This paper aimed to develop a solvent extraction and purification process to recover high-purified ${\beta}$-carotene from recombinant Escherichia coli. Cells harvested from the culture broth were treated through numerous steps: dehydration, solvent extraction, crystal formation and separation. To optimize the extracting condition, experiments were carried out to investigate the effect of cell disruption, temperature, organic solvents, solvent-biomass ratio on the yield of ${\beta}$-carotene extracted from cells. The result indicated that no significant differences of extraction yield were observed from cells with or without step of cell disruption. Among different extracting solvents, the highest extraction yield of ${\beta}$-carotene, 30.3 mg-${\beta}$-carotene/g-dry cells, was obtained with isobutyl acetate at solvent-biomass ratio 25 mL/g-dry cells at $50^{\circ}C$. Notably, in case of acetone, the extraction yield was quite low when using acetone itself, but increased almost up to the highest value when combining this solvent and olive oil. The purity of ${\beta}$-carotene crystals obtained from crystallization and separation was 89%. The purity degree was further improved up to 98.5% by treating crude crystals with additional ethanol washing.

Purpose: Recently incidence of VDT syndrome has gradually increased as extensive use of computers. VDT syndrome reported by VDT workers include musculoskeletal disorder, neuropsychiatric disoders and eye symptoms such as eye strain, tired eyes, irritation and blurred vision. The environmental factors of VDT syndrome include electromagnetic waves, size, brightness and lighting of computer screen, height of a monitor and a worktable, working hours, kind of task, distance between screen and workers, indoor humidity and temperature, indoor air contamination and ventilation. In this study, we investigated the environmental factors related to body symptoms and health effects included in VDT syndrome. Methods: Study subjects were total 120 persons (54 male, 66 female) with age from 19 to 28. We surveyed the body symptoms and physical discomfort when doing an activity in a short distance such as reading book or paper, computer work. The questionnaire included main body symptoms, self-consciousness symptoms of eye, satisfaction of working environment, pain of the wrist when using keyboard and mouse. Results: Most of people (70%) felt physical pain from long time work of computer, paper, electrical apparatus. They mainly complained pain of neck and low back (57.1%), eye (45.2%) and head (31%). With the environmental factors, 78.3% of the subjects complaint pain of eye from inappropriate illumination. Most of the symptoms included 'eye fatigue'(38.3%), 'dryness of eye'(31.9%) and 'blurred vision'(23.7%). Subjects in this study complained discomfort of their chairs and most of them experienced pain in the wrist when using keyboard or mouse. Conclusions: When people use electrical apparatus or work with paper, people would get their eye fatigue and feeling of physical fatigue because of not harmonizing various environmental factors such as light, space, posture, worktable with theirselves. Therefore, workers should develop preventive method such as self-control of adequate break time to avoid fatigue while VDT work. Work environment should be changed to ergonomic design for optimal visual environment to prevent musculoskeletal disorder through constant research.

To investigate the feasibility of refuse derived fuels (RDFs) combined of sewage sludge and combustible wastes such as substitutive fuels instead of a stone coal, several different RDFs made with different mixtures of sewage sludge and combustible wastes were analyzed by various experiments. The combustion characteristics for the RDFs were investigated by analyzing fuel gases, and heating values were also measured by a bomb calorimeter. The fundamental properties such as moisture contents, ratios of combustible materials, amounts of ashes, heavy metals, ratios of each chemical elements and heating values were analyzed in accordance with mixing ratios of wt(%) for researching the characteristics of the RDFs. $RDF_{k-1}$ was made of mixing materials which were dried sewage sludge, food wastes and combustible wastes. $RDF_{k-2}$ was made of mixing materials which were peat-moss, tar and sewage sludge. Combustion experiments were carried out at the optimal conditions which were m=2 under air-fuel condition and $850^{\circ}C$. The retention times in the combustor were set at 5, 10 and 15minutes. 50 g of RDFs was put in the combustor for each experiments. The ranges for heating values of $RDF_{k-1}$ with different mixing ratios were from 6,900 kcal/kg to 8120 kcal/kg. The ranges for heating values of $RDF_{k-2}$ with different mixing ratios were from 4,014 kcal/kg to 8,050 kcal/kg. As a result of this study, the heating values, moisture contents, components of chemical elements and mixing ratios of the materials in RDFs had big effects on the efficiency of the combustion. In $RDF_{k-1}$, the higher amounts of combustible wastes in the mixtures, the higher heating values, concentrations of $C_xH_y$ and amounts of ashes were produced. In $RDF_{k-2}$, the higher tar amounts in the mixtures caused the higher heating values, amounts of ashes, concentrations of CO gas and CxHy.

