Studies on Stream Ecosystem in the Bukhan River using Stable Isotopes (안정동위원소를 이용한 북한강 하천생태계 연구)

  • This study examined the epiphytes using stable isotopes. Separated epiphytes should represent the water quality. In study areas, where agricultural paddies were prevailing, biomass of epiphytes can increase nitrogen isotopes(${\delta}^{15}N$) and as a result decrease in carbon isotopes(${\delta}^{13}C$). Naerin stream and Inbuk upper areas showed higher nitrogen isotopes(${\delta}^{15}N$) than in Hangye and Buk stream. Also, stable carbon isotopes(${\delta}^{13}C$) in Naerin stream and Inbuk streams tended to increase than in Hangye and Buk stream. This can indicate the epiphytes can be a good indicator for water quality pollution.

Ecological Importance of Benthic Microalgae in the Intertidal Mud Flat of Yeongheung Island; Application of Stable Isotope Analysis (SIA) (영흥도 조간대 갯벌 저서미세조류의 생태적 중요성; 안정동위원소 분석 활용)

  • In order to reconstruct a benthic foodweb structure and assess the role of benthic microalgaes as a diet source for benthos, we analyzed the carbon and nitrogen stable isotopes of diverse benthos (bivalves, crustaceans, gastropods and fishes) and potential diets (particulate organic matter, sedimentary organic matter, benthic microalgae, seagrass, and macroalgaes) in the intertidal mudflat surrounding Yeongheung Island. The ${\delta}^{13}C$ values of the diets indicated wide ranges (- 26.5‰ to - 8.4‰) while benthos showed a small range of ${\delta}^{13}C$ values (-12.1‰ to - 17.8‰), although they were in the same range. Except for green algaes among the macroalgaes as well as sedimentary organic matter, ${\delta}^{15}N$ values of the diet candidates ($5.7{\pm}1.0$‰) were lighter in comparison to those of the benthos ($11.8{\pm}1.9$‰). Based on the ${\delta}^{13}C$ and ${\delta}^{15}N$ data, the benthos were classified into 3 groups, indicating a different diet and trophic position. But benthic microalgae is the most important diet source for all three benthos groups based on their stable isotope ratios, suggesting benthic microalgae should be a main diet to the intertidal ecosystem. Hence this study highlights that the biomass of benthic microalgae as biological resource should be evaluated for the management of the intertidal ecosystem of Yeongheung Island.

Foodweb of Aquatic Ecosystem within the Tamjin River through the Determination of Carbon and Nitrogen Stable Isotope Ratios (탄소 및 질소 안정동위원소비를 이용한 탐진강 수생태계 먹이망 연구)

  • To investigate foodweb of aquatic ecosystem in the Tamjin River, carbon and nitrogen stable isotopes ratios of aquatic organisms, as well as environmental indicators based on the water, were determined in this study. Various organisms such as fishes (Coreoperca kawamebari, Zacco platypus, Cobitis lutheri, and Pungtungia herzi) and periphyton (epilithon and epiphyte), and particulate- and coarse particulate organic matters (POM and CPOM) were collected in upper (Tamjin River, Yuchi Stream, and Omcheon Stream) and lower (TJ-1~TJ-5) reaches of Jangheung Dam. The nitrate concentration and ${\delta}^{15}N$ signature of POM and organisms (invertebrates and fish) were found to be more enriched toward the downstream section of the river. It was determined that allochthonous matter occurring from a tributary alters the chemical character of water, as well as the isotopic signature of organisms contained therein. Attached algae (ephilithon) were identified as a base component of the benthic foodchain further downstream.

Determination of the Origin of Particulate Organic Matter at the Lake Paldang using Stable Isotope Ratios (${\delta}^{13}C$, ${\delta}^{15}N$) (입자성 유기물의 안정동위원소비를 이용한 팔당호 수계내의 유기물 기원 연구)

  • Organic carbon and nitrogen stable isotope ratios of particulate organic matter (POM) were determined at 7 stations (6 stations in Han river and Paldang dam outflow station) from May to October 2013, in order to understand the origin of POM at the Han river. ${\delta}^{13}C$ values of POM in four stations showed significant seasonal changes season, but ${\delta}^{15}N$ values were enriched in around Kyeongan stream (K). POC, PN and Chl-a concentration showed a similar seasonal pattern in Kyeongan stream, with an apparent decrease from July to August. POC and PN concentration has a higher correlation with Chl-a concentration in Kyeongan stream (K). ${\delta}^{13}C$ and ${\delta}^{15}N$ values of POM has a lighter value during lower Chl-a concentration peroid, compared to other seasons. Our results revealed that Kyeongan stream (K) seemed to be influenced by substantial amount of organic manure or fertilizer input in 2013, compared to the previous year (2012). These results suggest that the analysis of stable isotope ratios is a simple but useful tool for the identification of organic matter origin in aquatic environments.

