Food Composition of Crab(Charybdis japonica) Preserved in Brine (민꽃게(Charybdis japonica)장의 식품 성분)

  • Pickled crabs made using Charybdis japonica are becoming part of our dietary habits. Yet research on this food is disappointingly scarce. It is generally accepted that the original food composition of pickled crabs defies scrutiny because various seasonings are added to soy for the production of pickled crabs in most regions. Accordingly, this study attempted to measure the food composition of the Charybdis japonica preserved in brine according to different storage days (10, 15, 20, 25, and 30 days) and different temperatures ($5{^{\circ}C}$, $15{^{\circ}C}$, and ambient temperature). It was found that Charybdis japonica is comprised of moisture (77.5%), ash (2.3%), crude protein (19.7%), and crude lipid (0.5%). The content of crude protein and crude lipid during the storage period tended to decrease over time.

Effect of Steroid Hormones and Intracellular $Ca^{++}$ on Taurine Transporter Activity in Murine Macrophage Cell Line (마우스 대식세포에서 스테로이드 호르몬과 세포내 $Ca^{++}$ 타우린수송체의 활성에 미치는 영향)

  • The activity of taurine transporter is affected by various extracellular stimuli such as ion, hormone and stress. To assess effects of steroid hormones antral cyclosporin A (CsA) on the taurine transporter activity, murine monocytic RAW264.7 cell line was stimulated with dexamethasone (DM), triamcinolone (TA), cortisone (CS), hydrocortisone (HCS), prednisone (PSN), prednisolone (PSL) and methylprednisolone (MPSL) in the presence of 12-0-tetradecanoylphorbol-13-acetate(TPA). Treatment of TPA on the cell line led to significant reduction of taurine transporter activity. However, in case of stimulation of the cells with steroid hormones in the presence of TPA, all of them recovered TPA-induced reduction of the taurine transporter activity. Treatment of the cells with CsA led to significant reduction of the taurine transporter activity. Ionomycin (IM) recovered the reduced taurine transporter activity by CsA, but failed in the presence of EDTA, a calcium chelating agent. These results showed that glucocorticoid hormone recovered TPA-induced reduction of taurine transporter activity and that IM recovered CsA-induced reduction of the transporter activity by increasing intracellular free $Ca^{++}$ concentration.n.

  • Taurine (2-ethaneaminosulfonic acid, $^+{NH}_3{CH_2}{CH_2}{SO_3^{-}}$) is endogenous amino acid with functions as modulator of osmoregulation, antioxidation, detoxification, transmembrane calcium transport, and a free radical scavenger in mammalian tissues. Taurine transporter(TAUT) contains 12 transmembrane helices, which are typical of the $Na^+$- and $Cl^-$-dependent transporter gene family, and has been cloned recently from several species and tissues. To analyze the expression of TAUT mRNA, one step RT-PCR was performed from human and mouse cultured cell lines and from various mouse tissues. The primers were designed to encode highly conserved amino acid sequences at the second transmembrane domain and at the fourth and fifth intracellular domains. RT-PCR analysis showed both of the human intestine HT-29 and mouse macrophage RAW264.7 cell lines expressed mRNA of TAUT. To define the expression patterns of the TAUT mRNA in the murine organs, RT-PCR was performed to detect cDNA representing TAUT mRNA from seven different mouse tissues. The TAUT was detected in all of the mouse tissues analyzed such as heart, lung, thymus, kidney, liver, spleen and brain. A large amount of transcript was fecund from heart, liver, spleen, kidney, and brain, while lung contained a very small amount of transcript.

Effects of Oral Taurine Supplementation on Plasma Concentration and Urinary Excretion of Free Amino Acids in Healthy Female Adults (타우린복용이 정상 성인여성의 혈장 유리아미노산 농도 및 소변내 배설에 미치는 영향)

  • Effects of oral taurine supplementation (6g/day) on plasma concentration and urinary of free amino acids were evaluated in healthy female adults. Among twenty five female volunteers(23.6$\pm$0.3 years old) participated in the taurine supplementation program, twenty four subjects successfully completed the two supplementation program. Plasma and urinary levels of free amino acids were determined by using an automated amino acid analyzer based on ion-exchange chromatography. Two weeks of taurine supplementation resulted in a 65% increase in plasma taurine concentration (p<0.001), Changes in fasting plasma amino acid concentrations followed by taurine supplementation were not spectacular, and were all within the normal range for human aldults. Taurine supplementation significantly elevated urinary methionine, asparagine, hydorxyproline and phosphoserine excretions(31~280%), and significantly decreased the urinary excretions of isoleucine, glutamate and serine compared to the values prior to taurine supplementation. For almost every individual amino acids, 24 hr urinary excretion level was significantly correlated to the urinary excretion value expressed as nmol/mg creatinine(p<0.001). A significant negative correlation found between plasma glutamine concentration and urinary glutamine excretion level suggests that the decrease in plasma glutamine concentration might be associated with the enhanced glutamine excretion in urine followed by taurine supplementation.

