Characterization of Erwinia tasmaniensis Isolated from Nuruk Producing Alginate Lyase (누룩으로부터 분리한 알긴산 분해 효소 생산 균주인 Erwinia tasmaniensis의 특성)

  • Oligosaccharides production showed various biological activities in vivo like functional foods and industrial materials utilized available within many practical applications which have obtained from the degradation of alginate. Alginate is rich in the main component of seaweeds especially the brown algae. We investigated what degrading alginate from seaweeds to make alginate oligosaccharides can utilize in various fields using enzyme secreting Erwinia tasmaniensis. In this study, we observed an optimal culture condition of E. tasmaniensis, and characteristics of alginate lyase secreting E. tasmaniensis. These bacteria, E. tasmaniensis, were isolated from Nuruk. In this case, a suitable growth factor for E. tasmaniensis was culture it for 36 h in broth media on concentration of 1.0% (w/v) alginate. The enzyme showed the highest level of alginate lyase activity when cultured on broth media containing 1.0% (w/v) sodium alginate for 72 h. Optimal condition of pH, temperature and duration time for alginate lyase activity were found to be pH 6.0, $20^{\circ}C$ and 60 min, respectively.

  • Among the 34 strains of Nicotinophiles selected in the previous experiments, strain NCT27 identified with Pseudomonas putida and strain NCT30 identified with Arthrobacter oxydans biotype nan thus were Investigated for optimization of growth conditions for nicotine degradation and other cultural characteristics. The compositions of optimized medium were to be following: $KH_2PO_4$ 2.Ogr, KCI 5.Ogr, $MgSO_4$.$7H_2O$ 20mg, $MnSO_4$.$6H_2O$ 0.2mg, $FeSO_4$.$7H_2O$ 1.Omg, Col$^{++}$ (Cobalt Acetate),2.O$\gamma$, N1$^{++}$ (NiSO4,6H2O) 0.5$\gamma$, and yeast extract 80mg per liter. The optimum initial concentrations of nicotine for growth were 0.4% for Pseudomonas and 0.1% for Arthrobacter, respectively. The optimum temperature and pH were 3$0^{\circ}C$ and 7.0 for both of strains. The pH of culture medium of Pseudomonas was changed from acidic condition to basic one in going from the logarithmic growth phase to the stationary growth phase. In contrast with Pseudomonas, it remained constant in case of Arthrobacter. The growth of Arthrobacter was completely inhibited in the nicotine concentration of 0.7&. However, Pseudomonas could grow even in the nicotine concentration of 1.0%. Moreover, it could grow successfully in the tobacco extract media as well as media containing carbon and nitrogen sources other than nicotine. The maximum rates of nicotine degradation were to be 1.22 gr./hr./liter for Pseudomonas and 0.186 gr./hr./liter for Arthrobacter, respectively.

  • The optimal culture conditions on bacteriocin producing of Lactobacillus sp. FF-3 isolated from Korean Dongchimi, were studied for enhancing its production with regard to environmental and nutritional factors. The optimal cultivation time, initial pH and temperature were 21 hours, pH 7.0 and 30∼37$^{\circ}C$ respectively. Optimal compositions of culture medium for bacteriocin production were glucose 3% as carbon source, tryptone 4% as nitrogen source, and manganese sulfate 0.005% as inorganic salt with other basal components. The maximum antimicrobial activity was 484 BU/mL under the optimal culture condition.

Optimization of biomass production of Acetobacter pasteurianus SRCM101388 (Acetobacter pasteurianus SRCM101388 바이오매스 생산 최적화)

