The immunostimulating effects of egg white derivatives (EWD) on the phagocytic response of feline peripheral blood polymorphonuclear cells (PMN) as well as mono- nuclear cells (MNC) were examined. The phagocytic activity was analyzed by a flow cytometry system. The EWD showed directly an enhanced effect on the phagocytic response of MNC but not PMN. The phagocytic activity of MNC was enhanced by culture supernatant from MNC and PMN treated with EWD, respectively. Similarly, the phagocytic activity of PMN was enhanced by culture supernatant from MNC but not PMN treated with EWD. It was, therefore, indicated that the enhanced phagocytic activity of feline PMN could be mainly mediated by humoral factor(s) released from MNC treated with EWD. These results suggested that EWD could enhance the phagocytosis of feline peripheral blood phagocytes.
NADPH oxidase dependent superoxide generation and phagocytosis in neutrophils stimulated with opsonized zymosan or heat aggregated IgG were coincided with the process of calcium uptake. The responses in activated neutrophils were enhanced with increasing concentrations of extracellular calcium and these effects were significantly inhibited by calcium chelators, EGTA and EDTA. The superoxide generation in activated neutrophils was reduced by dantrolene and chlorpromazine. Calcium antagonists, bepredil, diltiazem, verapamil, nifedipine and nimodipine effectively inhibited the calcium uptake, superoxide generation and phagocytosis in activated neutrophils, and NADPH oxidase activity was also inhibited. The results suggest that calcium antagonists may inhibit the superoxide generation and phagocytosis in activted neurtophils by the inhibition of calcium influx and by the action on intracellular redistribution of calcium and NADPH oxidase system.
Three viral strains, causing CPE in porcine alveolar macrophage cell, were isolated from aborted fetus, serum from young pig showing blue-ear sign and lung of suspected pig, respectively. The differential diagnostic results showed no characteristics of Aujeszky's disease virus(ADV), hog cholera virus (HCV), Japanese encephalitis virus(JEV), porcine parvovirus(PPV) and encephalomyocarditis virus (EMCV). However, positive reactions were demonstrated by IFA using monospecific porcine antibodies against Lelystad virus. When the paired sera of experimentally inoculated swine with one of isolate, KPRRSV-l were tested by IPMA, the result indicated that the isolate was related to United States isolate than European LV.

Carboxyethylgermanium sesquioxide(Ge-132)를 투여용량(300,600, 900mg/kg)과 투여일을 변화시켜 정상마우스와 cyclophosphamide(CY)로 처리한 마우스에 경구투여한 후, Ge-132가 CY 에 의해 변화된 마우스의 면역기능에 미치는 효과를 실험하였다. SRBC 항원 주사전, 동시 또는 후에 Ge-132를 투여시 SRBC에 대한 적혈구 응집소가와 비장세포의 용혈반 형성세포(PFC)수가 항원 주사일과 관계없이 용량의존적으로 증가되어 체액성 면역반응을 강화시켰으며, 마크로파지의 탐식능도 항진시켰다. 그러나 DNFB에 대한 접촉성 지연형 과민반응은 DNFB 감작일과 투여용량에 관계없이 Ge-132투여로 억제되었다. CY를 투여한 마우스에 Ge-132를 병용 투여한 군이 CY 단독 투여군에 비하여 적혈구 응집소가와 PFC수 및 혈중 탄소입자의 제거율이 현저하게 증가되어, CY로 억제된 체액성 면역반응과 마크로파지의 탐식능이 항진된 것으로 나타났다. DNFB 감작 전 CY 투여로 증폭된 접촉성 지연형 과민반응이 Ge-132 병용투여로 감소되어 CY로 유도된 면역독성 반응에 대한 Ge-132의 저지효과를 시사해 주었다.
Activation of macrophages plays an important role in the host-immune system. In this study, we investigated the functional roles and related signaling mechanism of hot-water extracts of bee pollen (BPW) in RAW 264.7 macrophages. Since BPW did not exert cytotoxicity at concentrations ranging from 62.5 to $250{\mu}g/mL$ in macrophage cells, a concentration of $250{\mu}g/mL$ was used as the maximum dose of BPW throughout subsequent experiments. BPW increased inducible nitric oxide synthase-mediated nitric oxide production in a concentration-dependent manner. Additionally, BPW was found to induce macrophage activation by augmenting the expression of cell surface molecules (cluster of differentiation; CD80/86, and major histocompatibility complex; MHC class I/II) and production of pro-inflammatory cytokines (tumor necrosis $factor-{\alpha}$, interleukin-6, and $IL-1{\beta}$) through mitogen-activated protein kinase and nuclear $factor-{\kappa}B$ signaling pathways in RAW 264.7 macrophages. Taken together, our results indicate that BPW could potentially be used as an immunomodulatory agent.
