Optimum Conditions of Freezing Lyophilization and Bioluminescence Activity Recovery for Environmental Applications Using a Recombinant Strain (유전자 재조합 균주를 환경에 적용하기 위한 (동결) 건조 및 활성회복 조건 최적화)

  • Bioreporter bacteria, such as recombinant bioluminescent bacteria, have been used for the detection of specific compounds in complex environmental media. In this study, optimum conditions for the preparation and application of deep-freezed and Iyophilized recombinant bioluminescent strain KG1206 were investigated for the future application on contaminated environmental sites. Genetically engineered microorganism, Pseudomonas putida mt-2 KG1206, contains TOL plasmid and the plasmid inserted $P_{m}$, promoter on the upper part of lux gone in vector pUCD615, and m-toluate and benzoate are considered direct inducers for bioluminescence. Optimum conditions determined for the preparation and application of the deep-freezed and lyophilized strain were followings: cryoprotective agent (24% sucrose), lyophilization time (12 hrs), strain concentration ($OD_{600}=0.6$), reconstitution for freezed strain (quick reconstitution at $35^{\circ}C$), reconstitution for lyophilized strain ($3{\sim}6$ hrs exposure on LB medium), carrying conditions (keep at $20^{\circ}C$ after reconstitution). These results demonstrate the feasibility of deep-freezed or lyophilized state of genetically engineered bioluminescent strain for environmental usage.

Selection of Optimum Pebbles Size in Sewage Treatment Plant by Natural Purification Method (자연정화공법에 의한 하수처리장에서 최적 여재 선정)

  • An investigation was carried out to evaluate the effects of pebble size on the small-scale sewage treatment apparatus by natural purification method that consisted of aerobic and anaerobic plot. pH and EC in both plots varied minimally regardless of pebble size. DO in aerobic plot increased as the pebble size decreased. That in the anaerobic plot was slightly less in comparison with that of the aerobic plot but varied minimally, $2.4{\sim}5.1\;mg/L$ regardless of pebble size. Under all experimental conditions, removals of BOD, COD and turbidity in anaerobic plot were more than 98, 91 and 98, 98% respectively. Removals of T-N and T-P increased as pebble size decreased. Under all experimental conditions, removals of T-N and T-P in anaerobic plot were about $45{\sim}59$ and $480{\sim}96%$, respectively. Judging from the above results, it should be considered that the optimum pebble size and pebble permeability in both plot was $2{\sim}4$ and $0.1{\sim}4\;mm$, respectively.

Optimizing Coagulation Conditions of Magnetic based Ballast Using Response Surface Methodology (반응표면분석법을 이용한 자성기반 가중응집제의 응집조건 최적화)

  • As a fundamental study to apply the new flocculation method using ballast in water treatment process, the optimal conditions for general and ballast coagulant dosage, and pH, which are known to have a significant influence, were derived by response surface methodology. Poly aluminum chloride (PAC) and magnetite ballast were used as a general coagulant and ballast, respectively. Coagulation experiments were performed by jar-tester using the kaolin based synthetic water. The effects of three independent variables (pH, PAC, and ballast) on response variables (turbidity removal rate and average settling velocity of flocs) and the optimum condition of independent variables to induce the optimum flocculation were obtained by 17 experimental conditions designed by Box-Behnken procedure. After performing experiments, the quadratic regression model was derived for each of response variables, and the response surface analysis was conducted to explore the correlation between independent variables and response variables. The $R^2$ values for the turbidity removal rate and the average settling velocity were 0.9909 and 0.8295, respectively. The optimal conditions of independent variables were 7.4 of pH, 38 mg/L of PAC and 1,000 mg/L of ballast. Under these conditions, the turbidity removal rate was more than 97% and the average settling velocity exceeded 35 m/h.

