Isolation and Characterization of Bacillus Strain as a Potential Biocontrol Agent (환경친화적 미생물농약으로서의 잠재성을 가진 세균의 분리 및 특성)

  • In this study, to retain a stable bacterial inoculant, Bacillus strains showing antifungal activity were screened. The improved production, antifungal mechanism, and stability of the antifungal metabolite by a selected strain, AF4, a potent antagonist against phytopathogenic Botrytis cinerea, were also investigated. The AF4 strain was isolated from rhizospheric soil of hot pepper and identified as Bacillus subtilis by phenotypic characters and 16S rRNA gene analysis. Strain AF4 did not produce antifungal activity in the absence of a nitrogen source and produced antifungal activity at a broad range of temperatures (25-40℃) and pH (7-10). Optimal carbon and nitrogen sources for the production of antifungal activity were glycerol and casein, respectively. Under improved conditions, the maximum antifungal activity was 140±3 AU/ml, which was higher than in the basal medium. Photomicrographs of strain AF4-treated B. cinerea showed morphological abnormalities of fungal mycelia, demonstrating the role of the antifungal metabolite. The B. subtilis AF4 culture exhibited broad antifungal activity against several phytopathogenic fungi. The antifungal activity was heat-, pH-, solvent-, and protease-stable, indicating its nonproteinous nature. These results suggest that B. subtilis AF4 is a potential candidate for the control of phytopathogenic fungi-derived plant diseases.

Diminution of Pesticide Residues on Crops and Soil by Accelerated Photolysis (광분해(光分解) 촉진(促進)에 의한 작물(作物) 및 토양중(土壤中) 농약잔유물(農藥殘留物)의 경감(輕減)에 관한 연구(硏究))

  • In an effort to reduce artifically the residual pesticides on crop and soil by accelerated photolysis,some 40 among the naturally occurring and synthetic coumpound were screened for photosensitization and/or photocatalysis and six promising chemicals were selected.The fungicides and the four selected photosensitizers and/or photocatalyst (PS) were applied to each crop.The results obtained are summarized as follows. 1. The wavelengths of maximum absortion (${\lambda}$max) and the molar absorptivites (${\varepsilon}$max) of procymidone,vinclozolin,and carbendazim in acentone were all 209 nm and 853,854,and 8740 respectively. 2. Of the 40 naturally-occuring and synthetic compounds screened,six promising ones were selected and designated as PS-1 (aromatic ketone),PS-2(aromatic amine)PS-3(quinone) ,PS-4 (inorganic compound),PS-5(organic acid salt) and PS-6(semiconductor photocatalyst). 3. In the accelerated photolysis of pesticide in soil by applying PS ,procymidone was decoposed rapidly by virtue of PS-2,being 59% of the control 3 days after application. 4. The vinclozolin residue in soil was reduced to 71% and 21% of the control 1 and 15 days,respectively,after PS-2 application. 5. The photolysis of carbendazim in soil was not accelerated by any of the PS tested. 6. The pesticide residues on the crop were prominently reduced by PS application.The procymidone residue on tomato was reduced to 47% of the control 15 days after PS-1 application and that on red pepper reduced to 57% 15 days after PS-2 application. 7. Vincrozolin residus remaining on tomato 1 and 15 days after PS-2 application were 38% and 56% of the control whereas those on the red pepper were 82% and 64%,respectively. 8. PS-2 was the most effective for the accelerated photolysis of carbendazim residues remaining on tomato, whereas on red pepper, the four of PS tested were all effective, but did not make much difference between them. This might be due to the shielding of sunlight by the leaves of red pepper not to exert the photosensitizing effect of PS-2 to the full.

