Analysis of Azodicarbonamide in Food Products by High Performance Liquid Chromatography (HPLC를 이용한 식품 중 아조디카르본아미드 분석)

  • This study was conducted to establish a method to analyze azodicarbonamide (ADA) in wheat flour. A new method using high performance liquid chromatography (HPLC) was developed for the determination of ADA in wheat flour. The recovery rate was 91.93~97.54%. The limit of detection for ADA was 0.02 mg/kg and the limit of quantification was 0.05 mg/kg. The monitoring results for ADA contents using the established methods showed that it was detected as the low value of 0.95 mg/kg in one of 51 flour samples (detection rate : 2%), but not detected in 59 bakery samples. The detected ADA level was suitable to its usage standard, compared to the standard (45 mg/kg). Although the detection rate was very low, the established analytical method of ADA will contribute to the management of ADA in processed foods such as wheat flour and bakery.

Survey on Fusarium Mycotoxin Contamination in Oat, Sorghum, Adlay, and Proso Millet during the Harvest Season in Korea (귀리, 수수, 율무, 기장의 수확기에 발생하는 Fusarium 곰팡이독소 오염도 조사)

  • A total of 244 cereal samples (oat, sorghum, adlay, and proso millet) were collected from fields to examine the contamination of Fusarium mycotoxins in cereals during harvest season in 2017 and 2018. The contamination levels of deoxynivalenol (DON), nivalenol (NIV), and zearalenone (ZEA) were analyzed individually by using the immunoaffinity column clean-up method with ultra performance liquid chromatography, and fumonisins (FUM) were analyzed by using the QuEChERS method with liquid chromatography-mass spectrometry. Highest level of NIV contamination (120.0-3277.0 mg/kg) was observed in oat samples among the analyzed cereals. In the adlay samples, DON contamination was the highest (maximum level 730.0 ㎍/kg). The proso millet samples had a high frequency of detection of NIV and ZEA (61.5% and 57.9%, respectively), but the levels were low (average detection level of NIV, 75.6 ㎍/kg, for ZEA, 21.5 ㎍/kg). Among the cereal samples, sorghum had the highest contamination frequency of DON, ZEA, and FUM, and the co-occurrence of Fusarium mycotoxin was 70.0%, which was higher than the average of 29.9%. In order to safely manage Fusarium mycotoxin levels in cereals, continuous research on the development of contamination prevention technologies together with monitoring of mycotoxin contamination is needed.

  • Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections, and infections of the body surface. Many analytical methods have been used to study oriental herbal medicines, such as thin-layer chromatography, column liquid chromatography, and high performance liquid chromatography (HPLC). In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out in order to separate pure compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 mL/min in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. The $^1H$ NMR spectrum revealed that the resonances at ${\delta}$ 4.10 and 4.20 ppm corresponded to three protons ($-OCH_3$), whereas those at ${\delta}$ 6.10 ppm corresponded to two protons ($-OCH_2O-$). Further, two aromatic protons (H-11 and H-12) conveys a doublet-doublet pattern. The H-11 doublet and H-12 doublet appear at ${\delta}$ 7.98 and 8.11, respectively. The $^{13}C$ NMR. spectrum showed a tetrasubstituted with a methylenedioxy group at C2 and C3, and two methoxy groups at C9 and C10. The chemical structure of the berberine was identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.

