  • Sparassis latifolia is one of the most expensive mushrooms in Korean market owing to its high ${\beta}$-glucan content and immunoactivity. However, because of the long cultivation period and high contamination rates, it has low production efficiency. Therefore, we first need to establish the optimum conditions for liquid spawn production to increase its production efficiency. As a result of experiments, molasses culture medium was selected for mycelial growth. Also, the optimum sugar content for molasses and amount of aeration used were approximately 8 Brix% and 0.3~0.6 vvm, respectively. Mycelial dry weight increases, while the medium decreases, as the incubation period increases. Therefore, to achieve maximum production efficiency, the incubation period of 9 to 11 days is appropriate.

Performance of an Intermittent Aerated Pilot-scale Reactor Vessel for Commercial Composting (상업용 퇴비화를 위한 간헐통기식 파이로트 규모 반응조의 성능)

  • Mixtures of hog manure slurry and sawdust were composted by an intermittent aeration method to verify the performance evaluation of pilot-scale reactor vessels during composting high rate (decomposition) process. Instrumentation was designed to measure temperatures in compost, oxygen and carbon dioxide concentration, air flow rates, and ammonia gas emitted. It was found that ammonia concentration during composting high rate decreased more quickly to the allowable range of 34-40 ppm after 14days at near the optimal levels (II) than in the case of lower levels (I). The influence of the optimal levels (II) such as moisture content (55-65%), C/N ratio (20-40), pH (7-8) and temperature in compost (<$60^{\circ}C$) on the reduction of ammonia gas was considerable for commercial composting.

  • To enhance the productivity of recombinant protein, ${\beta} -glucuronidase(GUS)$, by transgenic Nicotiana suspensions, perfusion culture was carried out in a 5-L stirred tank bioreactor. Maximum cell density of 20.0 gDCW/L was obtained and GUS activity was noticeably affected by medium composition, such as salt concentration. Effect of headspace purging by $CO_2-enriched$ air was also investigated. Aeration of $CO_2(5%)-enriched$ air with 0.2 vvm significantly promoted the cell growth in lag pahse and increased volumetric GUS activity from 0.69 U/mL to 5.76 U/mL. In contrast, aeration of $CO_2(5%)-enriched$ air with 0.1 vvm did not affect the cell growth but enhanced the GUS acitivity from 3.24 to 5.25 U/mL at the 4th day.

An experimental analysis of the sound reduction characteristics of air transparent noise barriers (통기형 방음벽의 음향감쇠 특성에 대한 실험적 분석)

  • The present study describes the acoustical characteristics of the new noise barriers which can control not only noise but also wind pressure by allowing air flow through barriers. In order to investigate the sound reduction index of the air transparent noise barrier, 17 models in total were examined with various size of openings and the volume of the resonators. As a result, it was found that the sound reduction index varies with the volume of the resonator and the area of the openings. Also, it was revealed that double layer of units has more sound reduction index than the single layer of unit at the frequency band from 400 Hz to 1250 Hz. This denoted that physical features of openings and resonators affect the sound reduction index of the air transparent noise barrier.

Biodegradation of JP-8 in soil column by Rhodococcus fascians isolated from petroleum contaminated soil (유류 오염 토양에서 분리된 Rhodococcus fascians 를 이용한 토양 column에서의 JP-8의 분해)

  • The environmental contamination by organic pollutants is a widespread problem. The most widely distributed pollution can be attributed to oil contamination. Bioremediation, the use of microorganism or microbial processes to degrade environmental contaminant, is one of the new technologies. The objective of the present study is to study the degradation of JP-8 in soil by microorganism. The degradation of JP-8 was analysed by TPH using gas chromatography. Rhodococcus fascians isolated from the petroleum contaminated site was applied for the degradation of JP-8 in the soil column system. Air flow rate of 30 ml/min was sufficient to degrade JP-8 in the soil column as much as 70% of JP-8 in the soil column. The addition of nitrogen source resulted in the increase in JP-8 degradability to 75% of JP-8 and the C:N ratio for JP-8 degradation was 100:10.
