Effects of pH and Nitrogen sources on the Pullulan Production by Aureobasidium pullulans (Aureobasidium pullulans에 의한 플루란 생산에서 pH와 질소원의 영향)

  • In this study, the effects of nitrogen sources and pH on pullulan production were investigated. As a result, the best nitrogen source in pullulan production by Aureobasidium pullulans was shown to be peptone and its product yield was 62%. Optimum concentration of carbon/nitrogen source ratio was 50/0.15 and the production of pullulan was inhibited at ratios higher than 50/0.15. Aureobasidium pullulans had produced 29.1, 27.4 and 26.5 g/L pullulan, respectively in media I, II, and III containing mixed nitrogen sources. This result showed that pullulan could be produced efficiently from mixed nitrogen source. It was found that pullulan yield with pH control was higher than that with no pH control. In fedbatch fermentation, pullulan yield obtained with a feeding rate of 0.05 N-g/L for nitrogen source was higher than that without nitrogen source feeding.

  • Biosynthesis of curdlan( ${\beta}$-1,3 glucan) was shown by fluorscence on cellufluor medium. The highest production of curdlan was produced when glucose was used as a carbon source and ($NH_4$)$_2$$SO_4$ was used as a nitrogen source. ${\beta}$ -form of curdlan was detected in the fingerprint region (890 $cm^{-1}$) by FT-IR spectrum and shown homogeneous ${\beta}$ -1,3 glucan by $^{13}C$ NMR spectrum ($C_1$-103 ppm, $C_2$-73.2 ppm, $C_3$-86.4 ppm, $C_4$-68.7 ppm, $C^{5}$-76.63 ppm, $C_{6}$-61.2 ppm). Transition of structure from triple helix coil form to random coil form was appeared at 0.1 ∼0.25 M NaOH concentration. It was shown that natural curdlan is a triple helix form in neutral but becomes weak in alkaline condition.

Effect of Medium Components on the Production of Lovastatin by Aspergillus terreus (Aspergillus terreus에 의한 Lovastatin 생산에 배지성분이 미치는 영향)

  • The biosynthesis of Lovastatin, a cholesterol lowering agent formed by the filamentous fungus Aspergillus terreus, was investigated in shaking flask. The effects of essential elements in the experimental medium such as carbon, nitrogen, phosphate sources, and amino acids were examined to increase Lovastatin productivity. Lovastatin production in shaking flasks was 68 mg/L in the used medium. Effect of carbon source on Lovastatin production was performed. As a carbon source in the medium, 45 mL/L of glycerol increased the Lovastatin production up to 256 mg/L, which was found to be improved almost 3.5 times in comparison with that in common medium. The optimum concventration of peptonized milk as nitrogen source was obtained 30g/L on Lovastatin production. The severe inhibition of the cell growth and the Lovastatin production were observed in shaking flasks conducted at the medium contained ammonium carbonate as a nitrogen source. Lovastatin production various concentrations of several phosphate compounds was also examined. The addition of either potassium phosphate diabsic or sodium phosphate dibasic increased the Lovastatin production and the optimal level of potassium phosphate dibasic was 6 g/L. Even though Lovastatin contain methionine-derived methyl group, L-methionine and DL-methionine tend to diminish the Lovastatin production. Among the amino acids, L-histidine and L-tryptophan had a remarkable enhancing effect on the Lovastatin production. The optimal concentration of L-histidine and L-tryptophan was 6g/L.

Ecological Importance of Benthic Microalgae in the Intertidal Mud Flat of Yeongheung Island; Application of Stable Isotope Analysis (SIA) (영흥도 조간대 갯벌 저서미세조류의 생태적 중요성; 안정동위원소 분석 활용)

