Characterization of Human ${\beta}-Carotene$ 15,15-dioxygenase Isolated from Recombinant Escherichia coli (유전자 재조합 기술에 의하여 제조된 인간 ${\beta}-carotene$ 15,15'-dioxygenase의 반응특성)

  • Characteristics of human ${\beta}-carotene$ 15,15'-dioxygenase isolated by recombinant DNA technology was elucidated. Optimal pH and temperature were 9.0 and $40^{\circ}C$, respectively. Enzyme activity was temperature-sensitive. Enzyme was stable at pH 6.0-9.0 for 24 hr and under $5^{\circ}C$. Half-life of enzyme at $35^{\circ}C$ was 40 min. Crude preparations of enzyme were inhibited by ferrous ion-chelating agent and sulfhydryl-binding agent. GSH offsets inhibitory effect of PCMB. With increasing substrate concentrations, enzyme activity gave typical Michaelis-Menten curve, Based on Hanes-Woolf plot of data, $K_{m}\;and\;V_{max$ were $3.39{\times}10^{6}\;M\;and\;1.2\;pmol/mg$ protein/min, respectively.

Removal of Cd(II) by Cation Exchange Resin in Differential Bed Reactor (미분층반응기에서 양이온 교환수지에 의한 카드뮴(II)의 제거)

  • In this study, in order to remove Cd(II) from aqueous solutions, strongly acidic cation exchange resin(SK1B) by Diaion Co. was employed as an adsorbent. Experiments were mainly performed in two parts at room temperature($25{\pm}5^{\circ}C$) : batch tests and adsorption kinetics tests. In batch tests adsorption equilibrium time, pH effects, temperature effects, several adsorption isotherms, and finally desorption tests were examined. In differential bed tests, an optimum flow rate and an overall adsorption rate were obtained. In the batch experiment, adsorption capability increased with pH and became constant above pH 6 and adsorption quantity increased with temperature. Batch experimental data found that Freundlich and Sips adsorption isotherms were more favorable than Langmuir adsorption isotherm over the range of concentration (5~15ppm). The desorbent used in the desorption test was hydrochloric acid solution with different concentrations(0.01~2N). The degree of regeneration increased with concentration of desorbent and decreased slightly with the number of regeneration. In the continuous flow process using a differential bed reactor, the optimum flow rate was $564m{\ell}/min$ above which the film diffusion resistance was minimized. The overall adsorption rate for the removal of Cd(II) by cation exchange resin was found as follows ; $r=1.3785C_{fc}^{1.2421}-2.0907{\times}10^{0.0746C_i}\;q_e^{0.0121C_i-0.0301}$

The Processing Conditions of Extracts from Rapid Fermented Anchovy Sauce (속성멸치간장엑기스분의 가공조건)

  • AS a part of investigation to use anchovy more effectively as food source, this work was undertaken the processing conditions of extracts from rapid fermented anchovy sauce. To prepare rapid fermented products, the chopped anchovy was mixed with 50% water (w/w), and then hydrolyzed by autolysis or addition of koji under different conditions of hydrolysis. The optimal conditions for hydrolysis of chopped anchovy were revealed $50^{\circ}C$, 6 hours, pH 8.0 by autolysis, and ,$50^{\circ}C$, 6 hours, pH 7.0 by addition of 10% koji, respectively. But, pH control was not much beneficial in increasing yield. The effect of soybean protein isolote for improvement of bitter taste was also tested. The reasonable amount of added soybean protein isolate was revealed 5% on the weight basis of the chopped anchovy. The reaction mixture hydrolyzed under optimal conditions were added with 1% onion powder (w/w), 1% garlic powder(w/w) and 1% red pepper powder(w/w) for masking fishy odor, inactivated for 20 min at $100^{\circ}C$, and then centrifuged for 20 min at 4,000 rpm. The supernatant liquor was filtrated and evaporated to 50%(v/v). finally, table salt was added for bateriostatic effect and characteristic taste of rapid fermented products. the reasonable amount of added table salt was reversed 15% on the volume basis of the evaporated liquor. The hydrolysis ratio of product made by addition of water, product made by addition of koji and water, and product made by addition of soybean protein isolate, koji and water hydrolyzed under optimal conditions were 58.4%, 82.1% and 86.2%, respectively.