Robust Eye Localization using Multi-Scale Gabor Feature Vectors (다중 해상도 가버 특징 벡터를 이용한 강인한 눈 검출)

  • Eye localization means localization of the center of the pupils, and is necessary for face recognition and related applications. Most of eye localization methods reported so far still need to be improved about robustness as well as precision for successful applications. In this paper, we propose a robust eye localization method using multi-scale Gabor feature vectors without big computational burden. The eye localization method using Gabor feature vectors is already employed in fuck as EBGM, but the method employed in EBGM is known not to be robust with respect to initial values, illumination, and pose, and may need extensive search range for achieving the required performance, which may cause big computational burden. The proposed method utilizes multi-scale approach. The proposed method first tries to localize eyes in the lower resolution face image by utilizing Gabor Jet similarity between Gabor feature vector at an estimated initial eye coordinates and the Gabor feature vectors in the eye model of the corresponding scale. Then the method localizes eyes in the next scale resolution face image in the same way but with initial eye points estimated from the eye coordinates localized in the lower resolution images. After repeating this process in the same way recursively, the proposed method funally localizes eyes in the original resolution face image. Also, the proposed method provides an effective illumination normalization to make the proposed multi-scale approach more robust to illumination, and additionally applies the illumination normalization technique in the preprocessing stage of the multi-scale approach so that the proposed method enhances the eye detection success rate. Experiment results verify that the proposed eye localization method improves the precision rate without causing big computational overhead compared to other eye localization methods reported in the previous researches and is robust to the variation of post: and illumination.

An Analysis of the Determinants of Employment and Wage of New College Graduates (신규대졸자의 취업 및 임금수준 결정요인 분석)

  • This study examines the determinants of employment and wage of new college graduates by using Youth Panel Data(2003-2005) of the Work Information Center, and seeks assignments for mitigating unemployment and wage disparities of new college graduates. Results are summarized as follows. First, an analysis of the determinants of employment shows that the Kyunggi Inchon district in school locations, higher school records, and qualification certificates positively affect the employment rate, while the private college group in the non-capital area negatively affects the employment rate. Second, an analysis of determinants of standard employment demonstrates that the Kyunggi Inchon district in school locations, higher school records, qualification certificates, and the major group of medical science, pharmacy, nursing science and health science, and the major group of education positively affect the employment rate, while the private college group in the non-capital area, the junior college groups in the capital and non-capital areas negatively affect the employment rate. Third, an analysis of determinants of nonstandard employment shows that the junior college graduation in scholarly attainments, the junior college groups in the capital and non-capital areas positively affect the employment rate, while the private college group in the non-capital area negatively affects the employment rate. Fourth, an analysis of the determinants of wages demonstrates that male in sex, the older in ages, the major group of medical science, pharmacy, nursing science and health science, and the major group of education positively affect the wages, while nonstandard employment, Kyunggi Inchon and Cholla districts in school locations negatively affect the wages. These results suggest several implications. First, college education should be reformed to cultivate professional manpower who are required by industries. Second, alternative measures to mitigate sex discrimination in labor markets should be prepared. Third, the process of attaining qualification certificates should be reformed in order that it is actually connected to the abilities of work performances and the improvement of productivity. Fourth, a locally balanced development must be realized through the decentralization of industries. Fifth a systematic and comprehensive program need to be prepared to promote the employment of new college graduates.

Variation in Pod Shattering in a RIL Population and Selection for Pod Shattering Tolerance in Soybean [Glycine max (L.) Merr] (콩 RIL 집단의 내탈립성 변이 탐색 및 유망계통 선발)

  • Pod shattering during the maturing stage causes a serious yield loss in soybean. It is the main limiting factor of soybean cultivation and mechanization. It is important to develop varieties suitable for mechanical harvesting and to develop energy-efficient agricultural machinery to save labor and costs. 'Daewonkong,' developed by the National Institute of Crop Science (NICS) in 1997, is an elite cultivar that occupies more than 80% of the soybean cultivation area in Korea because of its strong tolerance to pod shattering. The objectives of this study were to investigate the variation in pod shattering degree in a RIL population developed from a 'Daewonkong' parent and to select promising lines with pod shattering tolerance. 'Daewonkong' demonstrated a high level of tolerance to pod shattering compared to the 'Tawonkong' and 'Saeolkong' varieties, with no shattered pods after 72 hours of drying. Screening of pod shattering showed a clear distinction between the tolerant and susceptible varieties. Also, the distribution of shattering pod ratio in the two populations showed a similar pattern for three years. The promising lines with pod shattering tolerance included 27 lines in the 'Daewonkong'×'Tawonkong' population and 21 lines in the 'Daewonkong'×'Saeolkong' population. The promising lines are expected to be widely used as breeding parents for creating soybean cultivars with pod shattering tolerance.

