Optimization of artificial cultivation of Tremella fuciformis in closed culture bottle (흰목이버섯 대량생산을 위한 용기내 재배 최적화 연구)

  • The stromatal forms of T. fuciformis and the mycelia of Hypoxylon sp. were collected. The DNA sequence in the ITS region of the 5.8S ribosomal genes of isolated strain KG103 was very similar to that of T. fuciformis AF042409 with a homology of over 98% in the EMBL/GenBank database through BLAST searching. A second isolate, No KG201, one of the symbiotic strains for cultivating T. fuciformis also exhibited high homology with Annulohhypoxylon stygium AJ390406. Potato Dextrose Medium exhibited the best mycelial growth of 14 mm/14 days and 85 mm/14 days for T. fuciformis and its symbiotic fungi, respectively. Optimum culture conditions for the micelial growth were pH 5 at $25^{\circ}C$. For the optimization of artificial cultivation of T. fuciformis in bottle with sawdust medium, several conditions such as type of sawdust, supplements, pH, moisture content, and incubation temperature were investigated. T. fuciformis and symbiotic fungi showed fast mycelial growth on corn cob media (77 and 52%) followed by oak tree sawdust and cotton seed meal. The optimal temperature for mycelial growth of T. fuciformis and symbiotic fungi on corn cob media was $25^{\circ}C$ at 55% of moisture content.

Morphological and Physiological Characteristics of Amanita hemibapha subsp. hemibapha (Berk. & Broome) Sacc. (달걀버섯의 형태 및 생리적 특성)

  • This study was conducted to investigate optimum cultural conditions for Amanita hemibapha isolated in Korea and its hyphae characteristics. Micrographs shows the presence of clamp connection. A. hemibapha grows as mycelial form(M-phase) 2-4 ${\mu}m$ and yeast-like form(Y-phase) 7-8 ${\mu}m$. The fungal spores were broadly elliptical and papillate, 8-11 ${\times}$ 6-9 ${\mu}m$ in size. The nucleotide sequence analysis of the ITS of nuclear ribosomal DNA from sporocarps and in-vitro-grown mycelium supported the fungal species is Amanita hemibapha. A. hemibapha showed sequence similarity in the ITS rDNA with A. caesarea(97.5) and A. jacksonii(98.5%) which are morphologically similar species to A. hemibapha. The optimal pH and temperature for mycelial growth of A. hemibapha were pH 6.0 and $28^{\circ}C$, respectively. The fungal species showed best growth in SYP and GYS medium. A. hemibapha grew well with mannitol and glucose as carbon sources and peptone as a nitrogen source.

Characterization of Bacteriocin Production by Lactococcus sp. J-105 Isolated from Kimchi (김치에서 분리한 Lactococcus sp. J-105가 생산하는 Bacteriocin의 특성)

  • A bacteriocin-producing strain, J-105, was isolated from Kimchi and identified as Lactococcus sp. The optimum conditions for the bacteriocin production from the isolated microorganism were evaluated. For maximum yield of bacteriocin from Lactotoccus sp. J-105, the cell should be harvested at the early stationary phase and temperature, pH and NaCl concentration should be $25^{\circ}C$, pH 8.0 and without the addition of NaCl, respectively. Maltose should be used as a carbon source and organic nitrogen such as polypeptone should be used as a nitrogen source for the best yield. The bacteriocin from isolate was inhibitory against Acetobacter aceti, Bacillus subtilis and several strains of lactic acid bacteria. The bacteriocin of J-105 was sensitive to pepsin, but stable for heat treatment. It was stable even at autoclaving temperature for 15 min.

  • We investigated the growth parameters of Candida parapsilosis isolated from spoiled red ginseng extract. The optimum pH range for C. parapsilosis was 6.0, whereas the minimum and maximum pH values that permitted growth were 3.0 and 11.0, respectively. For cells grown in PG medium plus 0 and 60% sucrose, the optimum water activity(Aw) values were 0.98 and 0.97, respectively. The optimum temperature for C. parapsilosis were 3$0^{\circ}C$ at an Aw of 0.90 in 4.8% potato dextrose broth with 18% sucrose (PGS). Cations inhibiting the growth of C parapsilosis were L $i^{+}$ $Ca^{2+}$, $Mg^{+2}$, $K^{+}$ in decreasing order, while anions were S $O_4$$^{2-}$, N $O_3$$^{[-10]}$ , C $l^{[-10]}$ .TEX> .

  • This study was carred out to get the basic conditions for the mycelial growth of Morchella esculenta in shaking flask culture. The optimal temperature and initial pH of mycelial growth of Morchella escuzenta were 25$\pm$1$^{\circ}C$ and 6.5, respectively. The optimal medium was BG medium. Among the carbon sources tested, fructose was favorable for the mycelial growth and optimal fructose concentration was 5.0% (w/v). As nitrogen sources, peptone and NH$_4$Cl appeared to be favorable and optimal concentration was 4.0% [(w/v), ratio of 1:1].