Microarray which enables us to obtain hundreds and thousands of expression of gene or genotype at once is an epoch-making technology in comparative analysis of genes. First of all, we have to measure the intensity of each gene in an microarray image from the experiment to gain the expression level of each gene. But it is difficult to analyze the microarray image in manual because it has a lot of genes. Meta-gridding method and various auto-gridding methods have been proposed for this, but thew still have some problems. For example, meta-gridding requires manual-work due to some variations in spite of experiment in same microarray, and auto-gridding nay not carried out fully or correctly when an image has a lot of noises or is lowly expressed. In this article, we propose Hierarchical Grid Alignment algorithm for new methodology combining meta-gridding method with auto-gridding method. In our methodology, we necd a meta-grid as an input, and then align it with the microarray image automatically. Experimental results show that the proposed method serves more robust and reliable gridding result than the previous methods. It is also possible for user to do more reliable batch analysis by using our algorithm.
Generally, design flood for a hydraulic structure is estimated using statistical analysis of runoff data. However, due to the lack of runoff data, it is difficult that the statistical method is applied for estimation of design flood. In this case, the synthetic unit hydrograph method is used generally and the models such as NYMO method, Snyder method, SCS method, and HYMO method have been widely used in Korea. In this study, these methods and KICT method, which is developed in year 2000, are compared and analyzed in 10 study areas. Firstly, peak flow and peak time of representative unit hydrograph and synthetic unit hydrograph in study area are compared, and secondly, the shape of unit hydrograph is compared using a root mean square error(RMSE). In Nakayasu method developed in Japan, synthetic unit hydrograph is very different from peak flow, peak time, and the shape of representative unit hydrograph, and KICT method(2000) is superior to others. Also, KICT method(2000) is superior to others in the aspects of using hydrologic and topographical data. Therefore, Nakayasu method is not a proper in hydrological practice. Moreover, it is considered that KICT model is a better method for the estimation of design flood. However, if other model, i.e. SCS method, Nakayasu method, and HYMO method, is used, parameters or regression equations must be adjusted by analysis of real data in Korea.
1. The sericin solubility increased rapidly as the increase of water M-alkalinity. 2. The acidity of the treated water was nutralized at the over 25ppm of M-alkalinity. 3. The more M-alkalinity of the sample water is, the more M-alkalinity was found after cocoon treat. 4. The total hardness of sample water seemed to be droped as M-alkalinity increased. 5. The sericin solubility also seemed to be droped as the increase of water acidity. 6. In case of treat finish with cocoon, the acidity and total hardness seemed to increase as the acidity of the water increased, but M-alkalinity was nutralized at 20~40 ppm of water acidity or the M-alkalinity could not be found in case over 40ppm of acidity. 7. In case increase of iron component with sample water, sericin solubility seemed to drop down, and mangan component showed the same nature but dull drop. 8. After cocoon was treated with water, acidity, M-alkalinity and total hardness were increased by the extraction from cocoon shell because of pH and treating temperature but not because of iron componnent. Mangan component, however, affected as to increase of acidity and total hardness but to decrease for M-alkalinity. 9. In case change of M-alkalinity and total hardness, sericin solubility has increased also. 10. In case constant pH and total hardness, the more M-alkalinity is, the more sericin solubility was found. 11. In case constant pH, total hardness, and M-alkalinity, the more acidity is, the less sericin solubility was found. 12. In case constant pH(6.8) and M-alkalinity, the more total hardness is, the less sericin solubility was found. 13. Though the combination of water, high solubility water, medium solubility water and low solubility water were prepared. The high solubility water desolved sericin 2.2% more than low solubility water. And the medium solubility water desolved sericin as much as 2.4~2.9%. 14. It was found that the most important factors for filature water are pH, M-alkalinity, acidity and total hardness which may need more words for optimum filature water development. 15. In case of repeat use of water, the buffer action of water has increased so that the sericin solubility to be decreased.

When Aphidius colemani mummies were stored at four different temperatures (6, 8, 10 and 12 ± 1℃), they could be stored at 8℃ for 10 days with over 50% adult emergence. A number of mummies of Rhopalosiphum padi parasitized by A. colemani adults emerged from mummies after 8℃ storage clearly reduced with increasing duration of storage and especially declined with storage longer than 13 days showing < 20% parasitism. However, there were no differences in aphid parasitisms by A. colemani on 3 to 10 days after storage. Over 3 day storage at 8℃ was adversely affected on survival of A. colemani adults. Therefore, cold storage of A. colemani adults is considered as an unsuitable method for mass production of biological control agent. When 2-day-old Meteorus pulchricornis cocoons were stored at 6, 8 and 10 ± 1℃, it was possible to be stored up to 63 days at 8℃, showing the highest emergence. With increasing of cold storage duration at 8℃, parasitism on 2nd and 3rd larval instar of Spodoptera litura by stored M. pulchricornis significantly reduced. However, parasitic rate by M. pulchricornis stored for 2 weeks was similar to that by 1 week stored ones. Therefore, M. pulchricornis cocoons could be stored up to 2 weeks at 8℃ for stockpiling before release.