The Method of the Cultivation of Taste and the Possibility of the Edification of Personality & the Cultural Development Through It: The Approach to Analyzing the Examples of the Judgment of Negative Taste in Kant's Critique of Judgment(§§32-33) (취미 도야의 방식과 이를 통한 인성의 교화 및 문화발전의 가능성: 칸트의 『판단력비판』 §§32-33 부정적 취미판단의사례 분석을 중심으로)

  • This essay attempts to reveal how taste spontaneously cultivates and why it is necessary for cultivating taste to edify our personality and to develop culture. It is a key of the solution of the problems that taste always reflects its judgment through pleasure. Because the grounds of the universal validity of the judgment of taste are found, whenever taste tests the validity of its own judgment, the so-called 'delight of discovery' makes taste cultivate itself. For having the moral personality, we need to practice spontaneously the morality of our own behaviour and for judging whether an artwork to represent the period is succeeded or not, we need to have a high insight to select the cultural heritage. But the autonomous thinking can delightfully be made a habit, judging the beauty of artworks. In the main body of this essay, it is determined from the three examples of the negative judgment of taste which Kant suggested in deduction. According to Kant, the negative judgment of taste means that the beautiful work is displeased, but what it asserts is that taste is cultivated. I formalize the methods of reflection of taste revealed in three negative judgments of taste into'resisting', 'indicating of error', 'self-retracting'.(Chapter 2) And from this, I emphasize the necessity to cultivate taste in the way that these methods of the cultivation of taste can affect building our personality by stimulating our reason to have interest in moral(Chapter 3) and in the way that taste directly judges the product of cultural succession.(Chapter 4) In the end of last chapter, I examine further essentially the method of the reflection of taste, to inquire into how to enable it.(Chapter 5) Especially, I try to illuminate its grounds through Schiller's concept of the "impulse of amusement(Spieltrib)", because his explanation helps us to understand the dynamics of taste's delight of discovery. Although the abilities of mind conflict with each other, taste has the characters that it reflects to encourage them for each other and that it is vitalized by its own activity. We, as it were, can pleasantly handle two tasks, because taste makes the impulse of amusement from conflictive impulses in mind. In conclusion, I state that we have to experience directly the impulse of amusement like creative artist, because it is maximized from creation.

Can the Sasang Constitutional Type Trait Act as an Independent Risk Factor for Dyslipidemia? (사상체질이 이상지질혈증의 위험인자가 될 수 있는가?)

  • Objectives: This study was designed to investigate the correlation between Sasang constitution and dyslipidemia and the possibility that Sasang constitution can be a risk factor of dyslipidemia. Methods: This study was a retrospective cross-sectional study based on chart review. Seventeen thousand sixty-nine charts which were collected from May 2007 to June 2010 in International Healthcare Service Center, Kyung Hee University Medical Center, were reviewed for the study. Fifteen thousand two hundred fifty charts including Sasang constitutional diagnosis and lipid profiling were selected and we examined the correlation between Sasang constitution and dyslipidemia. Results: After adjusted for body mass index, the levels of total cholesterol and triglyceride were highest in Taeeum group, and lowest in Soeum group. High-density lipoprotein was highest in Soeum group, and lowest in Taeeum group. After adjusted for age, sex, and body mass index, the odds ratios for 'high triglyceride' were 1.716 (1.411~2.087) in Soyang group and 2.021 (1.650~2.475) in Taeeum group compared to Soeum group. The odds ratio for 'high low-density lipoprotein' was 1.229 (1.006~1.501) in Soyang group compared to Soeum group. The odds ratios for 'low high-density lipoprotein' were 1.195 (1.033~1.381) in Soyang group and 1.414 (1.212~1.649) in Taeeum group compared to Soeum group. Conclusions: The lipid profiles and dyslipidemic risk differed across constitutional types, showing constitution type-specific patterns, so that we could infer the possibility that Sasang constitutional types can be a risk factor of dyslipidemia.

