Functional Properties of Lupinseed Protein Concentrate (루우핀콩 단백질 농축물(LPC)의 식품기능성)

  • The functional properties of lupin seed protein concentrate (LPC) were examined and compared to those of soybean protein isolate (SPI) and Na-caseinate. LPC-50, of which protein level was 50%, was prepared by a two phase solvent (hexane: alcohol: water= 10:7:3) extraction method. LPC-70 was made from LPC-50 by removing the fractions solubilized by carbohydrate decomposing enzymes. The solubilities of LPC-50 and LPC-70 were similar to that of of SPI but slightly higher at pH 4-5, and less susceptible to the added salt. The apparent viscosity of LPC increased exponentially as the concentration increased over 6% level, and the change was similar to that of Na-caseinate. LPC showed strong pseudoplastic non-Newtonean flow behavior, which was similar to that of SPI The emulsifying capasity of LPC-70 was similar to that of SPI when salt was added. The foaming capacity of LPC was comparable to that of SPI. LPC showed high oil and water absorption capacities, which increased as the protein level was elevated. LPC-70 showed the highest oil absorption capacity of all the samples tested.

Levels of Polycyclic Aromatic Hydrocarbons in Fish, Shellfish and their Processed Products (국내 유통 어패류 및 가공품 중 Polycyclic Aromatic Hydrocarbons 함량)

  • Cocentrations of PAHs [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h.i)perylene, indeno(1,2,3-c,d)pyrene] in fish (n=120), shellfish(n=50) and their products (n=35) were estimated by saponification and extraction with n-hexane, clean-up on Sep-Pak Florisil Cartridges and HPLC/FLD. Overall recoveries for eight PAHs spiked into samples ranged from 90 to 106%. Mean level of benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene and indeno(1,2,3-c,d)pyrene were not detected, 0.01, 0.04, 0.07, 0.05, 0.004, 0.0008 and 0.06ng/g, respectively, similar to those reported by other countries.

  • Optimal extraction, separation, and capillary rinsing conditions for capillary electrophoresis (CE) were established to identify the cultivation site (domestic vs. foreign) of Job's-tears (Coix lachrymajobi var. mayuen) using 240 samples (domestic sample n = 121, foreign sample n = 119). Job's-tears was extracted with 30% ethanol and separated on a $50-{\mu}m-I.D.$ untreated fused-silica capillary. Optimal analytic conditions were: temperature, $45^{\circ}C$; voltage, 15 kV; detector rise time, 0.1 sec; and pressure injection, 20 sec. Separation of peak investigated using 0.1 M phosphate buffer (pH 2.5) containing 0.05% hydroxypropylmethylcellulose (P buffer) revealed the optimal separation buffer was P buffer containing 26 mM hexane sulfonic acid with 30% methanol. Under the optimal conditions established for CE, the average correct identification percentage of domestic or foreign Job's-tears was 82%.

Characteristics of Protease Produced by Rhizopus stolonifer, Rhizopus oryzae and Absidia corymbifera from Korean Traditional Meju (한국 전통 메주 유래의 Rhizopus stolonifer, Rhizopus oryzae 및 Absidia corymbifera가 생성하는 Protease의 특성)

  • Pretense production and its characteristics were investigated for Rhizopus stolonifer, Rhizopus oryzae and Absidia corymbifera which were isolated from Korean traditional meju. The optimum culture conditions of the strains for the production of protease in basic medium [wheat bran : 1% glucose solution=1 : 1 (w/v)] were 3$0^{\circ}C$ and 4 days. The optimum pH and temperature for the enzyme activity of crude enzymes produced by Rhizopus sto-lonifer, Rhizopus oryzae and Absidia corymbifera were pH 6.0 and 5$0^{\circ}C$, respectively. The enzymes were relatively stable at pH 4.0~7.0, at temperature below 4$0^{\circ}C$, and at NaCl concentration lower than 16%. The $K_{m}$ value for Hammastein casein was 3.3$\times$10$^{-4}$ , 0.75$\times$10$^{-4}$ and 1.3$\times$10$^{-4}$ M, and $V_{max}$ value was 17.2$\mu\textrm{g}$/min, 9.4$\mu\textrm{g}$/min and 7.8$\mu\textrm{g}$/min, respectively.y.

