Radioiodine Labeling of Insulin Using Dimethylsulfoxide as a Labelling-Aid

  • Using dimethylsulfoxide (DMSO) as a labelling aid, insulin--$^{126}$ I of radioimmunoassay use has been effectively prepared. A small amount of DMSO was added to usual labelling mixture ana the reaction time was controled. The labelled insulin obtained in such a way showed improved bindabilities to the antibody and thus expressed larger dose-gradients in the plots of standard dose-response curves even though the labelling rate was decreased to some extent. However, by extending the reaction time to about 1 min, average labelling yield of 30% could be obtained. The average increase of bindability (B/F) in definite antiserum dilution was 2.5 comparing with 1.5 obtained in the absence of DMSO. Thus, the net bindability increase was 70% of those obtained in tile absence of DMSO. By means of a NMR spectrometry, it has been confirmed that the DMSO in the labelling mixtutre is converted to dimethylsulfone by chloramine-T. The results, generally agreed with the Stags's postulation, were discussed in view of a competitive oxidation of DMSO with disulfide linkages of the insulin molecule by the chloramine-T.

A Study the THMs Formation by Chloramination Disinfection (클로라민 소독에 의한 THMs 생성에 관한 연구)

  • This study was performed to investigate THMs formation by chloramination as a secondary disinfection in drinking water distribution system. As the $Cl_2/NH_3-N$ ratio increased from 1:1 to 4:1, the THMs concentration had actually no change in the breakpoint curve of pH 6, 7. At pH 8, THMs level was not augmented at between 1:1 and 5:1. In the $Cl_2/NH_3-N$ ratio of more than 5:1 and 6:1 respectively, THMs level was raised. Only the chloroform of THMs was insignificantly detected by both preformed chloramine and preammoniation applications, which was probably attributed to the stoichiometrically unstable equilibrium in the preparation of chloramine, whereas $CHCl_3$, $CHCl_2Br$ and $CHClBr_2$ of THMs were found in the application of free chlorine. However it turned out that the THMs levels by chloramine was incomparably inferior to that of free chlorine.

  • 인슈린-$^{125}I$를 클로라민-T법을 써서 $30{\sim}35%$의 표지 수율로 얻었으며 그 비방사능은 대략 100uCi/ug이었다. 생성물을 방사면역 측정용으로 표준화하기 위해 최적 표지부분을 전분젤 전기영동, 래디오오토그래피 및 인슈린 항체와의 인큐베이션등으로 선정하였다. 선정된 표지 인슈린으로 표준화한 결과 $50{\mu}U/ml$까지의 인슈린농도에 대해 항체와의 결합 인슈린-$^{125}I$(B)와 미결합 인슈린-$^{125}I$ (F)와의 비 (B/F)가 $0.2{\sim}1.6$으로써 매우 가파른 표준 dose-response곡선을 보여 주었다. 인슈린 방사면역 측정킷트를 만들어 그 사용 가능기간을 측정하였으며 일상 생산방법을 확립하였다.

  • 난포자극호르몬(HFSH)을 클로라민 티를 사용하여 방사성요오드로 표지하였으며 평균 표지수율은 대략 65%이었다. 표지호르몬을 방사면역측정용으로 사용하기 위하여 전분젤 전기영동과 젤여과법으로 분리정제하고 그 분리정제효과를 분석한 결과 젤 여과법이 분리시간, 간편성, 항체와의 결합력 등으로 보아 우수함을 알 수 있었다. 한편 유리표지호르몬과 항체와 결합한 표지호르몬의 비율를 결정하기 위하여 이중항체법을 크로마토전기영동법과 비교하여 본 결과 이중항체법에 의해서만 효과적 비율결정이 가능하였다.

The Inactivation and Microbial Regrowth Inhibition of Heterotrophic and Nitrifying Bacteria by Chloramination (클로라민 소독에 의한 종속영양세균과 질산화세균의 불활성화 및 재성장 억제)

  • This study was performed to evaluate the inactivation and microbial regrowth of heterotrophic and nitrifying bacteria using chloramine as a secondary disinfectant for drinding water distribution system. Three sets of the three reactors filled with the $Cl_2/NH_3-N$ ratio of 3:1, 4:1, and 5:1 were used in these experiments. Chloramine concentration were applied to each set of the reactors with $1mg/\ell$,\;2mg/\ell\;and\;3mg/\ell$, respectively. For the set with elapsed time and reached to zero level after 7 days. Heterotrophic bacteria remarkably increased and nitrification through the experimenatal period (21 day). Furthermore the regrowth of heterotrophic bacteria and nitrification were not found. More than $2mg/\ell$ of chloramine with $Cl_2/NH_3-N$ ratio of 3:1, the nitrification could be inhibited by 2 days of contact time.