Antioxidant Activities of Extracts from Ligustrum ovalifolium H. Leaves (왕쥐똥나무잎 추출물의 항산화 활성)

  • The free radical scavenging activities of extracts from Ligustrum ovalifolium H. leaves (LOH) as well as various antioxidant activities such as ferric reducing antioxidant power (FRAP), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, reducing power and lipid peroxidation inhibition were evaluated by electron spin resonance (ESR). The total polyphenol and flavonoid contents of the water and ethanolic extracts from LOH were $105.5{\pm}1.31$ and $102.1{\pm}1.82$ mg gallic acid equivalent/g extract, respectively, and $84{\pm}1.72$ and $82.8{\pm}1.65$ mg catechin equivalent/g extract. In addition, $IC_{50}$ values for the 1,1-diphenyl-2-picryldrazyl (DPPH), alkyl, and hydroxyl radical scavenging activities of the water and ethanolic extracts were $0.021{\pm}0.002$ and $0.010{\pm}0.003$ mg/mL, $0.011{\pm}0.003$ and $0.012{\pm}0.002$ mg/mL, and $0.395{\pm}0.002$ and $0.443{\pm}0.002$ mg/mL, respectively. The ABTS radical scavenging activities of the water and ethanolic extracts from LOH and BHT were $0.073{\pm}0.12$, $0.130{\pm}0.06$ and $1.461{\pm}0.02$ mM Trolox equivalent/mg extract, respectively. The FRAP values of the extracts from LOH were higher than those of BHT, which was used as a positive control. The LOH extracts showed strong inhibitory effects on lipid peroxidation as measured by ferric thiocyanate (FTC) and thiobarbituric acid (TBA) assay compared to that of ${\alpha}$-tocopherol. Using MTT assay on human liver cells (Chang), extracts from LOH showed no toxicity at a concentration of 0.5 mg/mL. These results indicate that the LOH extracts possessed antioxidant activity.

Optimal hot water extraction conditions of fermented Polygonum multiflorum root by Lentinula edodes pegler mycelials using response surface methodology (반응표면 분석법에 의한 표고균사체발효 적하수오 열수 추출조건의 최적화)

  • This study was performed to determine optimal extraction conditions of fermented Polygonum multiflorum root by Lentinula edodes (JMI10079) Pegler mycelials using response surface methodology. The independent factors were extraction temperature (X1: $40-100^{\circ}C$), extraction time (X2: 2-10 hrs.), and the ratio of water to sample (X3: 33-100 mg/mL). Their effects were assessed on dependent variables of the extract properties, which included soluble solid contents (Y1), $^{\circ}Brix$ of sample extract (Y2), total polyphenol content (Y3), total flavonoid content (Y4), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical cation scavenging activity (Y5) and 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (Y6). The experimental data obtained were fit to a second-order polynomial equation. The optimal extraction conditions for fermented P. multiflorum root were: X1: $91.22^{\circ}C$, X2: 7.72 hrs, and X3: 39.71 mg/mL.

Evaluation of free radical scavenging and pancreatic lipase inhibitory effects of Aquilaria agallocha extracts (침향 추출물의 라디칼 및 췌장 지방분해 효소저해 활성 평가)

  • In a continuing screening of selected medicinal plants native to South Korea, the antioxidant and pancreatic lipase inhibitory activities of an aqueous methanolic extract from the heartwood of Aquilaria agallocha were investigated. Eighty percent of the methanolic extract of A. agallocha was further divided into $CH_2Cl_2$, EtOAc and n-BuOH in order to yield four solvent-soluble portions, namely $CH_2Cl_2$-soluble, EtOAc-soluble, n-BuOH-soluble and $H_2O$ residue. The antioxidant properties were evaluated by employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals, while the anti-obesity efficacy of A. agallocha extracts and solvent-soluble portions were tested by porcine pancreatic lipase assay. All tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. Among the tested extracts and solvent-soluble portions, the $CH_2Cl_2$-soluble portion showed much higher radical scavenging activity and pancreatic lipase inhibitory properties when compared with other solvent-soluble portions. This result suggested that there was a significant relationship between the total phenolic content and biological efficacies, and A. agallocha extract might be considered as a new potential source of natural antioxidants and as a pancreatic lipase inhibitory source. A more systematic investigation of this biomass will be performed for further investigation of activity against antioxidative and anti-obesity effects.