Taurine Content in Korean Foods of Plant Origin (한국인 상용 식물성 식품의 타우린함량 조사)

  • Taurine content in Korean foods of plant origin was determined for 118 commonly used food items including cereals, potatoes, pulses, nuts, seeds, vegetables and fruits. Taurine concentration in food sample was analyzed using an automated amino acid analyzer(Biochrom 20, Pharmacia LKB) based on ion-exchange chromatography. Taurine was frequently detected in plant kingdom in much lower concentrations(1/100~1/1000) than those found in marine lifes and mammals. Glutinous rice, glutinous millent and sorghum did not contain taurine, while 0.7~3.9mg taurine/100g dry wt were detected in rice, barley and their products. Potatoes and sweet potatoes contained 0.3~1.2mg taurine/100g wet wt, and seasame seeds, perilla seeds, almonds, walnuts and gigko nuts contained 0.7~3.0mg taurine/100g wt. Taurine concentration was undetectable in most of the pulses, and in a large number of vegetables. Garlic bulbs, eggplants, green peppers, lotus roots, and cabbages have a relatively high level of taurine(around 1mg taurine/100g wet wt) among vegetables. Taurine was absent or found in very low levels(<1mg taurine/100g wet wt) in most of the commonly used fruits.

Changes in Composition of Fish Meat by Thermal Processing at High Temperatures (고온가열처리에 의한 어육성분의 변화)

  • The mackerel meat was packed in square No 3B can, and thermally processed at $121.1^{\circ}C$ to reach Fo values of 5, 10 and 15. On heating at $121.1^{\circ}C$, pH and volatile basic nitrogen content increased markedly, and histamine was detected at concentrations of 3.98 mg/100g in raw meat, 10.45 mg/100g in Fo 5 sample, 7.54 mg/100g in Fo 15 sample, respectively. Total free amino acid content of mackerel meat decreased with increasing Fo values. Little changes were observed in free amino acid composition, but histidine decreased significantly during heat treatment. Degradation of inosine monophosphate was proceeded during preparation and thermal processing of samples, and trimethylamine oxide changed to trimethylamine almost equivalently. In changes of lipid components, peroxide and carbonyl values were increased markedly during heat treatment. When the sample was heated at the Fo values of 5 or higher, content of non-polar lipid(NL) was increased while that of polar lipid(PL) was decreased, and percentages of polyenes such as 22 : 6 decreased with increasing Fo values. The decomposition of polyunsaturated fatty acids in PL were much greater than those in total lipid and NL.

Quality Characteristics of Kimchi Fermented with Abalone or Sea Tangle Extracts (전복과 다시마 추출물을 첨가한 김치의 발효 특성)

  • This study evaluated the quality characteristics of fermented abalone-kimchi or sea tangle-kimchi during 28 days of storage at $4^{\circ}C$. Abalone and sea tangle were added as extracts compared to a control. We analyzed lactic acid bacteria, pH, acidity, salinity, reducing sugars, free sugars, organic acids, free amino acids, and sensory characteristics. The lactic acid bacteria levels were 4.4~5.3 log CFU/g on day 0, 6.8~7.1 log CFU/g after 7 days, and continued to rise during the fermentation period. The salinity increased slightly over time. As fermentation progressed, pH and reducing sugars, free sugars, and organic acid contents decreased. Free sugars, in particular, dramatically decreased about 70% compared to the initial day. The total free amino acid content was higher in abalone-kimchi. In terms of free amino acid contents, taurine was high in abalone-kimchi, while glutamic acid and aspartic acid content was high in sea tangle-kimchi. The sensory evaluation results show that abalone-kimchi and sea tangle-kimchi had higher scores for their savory taste and overall acceptance compared to the control.

Optimizing Maillard Reaction for Development of Natural Seasoning Source Using Oyster Hydrolysate (굴 가수분해물을 이용한 천연조미소스 개발을 위한 마이얄 반응의 최적화)

  • The oyster is called "milk of sea" which is abundant in taurine, glycogen, cellenium. It could be used in making natural source. Recently, consumers have more interest in natural source because of their diverse preference and its special taste. The goal of this study is to optimize maillard reaction condition for manufacturing natural seasoning using oyster and oyster cooking drip hydrolysate. The result was judged by browning degree and pyrazine, which is flavor components when food heating. Hydrolysate and sugar react according primarily to type of sugar - glucose, xylose and fructose. Xylose was selected as best sugar of browning degree. In the case of sugar contents, all conditions over 1% of sugar contents are almost same. Therefore, the lowest 1% of sugar was selected as appropriate condition. According to the reaction with different temperature, browning degree and pyrazine contents had been increased over $60^{\circ}C$, but the product at $120^{\circ}C$had off-flavored. So, $100^{\circ}C$ is the best condition for the browning reaction. And in accordance with different reaction time, after 6 hours, there was no change in pyrazine and browning reaction. Therefore, to manufacture natural seasoning source, it is optimal to react xylose for maillard reaction at $100^{\circ}C$ for 6 hr with hydrolysate of oyster and oyster cooking drip.

