Pyrocatechase as a phenolytic dioxygenase was extracted from the benzoate-induced cells of a soil bacterium, a member of Pseudomonadaceae, and purified partially by DEAE-cellulose ion-exchange chromatography and Sephadex G-75 gel filtration. Final preparation of the enzyme yielding 200 fold purification over the crude extracts showed a specific activity of about 40 ${\mu}moles$ per minute per mg protein based on catechol as the substrate. The enzyme showed a very limited substrate specificity towards catechol for its catalytic activity. Based on the inhibition study with the substrate analogues, it was assumed that ortho dihydroxy groups on the aromatic ring may participate in the enzyme-substrate binding. The $K_m$ value for catechol was obtained as $1.9{\times}10^{-6}M$, and the optimum activity of the enzyme was obtained at the pH range of 7∼10 and $35^{\circ}C$. With SH-group blocking agents the enzyme was inhibited seriously. The activity of enzyme was also inhibited by the addition of some heavy metals, $Ag^+$ and $Cu^{2+}$, but was not affected by EDTA. General property of the enzyme was characterized and the possible nature of the enzyme active center was also discussed.
To find $\alpha$-glucosidase inhibitors produced by Actinomycetes, bacteria belonging to Actinomycetes were isolated from soil sample using Bennett's medium. The inhibitory activity induced by these bacteria on $\alpha$-glucosidase, which is the key enzymes far carbohydrates digestion and the prevention of diabetic complications, was investigated. A strain of these bacteria, PM718 potently inhibited $\alpha$-glucosidase activity in vitro.

This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.
Enzymatical properties of Streptomyces sp. S-45 producing chitinase and ${\beta}$-1.3-glucanase isolated from soil were investigated. Chitinase activity was 3.01(U/ml) and ${\beta}$-1.3-glucanase activity was 2.49(U/ml). The optimum medium for mycolytic enzyme production of strain was composed of 0.7% colloidal chitin, 0.3% glucose, 0.5% asparagine, 0.2% peptone, 0.01% NaCl, 0.01% $K_2HPO_4$ and 0.01% $MgSO_4{\cdot}7H_2O$ in intial pH 7.0. The optimal condition for mycolytic enzyme activities were: pH 6.5-7.0, $45-50^{\circ}C$. Enzyme activities were activated by metal ion as $10^{-2}M\;Co^{{+}{+}}$, $Cu^{{+}{+}}$, $Mn^{{+}{+}}$, $Al^{{+}{+}{+}}$ and $10^{-3}M\;Sn^{{+}{+}}$ but $Ag^{{+}{+}}$, $Hg^{{+}{+}}$ inhibited.
This study was conducted to investigate the effect of pyroligneous acids on urea hydrolysis for the purpose of inhibiting ammonia volatilization during urea fertilizer application. Different types of synthetic urease inhibitors have been searched and developed, but their use is limited due to varying inhibition effects on soil urease, and environmental problems. In this study, the effect of pyroligneous acids, a natural substance, on urea hydrolysis in soil was evaluated by analyzing inhibition of urease activity. Pyroligneous acids inhibited plant urease and microbial urease activity, as well as soil urease with various urease complex. In addition, pyroligneous acids exhibited non-competitive urease inhibition effect through urease kinetics and inhibited urea hydrolysis in the soil. This study showed that pyroligneous acids treatment with urea fertilizer decreases the loss of urea fertilizer, improves the efficiency of nitrogen application on plant and reduces the amount of nitrogen fertilizers applied in soil.
This study was performed to investigate effect of different soil managements on physical properties and microbial activities in volcanic ash citrus orchard soil. Experiment plots had managed to control weeds on soil for 4 years with clean cultivation (CCM) used with herbicide, natural sod cultivation (NSCM), kentucky blue grass sod cultivation (KBG). Soil samples were taken on October, in both 1998 and 2000 from 3 experimental plots. In NSCM, Soil hardness was lower at 11.8 mm than in CCM. And water stable Aggregation coefficient(>0.5 mm) was high at 26.7% compared with CCM. Soil bulk density and porosity showed no significant among the treatments. Soil acid phosphatase was high in sod cultivation plots and the amount of microbial biomass C was about twice higher at $525.4mg\;kg^{-1}$ in KBG than in CCM. Conclusionally, Sod cultivation improved soil physical properties such as aggregation, hardness and increased microbial activities compared with clean cultivation in citrus orchard soil. Soil total PLFA, acid phosphatase, and microbial biomass C contents were investigated on May in nonvolcanic ash citrus soil. Soil samples were collected at 5 sites each; convention cultivation grown with herbicide, natural sod cultivation grown with 1/2 chemicals, organic cultivation. That sites have been managed for 5 years over. PLFA contents were two times higher at $112.2n\;mol\;g^{-1}$ in organic cultivation than in convention cultivation. According to the PLFA indicator, Gram negative bacteria and actinomycetes in organic cultivation were high compared with convention cultivation, which were at 15.1%, 6.6%, respectively. Soil microbial biomass C contents was about twice higher in organic cultivation than in convention cultivation. Soil acid phosphatase was high at 17.6% in organic cultivation compared with convention cultivation.
