4품종의 알팔파 (Medicago sativa L.) 종자로부터 직접 캘러스를 유도하고, 형성된 캘러스로부터 식물체를 재분화하는 조건을 확립하였다. 공시품종중 "Vernal" 품종이 캘러스 유도 및 재분화에서 가장 우수한 것으로 판명되었으며, SH(Schenk and Hildebrandt), MS (Murashige and Skoog), N6 (Chu) 배지중에서 SH 배지가 캘러스 유도와 재분화시에 모두 유리한 것으로 나타났다. 캘러스 유도 및 증식시에는 2,4-D를 3mg/$\ell$ 농도로 첨가한 SH-3 배지에서 가장 효율이 좋았으며, kinetin을 첨가하거나 kinetin의 농도가 높을수록 캘러스 형성 정도는 급격히 저하되었다.(중략)저하되었다.(중략)
This experiment was canted out to obtain embryogenic callus and to understand developmental mechanism of somatic embryogenesis in Oenanthe javanica ($B_{L.}$) DC. experiments included the examination of explant source and media for embryogenic callus production and the observation of developmental pattern of embryogenic cells and non-embryogenic cells. Embryogenic calli were formed on zygotic pro-embryos together with their endosperms when they were cultured on Ms media containing 1.0mg/L 2,4-D. Embryogenic calli were also formed on the intact surface in vitro grown stem or petiole segmentsafrer 6-8 weeks of culture, whereas non-embryogenic calli were formed on cut surfaces of the stem and petiole after 2 weeks of culture. Non-embryogenic calli were rhizogenic in suspension and solid media culture.

Comparison results of callus formation ratio from seed explants, callus sizes, regeneration ratios from callus and regeneration efficiency [calculated by following formular; callus formation ratio(%) ${\times}$ regeneration ratio(%)/100] for 27 ochardgrass (Dactylis glomerata L.)varieties imported and Hapsung 2 developed in Korea are as follows; 1. Among ochardgrass varieties showing more than 50% callus formation ratios, the descending order of callus formation ratio after bedding the seed explants for 4 weeks was 93M>Sparta> Pizza>Condor>Lidaglo>Glorus>Hapsung 2>Frode. 2. The callus sizes after bedding for 4 weeks were in the range of $\Phi$ 0.43cm~4.2cm in which there was 10 times size difference between the largest one and the smallest one but most of them were between ${\Phi}2.5cm~4cm$. 3. The regeneration ratio from callus among varieties were in the range of 0~36% and descending order of the upper 6 varieties was Plano>Akimidori>Justus>Lidacta>Currie>Hall mark. 4. The regeneration efficiency which is calculated by the ratios of regeneration from seed explant numbers was between 0 to 17.4% among which Justus showed the highest value in the 4-week treatment. 5. The correlation between callus formation ratios and the callus sizes, callus formation ratios and regeneration efficiency, and callus sizes and regeneration efficiency were r=0.5765, r=0.6365 and r=0.6246, respectively in 4-week callus and all the correlations were significant on the 1% level. 6. In 6-week callus, the descending order callus formation ratios from seed explants fur the best 6 varieties was Condor>Sparta>93 M>Justus>Potomac>Lidaglo>Frode. 7. The callus sizes formed were between ${\Phi}1.5~5.7cm$ in which Sparta, the largest one of ${\Phi}5.7cm$ was five times larger than the smallest one. The callus size of the control variety, Hapsung 2 was ${\Phi}3.8cm$, which belonged to a larger size. 8. Regeneration ratio showed a great deviation among varieties from 6-week old calli by showing from 0% to 100% in which all the calli were regenerated in Piano while no callus was regenerated in Juno. 9. The range of regeneration efficiency was between 0~28% among varieties in which the values from 6-week callus treatment were larger than those from 4-week callus treatment. Especially, the value of Potomac in 6-week was 3 times larger than that in 4-week. 10. The correlation between callus formation ratios and the callus sizes, callus formation ratios and regeneration efficiency, and callus sizes and regeneration efficiency were r=0.5369, r=0.6683 and r=0.5937. respectively in 6-week callus, and all the correlations were significant on the 1% level.
This study was carried out to investigate the effect of Germaniwn(Ge) treatment in the culture media on the Ge absorption by the callus induced from brown rice cv. Dongjinbyeo. MS medium was more effective on the growth ratio of callus and the content of Ge and some inorganic elements except K in callus than N$_{6}$ medium. The more Ge treatment in the N$_{6}$ or MS medium, the more Ge absorption by the callus, but the growth ratio of callus and the content of Ca, Mg, and K in callus were decreased. The content of Fe, Mn, Zn, and Cu was increased under treatment up to 100~200mg /$\ell$ Ge, but tended to be decreased under treatment more than that of Ge concentration. Under treatment less than 150mg /$\ell$ Ge, GeO$_2$(inorganic Ge) was more effective on the Ge absorption by callus than Ge-132(organic Ge), but Ge-132 was more effective on the Ge ab-sorption by callus and the activity of callus in case of treatment more than 150mg /$\ell$ Ge. The lower pH of culture medium, the higher Ge content in the callus. When callus was cultured on medium supplemented with Ge and 0.1~1.0mM of citric acid or myo-inositol, content of Ge and some inorganic elements in callus, as well as growth and dry weight of callus, were tend to increase in comparison to control, but myo-inositol was more effective on them than citric acid.cid.
Callus from scale segments of Lilium longiflorum 'Gelia' was effectively induced and maintained from unorganized tissue on the semi-solid medium by 0.42% Bacto agar with MS basal salts and vitamins of SH medium supplemented with 0.5 mg/L 2, 4-D, 1.0 mg/L NAA, 0.3 mg/L BA, and 3% sucrose. More than 5% of high sucrose level had inhibiting effect on regeneration capacity of formed callus and decreased callus growth. Various combinations of nitrogen did not effective to proliferate the ELC (Embryogenic-like callus), but friability of callus was increased in the medium containing only nitrate as nitrogen source. 5 mL conditioned medium into 30 mL fresh medium was good for cell growth. However friable cell aggregates during suspension culture had to form hard callus which hindered to establish suspention culture system. Addition of 2 g/L casein hydrolysate increased callus growth and friability of the hard callus. As a result of anatomical observation of callus, organogenesis such as shoots, roots and bulblets was independently induced from callus tissue. Somatic embryogenesis from callus tissue could be observed with low frequency.