Neutrophils that influx into the alveolar spaces from circulatory bloods play a important role in pathogenesis of acute lung injury. During the acute inflammatory phase, in order to investigate the acceleration of macrophage phagocytosis to the neutrophils is able to reduce the neutrophil-derived acute lung injury, endotoxemia was induced by insufflation of lipopolysaccharide intratracheally and organic germanium was injected intraperitoneally after endotoxin treatment. At 5 h after endotoxin treatment, lung weight and BAL protein concentration are significantly increased (p<0.001) compared to sham, and that was remarkedly decreased (p<0.001, p<0.01) by injection of germanium. In addition germanium treatment resulted to decreased the number of alveolar PMNs and to increase the percentage of engulfed neutrophils by alveolar macrophages. These observations indicate that organic germanium may have a role of reduction to neutrophil-derived acute lung injury in endotoxemia.
The objective of this study was to examine the effect of fucoidan on the phagocytic capapcity and oxidative burst activity (OBA) of canine peripheral blood polymorphonuclear cells (PMNs). The phagocytic capacity and OBA of PMNs were evaluated simultaneously by using a flow cytometer. Fucoidan itself did not cause any direct effect on the phagocytic capacity and OBA of PMNs. However, the phagocytic capacity and OBA of PMNs were enhanced by the culture supernatant from PBMCs treated with fucoidan. The phagocytic capacity and OBA of PMNs were also increased by treatment with recombinant canine (rc) tumor necrosis factor (TNF)-${\alpha}$. The ability of the culture supernatant from fucoidan-treated PBMCs to stimulate the phagocytic capacity and OBA of PMNs was inhibited by addition of anti-rc TNF-${\alpha}$ polyclonal antibody (PAb) prior to the culture. The amount of TNF-${\alpha}$ in the culture supematant from PBMCs was shown to increase upon treatment of fucoidan as compared with that of vehicle-treated PBMCs culture supematant. The level of TNF-${\alpha}$ mRNA expression in PBMCs was also up-regulated by the fucoidan treatment. These results suggest that fucoidan has an immunoenhancing effect on the phagocytic capacity and OBA of canine PMNs, which is mainly mediated by TNF-${\alpha}$ released from fucoidan-stimulated PBMCs.
Macrophages play a crucial role in the host immune defense system. The current study investigated immunomodulatory activities induced by polysaccharides extracted from Cudrania tricuspidata (CTPS) fruits in murine macrophages and their role in signaling pathways. In macrophages, CTPS predominantly induced nitric oxide (NO), tumor necrosis factor-a, and interleukin-6 production. In addition, CTPS significantly up-regulated expression of the macrophage surface marker (CD80/86 and MHC class I/II). These results indicate that polysaccharides extracted from CTPS may potentially play an immunomodulatory role in macrophages via mitogen-activated protein kinases and nuclear factor-B signaling. These findings may be useful in the development of immune enhancing adjuvant materials obtained from natural sources.
The essential factor of acute respiratory distress syndrome (ARDS), an acute lung injury accompanied commonly by sepsis syndrome is accumulation of neutrophils in lung tissue. The study attempted to confirm whether a lung injury would be decreased with the anti-inflammatory effect of germanium by the treated germanium prior to the development of ARDS and whether nitric oxide influence in suppressing a lung injury. Test groups were divided in the following structure for experiment; CON that has been administered with sodium chloride to airway, LPS administered with endotoxin for 5 hours in the same amount and 5 hours of endotoxin administered Ge+LPS following 1 hours of pre-treated germanium. The result of a test using experimental animals, infilteration of neutrophils (p<0.001) in bronchoalveolar lavage fluid (BALF) was significantly decreased, the structure of lung tissue was preserved relatively well, and much neutrophils with distinct positive were observed on tunel staining which showed increase of apoptotic neutrophils in the pre-treated germanium group compare to the endotoxin administrated group. In observation of ultrastructural changes of cell in BALF, phagocytic alveolar macrophage was increased in alveolar space, the nucleus of most engulfed neutrophils were condensed, and some apoptosis neutrophils appears to be DNA fragmentation and effacement of cellular organelles were found in intercellular matrix in the pre-treated germanium group. However, the nitric oxide showed increase in all the groups excluding CON, and the nitric oxide effect such as degranulation diminishing of mast cells and apoptosis increase of neutrophils in the pre-treated group only. The situation appears that there was change in internal environment of the experimental animal by the pre-treated germanium before the nitric oxide is produced and the anti-inflammatory effect activated the pre-processed germanium by nitric oxide which activated following the change. Therefore, the nitric oxide created from macrophage in accordance with the pre-treated germanium appears to influence in alleviating a lung injury. Accordingly, acute lung injury is alleviated by the anti-inflammatory effect of germanium such as inhibition of neutrophils migration, induction of neutrophil apoptosis and increase of phagocytic function of phagocyte, and the nitric oxide produced from activated macrophage by germanium would influence in suppressing a lung injury.
