Optimization of the Production of an Immunostimulant from a Marine Bacterium (해양미생물로부터 면역증강물질의 생산 최적화)

  • A halophilic bacterium for the production of the immunostimulant was isolated from domestic marine, it was identified as Burkholderia sp. IS-203. The optimal conditions for the production of the immunostimulant were 1 % dextrose and 1 % yeast extract in artificial sea water for carbon and nitrogen sources, respectively. The initial pH and growth temperature for the prodution were 8.0 and $30^{\circ}C$ under the presence of oxygen, respectively.

  • The surimi processing from jack mackerel and white croaker muscle using acidic and alkaline solubilization was evaluated. The optimum pH for solubilizing protein in acidic and alkaline range was around 2.5 and 10.5, respectively. The optimum pH value for recovery of protein was around 5. The protein solubility was decreased with increase of salt. The homogenized speed and time for maximum solubility were below 9,500 rpm and 30s, respectively The optimum ratio of water to minced muscle was 6 by evaluating breaking force, deformation and whiteness of cooked gel. The protein yield of alkaline processing is higher than that of conventional processing. In addition, the waste water of conventional processing had high solid, nitrogen content and chemical oxygen demand compare to those of acidic and alkaline processing.

  • 1) Two xylanase (designed as A and B) of Myriococcus albomyces were purified from an extract of wheat koji culture. Puriscation steps included first ammonium sulfate fractionation followed successively by SE-Sephadex column chromatgraphy. DEAE-Sephadex column chromatography and gel filtration on Sephadex G-100 repectively. 2) The optimum pH and pH stability for crude xylanse were found to be pH 5.0 and pH 4.0-7.0 respectively. 3) The optimium temperature was found to he 5$0^{\circ}C$ and for the thermal statbility of xylanase, the enryme incubated at $65^{\circ}C$ for 60min did not affect their stability. 4) The purised xylanase A and B were considered as liquefying xylanase and saccharogenic xylauase repectively. 5) The Bylanase A was most active at pH 4.0 and range of pH 3.0-8.0 at 3$0^{\circ}C$ for six hrs. The B was most active at pH 5.0 showing stability range of pH 4.0 to 8.5 at 3$0^{\circ}C$ for 6 hrs. incubation respectively. The Optimum temperature of xylanase A and B were found to be 7$0^{\circ}C$ and $65^{\circ}C$ for 60min repectively.

A Study on the Cultural Characteristics of Pholiota nameko Mycelium (맛버섯 균사체의 배양 특성에 관한 연구)

  • This Study was carried out to investigate the optimal mycelial growth of Pholiota nameko. The optimal medium for the mycelial growth was ME medium. The optimal temperature and pH were $25^{\circ}C$$\pm$1 and 5.5, respectively. The modified optimal medium compositions were glucose 3% (w/v), malt extract 0.25% (w/v), yeast extract 0.25% (w/v), $KH_2PO_4$ 0.046% (w/v), $K_2HPO_4$ 0.1% (w/v), $MgSO_4$$7H_2O$ 0.05% (w/v). From the result of experiments on the optimal temperature, pH and nutritional requirements, the mycelial growth of modified optimal medium was higher than that of ME medium.

  • To study the cultural characteristics and wood rotting ability of the secondary mycellia of Armillaria mellea, it was cultivated on the various media. The optimal mycelial growth condition was 20~27 and pH 5.0~6.5 on PDB. A. mellea grew well on MEA, PDA and GP. Lactose and mannitol as carbon sources and glutamic acid as nitrogen sources were found to be effective as additives. A. mellea employed in this study have the characteristics of white rot types. Pine and oak wood were selected as candidates for sawdust substrate.