  • The nitrogen isotope value in both ammonium and nitrate ion were determined at 9 stations during both June and August 2016, in order to understand the origin of DIN at the Han river. ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values in 8 stations (CP, SB, MHC, P4, SJ, SBC, P2, SC) were no significant variation. However ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values in KK (Kyeongan stream) showed significant different in comparison with 8 stations, with an apparent increase of nitrogen isotope values. These results indicate that antropogenic nitrogen source influence on KK station. Also the ${\delta}^{13}C$ and ${\delta}^{15}N$ isotope ratio of phytoplankton (Diatom and Cyanobacteria) in KK (Kyeongan stream) showed heavier values, compared to other study stations. These results indicate that nitrogen isotope value in phytoplankton effects by different nitrogen source in study sites. These results suggest that the analysis of stable isotope ratios is a simple but useful tool for the identification of dissolved inorganic nitrogen origin in aquatic environments.

Decomposition Characteristic of Sedimentary Organic Matters by Bacteria (세균에 의한 퇴적유기물의 분해 특성에 관한 연구)

  • The Temporal variability in the food chain structure of bacteria in the sedimentary organic matter was investigated using stable isotope and fatty acid. Potential organic matter sources (Land plant, Marine POM, benthic microalgae, Riverine POM), sedimentary organic matter and bacteria were sampled in Gamo largoon and Nanakita estuary. The main objective of the present study was to determine food sources of bacteria along with temporal variability. Land plant (${\delta}^{13}C$ = -26.6‰ and ${\delta}^{15}N$ = 3.6‰) and Riverine POM (${\delta}^{13}C$ = -25.5‰ and ${\delta}^{15}N$ = 8.9‰) were isotopically distinct from benthic microalgae (${\delta}^{13}C$ = -16.3‰ and ${\delta}^{15}N$ = 6.2‰) and Marine POM (${\delta}^{13}C$ = -20.3‰ and ${\delta}^{15}N$ = 10.3‰). ${\delta}^{13}C$ values of sedimentary organic matter showed a distinct gradient in the range of -20.7‰ to -191‰. The stable carbon and nitrogen isotope values of bacteria in the study were -20.8‰ to -18.6‰ for ${\delta}^{13}C$ and 6.5‰ to 8.6‰ for ${\delta}^{15}N$. From this results based on stable isotope measurements showed that in the bacteria was found to be dominated by Marine POM and Benthicmicoralge during 0 to 20 day. Whereas, terrestrial plant and riverine POM showed little in fluence to bacteria during the experiment.

Isotopic Evidence for Ontogenetic Shift in Food Resource Utilization during the Migration of the Slipmouth Leiognathus nuchalis in Gwangyang Bay, Korea (광양만 주둥치(Leiognathus nuchalis)의 서식처 이동에 의한 먹이원 변동 파악을 위한 안정동위원소 분석기법 적용)

  • We investigated carbon and nitrogen isotope ratios (δ13C and δ15N) of the slipmouth Leiognathus nuchalis to reveal the effects of body size, feeding strategy and spatial distribution on the food resource utilization during the migration in the Seomjin estuary and Gwangyang Bay. The δ13C values of L. nuchalis caught in the Seomjin estuary where the salinity is lower than 30 psu were much lower than those in the deep-bay area of Gwangyang Bay. Such a spatial heterogeneity in δ13C values of the L. nuchalis clearly indicates active feeding within the estuarine habitat. In contrast, the δ15N values of L. nuchalis showed a consistency among sites, indicating that this species occupies identical trophic level across the whole area. The slipmouth distributed throughout the bay area, reflecting its euryhaline characteristics. However, the distribution pattern appeared to be separated according to body size into smaller individuals in the low-saline estuary and larger ones in the deep bay. Overall results support the plastic feeding strategy of the slipmouth from zooplanktonic (estuarine habitat) to epibenthic (deep-bay habitat) feeder during the migration between estuarine to deep-bay habitats.