Correlations of Plasma Taurine, Homocysteine and Serum Lipid Levels of Elderly Women (노년기 여성들의 혈장 타우린의 농도와 혈장 호모시스테인 및 혈청 지질농도와의 상관관계)

  • This study was conducted to assess blood components caused by metabolic syndrome increasing in postmenopausal elderly women. The blood samples of these subjects were analyzed to investigate the correlation of plasma taurine levels and plasma homocysteine levels, and serum lipid profiles. The subjects were 33 elderly women($72.8{\pm}4.4$ years). Their mean height, weight and BMI were $150.5{\pm}5.7\;cm$, $57.5{\pm}6.3\;kg$ and $25.4{\pm}2.5\;kg/m^2$. 16 women of this study subjects have been chronic diseases such as hypertension or diabetes. Their fasting blood glucose was $98.2{\pm}24.0\;mg/d{\ell}$, and their plasma total cholesterol (TC), HDL-C, LDL-C, triglyceride(TG) were $216.5{\pm}29.9$, $52.1{\pm}10.7$, $145.7{\pm}27.9$ and $141.2{\pm}59.6\;mg/d{\ell}$, respectively. Their blood lipid profiles were higher than the standard levels of metabolic syndrome, thus these levels of lipid profiles may play a role as risk factors on the elderly person. Plasma taurine level of the subjects was $278.5{\pm}48.1\;{\mu}mol/{\ell}$, and their plasma homocysteine level was $12.8{\pm}2.9\;{\mu}mol/{\ell}$. The concentration of plasma vitamin $B_{12}$ was significantly decreased by aging(p<0.05). The correlation of plasma homocysteine and plasma folate showed significantly negative(p<0.05). Thus, the decreased levels of plasma vitamin $B_{12}$ and folate by aging might affect on the plasma homocyteine concentration acting as a risk factor of cardiovascular diseases for elderly person. The correlation of plasma taurine and hemoglobin, and their platelet showed significantly positive(p<0.05). In conclusion, the diet on the elderly person is one of the important factors to prevent their health from chronic diseases. This study recommends that well balanced diets are needed for elderly person to keep their health and prevent from metabolic syndrome.

Dietary Intakes, Plasma Levels and Urinary Excretions of Taurine in Adolescents and Adults Residing in Seoul Area (서울지역 청소년 및 성인의 타우린 섭취량, 혈중 농도 및 소변내 배설량에 관한 연구)

  • As diverse physiological functions of taurine have been reported, taurine-containing health drinks and products are marketed worldwide for the treatment of various conditions such as improvements of liver, heart and circulatory functions or as an aid to athletic performance. Although animal studies have shown that taurine is fairly safe when supplemented in the diet for an extended period, the effective dose range of taurine for dietary supplements is in controversy. Reports on dietary taurine intakes have been sparse, and would serve as a guideline for determining an appropriate taurine dosage. The present study was aimed to estimate dietary intake level of taurine using the taurine content database of commonly used food stuffs established recently in our laboratory, and also to evaluate plasma concentration and urinary excretion of taurine in adolescents and adults residing in Seoul area. Dietary taurine intakes of the subjects were 219$\pm$16.9mg/day for 16-19 years old(n=123), 177$\pm$18.1mg/day for adults older than 20 years old(n=123). Male subjects(n=115) consumed 216$\pm$21.1mg of taurine/day, while female subjects(n=131) consumed 181$\pm$14.3mg of taurine/day(p<0.05). The level of dietary taurine intake was positively correlated with the levels of dietary intakes of energy, carbohydrate, total lipids, cholesterol, vitamin A, vitamin B$_1$, niacin, vitamin C, calcium, phosphorous, sodium and potassium at p<0.01, and with dietary intakes of iron and animal lipids at p<0.05, respectively. Plasma taurine concentration of subjects were 135$\pm$5.9$\mu$mol/L, which is considered to be within a normal range for healthy subjects. The subjects excreted 1158$\pm$72.7nmol/ of tarine mg creatinine in their urine, which is approximated as 150-170mg of taurine/24hr urine based on the assumption that 18mg creatinine/kg/day is excreted in the urine of healthy adults, and this would be about 80% of the daily taurine intake observed in the same subjects. Dietary taurine intake level was positively correlated with plasma taurine concentration, as well as with urinary taurine excretion corrected by creatinine excretion at p<0.05. The present study was the first report of taurine intake, and plasma concentration and urinary excretion of taurine in a Korean population so far, and these results would serve as an index for the future study evaluating taurine status in a diverse population within and outside Korea.
