Cold Tolerance Assessment of Ever Ground-cover Plants for Extensive Green Roof System (저관리형 옥상녹화를 위한 상록 지피식물의 내한성 평가)

  • This study was carried out to suggest an experimental base in selecting the cold tolerance of plants. The cold tolerance of the plants were subject to laboratory low temperature treatments and cold processing time were evaluated using both electrolyte leakage and regrowth test. The Logistic model of nonlinear regression analysis was used to evaluate the lethal temperatures that were predicted with the range of $-16.1{\sim}-24.4^{\circ}C$. The order of low-temperature resistance was Sedum reflexum > S. spurium > Ophiopogon japonicus > S. album > S. takevimense > Dianthus chinensis. At the lowest temperature of $13.4^{\circ}C$ the electrolyte leakage value of the plants were lower than 50% demonstrating that they could be applied stably to the roof installed in Korea during the winter with the lowest temperature of $-13.5^{\circ}C$.

Influence of Temperature on the Survival, Growth and Sensitivity of Benthic Amphipods, Mandibulophoxus mai and Monocorophium acherusicum (국내산저서단각류 Mandibulophoxus mai와 Monocorophium acherusicum의 생존, 성장 및 민감도에 대한 온도의 영향)

  • A series of experiments was conducted to evaluate the effects of temperature on the survival, growth and sensitivity of the benthic amphipods, Mandibuluphoxus mai and Monocnrophium acherusicum, which have been recently developed as new sediment toxicity testing species in Korea. The biological performance for each amphipod species was determined by the survival and growth rates at different water temperatures. The influence of temperature on the sensitivity to reference toxicant, Cd, was determined by the comparison of survival rates of amphipods exposed to control and Cd-spiked seawater at different temperatures. Temperature significantly influenced on the survival, growth and Cd sensitivity of both amphipods. Tolerable ranges of temperature for the >80% individuals of both M. mai and M. acherusicum with sediment substrates were mostly overlapped (13 to 22℃). The daily growth rates of M. mai and M. acherkisicum increased proportionally with temperature up to 20℃ and 25℃. respectively. Similarly, the sensitivities of M. mai and M. acheyusicum to Cd increased with temperature up to 20℃ and 15℃, respectively. Overall results showed that temperature is a substantially important factor potentially influencing the results of lethal and sublethal bioassays using the amphipods. Therefore, defining the adequate ranges of experimental temperature for the toxicity testing species is the pre-requisite for the development of standardized bioassay protocols.

Effects of Temperature and Nematode Concentration on Pathogenicity and Reproduction of Entomopathogenic Nematode, Steinernema carpocapsae Pocheon Strain (Nematoda: Steinernematidae) (온도 및 농도가 곤충병원성 선충, Steinernema carpocapsae 포천 계통 (Nematoda: Steinernematidae)의 병원성과 증식에 미치는 영향)

  • Ecological studies on entomopathogenic nematodes are required to increase control efficacy against target insect pests and to obtain basic information for mass production. Thus, effect of temperature and nematode concentration on infectivity and reproduction of Steinernema carpocapsae Pocheon and that of exposure time and soil depth on infectivity were examined using Galleria mellonella larvae. Infectivity and reproduction were examined at five temperatures, 13, 18, 24, 30 and 35$^{\circ}C$ with seven concentrations, 0, 5, 10, 20, 40, 80 and 160 infective juveniles (IJs)/larva. Temperature and nematode concentration influenced infectivity and reproduction of S. carpocapsae Pocheon. Although G. mellonella larvae were killed by S. carpocapsae Pocheon at all given temperatures and nematode concentrations, mortality was higher at 24$^{\circ}C$ than other temperatures. Lethal time of G. mellonella by S. carpocapsae Pocheon was shorter with increasing temperature and nematode concentrations. S. carpocapsae Pocheon was not established in G. mellonella at 13 and $35^{\circ}C$. Time for the first emergence from G. mellonella cadaver was longer $18^{\circ}C$ (about 20 days) than 24 and $30^{\circ}C$ (about 5 days). The highest number of progenies was obtained at $24^{\circ}C$ with 80IJs/1arva, i.e., $18.8$\times$10^4$IJs were produced from a larva. In the exposure time assay, G. mellonella death was recorded in 10 minutes when 300 IJs were inoculated per larva. When S. carpocapsae Pocheon was inoculated at the rate of $10^{9}$ IJs/ha to G. mellonella at the depth of 0, 2, 5 and 10 cm of sand columns, 100% mortality and similar sex ratio were observed but number of established IJs in cadaver was decreased with deepening the soil depth. The results indicated that optimum temperature for infectivity and reproduction of S. carpocapsae Pocheon was $24^{\circ}C$ In addition, S. carpocapsae Pocheon was effective to target insects within 5 cm from the soil surface.