Spatial Characteristic in Food Sources for Benthic Invertebrates in an Estuary Tidal flat: Carbon and Nitrogen Stable Isotope Analyses (안정동위원소 비를 이용한 하구 갯벌에 있어서 저서 무척추 동물의 유기물 기원의 공간적 특성)

  • The spatial variability in the food chain structure of an estuarine environment(Nanakita estuarine, Japan) was investigated using stable carbon and nitrogen isotope. Potential organic matter sources(TP:Terrstrial Plant, MPOM:Marine particulate organic matter, BMA:Benthic microalgae, EPOM:Estuarine particulate organic matter), sedimentary organic matter and benthic invertebrates(Nuttallia olivacea and Nereidae) were sampled at four locations with different tidal flat types(e.g. sanddy, sanddy-muddy and muddy). The main objective of the present study was to determine food sources of Nuttallia olivacea and Nereidae along with small-scale spatial variability within the community of benthic invertebrates. TP(${\delta}^{13}C=-26.6{\pm}0.76$ and ${\delta}^{15}N=2.7{\pm}0.31$) and EPOM(${\delta}^{13}C=-25.5{\pm}0.13$ and ${\delta}^{15}N=5.2{\pm}0.46$) were isotopically distinct from BMA(${\delta}^{13}C=-16.3$ and ${\delta}^{15}N=6.2$) and MPOM(${\delta}^{13}C=-19.6{\pm}0.08$ and ${\delta}^{15}N=8.9{\pm}1.70$). ${\delta}^{13}C$ values of sedimentary organic matter showed a distinct gradient in the range of -27.4 to -22.8‰ with a declining trend from the upstream to the downstream stations. The stable carbon and nitrogen isotope values of benthic invertebrates in the study site was -22.8 to -18.4‰ for ${\delta}^{16}C$ and 8.1 to 11.9‰ or ${\delta}^{15}N$, respectively. Mixing model(Isosource) calculations based on stable isotope measurements showed that benthic invertebrates of Nuttallia olivacea and Nereidae were found to be dominated by MPOM and BMA in stations. Whereas, TP and EPOM showed little influence to benthic invertebrates. The current result suggests that the different contribution for benthic invertebrates should be affected by both seasonal variation and physical factor among stations.

  • Stable Isotope Analysis(SIA) of carbon and nitrogen is useful tool for the understanding functional roles of target organisms in biological interactions in the food web. Recently, mixing model based on SIA is frequently used to determine which of the potential food sources predominantly assimilated by consumers, however, application of model is often limited and difficult for non-expert users of software. In the present study, we suggest easy manual of R software and package SIAR with example data regarding selective feeding of crustaceans dominated freshwater zooplankton community. We collected SIA data from the experimental mesocosms set up at the littoral area of eutrophic Chodae Reservoir, and analyzed the dominant crustacean species main food sources among small sized particulate organic matters (POM, <$50{\mu}m$), large sized POM (>$50{\mu}m$), and attached POM using mixing model. From the results obtained by SIAR model, Daphnia galeata and Ostracoda mainly consumed small sized POM while Simocephalus vetulus consumed both small and large sized POM simultaneously. Copepods collected from the reservoir showed no preferences on various food items, but in the mesocosm tanks, main food sources for the copepods was attached POM rather than planktonic preys including rotifers. The results have suggested that their roles as grazers in food web of eutrophicated reservoirs are different, and S. vetulus is more efficient grazer on wide range of food items such as large colony of phytoplankton and cyanobacteria during water bloom period.

Introduction of a New Method for Total Organic Carbon and Total Nitrogen Stable Isotope Analysis of Dissolved Organic Matter in Aquatic Environments (수환경 내 용존성 유기물질의 총 유기탄소 및 총 질소 안정동위원소 신규 분석법 소개)

  • Dissolved organic matter (DOM) is a key component in the biogeochemical cycling in freshwater ecosystem. However, it has been rarely explored, particularly complex river watershed dominated by natural and anthropogenic sources, such as various effluent facility and livestock. The current research developed a new analytical method for TOC/TN (Total Organic Carbon/Total Nitrogen) stable isotope ratio, and distinguish DOM source using stable isotope value (δ13C-DOC) and spectroscopic indices (fluorescence index [FI] and biological index [BIX]). The TOC/TN-IR/MS analytical system was optimized and precision and accuracy were secured using two international standards (IAEA-600 Caffein, IAEA-CH-6 Sucrose). As a result of controlling the instrumental conditions to enable TOC stable isotope analysis even in low-concentration environmental samples (<1 mgC L-1), the minimum detection limit was improved. The 12 potential DOM source were collected from watershed, which includes top-soils, groundwater, plant group (fallen leaves, riparian plants, suspended algae) and effluent group (pig and cow livestock, agricultural land, urban, industry facility, swine facility and wastewater treatment facilities). As a result of comparing characteristics between 12 sources using spectroscopic indices and δ13C-DOC values, it were divided into four groups according to their characteristics as a respective DOM sources. The current study established the TOC/TN stable isotope analyses system for the first time in Korea, and found that spectroscopic indices and δ13C-DOC are very useful tool to trace the origin of organic matter in the aquatic environments through library database.
