Estimation of Carbon Storages and Fluxes by Ecosystem Type in Korea (국내 생태계 유형별 탄소 저장 및 거동 산정 연구 현황 분석)

  • As climate change gets severe, the ecosystem acts as an important carbon sink, therefore efforts are being made to utilize these functions to mitigate climate change. In this study, we inventoried and analyzed the previous studies related to carbon storage and flux by ecosystem type (forest, cropland, wetland, grassland, and settlement) and carbon pool (aboveground and belowground biomass, dead wood, Litter, soil organic carbon, and ecosystem) in Korean ecosystems. We also collected the results of previous studies and calculated the average value of carbon storage and flux for each ecosystem type and carbon pool. As a result, we found that most (66%) of Korea's carbon storage and fluxes studies were conducted in forests. Based on the results of forest studies, we estimated the storage by carbon stock. We found that much carbon is stored in vegetation (aboveground: 4,018.32 gC m-2 and belowground biomass: 4,095.63 gC m-2) and soil (4,159.43 gC m-2). In particular, a large amount of carbon is stored in the forest understory. For other ecosystem types, it was impossible to determine each carbon pool's storage and flux due to data limitations. However, in the case of soil organic carbon storage, the data for forests and grasslands were comparable, showing that both ecosystems store relatively similar amounts of carbon (4,159.43 gC m-2, 4,023.23 gC m-2, respectively). This study confirms the need to study carbon in rather diverse ecosystem types.

  • For the purpose of optimizing poly-l3-hydroxybutyrate (PHB) production from Alcaligenes eutrophus, two-stage fed-batch culture was adopted. In this system, specifk growth rate was maximized during the first stage whereas specific production rate was maximized during the second stage. The optimal concentrations of glucose and ammonium chloride were 16.6 and 0.54 g/I in the growth stage and 20.0 and 0.07 g/l in the production stage, respectively. Proportional feedback control considering time lag was suggested for PHB production process and a simulator was developed for real-time control purpose.

  • By the successive enrichment culture, methanol-utilizing bacteria of 213 strains were isolated from soil samples collected from various places. Among them one strain showing excellent growth was selected. The organism isolated was obligate methylotroph and identified as Methylomonas methanolica on the basis of its mophological and physiological characteristics of the cell. The medium have been to be collected for the maximum biomass productivity. The microorganism was capable of growing satisfactorily on a medium containing only methanol 0.8% (v/v), ammonium sulfate 0.6%, magnesium sulfate 0.1%, phosphate salts, but did not require growth factor.

  • The effect of nutritional and cultural conditions for the production of anti-complementary polysaccharide from Pleurotus sajor-caju was studied. Maximum anti-complementary activity from its culture broth and mycelium was obtained by using lactose and maltose as a carbon source, respectively. Ammonium phosphate (monobasic) and serine were chosen for the highest activity among the inorganic nitrogen compounds and amino acids tested. Optimum temperature for the production of the anti-complementary polysaccharide was $25^{\circ}C$.

β-Glucosidase Formation In Cellulomonas sp. (Cellulomonas sp.의 β-글루코시다아제 생성)

  • To elucidate ${\beta}$-glucosidase formation on various carbon scurces by cellulolytic bact-eia, Cellulomonas sp. CS1-1, the strain was grown on Nutrient Yeast Broth, carboxymethyl cellulose, avicel and cellobiose using a Ouickfit FVIL fermentor operated in batch, and the growth characteristics on those substrates and ${\beta}$-glucosidase distribution of extra and intracellular enzyme components were studied. The results were: 1) ${\beta}$-glucosidase was always intracellular, and was formed under all growth conditions tested, ii) but levels of relative activities were higher when the culture was grown on cellobiose and on avicel, iii) the relative activities were always maximum during the growth phase of the organism irrespective of the substrate used.

  • The effects of liquid culture conditions and nutrient sources on the formation of protein-binding polysaccharide (PS) in Cordyceps militaris were examined. The formation amount of PS was increased in proportion to the growth rate of mycelium, in case of higher aeration or lower acidity. The optimum growth temperature of the mycelia was 25$^{\circ}C$ for the formation of PS. The optimum carbon source and nitrogen source were glucose and peptone, respectively. The ratio of C/N was optimal with 3% glucose to 0.5 % peptone. The sugar composition in the PS was greatly changed according to the carbon sources. The mycelium of Cordyceps militaris by liquid culture showed a higher electron donating ability than that by solid culture.