Effect of Cold Acclimation and Deacclimation on the Freezing Tolerance, Total RNA, Soluble Protein and Soluble Sugar in Chinese Cabbage (저온순화 및 탈순화가 배추의 내동성 및 total RNA, soluble protein, soluble sugar 함량에 미치는 영향)

  • This study was conducted to determine the change in freezing tolerance of Chinese cabbage (Brassica campestris L. ssp. penkinensis). Four-week old seedlings grown in a greenhouse at $25^{\circ}C$$\pm$0.5 were transferred to 5, 10 or 15$^{\circ}C$ condition for acclimation for one day, and then transferred to a lethal temperature of -3$^{\circ}C$ condition. Optimal acclimation temperatures were 5$^{\circ}C$ and 1$0^{\circ}C$. Freezing tolerance of leaf tissues was investigated during a week-long cold stress. Development of freezing tolerance was shown by survival ratio, and amounts of total RNA, soluble protein and soluble sugar. LT$_{50}$ value increased from -3 to -4$^{\circ}C$ after 5 days acclimation at 5 or 1$0^{\circ}C$, and this was considered to be the first indication of enhanced freezing tolerance. Plants, cold acclimated for five days, reached to a survival ratio of 60%. This increase in freezing tolerance was found to be associated with the increased levels of total RNA, soluble sugar and soluble protein. These metabolic changes imply the association of adjustment of growth and cell metabolism with low temperatures at the beginning of cold acclimation in chinese cabbage.e.

  • We studied expression patterns of thermotolerance gene (BcHSP17.6) in cabbages which was isolated from Chinese cabbage and we will attempt transformation of forage crops with the gene in order to increase thermotolerance of forage crops. Antiserum against a BcHSP17.6 protein was reacted with its antigen. With this antiserum, the accumulation of the 15- to 18-kD LMW HSPs under various heat shock (HS) conditions was quantified. The LMW HSPs began to be detectable at $35^{\circ}C$, and after 4 hours at $40^{\circ}C$ they were accumulated to a maximum level of 1.56 micrograms per 100 micrograms of total proteins in cabbage leaves and remained almost unchanged up to 24 hours after HS. Accumulation of the HSPs was reduced at temperatures higher than $40^{\circ}C$. We conclude that accumulation of these LMW HSPs are necessary for Chinese cabbages to survive at an otherwise lethal temperature.

Prediction of Heating Temperature of Jangjorim Food by Using Finite Element Method and Response Surface Methodology (유한요소분석법과 반응표면분석법을 이용한 장조림 식품의 가열온도 예측)

  • This Study was conducted to predict temperature profile of Jangjorim (boiled beef in soy sauce) food during retorting using the commercial NISA (Numerical Integrated Elements for System Analysis) program. NISA program is a good tool to simulate the temperature profile of a specific material based upon the finite element method. The cold point of Jangjorim food located not at the geometrical center but at 26.9 mm backward in y plane because specific heat of soy sauce was 20% higher than that of boiled beef. The effects of heat transfer coefficients on heat transfer during retorting process of Jangjorim were analyzed by response surface methodology (RSM). Independent variables were thermal conductivity of soy sauce, thermal conductivity of boiled beef, and convection heat transfer coefficient and dependent variables were temperature error and lethality error. Thermal conductivity of soy sauce was the most significant contributor among those (P<0.01).