Purification and Characterization of Radish Myrosinase (무우 Myrosinase의 정제 및 특성)

  • The purification of myrosinase from radish roots was performed using Concannavalin A-Sepharose affinity chromatography and gel permeation HPLC, which gave a 22-fold-purification(S.P.A.=39,000 units/mg) with 50% recovery, SDS-polyacrylamide gel electrophoresis showed a single major band, suggestive of a relatively pure myrosinase, and the M.W. of the enzyme determined on gel permeation HPLC was ca. 124K. The enzyme showed on optimum pH of 6.5 and was stable at pH 6 to 7 at room temperature, but unstable below pH 4. The enzyme possessed an optimum temperature of $37^{\circ}C$, and gave a Vmax value of $40\;{\mu}moles/mg{\cdot}min$ and a Km value of 0.12mM for sinigrin. The purified myrosinase was activated maximally by 0.6mM of ascorbic acid, but somewhat inhibited by more than 2 mM ascorbic acid. The activities of myrosinase present in the peelings and the peeled radish amounted to approximately 1,333 units/g and 140 units/g weight, respectively and the peelings contained much more myrosinase activity than the peeled radish.

Study of the Separation and Elution Behavior of Phenols as Priority Pollutants in Reversed Phase Liquid Chromatography (역상 액체 크로마토그래피에서 유기오염물질로서의 페놀류들의 분리 및 용리거동에 관한 연구)

  • The optimum condition for the separation of priority pollutant phenols using isocratic elution has been determined. The elution behavior of eleven phenols has been also studied to interpret the retention. The reversed phase liquid chromatographic methods were performed on a ${\mu}$-Bondapak $C_{18}$ column with methanol-water, acetonitrile-water, and THF water mixtures as mobile phases. The COF method, where Snyder's solvent triangle concept was combined with a mixture-design statistical technique, was used to optimize the strength and selectivity of solvents for the separation of phenols. The optimum solvent composition, which gives a complete separation of eleven phenols, was found to be $MeOH:ACN:H_2O$ = 7:40:53. The plots of ln k' vs. -${\Delta}H^{\circ}$ and ${\Sigma}{\pi}$ of phenols showed relatively good linearities. Effect of van der Waals volume, pi-energy and hydrogen bonding on the retention of phenols were investigated. The following equation with the correlation coefficient of 0.9927 for ACN-water solvent system was obtained; $log^{k'}=2.515{\times}10^{-2}VWV-1.301{\times}10^{-1}E-3.674{\times}10^{-1}$

Analytical Methods on the Determination of Active Ingredients for Hair Nourisher Products (HPLC를 이용한 양모제 유효성분의 동시분석법)

  • The hair nourisher products are used for prevention of hair loss and classified as quasi-drug in Korea. As concerns about hair loss has been increased, the demand for hair nourisher products has been growing. It is difficult to analyze their main ingredients because they contain various ingredients including natural plant extracts, vitamins, preservatives and exfoliators. The purpose of this study was to develop and validate simultaneous analytical methods of active ingredients in hair nourisher products such as nicotinamide, tocopherol, salicylic acid, dexpanthenol and benzyl nicotinate by HPLC. The active ingredients were separated on a $C_{18}$ column by using acetonitrile/phosphate buffer as a mobile phase, and detected at UV 220, 270 and 300 nm. The calibration curve showed linearity in the range of $12.5{\sim}800\;{\mu}g/mL$ and the recoveries were 97.3 ~ 103.5 % (RSD 0.9 ~ 2.8 %) in liquid matrix and 101.9 ~ 115.9 % (RSD 0.7 ~ 7.7 %) in shampoo matrix. Validated method was applied to hair nourisher products obtained from distribution market. Fortunately, all samples met their criteria. This study might be expected to provide the method for determining active ingredients in hair nourisher products and lead to promote a rapid market entry.

Gas Chromatographic and Mass Spectrometric Determination of Alcohol Homologues in the Korean Folk Sojues (Distilled Liquor) (기체크로마토그래피 및 질량분석법에 의한 민속 소주중의 알코올 동족체 분석)

