국 타인에 의존적이 되고 사회로부터 고립되는 심각한 기능장애를 일으키게 된다.(중략)

  • Iron has a vital role in the human body, including the central nervous system. Increased deposition of iron in the brain has been reported in aging and important neurodegenerative diseases. Owing to the unique magnetic resonance properties of iron, MRI has great potential for in vivo assessment of iron deposition, distribution, and non-invasive quantification. In this paper, we will review the MRI methods for iron assessment and their changes in aging and neurodegenerative diseases, focusing on Alzheimer's disease. In addition, we will summarize the limitations of current approaches and introduce new areas and MRI methods for iron imaging that are expected in the future.