Applicability of CATIS as a Postharvest Phytosanitation Technology against the Peach Fruit Moth, Carposina sasakii Matsumura (복숭아심식나방(Carposina sasakil)의 수확 후 소독 처리로서 CATTS 기술의 적용 가능성)

  • As an environment-friendly phytosanitary measure, CATTS (controlled atmosphere temperature treatment system) has been developed to kill several quarantine insect pests infesting subtropical agricultural commodities. This study tested any possibility to apply CATTS to apples to effectively eliminate the peach fruit moth, Carposina sasakii, which has been regarded as a quarantine insect from the imported countries. When the larvae of C. sasakii were directly exposed to $46^{\circ}C$ (an installed lethal temperature of CATTS), they showed a median lethal time at 14.66 min. Addition of high carbon dioxide to the temperature treatment enhanced the thermal limit susceptibility of C. sasakii to $46^{\circ}C$. CATTS device was constructed to automatically control $CO_2$ concentration and temperature with real-time monitoring both in the chamber and in the fruit. The larvae internally infesting apples were tested using the CATTS device and showed 100% lethality after 60 min exposure to a treatment of $46^{\circ}C$ under 15% $CO_2$ in the chamber. Relatively long exposure may be due to the deviation between the ramping temperature ($0.35^{\circ}C$/min) of the chamber and the ramping temperature (0.12-$0.23^{\circ}C$/min) inside apple fruit, where the tested larvae were located. This study suggests a possibility that CATTS can be applied as a quarantine measure to kill the larvae of C. sasakii locating inside the apples.

Biological Control of Apple Pests with Entomopathogenic Nematodes, Steinernema spp. (Steinernema 속 곤충병원선충을 이용한 사과원 병해충의 생물학적 방제)

  • Peach fruit month, smaller tea tortrix, and Melotontha incana are major pests of apple and apple trees throughout the country. In this work, we examined efficacies of entomopathogenic nematodes Steinernema carpocapsae and Steinernema glaseri against these apple pests. Steinernema carpocapsae showed 100% mortality after 24hr against peach fruit moth when it was applied on the larva with the concentration of 80 nematodes per larva, but Steinernema glaseri caused 83.3$\pm$5.8% mortality after 24hr at the concentration of 50 nematodes per larva. In the case of smaller tea tortrix, S. carpocapsae and S. glaseri caused 100%, 43.3$\pm$5.8% at the concentration of 50 nematodes per larva after 48 hr, respectively. However, 5~6 instar of Melotontha incana was not killed by treatments with S. carpocapsae and S.glaseri up to concentration of 200~800 nematodes per larva. The motility of nematodes in a soil increased as both inoculation concentration of nematode per larva and temperature increased. The mortality of G. mellonella by S. carpocapsae was 100% up to 10cm in depth and 56.7$\pm$5.8% at 10~15cm in depth when the temperature was $25^{\circ}C$ and 50 nematodes per larva were used.

Comparison of virulence by Acanthamoeba strains in a murine model of acquired immunodeficiency syndrome (면역결핍 마우스를 이용한 Acnnthamoeba 분리주별 병원성 평가)

  • The pathogenic potential of Acnnthamoebc strains was evaluated by experimental infection of murine AIDS (MAIDS) model. C57BL/6 mice were induced to immunocompromized state by intraperitoneal injection of LP-BM5 MuLV and revealed the typical splenomegalty and Iymphatic enlargement of axillar and inguinal regios on necropsy 4 weeks after viral infection. Although there was no significant difference in the mortality rate of MAIDS mouse according to the culture temperature, it was very different in the mortality rate from strain to strain of Accnthnmoebc. A. henIHi OC-3A strain isolated from the brain of a GAE patient showed !he highest mortality rate and A. culbertsoni A-1 strain from tissue culture was the second. KA/S3 and KA/S2 strains isolated from soil revealed very low virulence. The mice infected by intranasal inoculation of Acanthnmoebc showed relatively chronic course than intravenous inoculation. The gross findings of lungs and brains from infected mice were variable among mice. On the microscopic observations, the lungs showed much more severe inflammation and necrosis than the brains microscopically. This MAIDS model would be useful to study the opportunistic protozoan infections of AIDS patients. In the light of these results. the pathogenic potential and the virulence of Acnnthamoebo may be determined genetically.