Growth Characteristics according to GA3 Treatment of Angelica acutiloba (Siebold & Zucc.) Kitagawa (일당귀의 GA3 처리에 따른 생육특성)

  • 본 연구의 목적은 약용과 식용으로서의 활용도가 높은 일당귀[Angelica acutiloba (Siebold & Zucc.) Kitagawa]의 GA3농도에 따른 생육특성을 확인하고, 최적의 재배기술 개발 및 지속가능한 안정적인 작물생산에 대한 기초자료의 제공에 있다. 연구재료는 강원도 태백시에 위치하는 일당귀 재배농가에서 2021년 10월에 채종한 종자를 구입하였고, 영주시 풍기읍에 위치한 연구온실에서 연구를 수행하였다. 일당귀의 종자를 증류수에 48시간 침지 후 GA3농도 4조건(50 ppm, 100 ppm, 500 ppm, 1,000 ppm)에 24시간 처리 후 72구 포트에 파종 후 출아율과 생육특성을 조사하였다. 연구결과 일당귀 종자는 16일이 경과한 시점부터 출아를 시작하였고, 500 ppm처리구에서 73.3%로 가장 높은 출아율을 나타냈다. 생육특성조사는 출아 후 40일(1차)과 80일(2차) 경과된 시점에서 총 2회를 수행하였는데 1차 생육조사에서는 주근길이와 직경을 제외한 모든 생육특성에서 처리구별 생육의 차이가 확인되지 않았으나 2차 생육조사에서는 지상부와 지하부의 모든 생육특성에서 처리구별 생육의 차이가 확인되었다. 잎수(6.3개)와 주근길이(4.26 cm)는 500 ppm처리구에서 가장 높은 생육량을 나타냈고, 지상부 높이(21.81 cm), 잎길이(9.0 cm), 잎너비(10.77 cm), 주근직경(5.71 mm)은 1,000 ppm처리구에서 가장 높은 생육량을 나타냈다. 따라서 GA3 처리에 따른 일당귀의 생육특성은 생육초기보다 후기로 갈수록 발현량이 높게 나타나는 것으로 판단된다. 또한 저농도의 GA3처리보다 500-1,000 ppm 사이의 처리농도가 적합한 것으로 판단되며, 추후 일당귀의 유용성분 분석을 통한 이화학적 연구가 추가된다면 최적의 재배기술 확립을 통한 안정적인 작물 생산에 기여할 수 있을 것이라 사료된다.

Optimization for the Alcohol Fermentation of Hydrolyzed Vegetable Protein(HVP) Soy Sauce by Saccharomyces Rouxii (Saccharomyces rouxii에 의한 아미노산 간장의 알코올 발효)

  • In order to improve the flavor quality of a soy sauce, hydrolyzed vegetable protein(HVP), it was subjected to ethanol fermentation by Saccharomyces rouxii and the effect of several environmental factors on the alcohol fermentation of S. rouxii in HVP was investigated. The NaCl content of HVP affected significantly on the growth of S. rouxii, showing growth inhibition above the value of 6%(w/v). The growth of S. rouxii was not inhibited by the coloring materials of HVP. The proper initial concentration of glucose for the growth of the yeast was ranged from 15%(w/v) to 25%(w/v). The optimal temperature for the growth and alcohol production was $25^{\circ}C$. The growth increased with the increasing rate of aeration, while alcohol concentration of fermented HVP showed its maximum value of 4.2%(w/v) at the aeration rate of 0.5 vvm.

Antipathogenic Activity of Bacillus amyloliquefaciens Isolated from Korean Traditional Rice Wine (막걸리로부터 분리된 Bacillus amyloliquefaciens 균주의 항균 활성)

  • The presence of bacterial strains showing antagonistic activity to common pathogens found in a variety of fermented foods in Korea was explored. A bacterium inhibiting the growth of pathogens such as Aspergillus terreus (KCTC6178), A. flavus (KCTC6984), Staphylococcus aureus (KCCM12214), Escherichia coli O157:H7 (KCCM40406), Bacillus cereus (KCTC1012), Cryptococcus neoformans (ATCC208821), Salmonella typhimurium (ATCC19430), and Listeria monocytogenes (KCTC3569) was isolated from Makgeolli, a Korean traditional rice wine. The strain showing high antipathogenic activity was identified as B. amyloliquefaciens based on the nucleotide sequence of the 16S ribosomal RNA gene. Compared with B. amyloliquefaciens KCTC1660, whose genome has been sequenced, the isolate exhibited significantly low activities of starch-degrading enzymes and high resistance to high temperature and low pH.