An Edible Alginate Microcapsulation of Entomopathogenic Nematode, Steinernema carpocapsae (알지닌캡슐을 이용한 곤충병원선충(Steinernema carpocapsae)의 섭식유도형 제제화 기술)

  • Field application of the entomopathogenic nematode, Steinernema carpncapsae, is limited by its susceptibility to UV irradiation and desiccation especially at leaf spray control. This study was conducted to develop the control technique using alginate biocapsulation of the nematodes against the beet armyworm, Spodoprera exigua and the tobacco cutworm, Sp. litura that are normally infesting hosts above ground level. The alginate capsules including infective juveniles gave significant feeding toxicities to the larvae of the two lepidopteran species. The lethality followed a typical sigmoid dose-mortality pattern with increase of the nematode densities embedded in the capsules. Moisture content in the capsule was critical to the survival of the infective juveniles. More than 80% nematodes could survive above 10% moisture content remained in the capsule. Remaining moisture content within the capsule was dependent on relative humidity, ambient temperature, and capsule size, but not on citric acid reaction time during capsule formation. More than 80% of infective juveniles in the alginate capsules could survive in distilled water at 15$^{\circ}C$ for 60 days. When these nematode capsules containing welsh onion extract as another phagostimulant were applied on the 3rd instar larvae of Sp. exigua infesting peanut plants, they resulted in about 90% control efficacy. These results indicate that the alginate capsulation can be used for leaf-spray agent of the entomopathogenic nematodes as well as for improved storage purpose.

Electrochemical Characteristics of Microporous Polymer Electrolytes Based on Poly(vinylidene-co-hexafluoropropylene) (PVdF계 미세기공 고분자 전해질의 전기화학적 특성)

  • In order to develop polymer electrolyte for lithium/sulfur batteries, highly microporous P(VdF-HFP) membranes were prepared by phase inversion method. Porous structure was controlled by extracting NMP with mixture of deionized water and methanol. Porous structure of the membranes was observed with SEM. Polymer electrolytes were prepared by soaking the porous membranes in 1M $LiCF_3SO_3-TEGDME/EC$. The ionic conductivity of polymer electrolyte was found to be at high as $2\times10^{-3}S/cm$ when the polymer membrane extracted by $80\%$ methanol was used. The microporous polymer electrolyte optimized in this work displayed high ionic conductivity, uniform pore size, low interfacial resistance and stable ionic conductivity with storage time. The ionic conductivity of polymer electrolytes was measured with various lithium salts, and the conductivity showed $3.3\times10^{-3}S/cm$ at room temperature when $LiPF_6$ was used as a lithium salt.

Quality and Volatile-Flavor Compound Characteristics of Hypsizigus marmoreus (느티만가닥 버섯의 품질 및 휘발성 향기 성분 특성)

  • Hypsizigus marmoreus is a wild mushroom commonly consumed in South Korea due to its beneficial effects on health. In the present study, the general chemical and inorganic-element composition, the total amino acid contents, and the volatile-flavor compounds of H. marmoreus were investigated for food uses. The proximate compositions consisted of 60.1% carbohydrate, 32.0% crude protein, 8.98% moisture, 5.0% ash, and 2.0% crude lipid. The minerals in H. marmoreus were found to be as follows; potassium (429.5 mg), phosphorus (101.9 mg), sodium (20.3 mg), magnesium (54.86 mg), calcium (2.7 mg), zinc (0.8 mg), iron (0.7 mg), manganese (0.2 mg), and copper (0.1 mg), based on 100 g of mushroom dry weight. Seventeen kinds of total amino acids were found in H. marmoreus, with the glutamine acid content being the highest (2,340 mg/100 g), followed by the asparagine, serine, arginine, and leucine contents. The volatile-flavor components of H. marmoreus were collected via simultaneous steam distillation extraction (SDE), and were analyzed via gas chromatography-massspectrophotometry (GC-MS). A total of 17 volatile-flavor compounds were identified, including eight aldehydes, seven alcohols, one acids, and one other compounds. The most abundant compound was 2,3,6-trimethy1 pyridine, which accounted for more than 40% of the total volatiles; other important compounds were 1-octen-3-o1, buty1hydroxytoluene (BHT), isoocty1 phthalate, 3-octanal, 1-undecanol, and 2-amylfuran. These results provide preliminary data for the development of H. marmoreus as an edible food material.