  • This paper collect sleep environment data of bedroom to sleeping, and analyzing the relationship between conditions with obtained data and sleep. We provide the optimal sleep environment of individual by extracting the simulation model based on it. The experiments was using temperature/humidity sensor(SHT11) and ambient light sensors(GL5507). For extraction of tossing and turning, we use difference image method in motion extraction from video. In addition, the information of weight can affect to sleep, it was entered such as ratio of fatigue, drinking, empty stomach. As a result, we are able to extract the optimal sleep environment. The future, we will try to improve to help to lead more pleasant daily life providing proper indoor environment changes depending on the situation even a partial of organic ubiquitous living environments such as eating, work ete. as well as certain sleep circumstances.

Optimization of Growth Environment in the Enclosed Plant Production System Using Photosynthesis Efficiency Model (광합성효율 모델을 이용한 밀폐형 식물 생산시스템의 재배환경 최적화)

  • This study was aimed to assess the effects of microclimate factors on lettuce chlorophyll fluorescent responses and to develop an environment control system for plant growth by adopting a simple genetic algorithm. The photosynthetic responses measurements were repeated by changing one factor among six climatic factors at a time. The maximum Fv'/Fm' resulted when the ambient temperature was $21^{\circ}C,\;CO_2$ concentration range of 1,200 to 1,400 ppm, relative humidity of $68\%$, air current speed of $1.4m{\cdot}s^{-1}$, and the temperature of nutrient solution of $20^{\circ}C$. In PPF greater than $140{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, Fv'/Fm' values were decreased. To estimate the effects of combined microclimate factors on plant growth, a photosynthesis efficiency model was developed using principle component analysis for six microclimate factors. Predicted Fv'/Fm' values showed a good agreement to measured ones with an average error of $2.5\%$. In this study, a simple genetic algorithm was applied to the photosynthesis efficiency model for optimal environmental condition for lettuce growth. Air emperature of $22^{\circ}C$, root zone temperature of $19^{\circ}C,\;CO_2$ concentration of 1,400 ppm, air current speed of $1.0m{\cdot}s^{-1}$, PPF of $430{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and relative humidity of $65\%$ were obtained. It is feasible to control plant environment optimally in response to microclimate changes by using photosynthesis efficiency model combined with genetic algorithm.

광-펜톤 반응을 이용한 Rhodamine B의 색 제거

  • 수용성 안료인 RhB를 대상으로 광-펜톤 공정의 최전 운전조건을 구하고, 광-펜톤 공정을 구성하는 개별 공정을 비교한 결과 다음의 결론을 얻었다. 광-펜톤 공정의 최적 $Fe^{2+}$$H_2O_2$ 투입량은 각각 0.0031 mmol과 0.625 mmol이었으며, 최적 pH는 3으로 나타났으나, 7이하의 pH 범위에서는 RhB 색 감소에 큰 영향을 미치지 않는 것으로 나타났다. 초기 반응 속도에 가장 큰 영향을 주는 인자는 UV 광 전력 > $H_2O_2$> 철염의 순으로 나타났다. 80분간의 반응시간 경과 후 최종 RhB 농도를 고찰한 결과 UV 광 전력이 낮을 경우 색도가 다 제거되지 않기 때문에 광-펜톤 공정에서 UV 광 전력이 색 제거에 대한 가장 큰 인자라고 사료되었다. 광-펜톤 공정의 개별 공정인 UV, $H_2O_2$, 펜톤을 이용하여 RhB의 농도감소를 고찰한 결과 초기 반응속도상수는 펜톤 공정의 빠른 초기 반응로 인해 펜톤 > UV >$H_2O_2$로 나타났으나, 최종 RhB 농도를 고려할 경우 UV> 펜톤 > $H_2O_2$로 나타났다.