Synthesis of Alcohol-oxidase in Pichia pastoris on Various Carbon Sources (여러가지 탄소원에 의한 Pichia pastoris의 Alcohol-oxidase 생성)

  • The regulation of the synthesis of alcohol-oxidase(E. C. 1. 1. 3. 13) was investigated in the methanol-utilizing yeasts during growth on different carbon sources. For this experiment, Pichia pastoris CBS 2612 and Pichia pastoris CBM 10 were cultured in mineral salt medium by changing its carbon sources. The production of alcohol-oxidase was varied by the carbon sources. For example, alcohol-oxidase was undetectable in all strains submitted to the test in the medium with glucose, but its production was rapidely increased when the carbon source was changed from glucose to methanol after 48hrs of incubation. Moreover, this enzyme was not synthesized during growth on the primary aliphatic alcohols alone(ethanol, propanol, butanol or pentanol) or on the mixed substrates(0.5% methanol+0.5% primary aliphatic alcohols). When cells were grown on the various carbon sources(glucose, xylose, lactose, glycerol, galactose, saccharose, sorbose, lactic acid or acetic acid), The alcohol-oxidase activity was detected a very little amounts. These carbon sources together with methnol yieled far better synthesis of alcohol-oxidase than in case of carbon sources alone. Especially, the alcohol-oxidase activity of the cells grown on sorbose, lactose or lactic acid together with methanol was far better or similar than that of cells grown on methanol alone. The apparent Km values for the methanol of Pichia pastoris CBS 2612 and Pichia pastoris CBM 10 enzymes were 1.92 and 210 mM, respectively. It is also active towards alcohols of shorter alkyl-chain length than $C_7$, insaturated alcohols(allylalcohol, crotyl-alcohol) and secondary alcohols (iso-amylacohol, iso-butylalcohol). The affinity of alcohol-oxidase for this alcohols decreased with the increasing length of the alkyl-chain.

Mycelial Growth of Ectomycorrhizal Fungi by Different Carbon Sources in Liquid Culture (액체배양 시 탄소원의 종류에 따른 외생균근균의 균사생장 특성)

  • Four ectomycorrhizal fungi were tested for their ability to utilize seven carbon sources in modified Melin-Norkrans medium. After 60 days of mycelial culture, the highest mycelial growth in Hygrophorus russula (KFRI 1987), Sarcodon aspratus, Leccinum extremiorientale (KFRI 1194), and Tricholoma matsutake (KFRI 1256) was observed with use of dextrin used as a carbon source. H. russula, S. aspratus (KFRI 1676), and L. extremiorientale showed the lowest mycelial growth on nutrient medium with pectin. The utilization of homoglycans (starch, dextrin) in seven strains (except for T. matsutake KFRI 1256) was higher than that of heteroglycan (pectin). The final pH values of all culture media were decreased by pH 1.1~3.0 compared with the initial pH values of culture media. The dominant color of mycelia was white and varied according to the carbon sources (yellow, brown, and purple) in some strains. A single colony was observed in L. extremiorientale cultured in liquid media containing four or five different types of carbon sources, whereas multiple colonies were formed in liquid media containing six different types of carbon sources by six strains.

  • This study was done to improve the effectiveness of nitrification and denitrification using the aeration-anoxic combination method using CFSTR(continuous-flow stirred-tank reactor) attached with an anoxic reactor filled with a media. In order to calculate the concentration of nitric acid within the aeration tank proportional to the anoxic rate within the reactor, supernatant within the inflow and precipitation tanks were influxed into the anoxic reactor. The rate of nitrogen removal was calculated using the concentration of inflow and flow of returned supernatant. From the results of this experiment, the carbon source needed in the anoxic reactor came from the inflow so that anoxification was achieved completely using the inflow source without the introduction of an external carbon source. However, as the ratio of nitric acid becomes large in inflow and nitric acid flow, the carbon source within the input source decreases so that the concentration of carbon source is important.

  • Sisomicin, which is one of aminoglycoside antibiotics, was produced by Micromonospora inyoensis. The effects of carbon sources on sisomicin production were studied in batch cultures. Starch, dextrin and maltose were good carbon sources for the production of sisomicin. However, when glucose was used, the antibiotic productivity decreased significantly due to a carbon catabolite regulation. The carbon catabolite regulation depends mostly on carbon catabolite repression, but not on carbon catabolite inhibition. On the other hand, the growth-production curves of batch cultures show that sisomicin is produced most actively during the idiophase.