Although urban soils must be well understood in order to ensure their conservation and optimum use, these intensively managed and disturbed soils have not been extensively investigated up to now. Urban soils from forest, lawn, streetside, and bare ground and under pavement in Inchon had high bulk density as a result of widespread trampling-induced soil compaction. The various urban soils including forests showed lower water content and higher temperature as compared with rural forest soil. Chemically, soils from urban areas had an unusual neutral pH and low organic matter content. Total bacterial numbers in urban soils was only 5∼50% of that in the rural forest soil. An analysis of stepwise multiple regression revealed that soil organic matter was the most important predictor variable on total bacterial number. The dehydrogenase activity of most urban soils was not significantly different from that of rural forest soil, whereas the microbial activity of soils under pavement was lower. Our investigations show that inadequate organic matter of highly compacted soils has adversely affected the abundance of microorganisms involving nutrient cycling in urban soils.
This study was conducted to figure out the relationships among soil chemical properties and bacterial biomass related to denitrification and sulfur-reducing and the activity of dehydrogenase, and ultimately to consider the usefulness of dehydrogenase activity as a tool for evaluating the dynamics of forest soil ecosystem. Four sites were selected for the collection of soil samples within two regions(Onsan industrial estate as a polluted region and Mt. Mani at Kanghwa island as a clean area) with two forest types (coniferous and deciduous stands). The soils of Mt. Mani showed higher amount of organic matter, total nitrogen and available phosphorus than those collected from Onsan industrial estate, which indicated that the soils were more beneficial for microbial growth than those of Onsan. The dehydrogenase activity was more sensitive than the denitrifying bacteria or sulfur-reducing bacteria since the activity was significantly different between the regions and season while the two bacterial biomass were not significantly different between the two regions. In addition, the dehydrogenase activity showed relatively high correlation coefficients with organic matter(r=0.53, p=0.004), total nitrogen(r=0.41, p=0.008) and C/Ava. P-ratio(r=-0.52, p=0.001), which was thought to be closely related with microbial activity. Thus, the dehydrogenase activity was thought to be a useful index of soil ecosystem dynamics with considering that the technique need to be applied with the same soil texture for the comparison of the activity as other researchers indicated.
In this research, 3-hydroxypyridine(2-HP) metabolic ability of starving Arthrobacter crystallopoietes cell and the effect of humic acid on the metabolism of this starving cell were evaluated. 2-HP metabolic ability of exponential phase cell (acclimated cell) was much higher than that of lag phase cell (unacclimated cell) during starvation period. After 3 days of starvation, 2-HP half-life of the acclimated cell was 14 hours and that of the unacclimated cell was 46.5 hours. Humic acid enhanced the stability of 2-HP monooxygenase of starving co]1 and, after 2 days of starvation, the residual activity rate of this enzyme of the microbial cell starved in humic acid solution was 12% while the rate for control condition was 1.5%. After 14 days of starvation, 2-HP half-life for control condition was 43 hours and that for humic acid condition was 1.25 hour.
In an effort to develop a potent system for the production of various dp (degree of polymerization) chitooligosaccharides, 32 enzymes or microbial systems were screened for chitosanolytic acitivity using chitosan as a substrate. The efficiency of each enzyme system was evaluated by the changes of turbidity and viscosity of chitosan solution, the amount of precipitate and the reducing sugar-producing activity in the enzymatic reaction mixture. Based on these assay methods for the chitosanase activity, Bacillus sp. P2l out of 32 screened systems showed highly potent endochitosanase, which was comparable with a commercially available enzyme (E7). Chitooligosaccharides of dp 3-7 were separated by TLC as major enzymatic reaction products, suggesting that the chitosanase from Bacillus sp. P2l be endo-splitting type.