This study was conducted to investigate the effects of copper in the efficiencies of callus formation and plant regeneration of orchardgrass (Dactylis glomarata L.). Seeds were cultured on MS medium containing $2\;mg/{\ell}$ 2,4-D and different concentrations ($0.1-100\;{\mu}mol$) of copper sulfate. Plant regeneration was achieved on N6 medium containing $1\;mg/{\ell}$ NAA, $5\;mg/{\ell}$ kinetin and $0.1-100\;{\mu}mol$ of copper sulfate. Callus formation was not affected by copper incorporation into MS medium. However, the efficiency of plant regeneration was promoted by copper and the maximum efficiency was obtained when $70\;{\mu}mol$ copper was incorporated in the culture medium. The average number of regenerated plants from the seed-derived callus was also increased by copper.
This study was conducted to establish a regeneration system of plantlets through callus culture induced from bulb scales of Lillium‘Gelria’. Friable callus was induced very easily from bulb scales, and grew vigorously on medium lacking growth regulators. In media with 0.5∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA, 100% of explants produced callus. Proliferation of callus was actively occurred on media containing 0.1 ∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA. Callus proliferation and regeneration of bulblets from callus were occurred simultaneously. Light condition was more effective for the callus proliferation and solid medium was better than liquid medium. Althrough callus was proliferated vigorously on media containing 0.1 ∼ 1.0 mg/L BA and NAA, the frequncy of plantlet regeneration was better on medium without growth regulators, then on medium with 0.1 mg/L BA and NAA.

This study was carried out to establish improved techniques on organogenesis from callus culture of Gladiolus. Organogenesis from the callus was effective in the half strength of MS solid medium without 2,4-D at 15 $^{\circ}C$ under 24 hours of daylength. Formation of adventitious root was most effective in the liquid shaking culture, and adventitious shoot induction was effective in the liquid stationary culture. From these results, we could find optimal culture conditions for redifferentiation from callus, in addition, liquid shaking culture revealed as more useful when compared with that of solid culture method for the redifferentiation of callus in Gladiolus `Topaz'.
In an attempt to optimize the in vitro-regeneration conditions necessary for the genetic manipulation of tomato species, we examined 'Ailsa Graig' cultivar of Lycopersicon for regeneration ability. The basal medium used for callus formation and shoot regeneration was MS (MS + vitamin) supplemented with six combinations of zeatin 2 mg/l, zeatin 2 mg/l + IAA 0.1 mg/l, zeatin 2 mg/l + IAA 0.5 mg/l, zeatin 4 mg/l, zeatin 4 mg/l + IAA 0.1 mg/l and zeatin 4 mg/l + IAA 0.5 mg/l. When all conditions tested were considered, however, only zeatin 2 mg/l was shown to be the best in shoot regeneration. The morphological characterization from in vitro-cultured callus of Lycopersicon esculentum L. var. 'Ailsa Craig' was investigated with scanning electron microscope (SEM). The surfaces of in vitro-cultured callus had well-defined epidermal cell in condition of zeatin 2 mg/l, but those of different treatments were twisted. These results suggested that shape of callus was involved in efficiency of shoot regeneration in tomato 'Ailsa Craig'.