  • Optimization of the medium and fermentation conditions for erythritol production has been studied. We have found that the optimal carbon source was glucose at the concentration of 400 g/L. The optimal temperature was 3$0^{\circ}C$ with excessive aeration. Improved erythritol productivity was achieved by reducing the yeast extract from 5 g/L to 3g/L while adding 2.7 g/L urea, 1.79g/L $K_2HPO_4, and 0.18g/L MgSO$_4$. 7$H_2O. The erythritol productivity increased from 0.747 g/L/h to 1.071 g/L/h and the yield increased from 31.4% to 45.2%. The byproduct glycerol was reduced from 96.6g/L to 45.7g/L as well. We have performed 5L fermentation with and without the pH control. The erythritol productivity with the pH control was about 30% lower than that without pH control. Excessive foaming of 5L fermentation has been observed during fermentation.

  • - Ethanol production by calcium alginate-immobilized baker's yeast was studied in the continuous shaked-flask reactor (CSFR) using glucose medium as a feed. Immobilized cells were stable at 30~$37^{\circ}C$ and pH 4~8. Fermentation characteristics of immobilized baker's yeast were examined changing the initial glucose concentration employed were 50, 100 and 150 g/l, respectively. It was investigated that the influent glucose concentration and the dilution rate have an influence on the ethanol fermentation characteristics at steady state in continuous culture of immobilized baker's yeast. The optimum conditions for high ethanol productivity and low residual glucose output in ethanol prodution were shown to be 0.2 h ' for the dilution rate and 150 g/l for the influent glucose concentration. The maximum ethanol productivity, ethanol yield, specific growth rate and glucose conversion rate were around 7.12 g/$l\cdot h$, 0.23, 0.366 g/$l\cdot h$ and 78.43, respectively.

  • The purpose of this study is to looking for the optimal condition of methane production enhancement. The conditions tested for increasing methane production were temperature. pH. and various carbon sources including methanol. formic acid. sodium acetate. succinic acid. and glucose. As a result, optimal temperature was 55 .C and optimal pH was around neutral condition. And methanol seemed to be best carbon source which can drastically increase methane production.

Identification and Characterization of Alginate Lyase Producing Pseudomonas sp. N7151-6 (Alginate Lyase 생산 균주 Pseudomonas sp. N7151-6의 분리 및 특성)

  • A Gram-negative, alginate lyase-producing bacterium was isolated from the Haeundae Coast, Korea. The isolated strain N7151-6 produced alginate lyase. The optimal temperature and pH for growth were found to be $30^{\circ}C$ and pH 8.0, respectively. This strain can be grown at the NaCl concentration of 0-7% (w/v). Analysis of 16S rDNA sequence and physiological profiling indicated that the strain N7151-6 belonged to Pseudomonas sp. The enzyme alginate lyase produced by Pseudomonas sp. N7151-6 was partially purified by ultrafiltration (MWCO= 30 kDa). The optimum pH and temperature for the activity of the purified enzyme were found to be 7.0 and $30^{\circ}C$, respectively. The enzyme was stable at the pH range of 5.0-9.0 and temperature range of $23-30^{\circ}C$. The total activity of alginate lyase produced was reached about 110 unit/L.

Characterization of Bacteriocin Production by Lactococcus sp. J-105 Isolated from Kimchi (김치에서 분리한 Lactococcus sp. J-105가 생산하는 Bacteriocin의 특성)

  • A bacteriocin-producing strain, J-105, was isolated from Kimchi and identified as Lactococcus sp. The optimum conditions for the bacteriocin production from the isolated microorganism were evaluated. For maximum yield of bacteriocin from Lactotoccus sp. J-105, the cell should be harvested at the early stationary phase and temperature, pH and NaCl concentration should be $25^{\circ}C$, pH 8.0 and without the addition of NaCl, respectively. Maltose should be used as a carbon source and organic nitrogen such as polypeptone should be used as a nitrogen source for the best yield. The bacteriocin from isolate was inhibitory against Acetobacter aceti, Bacillus subtilis and several strains of lactic acid bacteria. The bacteriocin of J-105 was sensitive to pepsin, but stable for heat treatment. It was stable even at autoclaving temperature for 15 min.