  • 진화 알고리즘과 인공신경망은 생물학에서 비롯되어 컴퓨터과학 분야에서 응용되고 있는 문제해결 방법이다. 본 연구는 게임 환경에서 크기를 자율적으로 설정하여 생성할 수 있는 미로를 구성하고, 주어진 미로의 시작점으로부터 목적지까지 유전 알고리즘과 인공신경망을 이용하여 경로탐색을 하는 것에 대한 연구이다. 자동 생성된 미로가 특정 크기 이상으로 커지게 되면, 진화 알고리즘은 무작위적인 값에 의해서 결정되는 것으로 수렴한다는 결론을 얻었고, 인공신경망을 이용한 결과는 진화알고리즘 보다 미로의 경로탐색 문제해결에 적합한 결과를 보여주었다. 또한 어떤 방향이 최적경로인지 아닌지를 미리 알 수 있는 특수한 조건에서는 각 유전인자를 최적값인지 아닌지 표현하는 방법으로 효율적인 진화 알고리즘을 사용할 수 있다는 것을 제안하였다.

Analysis of Real-time, Shortest-Path Finding Algorithms for Unknown Environments (미지의 공간상의 실시간 최단 경로 탐색 알고리즘에 대한 분석)

  • 본 논문은 미지의 공간 그래프에 대한 실시간 최단 경로 탐색 알고리즘인 $RTA{\ast}$ 알고리즘과 $PHA{\ast}$ 알고리즘을 소개하고, 예제 그래프를 통해 이들의 특성을 설명한다. 또 3차원 온라인 게임 환경인 언리얼 토너먼트(Unreal Tournament) 게임과 Gamebots 시스템을 이용하여 두 알고리즘 간의 탐색 최적성과 효율성을 비교하였다. 이 실험을 통하여 $PHA{\ast}$ 알고리즘은 언제나 최선의 경로를 탐색하지만 이동경로가 많고, 시간이 많이 걸릴 수 있으며, $RTA{\ast}$는 최적의 경로 혹은 차선 경로를 빠른 시간 내에 탐색함을 확인 하였다. 따라서 이러한 특성을 바탕으로 주어진 탐색 환경의 조건에 따라 두 알고리즘을 선택적으로 적용함으로써 보다 높은 효과를 얻을 수 것으로 기대한다.

  • 최적 위치별 퇴적토에 대한 토양 특성 및 수목 식재용 유기토양 활용 가능성을 알아보고 각 시료의 수분, 유기물 함량 등에 따른 적절한 혼합을 통하여 식물 발아 성장의 최적화를 온실에서의 식물재배 실험을 통하여 달성하고자 한다. 또한 식재용 토양 활용 가능성을 알아보기 위한 기본 물성 및 유해성 평가를 하여 중금속이 식물 성장에 어떤 영향을 미치는가를 고려하여 재활용 여부를 알아보았다. 퇴적토를 이용한 식물의 발아 및 생장 실험은 원퇴적토와 모래와의 혼합비를 달리하고 다른 불순물이 함유되지 않도록 하여 발아일수와 발아적온, 생육적온 등 최적 조건을 팬지와 금잔화에 대하여 실험하였다.

A Study of Environmental Conditions of Survival Rate and Relative Growth Rate in Female Gametophyte of Undaria pinnatifida for Toxicity Assessment (생태독성평가를 위한 미역(Undaria pinnatifida) 암배우체 생존율 및 상대성장률의 환경조건 연구)

  • The ecotoxicity test method using Undaria pinnatifida spore is challenging to use throughout the year. Since U. pinnatifida female gametophytes can be cultured in the laboratory, they can be used for ecotoxicity testing at any time. Changes in female gametophyte survival rate and relative growth rate in U. pinnatifida exposed to various environmental conditions were analyzed. The female gametophyte of U. pinnatifida was exposed to salinity (5~40 psu), temperature (5~30℃), pH (4~10), and light intensity (0~120 μmol photon m-2 s-1). Based on the highest average value, the survival rate of female gametophyte was highest at a temperature of 20℃, salinity 27.5 psu, pH 8, and light intensity 30 μmol photon m-2 s-1. And the relative growth rate was highest at a temperature of 15℃, salinity 35 psu, pH 9, and light intensity of 60 μmol photon m-2 s-1. As a result of this study, the method using the optimal conditions for the survival rate and relative growth rate is expected to be a practical test method that can complement the current method.
