Qualitative Distribution of Stage-Specific Cuticle Proteins of Pieris rapace (배추흰나비(Pieris rupee) 발생 특이 큐티클단백질의 질적 분포에 관하여)

  • 배추흰나비의 조직과 발생시기에 따른 발생 특이 큐티클단백질의 질적 분포를 조사하기 위해 전기영동법, western blotting 및 autoradiography 법을 사용하였다. SDS-PAGE에서 유충기에는 4개, 용기에는 3개의 단백질이 발생 특이 큐티클단백질로 확인되었다. 전기영동적 이동도로 보아 유충기의 62 Kd 큐티클단백질은 지방체 표피, 혈림프에 모두 분포하며. 22 Kd, 16 Kd 단백질은 지방체와 표피, 그리고 19 Kd 단백질은 지방체에서 확인되고 있으며, 용기의 37 Kd 단백질은 표피에서. 28 Kd, 27 Kd 단백질은 표피와 지방체에 분포하는 양상을 보이고 있다 면역학적 방법으로 발생 특이 큐티클단백질의 동질성을 조사한 결과 용기의 27 Kd 단백질은 표피와 지방체에서 반응을 나타내며, 유충기의 22 Kd 단백질은 단지 표피에서만 미약하게 이의 동질성을 보여주고 있다 즉, 27 Kd 단백질은 지방체 22 Kd 단백질은 표피에서 기원하고 있어 발생 특이 큐티클단백질의 합성부위는 한 조직에서 기원하는 것이 아니라 각기 상이한 조직으로 부터 생성되는 것으로 사료된다.

Temporal and Spatial Role of Pupal Stage Specific Cuticle Protein in Artogeia rapae (배추흰나비 용기 특이 큐티클 단백질의 시공간적 역할)

  • Present study aims to investigate the topical distribution of pupal stage specific cuticle protein and its temporal and spatial role during the wing formation of Artogeia rapae. ArCP27(27 kd cuticle protein) was identified as pupal stage specific cuticle protein in cuticle tissues and has not shown any qualitative differences by local portions of body. ArCP27 maintained constant concentration just after pupal ecdysis to 5-day old pupal stage but thereafter decreased. In fat body, ArCP27 was found in both thoracic and abdominal fat body from the last larval to pupal stage. In wing cuticle, ArCP27 began to find from 5-day old pupal stage. Immunologically ArCP27 in thoracic and abdominal cuticle has the response against the ArCP27 at 5-day old pupa but since then has no response. But the antibody against ArCP27 has reacted to 5- and 7-day old pupal and adult wing protein. $^3H-leucine$ was not incorporated into ArCP27 in 5- and 7-day old thoracic and abdominal cuticle but was incorporated into ArCP27 in 7-day old wing cuticle and adult wing, suggesting that ArCP27 partly participates the wing cuticle formation by the process of digestion and reabsorption of old cuticle.

  • The physicochemical properties of damaged hair by irradiation of ultraviolet-B (UV-B) have been investigated by using transmission electron microscope and scanning electron microscope. The range of irradiation of hair irradiated for expectative 6 hours, 12 hours, 24 hours and 48 hours with stimulated ultraviolet ray. The treated hairs showed characteristic morphological damage pattern in the cornified cell of matrix and the cuticle following time past. The various sized vacuoles in the endocuticle of the cuticular cells was formed. The statistically significant differences in diameter of cuticular cell were observed in terms of tranverse swelling by formation of vacuoles. The hair cortex and matrix undergo long term exposure to UV-B radiation. The macrofibrils of cortex appeared to be affected most by UV-B, although the morphology and volume of melanin granule was not changed. The physicochemical destruction of hair matrix and cuticular cells is largely accelerated by long term irradiation of UV-B.

  • In this study, we observed the effects of repeated perms on the morphology of virgin hair of a healthy thirteen year-old girl with scanning electron microscopy. After the first treatment, the outer parts of cuticle cell were broken unevenly and roughly. Cuticle cells were lifted upward making a space. After the third treatment, cuticle cells were lifted off one another and the folded scales showed irregular surface areas. Broken pieces of cells were stuck on the surface and an empty hole was present in the endocuticle of the cytoplasm. We observed that cortex separated from cuticle layer more easilywith repeated treatments.

  • This study was investigated to observe morphological changes of two pre-astronauts' hair, male and female by electron microscopy and to analyze its tensile strength by using rheometer. The surface of those two pre-astronauts' hair, which were very rough and irregular, contained separated scales and destroyed remnants of cuticular cells. Also, there were many holes on the cytoplasm of the cuticular cells which forms the cuticle layer. The destruction begins when the endocuticle where the holes form gets destroyed. And then, The tensile strength of female pre-astronaut's hair was 14.60 mm which is 10% reduced, compared to that of the normal healthy hair. Thus, this result thought to be due to the prolonged change of the biorhythm and psychological instability of the pre-astronauts.