  • In order to reconstruct a benthic foodweb structure and assess the role of benthic microalgaes as a diet source for benthos, we analyzed the carbon and nitrogen stable isotopes of diverse benthos (bivalves, crustaceans, gastropods and fishes) and potential diets (particulate organic matter, sedimentary organic matter, benthic microalgae, seagrass, and macroalgaes) in the intertidal mudflat surrounding Yeongheung Island. The ${\delta}^{13}C$ values of the diets indicated wide ranges (- 26.5‰ to - 8.4‰) while benthos showed a small range of ${\delta}^{13}C$ values (-12.1‰ to - 17.8‰), although they were in the same range. Except for green algaes among the macroalgaes as well as sedimentary organic matter, ${\delta}^{15}N$ values of the diet candidates ($5.7{\pm}1.0$‰) were lighter in comparison to those of the benthos ($11.8{\pm}1.9$‰). Based on the ${\delta}^{13}C$ and ${\delta}^{15}N$ data, the benthos were classified into 3 groups, indicating a different diet and trophic position. But benthic microalgae is the most important diet source for all three benthos groups based on their stable isotope ratios, suggesting benthic microalgae should be a main diet to the intertidal ecosystem. Hence this study highlights that the biomass of benthic microalgae as biological resource should be evaluated for the management of the intertidal ecosystem of Yeongheung Island.

Study on the characteristics of transition metals for TSSG process of SiC single crystal (SiC 단결정의 TSSG 공정을 위한 전이금속 특성 연구)

  • In this study, a heat treatment experiment was conducted to select a new melt composition that can easily control the unintentionally doped nitrogen (N-UID) without degrading the SiC single crystal quality during TSSG process. The experiment was carried out for about 2 hours at a temperature of 1900℃ under Ar atmosphere. The used melt composition is based on either Si-Ti 10 at% or Si-Cr 30 at%, and also Co or Sc transition metals, which are effective for carbon solubility, were added at 3 at%, respectively. After the experiment, the crucible was cross-sectionally cut, and evaluated the Si-C reaction layer on the crucible-melt interface. As a result, with Sc addition, Si-C reaction layers uniformly occurred with a Si-infiltrated layer (~550 ㎛) and a SiC interlayer (~23 ㎛). This result represented that the addition of Sc is an effective transition metal with high carbon solubility and can feed carbon sources into the melt homogeneously. In addition, Sc is well known to have low reactivity energy with nitrogen compared to other transition metals. Therefore, we expect that both growth rate and Nitrogen UID can be controlled by Si-Sc based melt in the TSSG process.

Characteristics of Biological Nitrogen Removal for Low C/N Ratio Municipal Wastewater Using Methanol as an External Carbon Source in $A_2O$ Fluidized Media Process (유동여재 $A_2O$공정에서 외부탄소원으로 메탄올을 이용한 낮은 C/N비 하수의 생물학적 질소제거 특성)

  • This experiment was performed to evaluate the characteristics of BNR system performance, behavior of pollutants as organic and nitrogen at each basin and the effects of C/N ratio on biological nitrogen removal with methanol as an external carbon source for a low C/N ratio municipal wastewater. A lab-scale $A_2O$ system by employing the aerobic basin with the fluidized polyurethane media, which was $10{\sim}20$ mm rube type like a sponge, was used. The aerobic basin was hybrid type to be suspended and fixed biomass. The obtained results from this study were as follows; When no methanol was added, suspended biomass was 3 times more than that of the fluidized media in this system(total biomass 80 g). Biomass growed by an external carbon was firstly attached on media, and then suspended. $COD_{Cr}$ concentration for the effluent was a range of 13 to 29 mg/L regardless of pouring an external carbon. For nitrogen, the effluent concentration was $20.0{\sim}35.9mg/L$(removal efficiency; 18%) in case of no addition of an external carbon, but was $2.5{\sim}9.0mg/L$(removal efficiency ; $71{\sim}83%$) with addition of methanol. For the characteristics of pollutants removal, most of $COD_{Cr}$ were removed at the anaerobic basin when no external organic carbon was added, and were removed at the anoxic basin in case of adding external organic carbon but at the aerobic basin in case of adding excess external organic carbon. On the other hand, most TIN(total inorganic nitrogen) were removed at the anaerobic basin when no external organic carbon was added, but when an external organic carbon was added, they were removed at the anaerobic basin under unstable condition and at the anoxic basin under stable condition.

