The Removal of Algae and Phosphorus in Eutrophic Waters Using Various Filter Media (몇 가지 여재를 이용한 부영양수 내의 조류 및 인 제거효과)

  • In this study, the four different filter media (sponge, volcanic stone, activated carbon and magnesium hydroxide) were tested for the removal of algae and phosphorus in the two eutrophic water samples (natural water and artificial algal culture with BG-11 medium). These filter media were used in the column device as single or combined applications. The effect of the $Mg(OH)_2$ on phosphorus removal was examined using different particle sizes (<2 mm and >2 mm) and concentrations (0, 10, 50 and 100 g $L^{-1}$) of magnesium hydroxide. The removal efficiency of phosphate by magnesium hydroxide was increased with longer experimental time and higher concentration. However, there was no significant difference in the degree of phosphorus removal between any two particle sizes (1 mg P $L^{-1}$: F=0.109, P=0.685; 10 mg P $L^{-1}$: F=1.542, P=0.355). Among the four media, activated carbon showed the most potent effect on the removal of both algae and phosphorus. The highest removal efficiency of algae and phosphorus was obtained by combining four columns of each filter medium. Interestingly, integration of four filter columns showed higher removal efficiency than activated carbon alone. The highest removal efficiency by integrated filter columns seemed to be caused by a synergistic effect of combined activated carbon and magnesium hydroxide.

Simultaneous Determination of Tetracycline Antibiotics by 3-Phase Hollow Fiber-Liquid Phase Microextraction (HF-LPME) and HPLC-UV/Vis (3-상 속빈 섬유-액체상 미량추출법(HF-LPME)과 HPLC-UV/Vis을 이용한 Tetracycline류 항생제 동시분석)

  • A simple and efficient preconcentration method was developed using three-phase liquid phase microextraction prior to HPLC-UV for simultaneous extraction and determination of tetracycline antibiotics (tetracycline, oxytetracycline, and chlortetracycline). The tetracycline antibiotics were separated simultaneously on a column ($C_8$, $3.0{\times}150mm$, $3{\mu}m$) with high selectivity and sensitivity using gradient elution. Under optimized conditions (extraction solvent, heptanal; pH of donor, 9.0; pH of acceptor, 1.0; stirring speed, 700 rpm; NaCl salt, 0%; and extraction time, 60 min), enrichment factors (EF) were between 5.6 and 22.3. The limit of detection (LOD) and limit of quantitation (LOQ) in the spiked urine matrix were in the concentration range of $0.08{\sim}0.8{\mu}g/mL$ and $0.4{\sim}1.6{\mu}g/mL$, respectively. The calibration curves were linear within the range of $0.1{\sim}32{\mu}g/mL$ with the square of the correlation coefficient being more than 0.995. The precision (as a relative standard deviation, RSD) and accuracy (as a relative recovery) within working range were 1.3~9.1% and 84~118%, respectively.

Application as a Cosmeceutical Ingredient of Euryale ferox Seed Extract (가시연꽃 종자 추출물의 화장품 원료로서의 특성)

  • In our search for the natural cosmetic ingredients, we found that Euryale ferox seed extract exhibited the strong antioxidative activity. Five active compounds were isolated from the ethyl acetate extract through various chromatographic methods and their structures were determined by NMR and MS spectral analysis. These compounds were identified as fucosterol (1), 3-(4-hydroxy-3-methoxybenzyl)-4-[(7'R),5'-dihydroxy-3'-methoxybenzyl]tetrahydrofuran (2), resorcinol (3), pyrogallol (4) and 4-O-methylgallic acid (5).We evaluated the antioxidative, antielastase activities and melanogenesis inhibitory effects of these compounds. The $SC_{50}$ values of compounds 2 ~ 5 for free radical scavenging activity were $17.0\;{\sim}\;100.2\;{\mu}M$ and especially compounds 4 and 5 were 6-fold more effective than ferulic acid as a positive control. And compounds 2 ~ 4 inhibited human neutrophil elastase with $IC_{50}$ values of $18.8\;{\sim}\;78.2\;{\mu}M$ and compound 3 also inhibited melanin synthesis in B16F10 melanoma cells with an $IC_{50}$ value of $492.8\;{\mu}M$. These results suggest that Euryale ferox extract having a lot of various active ingredients may be useful as a natural multi-functioning agent.