Korean wheat semolina (FS: fine semolina) with similar characteristics to durum wheat semolina was substituted at rates of 0, 10, 20, 30, 40, and 50% in pasta dough and the physical and cooking characteristics were investigated for making optimal pasta. Water absorption of the dough increased with the 10, 20, and 30% substitution ratio of FS. Development times were high with >30% substituted FS. This result positively influenced an increase in production and the preparation of the fresh noodle pasta. Furthermore, soft textured fresh noodles could be made due to the decrease in stability and increased weakness of the >30% substituted FS. The amylograph gelatinization characteristics of Korean wheat semolina exhibited an increase of gelatinization temperature and decrease of maximum viscosity when compared with durum wheat. The handling property of the dough showed more than 4 points in all sample groups. Weight and volume decreased and turbidity and cooking loss increased according to the increasing amount of substituted FS. However, samples with ${\leq}$ 30% FS substitution ratio had similar volumes and cooking losses when compared to the control. The L- and a-values increased and the b-value of color decreased as more FS was added. In a texture analysis, the hardness of the cooking noodles showed a low value with the >30% substituted FS. Springiness, gumminess, and chewiness exhibited a high value. In the results of a sensory evaluation, overall acceptability was high score with more than 7 points for the 30% added FS. The preferences for pasta colors were divided into white, which is similar to the Korean traditional noodle, and yellow, which is similar to durum wheat. Flavor and taste were not affected by substituting with FS. Low hardness and high chewiness was the most preferred noodle. These results suggest that >30% substituted FS was suitable for increasing quality and organoleptic qualities of Korean wheat pasta.

Textile industry is considered as one of the most polluting sectors in terms of effluent composition and volume of discharge. It is well known that the effluents from textile dying industry contain not only chromatic substances but also large amounts of organic compounds and insolubles. The azo dyes generate huge amount of pollutions among many types of pigments. In general, the electrochemical treatments, separating colors and organic materials by oxidation and reduction on electrode surfaces, are regarded as simpler and faster processes for removal of pollutants compared to other wastewater treatments. In this paper the electrochemical degradation characteristics of dye wastewater containing CI Direct Blue 15 were analyzed. The experiments were performed with various anode materials, such as RuO2/Ti, PtO2/Ti, IrO2/Ti and graphite, with stainless steel for cathode. The optimal anode material was located by changing operating conditions like electrolyte concentration, current density, reaction temperature and initial pH. The degradation efficiency of dye wastewater increased in proportion to the electrolyte concentration and the current density for all anode materials, while the temperature effect was dependent on the kind. The performance orders of anode materials were RuO2/Ti > PtO2/Ti > IrO2/Ti > graphite in acid condition and RuO2/Ti > IrO2/Ti > PtO2/Ti > graphite in neutral and basic conditions. As a result, RuO2/Ti demonstrated the best performance as an anode material for the electrochemical treatment of dye wastewater.

In the enzymatic hydrolysis of rice straw and wood meals using extra-cellular enzymes from Fomitopsis palustris, key factors which enhanced the sugar conversion yield were investigated in this work, such as enzyme production and enzyme reaction conditions, surfactant effects, and the surface structure of substrates. F. palustris cultured with softwood mixture produced 12.0 U/$m{\ell}$ for endo-${\beta}$-1,4-gulcanase (EG), 116.68 U/$m{\ell}$ for ${\beta}$-glucosidase (BGL), 18.82 U/$m{\ell}$ for cellobiohydrolase (CBH), and 13.33 U/$m{\ell}$ for ${\beta}$-xylosidase (BXL). These levels of BGL, CBH, and BXL activities were two to four folds more than enzyme activities of F. palustris cultured with rice straw. The optimum reaction conditions of cellulase-RS which produced by F. palustris with rice straw and cellulase-SW which produced by F. palustris with softwood mixture were pH 5.0 at $45^{\circ}C$ and pH 5.0 at $50^{\circ}C$, respectively. The sugar conversion yield of cellulase-SW had the highest value of $40.6{\pm}0.6%$ within 72 h when rice straw was used as substrate. By adding 0.1% Tween 20 (w/w-substrate), the sugar conversion yield of rice straw was increased to 44%, which was about four fifths sugar conversion yield of commercial enzyme, Celluclast 1.5L (Novozyme A/S). A low crystallinity and an intensive fibril surface observed by the scanning electron microscope may explain the high sugar conversion yield of rice straw.