Characterization of Extracellular Cholesterol Oxidase Produced from Soil Microorganism (토양 미생물로부터 생산된 Extracellular Cholesterol Oxidase의 특성)

  • Cholesterol oxidase catalyses the conversion of cholesterol to 4-cholesten-3-one. This enzyme has been used for clinical assay of human serum cholesterol and for reduction of cholesterol level in foods and feeds. In order to search the microorganism which has a high extracellular and stable activity of cholesterol oxidase, soil microorganisms were screened. As a result, the one with the highest extracellular cholesterol oxidase activity was obtained and named as the BEN 115. The BEN 115 strain was identified as one of the Nocardia species based on our taxonomic studies. The cholesterol oxidase from this strain was shown to have two bands of extracellular proteins on SDS-PAGE and Western blot. Their molecular masses were estimated to be about 55 and 57 kDa, respectively. In addition, this cholesterol oxidase was considerably stable at the broad range of pH $3.5{\sim}9.5$ and at the temperature of $25{\sim}55^{\circ}C$. The optimum pH and temperature of this cholesterol oxidase were pH 5.5 and $35^{\circ}C$, respectively. The activity of extracellular cholesterol oxidase could be enhanced 1.6 to 2.0 folds by the addition of nonionic detergent such as Triton X-114, Triton X-100, or Tween-80 into the culturing broth. The substrate specificities against campesterol, sitosterol and stigmasterol were measured to be 50%, 50%, and 27%, respectively, compared to the cholesterol. These results suggest that Nocardia sp. BEN 115 may be useful as a microbial source of cholesterol oxidase production.

Mycelial characteristics for the artificial cultivation of Inonotus obliqua (Pers.) Pilot (차가버섯(Inonotus obliqua) 인공재배를 위한 균사 배양적 특성)

  • The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Inonotus obliqua (chaga) are as follows. 1) Mycelial growth and density oft, oblique were the highest in the medium of BDA (Birch Dextrose Agar; 66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA (pH 7.0), CHA, and MEA (pH 4.7). 2) Optimal temperature for the mycelial growth and density of 1. obliqua were shown to be 30$^{\circ}C$, but the mycelia were dead at 40$^{\circ}C$. the mycelial growth and density of KNAC3005 strains was the highest at 30$^{\circ}C$(66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and 5$^{\circ}C$. 3) Optimal pH for the mycelial growth and density were revealed to be 6.0 (88.4mm/10d). Above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of 40 were maltose (331mg/25$m\ell$/15d), peptone (347mg/25$m\ell$/15d), and glutamic acid (357mg/25$m\ell$/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40.

Optimization of Media Composition and Cultivation for the Mycelial Growth of Agrocybe Cylindracea (Agrocybe Cylindracea의 영양배지 조성 및 배양조건의 최적화)

  • Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.

  • A bacterial strain producing a novel endo-inulinase, hydrolysing inulin into oligosaccharides was isolated from soil and identified as Arthrobacter sp. S37 The enzyme production was induced by inulin and jerusalem artichoke extract. The maximum enzyme production was obtained with medium containing 1.5% jerusalem artichoke extract, 1.0% yeast extract, $0.5%\;NaNO_3,\;0.05%\;MgSO_4{\cdot}7H_2O,\;0.05%\;KCl,\;0.0016%\;FeCl_3{\cdot}6H_2O\;and\;0.05%\;KH_2PO_4$. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and 8.0, respectively. Under the optimum condition, the enzyme activity in the culture broth reached at maximum, 10.8 units/ml after cultivation for 24 hours.

  • The purpose of this study is to obtain an optimum condition of the concentrate by floatation using Australian low grade copper ore of suitable particle size by crushing, grinding and sizing. The effect of the dosage of collector, activator, depressant and change of acidity on the floatation was investigated. The floatation conditions comprising of pH 9.0 acidity, 500g/t $CuSO_4{\cdot}5H_2O$ activator, 500g/t K.E.X. collector, 1500g/t $Na_2SO_3$ depressant and 25wt.% plup density were optimized. Grade and recovery of copper in the concentrate obtained by froth floatation under optimum conditions were 4wt.% and 65wt.% respectively.

  • After series of purification by means of ammonium sulfate fractionation, the 1st and 2nd DEAE-Cellulose, DEAE-Sephadex A-50, and Sephacryl S-200 superfine gel filtration, the activity of extracellular adenine deaminase from Streptomyces sp. J-350P increased 1764 fold and the yield was 0.3% of original activity. The enzyme was stable at the pH range 6.5 to 8.5 and at up to 5$0^{\circ}C$. The optimum pH and temperature of the enzyme were around 6.5 and 35$^{\circ}C$. The molecular weight ol the enzyme was estimated as 36, 000 by calibrated Sephacryl S-200 superfine column chromatography.

  • For the biological production of hydrogen, a new fermentative hydrogen-producing bacterium, Enterobacter sp. SNU-1453, was isolated from a domestic landfill. During the culture of this bacterium, pH significantly decreased with the accumulation of various organic acids, and consequently this inhibited the production of hydrogen. It was found that the metabolic flux in this bacterium depended on the pH and affected the hydrogen production. A butanediol pathway was dominant during the fermentation when pH was not controlled. By controlling the pH at 7 this pathway can be shifted to a mixed acid pathway, which is favorable to the production of hydrogen.