Analytical Method Development of Avicularin and Quercitrin in Astilbe chinensis Extract Using HPLC (HPLC를 이용한 노루오줌 추출물의 Avicularin 및 Quercitrin 분석법 개발)

  • This study attempted to eatablish a high performance liquid chromatography (HPLC) analysis method for the determination of avicularin, quercitrin as a part of the quality control for the development of functional cosmetic materials from Astilbe chinensis extract. HPLC was performed on a Capcell Pak C18 MGII column with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at 30 ℃. The analyte was detected at 254 nm. The HPLC method was performed in accordance with the international conference on harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation of avicularin and quercitrin were 0.094 and 0.285 mg/mL, 0.031 and 0.095 mg/mL respectively. Calibration curves showed good linearity r2 > 0.99990 for avicularin and r2 > 0.99994 for quercitrin. Precision of analysis was satisfied with less than 0.59% for avicularin and 0.63% for quercitrin. Recoveries of quantified compounds ranged from 100.97 to 101.77% for avicularin and 100.18 to 100.32% for quercitrin. These result indicated that the established HPLC method is very useful for the determination of marker compounds in A. chinensis extracts.

Anti-oxidative and Anti-bacterial Constituents from the Extracts of Rhododendron weyrichii Leaves (참꽃나무 잎 추출물 유래 항산화 및 항균 활성 성분)

  • In this study, we investigated anti-oxidative and anti-bacterial constituents from Rhododendron weyrichii leaves. DPPH and ABTS+ radical scavenging activities were screened for the ethanol extract and solvent fractions. Potent scavenging activities were appeared from the extract, ethyl acetate (EtOAc) and n-butanol (BuOH) fractions. Upon the anti-bacterial tests using Staphylococcus epidermidis and Propionibacterium acnes, extract, n-hexane (Hex) and EtOAc fractions showed strong activities. To isolate the active constituents, the EtOAc fraction was further purified to afford five phytochemicals; ursolic acid (1), corosolic acid (2), asiatic acid (3), astragalin (4) and isoquercetin (5). All of the compounds 1-5 were isolated for the first time from this plant. Among the isolates, the compound 4 and 5 showed strong DPPH and ABTS+ radical scavenging activities. Also, compound 3 exhibited the most potent anti-bacterial activity. In addition, the content of astragalin isolated from this plant was determined by UPLC and the quantity was about 8.1 mg/g for the 70% ethanol extract and 34.8 mg/g for the EtOAc fraction. Based on these results, it is concluded that R. weyrichii extract could be potentially applicable as anti-oxidative and anti-bacterial ingredients in cosmetic formulations.

Anti-inflammatory and Anti-bacterial Active Ingredients Derived from the Extract of the Leaves of Hydrangea Petiolaris (등수국 잎 추출물 유래 항염 및 항균 활성 성분)

  • In this study, the anti-inflammatory and anti-bacterial activities of the extracts from the leaves of the Hydrangea petiolaris were identified, and the chemical structure was identified by separating the active ingredient. As the result of the anti-inflammatory activity experiment using RAW 264.7 cells, it was confirmed that the n-hexane (Hex) and ethyl acetate (EtOAc) fractions inhibited the production of nitric oxide (NO) and the expression of iNOS protein in a concentration-dependent manner without cytotoxicity. In addition, the n-Hex and EtOAc fractions reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6). Upon the anti-bacterial tests using Staphylococcus epidermidis and Cutibacterium acnes, the extract, n-Hex, EtOAc and n-butanol (BuOH) fractions showed potent activities. In order to isolate the active constituents, the n-Hex and EtOAc fractions were further purified to afford four phytochemicals; phytol (1), corosolic acid (2), asiatic acid (3) and 1-O-p-coumaroyl-β-D-glucopyranoside (4). All of the compounds 1 - 4 were isolated for the first time from this plant. In addition, the contents of isolated compounds were determined by HPLC and the quantity of phytol (1) was 27.8 mg/g for the 70% EtOH extract. Based on the above research results, it is believed that it will be possible to develop a natural cosmetic material that has anti-inflammatory and anti-bacterial effects using the extract of H. petiolaris leaves.