  • This work is undertaken to determine alcohol homologues so-called fusel oil that may be present in the Korean folk sojues (distilled liquor) made from grains and to describe sample preparation and analytical method by GC-FID-MS. Solid phase extraction method for sample preparation by using porous styrene divinyl benzene polymer (Porapak Q) was compared with steam distillation and solvent extraction method. Retention behaviors of homologous series of alcohols were also studied. Log values of retention time, molecular weight, boiling point, and capacity factor of alcohols showed linear correlations to the carbon number of an alcohol, to the oven temperature, and to the dielectric constant. Components such as methyl alcohol, n-propyl alcohol, isobutyl alcohol, isopentyl alcohol, and phenethyl alcohol have been identified. The more amount of isopentyl alcohol than other alcohols are contained in the Korean folk sojues, while that of n-propyl alcohol are contained in Chinese kaoliangchiew. Degree of similarity or dissimilarity and classification of the individual samples were discussed using multivariate statistical analysis(principal components analysis) based on GC data.

Separation of Waste TNT and RDX Mixture Using SMB Process (SMB 공정을 이용한 폐기 TNT와 RDX 혼합 용액의 분리)

  • Currently, researches on recycling and reuse of waste energetic materials have recently gained a great attention from advanced countries due to ever tightening environmental regulations. In this study, as a part of a recycling technology, the experiments and dynamic simulation of simulated moving bed (SMB) process were performed to efficiently separate TNT and RDX from their mixture, which are main components of ammunition. In order to determine the operation zone of SMB process, the retention times of TNT and RDX were measured using HPLC at different flow rates and the adsorption equilibrium of each component was obtained by using a moment method. According to the adsorption equilibrium and the triangle theory of SMB process, four operation points were determined and separation experiments were carried out by the SMB process using the solvent consisting of acetonitrile and water. Two different mixing ratios (6:4 and 1:1) of acetonitrile and water were chosen for the experiment due to the great impact of mixing ratio of the solvent on separation. The performance of SMB process was evaluated by purity, recovery, productivity and solvent consumption. Pure TNT and RDX were successfully obtained from the SMB process and the dynamic simulation for the SMB process agreed well with the experimental results. Therefore, the dynamic model could be applied for predicting the dynamic behavior of the SMB process and designing a large scale SMB process.

Two Phenolic Compounds Isolated from Umbilicaria esculenta as Phospholipase $A_2$ Inhibitors (석이로부터 분리한 페놀성 화합물의 phospholipase $A_2$ 저해활성)

  • Two depsides, medicinal herb products isolated from the methanol extract of Umbilicaria esculenta, inhibited human synovial fluid Phospholipase $A_2\;(PLA_2)$ ($IC_{50}$ of 0.22 and 0.26 mM, respectively). In the course of screening for antiinflammatory compounds from natural products, we successfully isolated two depsides $PLA_2$ inhibitory compounds, Orcinol and methyl orsellinic acid. The compounds were identified as orcinol and methyl orsellinic acid on the basis of various NMR studies including $^1H,\;^{13}C$ and DEPT experiments.

Inhibition of Cell Adhesion by Noncyclic Triterpenoids from Alpinia katsumadai (초두구로부터 분리한 비환식 트리테르페노이드계 화합물의 세포 접착인자 저해활성)

  • The present study purified two noncyclic triterpenoids, compound 1 and compound 2, that intervene interaction of ICAM-1 and LFA-1 from ethanol extracts of Alpinia katsumadai. The compound 1 and 2 inhibited adherence of sICAM-1 to THP-1 cells with an $IC_{50}$ of $7.59{\mu}g/m{\ell}$$6.98{\mu}g/m{\ell}$, respectively, without affecting viability of the cells. The compound 1 and 2 also inhibited interaction of CHO-ICAM-1 cells with THP-1 cells with an $IC_{50}$ $6.7{\mu}g/m{\ell}$$5.5{\mu}g/m{\ell}$, respectively. These findings suggest that the noncyclic triterpenoids from Alpinia katsumadai have inhibitory activities against cell adherent molecules. The present study proposes that noncyclic triterpenoids from Alpinia katsumadai can applied to therapeutic approaches to the diseases that are associated with adhesion of inflammatory cells.