  • In this study, culture conditions were optimized to confirm the feasibility of Acetobacter pasteurianus as a starter for fermentation vinegar. Acetobacter pasteurianus strain can be used as a food ingredient. The optimal temperature and pH conditions of the selected Acetobacter pasteurianus SRCM101388 were 28℃ and pH 6.00, respectively. The response surface methodology (RSM) was used to optimize the composition of the medium, and Plackett-Burman design (PBD) was used to obtain the effective selection of culture medium, resulting in that glucose, sucrose, and yeast extract had the highest effect on increasing biomass. The optimal concentration, which was performed by central composite design (CCD), were determined to be 10.73 g/L of glucose, 3.98 g/L of sucrose, and 18.73 g/L of yeast extract, respectively. The optimal concentrations of trace elements for the production of biomass were found to be 1 g/L of ammonium sulfate, 0.5 g/L of magnesium sulfate, 2 g/L of sodium phosphate monobasic, 2 g/L of sodium phosphate dibasic, and the final optimized medium was pH 6.10. When incubated in a 5 L jar fermenter, the SRCM101388 strain showed a faster-dissolved oxygen (DO) reduction at a lower agitation rate (rpm), and it was able to grow even at reduced DO level when aeration was maintained. The amount of final biomass produced was 2.53±0.12×109 CFU/mL (9.40±0.02 log CFU/mL) when incubated for 18 hours at 150 rpm, 0.5 vvm, pH 6.0, and 28℃.

The Optimal Conditions for Fibrinolytic Enzyme Production from Streptomyces sp. JK-20 (Streptomyces sp. JK-20유래 혈전용해효소의 생산조건)

  • An actinomycetes which produces fibrinolytic enzyme was isolated from soil. Characteristics of the isolated strain and the optimal conditions for the productions of fibrinolytic enzyme were summarized as follows; The fibrinolytic enzyme production strain generates gray airmycelium and had about 0.6~0.8$\times$0.4~0.8${\mu}{\textrm}{m}$ cylindrical spore, smooth surface and formed spore chain of 10~40 spores. We have identified this strain as Streptomyces sp. JK-20. This strain was able to grow up at 20~32$^{\circ}C$ and its optimum growth temperature and pH was 24$^{\circ}C$ and pH 6.0, respectively. The optimal conditions for porducing fibrinolytic enzyme; carbon source, nitrogen source, metal ions and phosphorous sources was 1% xylose, 0.5% yeast extract, 0.5% polypepton, 0.1% MgSO$_4$.7$H_2O$ and 0.1% NaH$_2$PO$_4$.2$H_2O$, respectively. This strain showed the highest productivity of fibrinolytic enzyme after the fourth day under such optimal culture conditions.

  • To improve extraction of insoluble proteins and functional properties of abolished proteins by protease. It was found that the optimum pH, optimum temperature, optimum treatment time and optimum unit of enzyme far extraction of protein were pH 9.0, $60^{\circ}C$, 8 hrs, 40 units. The foaming capacity and foaming satbility of sesame meal protein after treatment of enzyme were especially higher than control. The emulsion capacity and emulsion satbility of sesame meal protein were higher than control. Coil absorption as well as water absorption capacities of sesame meal protein were higher than control.

  • This study was excuted to decide the physiological characteristics of Lentinus lepides. L. lepides was tested to select pertinent substract, temperature and pH range for the growth. Mycelial growth of Lentinus lepides was mostly supported on MYPA among other tested synthetic or semi-synthetic media. The temperature range for pertinent mycelial growth was about 22~32$^{\circ}C$ and mostly stimulated at $25^{\circ}C$. And the pertinent pH range of MYPA was 3.0~4.0. The required carbon and nitrogen source for mycelial growth of Lentinus lepides was tested. The mycelial growth was mostly stimulated by soluble starch at content. The carbon sources for pertinent mycelial growth was maltose. And the nitrogen source for pertinent mycelial growth was peptone.

  • 1. The preparation and some enzymatic properties of crude alkaline protease from a thermophilic mold, Myriococcum sp. was investigated. 2. Optimum pH for the hydrolysis of casein was 9.0 at $50^{\circ}C$ for 10 minutes. Optimum temperature was $55^{\circ}C$ at pH 9.0 for 10 minutes. The enzyme was highly stable at the range of pH 6.0 to 11.0 at $30^{\circ}C$ 3. The alkaline protease in the culture filterate was isolated two fractions by elution column chromatography on DEAE-Cellulose.

  • Effect of various factors on the phenol degradation by activated sludge immobilized with the photocrosslinked resin were investigated. The optimum pH on the degradation of phenol in both free and immobilized activated sludge was 7. A higher rate of phenol degradation was observed when a bead size, vas smaller. The phenol degradation in the free activated sludge was inhibited at the 3000 mg/L of phenol, while that in the immobilized activated sludge was maintained at the same concentration for 28 hrs without an inhibition.