P-wave Velocity Analysis Around the BSR Using Wide-angle Ocean-bottom Seismic Data (해저면 광각 탄성파 탐사자료를 이용한 BSR 부근의 P파 속도 분석)

  • In April 2008, KIGAM carried out an ocean-bottom seismometer (OBS) survey in the central Ulleung Basin where strong bottom simulating reflectors (BSRs) were revealed from previous surveys and some gas-hydrate samples were retrieved by direct sampling. The purpose of this survey is to estimate the velocity structure near the BSR in the gas hydrate prospect area using wide-angle seismic data recorded on the ocean-bottom seismometers. Along with the OBS survey, a 2-D seismic survey was performed whereby stratigraphic and preliminary velocity information was obtained. Two methods were applied to wide-angle data for estimating P wave velocity; one is velocity analysis in the $\tau$-p domain and the other is seismic traveltime inversion. A 1-D interval velocity profile was obtained by the first method, which was refined to layered velocity structure by the latter method. A layer stripping method was adopted for modeling and inversion. All velocity profiles at each OBS site clearly show velocity reversal at BSR depths due to the presence of gas hydrates. In addition, we could confirm high velocity in the column/chimney structure.

Development of Analytical Method for Rutin in Buckwheat Plant using High Performance Liquid Chromatography (메밀 식물체 함유 Rutin의 HPLC 분석방법 개발)

  • This experiment was conducted to know the appropriate methods for extraction and determination of rutin contained buckwheat plants. The efficient HPLC analytical condition of rutin contained buckwheat plants was developed. The gradient elution employed a $250mm\times4.6mm$ i.d. Tosoh ODS 120T column. The gradient system was used two mobile phases. A gradient elution was performed with mobile phase A, consisting of $2\%$ Acetic $acid-45\%$ Acetonitrile, and mobile phase B, comprising $2\%$ aqueous acetic acid, and delivered at a flow rate of 1mL/min as follows: 0-18 min, $50-100\%$ A; 18-20 min, $100-50\%$ A; 20-22 min, $50\%$ A. The UV detection wavelength was set at 355 nm. The limit of detection (LOD) for rutin standard compound was 20 ng/mL. And, the higher content of rutin in the extracts was obtained by $80^{\circ}C$ reflex extraction for 120 min. from plants of buckwheat using ethanol.

Changes of Adsorption Capacity and Structural Properties during in situ Regeneration of Activated Carbon Bed Using Ozonated Water (오존수 산화를 이용한 활성탄 흡착탑의 현장 재생 시 흡착용량 및 구조특성의 변화)

  • An in situ regeneration of activated carbon bed using an ozonated water was studied in order for avoiding the carbon loss, contaminant emission and time consuming for discharge-regeneration-repacking in a conventional thermal regeneration process. Using phenol and polyethylene glycol (PEG) as adsorbates, the adsorption breakthrough and in situ regeneration with the ozonated water were repeated. These organics were supposed to degrade by the oxidation reaction of ozone, regenerating the bed for reuse. As the number of regeneration increased, the adsorption capacity for phenol was reduced, but the change was stabilized showing no further reduction after reaching a certain degree of decrement. The reduction of adsorption capacity was due to the increase of pore size resulting in the decrease of specific surface area during ozonation. The adsorption capacity of phenol decreased after the ozonated regeneration because the in-pore adsorption was prevalent for small molecules like phenol. However, PEG did not show such decrease and the adsorption capacity was constantly maintained after several cycles of the ozonated regeneration probably because the external surface adsorption was the major mechanism for large molecules like PEG. Since the reduction in the pore size and specific surface area for small molecules were proportional to the duration of contact time with the ozonated water, careful considerations of the solute size to be removed and controlling the contact time were necessary to enhance the performance of the ozonated in situ regeneration of activated carbon bed.

Numerical Modeling of Coupled Thermo-hydro-mechanical Behavior of MX80 Bentonite Pellets (MX80 벤토나이트 펠렛의 열-수리-역학적 복합거동 모델링)

  • Numerical simulations of CIEMAT column test in Spain are performed to investigate the coupled thermo-hydro-mechanical (THM) behavior of MX80 bentonite pellets using TOUGH2-FLAC3D. The heater power and injection pressure of water in the numerical simulations are identical to those in the laboratory test. To investigate the applicability of the thermo-hydraulic (TH) model used in TOUGH2 code to prediction of the coupled TH behavior, the simulation results are compared with the observations of temperature and relative humidity with time. The tendencies of the coupled behavior observed in the test are well represented by the numerical models and the simulator in terms of temperature and relative humidity evolutions. Moreover, the performance of the models for the reproduction and prediction of the coupled TH behavior is globally satisfactory compared with the observations. However, the calculated stress change is relatively small and slow due to the limitations of the simple elastic and swelling pressure model used in numerical simulations. It seems that the two models are insufficient to realistically reproduce the complex coupled THM behavior in the bentonite pellets.