Analysis and Decrease of Cyanogenic Glucosides in Flaxseed (아마인에 함유된 시안배당체의 분석과 저감화)

  • Linustatin and neolinustatin in flaxseed are called cyanogenic glucosides which produce acetone, hydrogen cyanide (HCN) and sugar when hydrolyzed. The HCN is the chemical whose toxin is being issued in food safety. Linustatin and neolinustatin were found in flaxseed by 206.5 and 174.2 mg/100 g, respectively using a RI-HPLC and LC/MSn analysis of $80\%$ methanol extraction. Also, HCN (137.38 mg/kg) was found in flaxseed distillate by autohydrolysis and IC analysis. More than 85$\%$ of linustatin, neolinustatin and HCN were removed when flaxseed was heated for more than 2 hours in $200^{\circ}C$. Especially, HCN was decreased to $98\%$ by heating only 30 min in $200^{\circ}C$.

Volatile Flavor Compounds of Korean Native Lilium (한국 자생나리의 휘발성 향기성분)

  • Volatile fragrance components in 5 kinds of Korean native Lilium were investigated and compared. The volatile components were extracted by SDE (simultaneous steam distillation and extraction) and identified by CC and GC-MS. As a result of the analysis of volatile aromatic ingredient of L. leichtlinii var. tigrinum Nickels., L. concolor var. parthneion Bak., L. tsingtauense Gilg., L. hansonii Leichtl., and L. amabile Palibin., using frozen materials, 60 kinds of volatile compound were identified, which were 28 aldehydes, 9 ketones, 8 alcohols, 5 esters, 5 acids, 3 furans and 2 others. The GC patterns of the aroma components of all samples resembled but the peak areas were different according to species, though all of them are Korean native Liliums.

Separation of 2,6-dimethylnaphthalene in Dimethylnaphthalene Isomers Mixture by Crystallization Operation (결정화 조작에 의한 Dimethylnaphthalene 이성체 혼합물 중의 2,6-dimethylnaphthalene의 분리)

  • Light cycle oil (LCO), one of the by-products of the catalytic cracking gasoline manufacturing process, contains a lot of valuable aromatics. In particular, 2,6-dimethylnaphthalene (2,6-DMN) contained in LCO has been becoming important as the basic material of polyethylene naphthalate plastic and liquid crystal polymer, etc. If it were possible to separate and purify the valuable aromatic hydrocarbons (such as 2,6-DMN) from LCO, which have only been used as fuel mixed with heavy oil, it would be very meaningful in terms of the efficient use of resources. We investigated the high-purity purification of 2,6-DMN by the combined method of melt crystallization (MC) and solute crystallization (SC). The enriched DMN isomer mixtures (concentration of 2,6-DMN : 10.43%) recovered from LCO by distillation-extraction combination and the crystal recovered by MC used as raw materials of MC and SC, respectively. The solvent of SC used was a mixture of methanol and acetone (60 : 40 wt%). The crystal of 2,6-DMN with a high-purity of 99.5% was recovered by MC-SC combination. We confirmed that the MC-SC combination was one of the very useful combinations for the high-purity purification of 2,6-DMN contained in the enriched DMN isomer mixtures.