  • This study was to evaluate disinfection efficiency of chlorine and chloramine as secondary disinfectants in the distribution systems. Indicator organism, HPC in the suspended and attached were measured for copper, galvanized steel, PVC, and carbon steel pipes. For suspended microorganism, the PVC pipe was markedly dense among the assessed pipe materials. The attached microorganism was markedly equivalent roughness of pipe materials. In copper and galvanized pipes, chloramine was more effective that free chlorine to disinfect suspended microorganism in the contact time of 2 hours. The contact time for the 99% inactivation of suspended microorganism by chloramine was longer than that of free chlorine. Regardless of pipe materials, chloramine was effective on both disinfection efficiency and 99% inactivation time for attached microorganism. In conclusion, chloramine which is good disinfectant for long contact time was recommended as secondary disinfectant in distribution system.

  • The disinfection effect of free residual chlorine and monochloramine on biofilm communities were investigated by CLPP (community level physiology profile) using Biolog GN plates. Low concentration of disinfectant, $0.5\;mg/\iota$ free chlorine and $1.0\;mg/\iota$ monochloramine, stimulated the growth of bacteria rather than disinfection. Bacterial concentrations were decreased at more than $1.0\;mg/\iota$ of disinfectants. CLPP was different with the type and concentration of disinfectant and sampling time. Common and different carbon sources were actively used with similar bacterial concentration in free chlorine and monochloramine. This represents the differences of bacterial communities with tap water contact times and disinfectant.

A Study on Formation Pattern of DBPs by Disinfection of Drinking Raw Water II (음용 원수의 염소소독에 의한 소독부산물 생성패턴에 관한 연구 II)

  • The formation pattern of DBPs (disinfection by-products) in raw water treated with hypochlorite, chlorine disinfectant was studied. TOC (total organic carbon), residual chlorine, turbidity and 14 DBPs in raw water from Han-river and Nakdong river during 1 ~ 14 days were determined. Total DBPs in Han river was 101.3 ng/mL (789.6 nM) after 7days and THMs (trihalomethanes) are the dominant portion of 68%. HAAs (haloacetic acids) and chloral hydrate were determined 19% and 10% respectively. In Nakdong river total DBPs was 98.4 ng/mL (678.6 nM) and dominant class was HAAs. (55.8 ng/mL, 57%) THMs(34%) and N-compounds like HANs (haloacetonitriles, 5%) and chloropicrin were increased. It may be explained that high concentration of NH4-N in Nakdong river react with chlorine produced chloramine and this formed different pattern of DBPs. As a result, total DBPs formation pattern depends on raw water and disinfectant and in generally the initial concentration of acidic HAAs was high and THMs was increased gradually.

  • This study was carried to investigate the characteristics of chloramination as a disinfection in drinking water distribution system. The raw water comes from midstream of Han river. In the range of pH 6~8, preformed chloramine of $Cl_2/NH_3-N$ ratio 5:1 had the HPC inactivation of more than 99% with lower pH and shorter contact time and available chloramine residual was decreased a little. In the chloramines of $Cl_2/NH_3-N$ ratio 3:1~5:1, the higher $Cl_2/NH_3-N$ ratio, the much inactivation of HPC was increased, but as contact time was longer, HPC inactivation of $Cl_2/NH_3-N$ ratio 3:1~5:1 were equaled. Bactericidal activity of three chlorine and postammoniation was influenced by free available chlorine completely and that of preammoniation was as follows : free chlorine ${\fallingdotseq}$ postammoniation>preammoniation>preformed chloramine.

Enzymatic Radioiodination of Insulin for Radioimmunoassay Use

  • Insulin was labelled with $^{125}$ I using lactoperoxidase as an oxidizing agent. The reaction product was purified via two stages: a starch gel electrophoresis(SGE) and a Sephadex gel filtration (SF). Upon comparison of the labelling yields and the bindabilities of the labelled insulin to its antibody, it has been found that the enzyme method shows higher yields (50%) and the better bindability to its antibody than the conventional chloramine-T method (35%). By checking the insulin blank labelling mixture with a SGE, a paper chromatography, and a radioautography technique, a by-product in the lactoperoxidase method has been identified. The separated fractions in SGE and SF were also analyzed and discussed.