Nutrient Distribution of Culm, Branches and Leaf in Phyllostachys bambusoides and Phyllostachys nigra var. nenosis (왕대와 솜대의 줄기, 가지, 잎에 양분 분포)

  • This study was carried out to determine effects of bamboo species on the distribution of nutrients in aboveground biomass of Phyllostachys bambusoides and Phyllostachys nigra var. nenosis, Damyanggun, Korea. The study site was established around 40-year-ago to produce bamboo culm and edible shoot production. Total 28 bamboos (14 P. bambusoides and 14 P. nigra var. nenosis) were cut to measure nutrient concentration of each bamboo component, such as culm, branches and leaf. Magnesium concentration in each bamboo component was significantly higher in the P. bambusoides than in the P. nigra var. nenosis. Nutrient concentrations except for calcium were significantly higher in the current-year-old bamboos than in the > 1-year-old bamboos. The nutrient concentration in leaf was generally highest in carbon, followed by potassium or nitrogen, phosphorus, calcium or magnesium. Total nutrient content in each bamboo component was significantly higher in the P. bambusoides than in the P. nigra var. nenosis. The nutrient content of bamboo biomass was the highest in carbon, followed by potassium, nitrogen, phosphorus, magnesium and calcium. The results indicate that P. bambusoides uptakes more nutrients compared with P. nigra var. nenosis during growth development.

Comparison of Morphological Characters and Stable Isotopes of Seeds between Wild Simulated Ginseng and Cultivated Field Ginseng (국내 산양삼과 인삼 종자의 형태적 형질과 안정성 동위원소 비교)

  • We compared the morphological characters and stable isotopes of seeds of wild simulated ginseng with those of cultivated field ginseng. Seeds were collected from ten areas for wild simulated ginseng and two areas for cultivated field ginseng. The length, width, thickness and number of seeds per gram were measured and the ratio of stable isotopes of carbon and nitrogen was analysed as well. There was a overlapping variation of morphological characters and the ratio of stable isotope of the wild simulated ginseng among study areas. In Geumsan area, compared with the seed of cultivated field ginseng, the seed of wild simulated ginseng was significantly small and light, and the ${\delta}^{15}N$(‰) of wild simulated ginseng was lower than that of cultivated field ginseng. However, it is somewhat impossible to compare the regional variation of the unique value of ${\delta}^{15}N$(‰) among study areas of wild simulated ginseng. We can suggest the value of ${\delta}^{15}N$(‰) can be used as detection factor for cultivation regime like chemical fertilization and organic farming.

  • 광물질(minerals)이란 일명 무기질이라고도 하는데 원자량 16이하인 수소(H), 탄소(C), 질소(N), 산소(O)를 제외한 나머지 원소들을 말한다. 지금까지 공부해온 영양소 즉 탄수화물, 지방, 단백질 및 비타민들은 유기화합물에 속하는 영양소들인데 비해서 광물질은 그것이 유기화합물의 일부 구성요소일 수도 있고 또는 무기화합물로 존재할 수도 있고 때로는 독립된 원소상태로 존재할 수도 있는데 우리가 영양소로서의 광물질을 언급할 때에는 화합물을 말하는 것이 아니고 그 화합물을 구성하고 있는 원소 그 자체를 의미한다. 중요한 영양소의 하나인 물($H{_2}O$)은 즉, 수소(H)와 산소(O)는 광물질로 분류되지 않는다. 한편 탄산칼슘(Ca$CO{_3}$)과 같은 화합물은 무기화합물로서 칼슘(Ca). 탄소(C) 및 산소(O)의 3가지 원소로 구성되어 있다. 동물체내에서 발견되는 광물질에는 30여 종류가 있는데 이들의 총 함량은 2$\~$5$\%$ 정도이다.

Establishment of Optimal Production Conditions of Transglucosidase Produced by Aspergillus niger (Aspergillus niger가 생산하는 transglucosidase의 최적 생산 조건 확립)

  • In this study, transglucosidase (TG), an enzyme produced by Aspergillus niger, synthesized isomaltooligosaccharide from ${\alpha}-(1{\rightarrow}4)$ linked substrates. The highest TG-producing A. niger KCTC6913 was selected from six kinds of species, and optimized TG producing conditions were established. Five different carbon sources (potato starch, sweet potato starch, corn starch, wheat starch, and dextrin) and three different nitrogen sources (yeast extract, malt extract, and beef extract) were tested to establish the carbon and nitrogen sources favorable for TG production. Measurements of TG activity after an initial culture at pH 5.0 for 15 days revealed that potato starch and yeast extract, which are basic culture media, resulted in the highest TG activity. In addition, A. niger KCTC6913 increased TG production under aerobic conditions and a controlled carbon/nitrogen ratio. In conclusion, to evaluate TG activity in the established optimal medium, it is confirmed that the basal and potato dextrose broth medium were used as a control, and the highest TG production was measured, which was highlighted in the established optimal medium.
