Inhibition Effect of Against Angiotensin Converting Enzyme of Flavan-3-ols isolated Korean Green Tea (한국산 녹차로부터 분리한 Flavan-3-ol 화합물의 Angiotensin Converting Enzyme 저해 효과)

  • For the Purpose of utilizing tannins in the functional foods and crude drugs, the enzyme inhibition of tannins isolated from Korean green tea were determined. Acetone extract from Korean green tea showed inhibition effect against the angiotensin converting enzyme. The galloyl tannins showed higher inhibition activity against angioteosin converting enzyme than the nongalloyl tannins. In terms of stereo isomers, (-)-epicatechins had higher inhibition activity than the (+ )-catechins. The synergistic activity was also observed. Tannins isolated from Korean green tea appeared to be incompetitive inhibitor against the angiotensin converting enzyme.

Optimum Soaking Condition of Raw Soybean for Meju Preparation (메주 제조를 위한 원료 대두의 최적 침지 조건)

  • In order to establish scientific foundations for the production of meju, optimum soaking conditions of raw soybean were investigated. 100% hydration of raw soybean by soaking conditions were observed in 17 hrs at 5$^{\circ}C$, 15 hrs at 1$0^{\circ}C$, 11 hrs at 15$^{\circ}C$, 9 hrs at 2$0^{\circ}C$, 7 hrs $25^{\circ}C$, 5 hrs 3$0^{\circ}C$, 4 hrs at 4$0^{\circ}C$ and 3 hrs at 5$0^{\circ}C$. Exudation amounts of total free amino acids and free sugars were the least when soybean was soaked for 2$0^{\circ}C$ for 9 hours.

  • This study was to investigate antiallergic contact hypersensitivity of the polyphenol fractons isolated from persimmon leaves (Diospyros kaki folum). The result of the allergic contact dermatitis is a common skin disease resulting from specific immunologic sensitization due to topically applied allergens. Here the contact hypersensitivity was assayed and obdominal skin morphological changes including mast cells were examined. At contact hypersensitivity assay, less right ear swelling occured in the polyphenol fraction I, II and III groups compared to the control group. The number of mast cells was significantly decreased in the sample groups than the control group.

  • This study was conducted to investigate antiallergy of the polyphenol fractions isolated from persimmon leaves (Diospyros kaki folum). The result of the passive cutaneous anaphylaxis (PCA) reaction of expriment on anti-allergy, the allergy remarkably decreased in polyphenol fraction I, II and III groups in comparison with the control group. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.

  • The alkaline protease producing mold isolated from and identified as Aspergillus fumigatus. It was found that the production of alkaline protease reach to maximum was cultured for 3 days at $30^{\circ}C$. The enzyme was purified 86.13 fold and yield of the enzyme purification was 6.4%, The purification procedure include ammonium sulfate treatment, gelfiltration on Sephadex G-25, G-75, G-150 and DEAE-cellulose ion-exchange chromatography. When the purified enzyme was applied sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the molecular weight was estimated about 63000. This enzyme composed 17 amino acids and main amino acids of this enzyme were glycine and glutamic acid.

  • Optimum conditions for the enzymatic hydrolysis of isolated sesame meal protein were investigated. Optimum conditions by papain were $60^{\circ}C$, pH 6.0, 3% enzyme concentration to substrate and 1.5% substrate concentration, respectively. The optimum operating conditions using pepsin were $55^{\circ}C$, pH 9.0, 3% enzyme concentration to substrate and 1% substrate concentration. The optimum operating conditions using trypsin were $60^{\circ}C$, pH 9.0, 1% enzyme concentration to substrate and 1% substrate concentration.

  • To extract insoluble proteins and improve functional properties of sesame meal proteins was treated with phytase and protease from Aspergillus sp. It was found that the optimum pH, optimum temperature, optimum treatment time and optimum unit of enzyme for extraction of protein were pH $10{\sim}12$ (alkaline), $60^{\circ}C$, 11 hr. and 900 units of phytase and 60 units of protease, respectively. The foaming capacity, foaming stability, oil absorption and water absorption of sesame meal protein after treatment with phytase and protease were increased as compared to the control.

  • A highly viscous biopolymer from Bacillus coagulans CE-74 was purified and its rheological properties were studied The rheological properties of biopolymers produced by Bacillus coagulans CE-74 were studied at the temperature ranges with 20~8$0^{\circ}C$, at the concentration of 0.5~4.0%, at the pH ranges from 3 to 1 and at the shear rate fo 7.34~73.38 sec-1. The apparent viscosity of biopolymer was decreased with increasing shear rate, and thereby biopolymer showed pseudoplastic characteristics. Biopolymer solution showed a characteristic of non-Newtonian fluid properties. At the concentration of 1%, the consistency index and the flow behavior index were shown at 2.64 poise. sec11 and 0.8571, respectively. All dispersions were pseudoplastic fluids described accurately by Herschel-Bulkley model. The change of the biopolymer viscosity on pH showed the highest value at the pH 7.0 and it showed lower at acidic conditon that at alkaline condition comparatively.

  • Biological activities and anticarcinogenicity of Korean Pear peel were investigated, Electron donating activity and superoxide dismutase (SOD)-like activity of fraction II, III were up to 80% and 50-60% at 50 ppm, respectively, Inhibitory effects on xanthine oxidase were about 80% at 50 ppm, breast adenocarcinoma was about 60% at 2,000 ppm, higher III than II Inhibitory effect on prostate adenocarcinoma was about 23% at 500 ppm. In conclusion, Korean pear peel was expected to use as a functional material.

Effect of Polyphenol Compound from Korean Pear(Pyrus pyrifolia Nakai) on Lipid Metabolism (한국산 배로부터 분리한 Polyphenol 분획물이 지질대사에 미치는 영향)

  • This study was conducted to investigate the effects of the polyphenol fraction isolated from pear on the reduction of fat accumulation in rats fed on high fat diet for 5 weeks, which was examined by analyzing the lipid composition in serum, liver and feces biochemically. It was shown that the levels of total lipid and total cholesterol in serum were remarkably reduced in polyphenol fraction supplemented group as compared with the control group. The levels of total lipid and triglyceride were also significantly lower in polyphenol group than control group. The contents of total lipid, total cholesterol and triglyceride in feces were tended to be slightly increased in polyphenol group compared to control group. The total protein and albumin of polyphenol groups were lower compared to those of control group, which were not significant.