Study on the Tensile Strength of Bleached Hair (탈색모발의 인장강도에 관한 연구)

  • Human hair experience tensile forces during combing, bleaching and permanent dyeing processes. We have used rheometer to characterize bleached hair tensile response. Also, we investigated to morphological changes after tensile test of virgin hair and bleached hair using scanning electron microscopy. In tensile test, the tensile strength of virgin hair was $14.66g/cm^2$. They were measured as $12.95g/cm^2$, $12.61g/cm^2$ and $11.43g/cm^2$ for after the first, the second and the third trials of bleaching were done, respectively. Virgin hair had a higher tensile force than bleached hair. Tensile strength got lowered as more bleaching trials were done. In the observation of scanning electron microscopy, the fracture and lift-off of the cuticle outer layer were shown in virgin hair and bleached hair. The bleached hair were more lifted off at the outer cuticle layer than virgin hair. Consequently, chemical damage of bleaching agent caused weakening of the outer cuticle in the hair.

Effects of Aloe Extract on the Protection of Hair (알로에 추출물이 모발보호에 미치는 영향)

  • To protect healthy and beautiful hair, the hair protection effect was tested by adding an aloe extract from the hair solution. The treatment groups were grouped into four categories: Group II (Famma Treatment Only), Group III (Oxidase+aloe), Group IV (Reduce+aloe), and Group V (Oxidase+aloe, Release+aloe). The test hair is collected from women's hair in their 20s, 30s and 40s who have not undergone chemical treatment such as permanent and dying for one year and use 100% aloe stock solution. In order to evaluate hair thickness, cuticles, tensile strength, elongation and formation rate, the experiment environment was 28.9 degrees and 72% humidity. All statistical tests were done at 5% significant level. Cuticle, elongation, and formation rate were significantly improved (P<.05) and cuticle was best in group V, elongation was group III. In conclusion, aloe added to perm treatment were effective in improving cuticle, elongation rate, and formation of hair.

Cuticle micromorphology of leaves of Quercus L. (Fagaceae) and its taxonomic implications (참나무속(Quercus L.) 식물 잎의 큐티클 미세형태의 분류학적 의미)

  • Cuticle micromorphology of 22 species of Quercus and outgroup were examined by the SEM. Twenty-two species selected each two or three species in all section of the genus Quercus. The genus Trigonobalanus and Alnus are selected as outgroups. Ten characters of the inner surface and eight characters of the outer surface of the cuticle have been described. Some characters, such as the present of papillae, arrangement of subsidiary cell, shape of anticlinal cell wall are considered important character for infrageneric classification. A parsimony analysis of 18 characters resulted in 72 most parsimonious trees with its lengths of 66 steps. The topology obtained from the analysis showed two major lineages. Subgenus Cyclobalanopsis formed one clade by 75% and subgenus Quercus formed another clade by 57% bootstrap value. Based on the cuticle morphology, the two subgenus delimitation of Camus was supported. However, sect. Erythrobalanus and sect. Cerris formed one group, and sect. Lepidobalanus formed polytomy.

Studies on the Preservative Condition and the Ultrastructure of Hair of Newly Found Sixteenth Century Mummy in Paju (파주에서 발견된 16세기 미라 머리카락의 미세구조과 보존상태에 관한 연구)

  • The preservative condition and ultrastructure on the mummified hair collected from newly found female mummy in Paju, were investigated by using scanning and transmission electron microscopes. The female mummy was found in september, 2002 during the traditional reburial process for the buried ones. The hair of 16th century mummy showed very intact appearances during observation with electron microscope. And the structures of the cortex, medulla and cuticle were well preserved. The cuticle layer was easily discernable, which are composed of six to seven cuticular cells. Each cuticular cells surrounded and thus seperated from its neighbors by intercellular membrane complex. In the cortex, many macrofibrils and some melanin granules between them were observed. We observed well preserved rod form macrofibrils running parallel along the direction of hair shaft. Especially, melanin granules were aggregated in the cortex which was adjacent to the cuticlu layer. As to the cause for the well-preservation of 440 year old hair sample, the presence of surface coat on the hair, which are composed of various materials. As calcium was included in the surface coat in Electron Dispersive X-ray Spectroscopy (EDS), the hardening process of the surface coat by calcium might inhibit the water or microorganism infiltration into the hair.
